1.The correlation between the hypertrophic ventricular tissue of SHR rats and ventricular arrhythmia occurred

Objective To explore the susceptibility differences of ventricular arrhythmia (VA ) in SHR rats with/without left ventricular hypertrophy(LVH) and the significance .Methods The experiments were performed on isolated hearts of 8‐week(the non‐LVH control group ,n=12) ,16 week‐old SHR(the LVH group ,n=12) and age‐matched Wistar Kyoto rats(Wistar ,the blank control group ,n=12) ,which were perfused in Langendorff mode with oxygenated Krebs‐Henseleit solution followed by a K+‐defi‐cient solution .The epicardial electrocardiogram was continuously monitored during all experiments .HE staining and collagen vol‐ume fraction(CVF)was used to evaluate the condition of LVH .Results Compared with the non‐LVH control group and the blank control group ,the low K+ induced ventricular arrhythmia occurred earlier with increased incidences and duration in the hearts of the LVH Group ,the incidences of ventricular tachycardia(VT) ,transient ventricular fibrillation(TVF) ,sustained ventricular fibrillation (SVF) and CVF were higher in the hearts of the LVH Group(P<0 .05) ,myocardial cell hypertrophy and myocardial cells intersti‐tial increased .Conclusion The ventricular arrhythmia occurred earlier with increased incidences and duration in the LVH tissue of SHR rats ,which implies that LVH is associated with VA .
2.MicroRNAs Differential Expression Patterns in the Hypertrophic Left Ventricular Tissue in Spontaneously Hypertensive Rats

Objective To investigate the molecular mechanisms for the development of cardiac hypertrophy in hypertension by scanning the expression profile of microRNA (miRNA) in the left ventricular hypertrophic tissue in spontaneously hypertensive rats(SHR)using miRNA microarray chip technique.Methods 17-week-old male SHR with left ventricle hypertrophy (LVH) (n=10) and 8-week-old male SHR without LVH were employed in this study (n=10).Left ventricular mass index (LVMI),HE staining and measurement of transverse diameter of myocardium (TDM) were used to evaluate LVH.The mRNAs of the two groups were extracted from left ventricle tissue and the differential expression of miRNA was detected by using miRNA microarray chip.Results LVMI and TDM were greater in rats with LVH vs without [LVMI(2.87?0.15)vs(2.17?0.14)mg/g;TDM(15.32?0.61)vs(12.01?0.43)?m,P
3.Laparoscopic adrenalectomy for adrenal tumors by transperltoneal or retroperltoneal approaches:a report of 41 cases

Objective To evaluate the effect of laparoscopic adrenalectomy for adrenal tumors by transperito-neal or retroperitoneal approaches. Methods Atotal of 41 laparoscopic adrenalectomy for adrenal tumor patients were pertormed,inchiding 16 patients by transperitoneal approach and 25 patients by retroperitoneal approach. The tumor size,operative time,intraoperative blood loss, postoperative hospital stay, postoperative ambulatory day and postopera-tive complications were observed. Results Of the 41 cases ,3 cases were transferred to open surgery and the other 38 cases were achieved successful operations. No significant differences( all P>0.05) were found in tumor size,intraop-erative blood loss, postoperative hospital stay, postoperative ambulatory day between the transperitoneal group and ret-roperitoneal group ,but there were significant differences( P<0.05) in operative time and intraoperative blood loss be-tween the two group. Conclusion Both the transperitoneal and retroperitoneal laparoscopic approaches are feasible and safe for adrenal tumors, and the latter has more advantages in operative time, but the operative approach still should be chosen based on the pathological changes,tumor size and position as well as the individual conditions of the patients.
4.Preventive effect of mitomycin C injection via great omentum vein during resection of intrahepatic cholangiocarcinoma against carcinoma recurrence

