1.Clinical control study of ondansetron, metoclopramide and haloperidol in the prevention of postoperative nausea and vomiting after neurosurgery

ObjectiveTo investigate the clinical difference of ondansetron,metoclopramide and haloperidol in the prevention of postoperative nausea and vomiting after neurosurgery.MethodsNinety patients with neurosurgery were divided by random digits table method into four groups:control group ( 18 cases ) treated with 10 ml 0.9% sodium chloride ;ondansetron group(24cases ) received ondansetron 4 mg;metoclopramide group (24 cases) with metoclopramide 10 mg and haioperidol group (24 cases) with haloperidol 2.5 mg.The efficacy and adverse reaction were compared among four groups.Results Compared with control group,ondansetron,metoclopramide and haloperidol could obviously inhibit the occurrence of postoperative nausea and vomiting after neurosurgery,the difference had statistical significance (P< 0.05).Total effective rate of ondansetron group [79.2%( 19/24)] was significantly higher than that of metoclopramide group [ 58.3% ( 14/24 ) ] and haloperidol group [ 54.2% ( 13/24 ) ] (P < 0.05 ).And total effective rate of metoclopramide group and haloperidol group had no significant difference (P > 0.05 ).The occurrence rate of adverse reaction of metoclopramide group [ 16.7%(4/24)] had no statistical significance compared with that of ondansetron group[8.3%(2/24)] and haloperidol group[ 12.5%(3/24)] (P > 0.05).ConclusionsOndansetron,metoclopramide and haloperidol can obviously inhibit the occurrence of postoperative nausea and vomiting after neurosurgery,and the effect of ondansetron is significantly better than that of metoclopramide and haloperidol.Therefore,it is necessary to use drugs for preventing postoperative nausea and vomiting for patients during neurosurgery.
2.Research on the correlation of COX-2 and renal cell carcinoma

ObjectiveTo explore the expression of Cyclooxygenase-2-specific(COX-2) in renal cell carcinoma and its relationship with angiogenesis.MethodsThe expression of COX-2,VEGF and bFGF in renal cell carcinoma specimens(85 cases) and adjacent normal tissue samples(48 cases) was measured by immunohistochemistry.Microvessel density (MVD) was evaluated by immunohistochemistry using a monoclonal antibody against CD34.ResultsThe positive expression rates of COX-2,VEGF and bFGF in adjacent normal tissue were 16.7%,20.8%,22.9%,and 77.6%,80.0%,74.1% in renal cell carcinoma,respectively,the difference,were significant ( all P <0.01 ).MVD was (55.47 + 18.47 ) Counts/field in renal cell carcinoma,and ( 20.42 ± 8.72) Counts/field in adjacent normal tissue,the difference was significant( P < 0.01 ).MVD in renal cell carcinoma which was positive expressed COX-2,VEGF,bFGF was higher than renal cell carcinoma which was negative expressed COX-2,VEGF,bFGF,the difference was significantly statistical( P < 0.05 or P <0.01 ).COX-2 expression and VEGF,bFGF in renal cell carcinoma expression was positively correlated( P <0.01 ).Conclusion COX-2 expression was elevated in renal cell carcinoma,and it was related closely with renal cell carcinoma angiogenesis.
3.Influence of nursing intervention on sleep quality of patients after cardiac intervention

ObjectiveTo investigate the effect of nursing intervention on sleep quality of patients after cardiac intervention. Methods140 patients undergoing cardiac interventional surgery in our hospital from January 2010 to January 2011 were chosen as the research object.They were divided into the observation group and the control group in accordance with the principle of randomization with 70 cases in each group.The control group was treated with conventional methods of nursing care of cardiology.The observation group was given with sleep- related nursing interventions on the basis of the routine care.The sleep quality score (PSQI)24 h and 1 week,2 weeks postoperation and treatment compliance 2 weeks postoperation were compared between the two groups. ResultsThe study showed that the PSQI score 24h postoperation showed no significant difference,but 1 week and 2 weeks after the operation,the score of the observation group was significantly lower than the control group.The full compliance rate in the observation group was significantly higher than the control group.The non-full compliance rate was significantly lower than the control group,the differences were statistically significantConclusionsNursing intervention can improve treatment compliance of patients,improve sleep quality of patients after cardiac intervention.It is conducive to the rehabilitation of patients and is worthy of clinical application.
4.Applied research of selective bronchial arterial embolization for severe hemoptysis

