1.Extracorporeal shock wave therapy plus enhanced load fibrin glue bone growth factors for nonunion or delayed osseous union of rabbit

Objective:To evaluate the effect of Extracorporeal Shock Wave combined percutaneous injection of enhanced fibrin glue loaded bone growth factor in accelerating the bone nonunion healing and promoting osteogenesis process .Methods: 48 radius model rabbits were randomly divided into four groups.Group A rabbits were treated with ESW;Group B rabbits were treated with ESW combined with FG;Group C rabbits were treated with ESW combined with FG/rhBMP-2/rhbFGF;Group D rabbits were treated with ESW combined with FG/rhBMP-2/rhbFGF/rhVEGF.After a combination therapy of 8 week,conventional X-ray radiography,optical in-spection organization form and biomechanical test were applied to evaluate the therapeutic effect .Results:Compared with the group B , the X-ray group C showed that the fracture line was fuzzy ,bony callus area began to increased ,fracture end range reduced.The histo-morphology showed that cartilage began to form , cartilage cell began to proliferate actively , and new blood vessels were forming surrounding fibrous connective tissue.Biomechanical determination found there was no statistical significance between the two groups ( P>0.05 ).FG/rhBMP-2/rhbFGF+ESW group showed the fracture line was still fuzzy , bony callus disappeared and apparent proliferative bone trabecula ,and biomechanical results showed that there was statistical significance between the two groups compared to the group B(P<0.01).FG/rhBMP-2/rhbFGF/rhVEGF+ESW group showed the fracture line was slightly fuzzy ,apparent proliferative bone trabecula in 4 week,and in 8 week,the fracture line was disappeared and moulding of healing was good.The histomorphology showed that the distribution was filled with fibrous connective tissue and many bone tissue .Biomechanical results showed that it could resistance to bending ,there was statistical significance between the two groups compared to the group C ( P<0.01 ).Conclusion: ESW therapy combined percutaneous injection of enhanced fibrin glue loaded bone growth factor can promote fracture healing and the method of combination is expected to be a good choice of non-surgical treatment of bone nonunion.
2.The determination of p53 protein in maxillary sinus carcinoma and its prognosis value

Objective:To determine whether p53 protein overexpression correlated with early tumor recurrence in carcinoma of maxillary sinus.Method:Forty-five patients with T2NoMo squamous cell carcinoma of maxillary sinus were treated with operation and/or radiotherapy . The median follow up time was 3 years. The ratio of local control was 62.2% (28 patients) , as control group ; The ratio of local recurrence and/or metastasis was 37.8% (17 patients),as experiment group . Cancer specimens were obtained before treatment.p53 protein overexpression, which indicated a mutated p53 gene ,was analyzed by immunohistochemistry, using the mouse monoclonal antibody DO-7.Result:Approximately 88.2% of the carcinoma that recurred locally overexpressed p53 protein , compared with only 28.6% of those with local control (P<0.01) .No significant relation was noted between p53 protein overexpression and pathohistologic grade .Intensity of staining did not predict tumor recurrece.Conclusion:The study demonstrated that the p53 protein is an important prognostic marker on gene level for maxillary sinus carcinoma.
3.Expression of a lysozyme from antheraea pernyi in pastoris and the effects of recombinant Ap-lysozyme on Streptococcus mutans

Objective: To construct the antheraea pernyi lysozyme(ApLZ) expression system using the methylotrophic yeast Pichia pastoris, to assay the antibacterial activity of the recombinant ApLZ against Streptococcus mutans. Methods:The ApLZ expression system was used in the expression and purity of Ap-lysozyme. The antibacterial activity of the recombinant ApLZ against Streptococcus mutans were assayed by using agar diffusion method. Results:Expression system of ApLZ was constructed using the methylotrophic yeast Pichia pastoris as a host. ApLZ was expressed correctly and secreted efficiently when the native signal sequence of ApLZ was used as secretion signal. The level of ApLZ expression in Pichia pastoris peaks at 96 h after the induction of sustaining 5 ml/L methanol. The molecular weight of the recombinant ApLZ is about 20 000. Conclusion:The recombinant ApLZ is active in inhibiting the growth of Streptococcus mutans.
4.Ring Artifact Location Algorithm Based on Edge Detection

Objective To investigate the Ring Artifact Location Algorithm based on edge detection to eliminate the ring artifact.Methods The Sobel operator was used to extract the ring edge,and then an algorithm,which combined scan line searching with line fitting,was applied to determining the ring center and its size.Finally the linear interpolation algorithm was used to get rid of the ring artifact.Results This method was very effective to eliminate the ring artifact of both phantom and clinical images.Conclusion This algorithm is faster and more effective than the usual one and its speed and accuracy are improved.
5.Inhibition of berberine on Ⅰ_(Kr), Ⅰ_(Ks) and Ⅰ_(K1) in thyroxine induced cardiomyopathic guinea pig ventricular myocytes