0.05).Multivariate analysis showed that the recurrence was related to the number and diameter of the tumors,with the hazard ratios being 1.877(P=0.001)and 1.391(P=0.029 4),respectively;injection of MMC had no influence on the recurrence.There was no significant difference in the time of disease-free survival(DFS)between the 2 groups,with the median time of DFS of MMC injection group being 5 months and of control group being 3 months after the first operation(P=0.125 3).The recurrence rates in the MMC group and control group were 65.79% vs 68.57%(P=0.800 4) 6 months after operation,81.58% vs 94.29%(P=0.099 0)1 year after operation,89.47% vs 97.14%(P=1.679 55) 2 years after operation and 92.11% vs 97.14%(P=0.344 7)3 years after operation.Conclusion: Injection of 10mg MMC via great omentum vein during resection of IHCC may have no definite effect on the recurrence of IHCC;however,perspective study is needed to verify this finding.
6.Determination of Emodin in Radix et Caulis Ventilaginis Leiocarpae by HPLC

Objective To establish a HPLC method for determining emodin in Radix et Caulis Ventilaginis leiocarpae. Methods Stationary phase was Agilent Eclipse XDB-C18 (4.6 mm?150 mm, 5 ?m), mobile phase was methanol-0.1%phosphoric (75∶25), detection wavelength was 285 nm, flow rate was 1.0 mL/min and column temperature was 30 ℃. Results There was good linearity in the range of 0.006 3~0.063 ?g for emodin (r=0.999 9). The average recovery of emodin was 102.0% with RSD=1.58%. Conclusion This method is rapid and accurate. It can be applied to control the quality of Radix et Caulis Ventilaginis leiocarpae.
7.Single-nucleotide polymorphisms of tumor necrosis factor-alpha gene are associated with severe adult community acquired pneumonia in Chinese

AIM: Tumor necrosis factor-?(TNF-?) participates in the establishment of inflammatory lesions in pneumonia.High production of TNF-? may relate to the severity of pneumonia.There have already been several studies examining the association between pneumonia and single nucleotide polymorphisms(SNPs) that affect cytokine productivity.SNPs of TNF-?,-1 031,-863,-857,-308 and-238 have been identified.The variant alleles of these SNPs have suggested to be related to high TNF-? production and the severity of pneumonia.Therefore,the aim of this study is to examine the association between the severity of pneumonia in Chinese and the following SNPs: five in the TNF-? gene promoter(-1 031,-863,-857,-308,-238).METHODS: A total of 117 Chinese individuals were enrolled in this study.They were 67 patients with pneumonia and 50 healthy subjects.TNF-? was genotyped by polymerase chain reaction-restriction fragment length polymorphism for all subjects.The frequency distributions of genotypes in different groups were analyzed by SPSS 11.5 program.RESULTS: Frequency of subjects who carried at least one variant allele in TNF-?-1 031,-863,-857,-308,-238 SNPs among pneumonia patients was significantly higher than that in healthy subjects.And frequency of subjects who carried variant allele in TNF-?-863,and-308 SNPs among severe adult community acquired pneumonia patients was significantly higher than that in common pneumonia patients.CONCLUSION: TNF-?-863 and-308 SNPs appear to be associated with severe adult community acquired pneumonia in Chinese populations.
8.Effect of benazepril combined with atorvastatin in treatment of chronic heart failure and its effects on level of plasma NT-proBNP

Objective To study curative efficacy of benazepril combined with atorvastatin in treatment of chronic heart failure and its effects on level of plasma N-terminal pro brain natriuretic peptide(NT-proBNP). Methods 90 patients of chronic heart failure who received therapy from January 2013 to January 2016 in the first people's Hospital of Xiangshan, Zhejiang. According to random number table, those patients were divided into the observation group and the control group with 45 cases in each group, on the basis of routine treatment, the control group was treated with benazepril, while the observation group was treated with atorvastatin on the basis of control group. After 2 months, the treatment effect was compared. Results After treatment, the left ventricular end diastolic diameter (LVIDd), left ventricular end systolic diameter (LVIDs) in the observation group were lower than the control group, left ventricular ejection fraction (LVEF) was higher than the control group, the difference was statistically significant (P<0.05), the level of NT-proBNP in the observation group was lower than the control group, the difference was statistically significant (P<0.05), 6 minute walking distance was better than the control group, the difference was statistically significant (P<0.05), the total effective rate in the observation group 91.11%(11/45) was higher than the control group 71.11%(32/45), the difference was statistically significant (P<0.05), during the follow-up of 6 months, the recurrence rate of heart failure in observation group 2.22%(1/45) was lower than the control group 20.00%(9/45), the difference was statistically significant (P<0.05). Conclusion Benazepril combined with atorvastatin is well for chronic heart failure, which can effectively reduce the level of plasma NT-proBNP and prognosis.
9.The effects of adenosine A2A receptor antagonist on the behavior and the striatal A2A receptor,metabotropic glutamate receptors protein expression in rats with levodopa-induced dyskinesia