Objective To observe the clinical curative effect of selective bronchial arterial embolization for severe hemoptysis.Methods Thirty-eight hospitalized patients with severe hemoptysis was enrolled in this study.Of which 31 patients were recurrent hemoptysis,27 patients were with bronchiectasis,4 patients were with bronchial lung cysts,4 patients had unknown causes,3 patients were with tuberculosis.All patients were diagnosed by chest imaging examination,fiber bronchoscopy,bronchial arteriography,and they were all treated by selective bronchial arterial embolization.Results The most times of embolization was 3,and bronchial arterial imaging were performed for vascular malformation.After having bronchial arterial embolization,35 patients were immediately released of bleeding.Postoperative 24 h,8 patients had hemoptysis again,of which 1 case was performed with conservative treatment and subsequently had pneumonectomy,5 patients had effective conservative treatment,2 cases were performed embolization again after failed conservative treatments.Hemoptysis completely disappeared within 1 week.The total effective rate was 97.4%(37/38).Patients were followed up for 1 year,of which 13 patients relapsed again,11 patients'annual and each time's quantity of hemoptysis reduced 50%.Two patients had pneumonectomy after a number of embolization.The total effective rate was 94.7% (36/38).After treatment,3 patients had mild chest pain in short time,2 patients had shoulder pain,3 patients had chest tightness,and 3 patients had subcutaneous passive congestion.After being treated for short-term,these symptoms disappeared eventually.Conclusion Selective bronchial arterial embolization for the treatment of acute or recurrent severe hemoptysis is very effective,and can avoid the risk of surgery.It is effective for recurrent cases and worthy of clinical application.

6.Application of high fibular osteotomy in the treatment of knee osteoarthritis.

OBJECTIVETo evaluate the short-and mid-term curative effect of high fibular osteotomy on knee osteoarthritis.
METHODSFrom October 2014 to July 2016, 76 patients with knee osteoarthritis were treated with high fibular osteotomy. There were 22 males and 54 females, ranging in age from 47 to 82 years old, with an average of 61.62 years old. Internal tenderness of the knee joint was examined preoperatively. Preoperative examination showed medial knee pain and medial positive McMurray sign. Preoperative AP and lateral X-ray films of the weight-bearing side of the knee showed the narrowing of the medial space. The knee medial space, femoral and tibial angle, VAS pain score and the American Knee Society(AKS) score were evaluated after operation.
RESULTSCompared with the preoperative one, the postoperative knee medial gap was widened obviously. The femur and tibia angle after operation was bigger than that before surgery. The VAS was improved from preoperative 5.70±1.56, to 3.70±1.03 at the 1st week after operation, 3.20±0.95 at the 1st month after operation, 2.35±0.99 at the 3rd month after operation, and 2.10±0.97 at the 1st year after operation. According to the AKS score, the preoperative pain score of knee joint was 14.45±1.76, the activity score of knee joint was 12.60±1.98, and the stability score was 12.15±1.72; after operation, the above scores were improved to 42.60±2.28, 21.80±2.14, and 20.85±2.16 respectively.
CONCLUSIONSThe high fibular osteotomy with less complications is simple. It can effectively alleviate the pain of the knee joint and improve the function of knee joint, and the clinical effect is satisfactory.
7.Protective effect of melatonin against oxygen-induced retinopathy: a study based on the HMGB1/NF-κB/NLRP3 axis.

OBJECTIVES:
To study the protective effect of melatonin (Mel) against oxygen-induced retinopathy (OIR) in neonatal mice and the role of the HMGB1/NF-κB/NLRP3 axis.
METHODS:
Neonatal C57BL/6J mice, aged 7 days, were randomly divided into a control group, a model group (OIR group), and a Mel treatment group (OIR+Mel group), with 9 mice in each group. The hyperoxia induction method was used to establish a model of OIR. Hematoxylin and eosin staining and retinal flat-mount preparation were used to observe retinal structure and neovascularization. Immunofluorescent staining was used to measure the expression of proteins and inflammatory factors associated with the HMGB1/NF-κB/NLRP3 axis and lymphocyte antigen 6G. Colorimetry was used to measure the activity of myeloperoxidase.
RESULTS:
The OIR group had destruction of retinal structure with a large perfusion-free area and neovascularization, while the OIR+Mel group had improvement in destruction of retinal structure with reductions in neovascularization and perfusion-free area. Compared with the control group, the OIR group had significant increases in the expression of proteins and inflammatory factors associated with the HMGB1/NF-κB/NLRP3 axis, the expression of lymphocyte antigen 6G, and the activity of myeloperoxidase (P<0.05). Compared with the OIR group, the OIR+Mel group had significant reductions in the above indices (P<0.05). Compared with the control group, the OIR group had significant reductions in the expression of melatonin receptors in the retina (P<0.05). Compared with the OIR group, the OIR+Mel group had significant increases in the expression of melatonin receptors (P<0.05).
CONCLUSIONS
Mel can alleviate OIR-induced retinal damage in neonatal mice by inhibiting the HMGB1/NF-κB/NLRP3 axis and may exert an effect through the melatonin receptor pathway.