Aim: To study the effects of berberine( Ber) on the rapidly activating component( Ⅰ_(Kr)), the slowly activating component(Ⅰ_(Ks)) of the delayed rectifier potassium current and the inward rectifier potassium current(Ⅰ_(K1)) in cardiomyopathic guinea pig ventricular myocytes. Methods: After guinea pigs were ip L-thyroxine 0. 5 mg/kg for 10 d, their hearts were cardiomyopathic. Then whole cell patch-clamp recording technique was used to observe the effect of 30 μmol/L Ber on the Ⅰ_(Kr), Ⅰ_(Ks) and Ⅰ_(K1) in cardiomyopathic guinea pig ventricular myocytes. Results: In cardiomyopathic guinea pig ventricular myocytes, Ber 30 μmol/L markedly inhibited Ⅰ_(Kr) and Ⅰ_(Ks) by 22. 8% and 29. 5% at + 10 mV and + 80 mV, respectively. The effect of Ber on Ⅰ_(Ks) was greater than that on Ⅰ_(Kr). Ber 30 μmol/L also inhibited the inward component of Ⅰ_(K1) by 29. 1% at + 120 mV, but the reverse potential of Ⅰ_(K1) was unaffected. Ber( 1-300 μmol/L) was shown to inhibit Ⅰ_(Kr) and Ⅰ_(Ks) in a concentration-dependent manner. Their IC_(50), were 76. 74 μmol/L and 55. 37 μmol/L, respectively. Conclusion: Ber inhibited Ⅰ_(Kr),Ⅰ_(Ks) and Ⅰ_(K1) in cardiomyopathic guinea pig ventricular myocytes, which may be important in understanding the antiarrhythmic effects of this drug.

Objective Insulin resistance is a possible cause of polycystic ovary syndrome(PCOS).It is presumed that ectonucleotide pyrophosphatase/phosphodiesterase 1(ENPP1) is associated with insulin resistance.The purpose of this study is to explore the expression of ENPP1 in granulosa cells of the ovary and its relationship with PCOS. Methods Twelve aliquots of follicular granulosa cells were isolated from 12 samples of the patients with PCOS and 22 aliquots from 22 samples of the patients without PCOS,respectively.ENPP1 expression was analyzed by reverse transcription polymerase chain reaction(RT-PCR) and in situ hybridization.The relative expression of ENPP1 mRNA was detected by real-time quantitative polymerase chain reaction(real-time PCR). Results ENPP1 was expressed in granulosa cells of the ovary.ENPP1 expression's 2~(-?Ct) in granulosa cells of PCOS was significantly higher than non-PCOS(1.67?0.89 vs.0.94?0.76,P=0.017).Conclusion ENPP1 expression plays a role in ovary function and may be closely associated with the development of PCOS.
7.Design of inpatient charging management system

To design an inpatient charging management system to optimize the patient accounting flow. The problems in all links were analyzed, and auditing mode and flow were modified. Under the existing HIS, the sys-tem was developed with added datasheets, determined elements, desired rules, .Net framework and C# language. The system made the prescription linked with the charging so that the doubtful items could be marked automatically. The system facilitates the fee auditing, shortens the waiting time of the patient, enhances the communication between the departments and promotes the quality of medical records.
8.Effects of different fibrin glue combination modes on the proliferation and viability of dental follicle cells.

OBJECTIVEThis study explores the effects of different fibrin glue combination modes on the survival, proliferation, and apoptosis of dental follicle cells (DFCs), as well as to evaluate the feasibility and effectiveness of fibrin glue as transplantation material.
METHODSThe membranes of surviving DFCs were marked using 3,3'-dioctadecyloxa carbocyanine perchlorate (DIO), and the cell number was counted by using ImageJ2x software. The apoptotic cells were marked with prodium iodide (PI).
RESULTSCompared with that of the 3D-2 and 2D-1 groups, the degradation speed of the 3D-1 group was the slowest. DFCs could survive and grow well in fibrinogen with a concentration of 15 mg · mL⁻¹ supplemented with thrombin with a concentration of 2 U · mL⁻¹. In particular, the 3D-1 combination mode was significantly conducive to cell proliferation and stretching.
CONCLUSIONFibrin glue can be used as an effective cell transplantation material. The different combination modes have certain effects on cell proliferation. The 3D-1 combination mode is more conducive to the survival and proliferation of DFCs than other modes.

9.Preliminary exploration of the influence factors of degenerate oligonucleotide primered PCR of genome DNA

Objective: To explore the influence factors of the degenerate oligonucleotide primered PCR(DOP-PCR). Methods: Genome DNA template from the mouse single oocyte or liver tissue were used to perform DOP-PCR. DOP-PCR was carried out with templates of different origin, different gradient dilution, with or without low melting point gel purified to wipe off the small fragment that might interfere with the following analysis, and then PCR of gene FTCD and CBS were carried out to evaluate the influence of these factors on the amplification efficiency and specificity. Results: Compared with genome DNA template from mouse liver, the template from single oocyte had the same efficiency and specificity but a minor yield and different gradient dilution of DNA template had no effect on the efficiency and specificity. Furthermore, there was a higher specificity in the low melting point gel-purified DOP-PCR product than in untreated ones. Conclusion: We have got a satisfactory result and increased specificity from DOP-PCR product purified with the low melting point gel. Single oocyte of mice could be used for further investigation of special genes detection by DOP-PCR and of an optimization in the yield of the products.
10.Clinical application of patella arthroplasty in total knee arthroplasty

Objective:To analyze and compare the clinical outcomes of total knee arthroplasty with patella arthroplasty and patella replacement. Methods: From May 1999 to January 2004, 251 knees in 203 patients (45 males and 158 females; age range: 25 to 91 years, average 62.8 years) with osteoarthritis (206 knees) and rheumatoid arthritis (45 knees) were evaluated using a questionnaire during physical examination. Totally 137 knees underwent patella arthroplasty and 114 underwent patella replacement, with a mean follow-up of 28.1 months. The patients were scored using KSS Score, Feller Score and British Orthopedics Association Patient-satisfaction Score. Results: Patella arthroplasty and replacement showed no obvious difference in post-operative improvement of the knee score, knee function score, patella function score and range of flexion and extent of knee. Compared with patella replacement group, patella arthroplasty group had a higher patient satisfaction rate (P