Objective To study the behavioural changes and biological effects of selective adenosine A2A receptor antagonist (CSC) in a rat model of levodopa(L-DOPA) -induced dyskinesia (LID).Methods The hemi-parkinsonian rat model was produced by stereotaxically injecting 6-OHDA to the right medial forebrain bundle. Rats were randomly divided into 4 treatment groups with a random number generating program to receive intraperitoneal injections twice daily for 21 days (n = 10): saline, L-DOPA at 25 mg/kg with benserazide at 6. 25 mg/kg, CSC at 2. 5 mg/kg alone and CSC at 2.5 mg/kg with L-DOPA at 25 mg/kg plus benserazide at 6. 25 mg/kg. Forepaw adjusting steps, abnormal involuntary movements (AIM) and rotational response duration were observed on 2, 9, 11,18 and 21 d. After sacrifice, the expression of adenosine A2A R and mGluR5 was observed by Western blot. Results Co-administration of LDOPA with CSC significantly increased the forelimb adjusting steps of parkinsonian rats during 21 days of treatment when compared to L-DOPA alone. CSC treatment alone increased the forelimb adjusting steps significantly. Co-administration of L-DOPA with CSC ( ( 11 ± 5 ) score) significantly decreased the AIM scores of limb and orolingual muscles when compared to L-DOPA alone (( 17 ± 4) score; t = 2. 44, P <0. 05). The subchronic L-DOPA treatment upregulated the striatal expression of adenosine A2A R and mGluR5. However, co-administration of L-DOPA with CSC reversed the shortening of the rotational motor response duration induced by L-DOPA administration during the period of the treatment and attenuated the LDOPA-induced upregulation of adenosine A2A R and mGluR5 expressions. Conclusions CSC improves motor function in a hemi-parkinson rat model, potentiates the antiparkinsonian effects with L-DOPA and partly attenuates LID. Co-administration of L-DOPA with CSC reverses the L-DOPA-induced upregulated expression of A2A R and mGluR5, indicating the involvement of both A2A R and mGluR5 in the onset and progression of LID. Adenosine A2AR antagonists may be promising drugs for treatment of LID.
10.Effects of AG1024 on hepatocellular carcinoma cell lines

Objective Tyrphostin AG1024(3-Bromo-5-t-butyl-4-hydroxybenzylidenemalonitrile) is a specific insulin like growth factor type Ⅰ receptor tyrosine kinase blocker,this study is to investigate the effect of AG1024 on the proliferation and apoptosis of hepatocellular carcinoma cell lines.Methods Treated with AG1024 on vailed concentrations(0~40 μmol/L),human hepatocellular carcinoma cel lines HepG2 and SMMC-7721 were observed for morphological and molecular biology changes,the effect of AG1024 on the cell lines proliferation invasion ability as well as apoptosis was evaluated. Results MTT showed that AG1024 dose-dependently inhibited the proliferation of hepatocellular carcinoma cells,flow cytometry suggested that AG1024 significantly promoted cell lines apoptosis,the cell invasion assay indieated that AG1024 significantly inhibited cell's invasion ability.RT-PCR showed over-expression of IGF-IR in liver cancer cells.and AG1024 dose-dependently increasedtheexpressionofcytochreme C. According totheresultsof Western, blotting,the phosphor-ERK and procaspase-3 were down-regulated while the total ERK remained unchanged. Conclusion AG1024 as a specific IGF-IR blocker blocks the downstream signaling cascade and thus inhibits the proliferation of hepatocellular carcinoma cells and induces cell's apoptosis.