Aim To investigate the effects of all-trans retinoic acid and benazepril on the expression of ?-smooth muscle actin in rats with glomerulosclerosis.Methods 80 Wistar male Rats were randomly assigned into the following groups: control group,model group,ATRA treatment group and benazepril treatment group,20 rats in each group.GS rats were uninephrectomized and injected with adriamycin(5mg?kg-1) after one week through the tail vein.All rats were sacrificed at the 12th week,GS was evaluated by glomerulosclerosis index(GSI) system.The expression of ?-SMA was assessed by reverse transcription-polymerase chain reaction(RT-PCR) and immunohistochemistry.Results Comparing with control group,the expression of ?-SMA mRNA and protein were decreased significantly in ATRA treatment group and benazepril treatment group(P0.05).Conclusions The postponed effects of ATRA and benazepril on GS were evident and equivalent.
9.Analysis of the correlation between sleep and mental health among primary and secondary school students in Shandong Province

Objective:
To investigate the sleep and mental health status of adolescents in Shandong Province, to explore the correlation between sleep and mental health, so as to provide a basis for adolescent physical and mental health management.
Methods:
From February to March 2023,a multistage stratified whole cluster sampling method was used to randomly select 3 cities in Shandong Province, one urban area and one township in each city, one junior high school and one senior high school in the urban area and the township, respectively, and then 4 classes were randomly selected from each grade level of each school, and all 3 667 students in the classes were surveyed by using the Pittsburgh Sleep Quality Index Scale (PSQI) short form and the Chinese Middle School Students Mental Health Inventory (MMHI-60).
Results:
The prevalence of sleep deprivation among adolescents in Shandong Province was 37.44%, and the detection rate of psychological problems was 46.41%. Adolescent psychology could be divided into four latent categories:psychological immune group (39.6%), psychological low risk group (12.2%), psychological medium risk group (13.2%) and psychological high risk group (35.0%). Multifactorial Logistic regression analyses showed that gender, school year, sleep duration and sleep quality were influencing factors for the psychological latent categories ( OR =1.39-9.55, P < 0.05 ), and that adolescents with sleep deprivation and poor sleep quality were more inclined to be classified as being in the psychological medium and high risk groups.
Conclusions
The sleep and mental health of adolescents in Shandong Province is not very good. Comprehensive prevention and control of psychological problems should not only focus on personality and psychological characteristics, but also need to be combined with the sleep condition of adolescents.
10. Exosomal miR-574-5p Promotes Proliferation, Invasion and Migration of Glioma Cells by Down-regulating LATS2

Exosomes are involved in invasion, migration and angiogenesis of tumor cells, and invasion is the main cause of death in glioma patients. Studies have shown that the exosomes secreted by tumor cells can carry miRNA into the receptor cells and regulate the biological functions of the receptor cells, such as proliferation, migration and invasion. miR-574-5p plays a key role in the occurrence and development of a variety of tumors. However, whether the exosomes derived from glioma cells express miR-574-5p and its role in the growth, invasion and migration of glioma cells have not been reported. This study investigated the mechanism of the exosomal miR-574-5p secreted from glioma cells in the process of cell proliferation, migration and invasion. The exosomes were characterized by electron microscopy, nanoparticle size tracking and Western blot. The results displayed that the extracted exosomes were round particles with a diameter of 30 ~ 100 nm. The internalization of exosomes was detected by immunofluorescence assay. The results showed that exosomes were internalized into LN229 cells; Bioinformatics and online data were used to screen the differentially secreted miRNA between LN229 and H4 glioma cells. The results showed that the differentially secreted miRNA was miR-574-5p, and large tumor suppressor 2 (LATS2) was predicted to be the target gene of miR-574-5p; Duel luciferase reporter assay confirmed that miR-574-5p was complementary to the 3'UTR region of LATS2; The transfection assay, qRT-PCR and Western blot was conducted to measure the relationship between miR-574-5p and LATS2. The results demonstrated that there was no significant difference in LATS2 mRNA levels between the control group and the group with miR-574-5P overexpression (P > 0.05), suggesting the regulatory effect of miR-574-5P on LATS2 was achieved by inhibiting its translation (P < 0.05). CCK-8, Transwell migration and invasion assays were conducted to explore the effect of miR-574-5p on proliferation, migration and invasion of LN229 cells. The results showed that overexpression of miR-574-5p could significantly promote the ability of proliferation, migration and invasion of LN229 cells (P < 0.05). In addition, Western blot was performed to measure the expression of kinase proteins involved in the LATS2/YAP signaling pathway, and the influence of the exosomes on this signaling pathway. The results revealed that the exosomes down-regulated the protein expression level of LATS2 and reduced p-YAP phosphorylation. In conclusion, the exosomal miR-574-5p can promote the proliferation, migration and invasion of glioma cells by down-regulating LATS2 and activating LATS2/YAP signaling pathway, which may provide a potential biomarker for the diagnosis and target for the treatment of glioma.
