1.Anti-oxidation Effects of Compound Chinese Medicine of Yiqiling on Hypertensive Rats

Objective To investigate the anti-oxidation effects of compound Chinese medicine of Yiqiling,on the spon- taneously hypertensive rats(SHR) and to explore the new target for improving the prognosis of hypertension.Method SHR were randomly divided into 2 groups:Yiqiling group and the control group.The serum content of malondiadehyde (MDA) and superoxide dismutase(SOD),and the urine content of 8-iso prostaglandin F2?(8-isoPGF2?) in two groups were measured.Examination of the gene expression profile was analyzed by using cDNA microarray.Three of the anti-oxidation genes were selected to undergo RT-PCR and quantitative real-time PCR.Results The 8-isoPGF2?urine level and MDA serum level were lower,and SOD serum level was higher in Yiqiling group than those in the control group,the differences of the above indexes had statistical significance.In Yiqiling group the expression of 354 genes was up-regulated and that of 121 genes was down-regulated in comparison with the control group.The results of the Hsphl,Hspca and Pam genes detected by RT-PCR and quantitative real-time PCR were consistent with those detected by the microarray examination.Conclusion Yiqiling has anti-oxidation effects and improvement on the prognosis of SHR,and its possible mechanism is related with the regulation of Hsphl,Hspca and Pam genes expression.
2.Application of Human Leukocyte Antigen Matching in Highly Sensitized Recipients of Renal Allografts

40%)were observed in HLA CREGs matching and outcome of post-transplantation.Results Patients with 0,1,2 or 3 mismatching(MM) of HLA CREGs+DR were 4(28%), 6(44%)and 4(28%)cases respectively according to the the rule of CREGs matching and no case had 3~6 MM.However the cases of 0,1,2,3 and 4 MM were 1(7%),3(21%),5(36%)and 5(36%)respectively by the standard of conventional HLA antigen matching,without 4~6 MM and only 4 cases had shared 0~1MM.Only 9 patients were developed into acute rejection, and were reversed by OKT3 treatment after transplantation.Renal function was returned to normal in all patients.Conclusions Using CREGs matching criteria would significantly increase the chance of recipients to receive well-matched kidney and provide more chance for waiting recipients.Suitable HLA matching could play an important role in reducing the incidence of acute rejection and improving graft survival in sensitized patients.
3.Diagnose and Treatment of Liver Functional Lesion After Kidney Transplantation

Objective To investigate the etiological diagnosis and therapy of liver function lesion after kidney transplantation.Methods By analyzing the etiological factor of 82 cases of liver function lesion,we provided a reasonable and operative therapy scheme in kidney post-transplantation.Results Of 82 cases,the number of drug toxicity and viral hepatitis B which result in liver functional lesion were 70 cases(85 5%) and 12 cases(14 5%) respectively.The former were all cured,8 cases in the latter were cured and the other 4 cases were died from acute hepatic failure.Conclusions The liver function lesion of post-transplantation should be diagnosed carefully and treated particularly.Morever,the adjustment of immuno-suppressive drugs and monitoring circlosporin A concentration should be done for the patients with liver function damaged.

5.Effect of Adenovirus-mediated p27kip1 Gene Transfer on Cell Cycle of Esophageal Carcinoma Cells

Objective To investigate effect of p27kip1 gene transfer mediated by adenovirus vector on the cell cycle of esophageal carcinoma cells. Methods The recombination adenoviral vector carrying p27kip1 gene (Ad-p27kip) was transfected to esophageal carcinoma Eca9706 cells. The effect of p21kip1 gene transfer on the cell growth suppression was measured by MTT method. Expression of p27kip1 was detected by immunocytochemical technique. Cell cycle was analysed by flow cytometry(FCM). Results After Eca9706 cells were transfected with Ad-p27kip1, p27kip1 expression increased, the cell growth was significantly inhibited and the ratio of G 0/G 1 phase cells increased and the cell number of G 2/M and S phases decreased. Conclusion p27kip1 gene transfer mediated by adenovirus can significantly inhibit the growth of esophageal carcinoma cells and cause G 1 phase blocking.
6.Immunoadsorption preventing hyperacute rejection of highly sensitized recipients of renal allografts

Objective To investigate immunoadsorption in the prevention of hyperacute rejection in highly sensitized recipients of renal allografts.Methods Immunoadsorption treatment was performed on 10 patients whose panel reactive antigen (PRA) was more than 40 %. The adverse effect and hyperacute rejection were observed.Results Two cases of acute rejection of renal allografts were found in highly sensitized recipients and reversed by the therapy of immunoadsorption and adjusting immuno-suppressive drugs.Conclusion Immunoadsorption treatment could effectively and safely prevent highly sensitized recipients from hyperacute rejection post-transplantation.

Objective To investigate the effects of inhibiting ubiquitin proteasome pathway(UPP) on proliferation of gastric carcinoma cells and the possible mechanism was discussed. Methods The gastric carcinoma cell strain SGC 7901 was treated with MG 132 to inhibit its UPP specially. The effect of growth suppression on cells was evaluated with MTT assay. Cell cycle and apoptosis were detected by flow cytometry(FCM). DNA fragment analysis was used for confirming the presence of apoptosis. The activity of telomerase was examined by TRAP PCR ELISA. Expression of p27kip1 was detected by immunocytochemical technique. Results MG 132 had great inhibitory effect on the growth of SGC 7901 cells. Objective To study the relationship between the imaging patterns of microvasculature change and histological diagnosis.MethodsOne hundred and thirty-seven patients with esophageal mucosa roughness,erosion,plaque,abnormal color and indentation in conventional endoscopy and thirty healthy volunteers were enrolled in this study.The magnifying endoscopy images were graded as four patterns by intraepithelial papillary capillary loop(IPCL) changes by NBI .The biopsies underwent pathologic evaluation.The imaging patterns of endoscopy and histological diagnosis were compared and statistically analyzed.Results137 patients were diagnosed by narrow-band imaging system with magnifying endoscopy combined miniprobe sonography.Among these patients,27 cases were pathologically diagnosed as squamous cancer including 15 cases of early esophageal cancer,21 cases were high grade intraepithelial neoplasia,23 cases were low grade intraepithelial neoplasia,66 cases were chronic inflammation.100% esophageal carcinoma and high grade intraepithelial neoplasia were Ⅳ,Ⅲ type IPCL,56.52% low grade intraepithelial neoplasia was Ⅲ type IPCL,43.48% wasⅡtype IPCL,90.91% esophagitis wasⅡtype IPCL.100% esophageal normal mucosa wasⅠtype IPCL.The difference was significant among esophageal carcinoma,high grade intraepithelial neoplasia and esophagitis,esophageal normal mucosa (P<0.05).ConclusionsThe imaging patterns of microvasculature change under NBI magnifying endoscopy were crucial in the diagnosis and identification of benign and malignant of esophageal disease.
10.Effects of Ubiquitin-proteasome Inhibitor MG-132on Apoptosis and Survivin Expression of Esophageal Carcinoma Cells

OBJECTIVE:To investigate effects of ubiquitin-proteasome inhibitor MG-132on apoptosis and survivin expression of esophageal carcinoma cells.METHODS:The esophageal carcinoma cells Eca9706were treated with MG-132,the growth inhibitory rate was determined with MTT assay,apoptosis was detected by flow cytometry,expression of survivin was detected by immunocytochemical technique.RESULTS:MG-132had obvious inhibitory effects on the growth of gastric car?cinoma cells,IC 50 of24hrs,48hrs,72hrs and96hrs were120.2,18.1,—12.2,and—16.9?mol/L respectively;Treated with5.0?mol/L MG-132for24hrs,48hrs,72hrs and96hrs,apoptotic rates of cells were(3.1?0.4)%、(31.7?3.5)%、(50.4?4.8)%and(66.6?6.2)%respectively;Expression of survivin was high in esophageal carcinoma cells and it was decreased in cells treated with MG-132.CONCLUSIONS:MG-132can significantly inhibit the proliferation of esophageal carcinoma cells and induce apoptosis,which might be associated with down-regulated expression of survivin.
