1.Clinical efficacy of sodium tanshinone IIA sulfonate in treatment of advanced schistosomiasis

Objective To evaluate the clinical efficacy of sodium tanshinone IIA sulfonate(STS)in the treatment of ad-vanced schistosomiasis. Methods Fifty cases with advanced schistosomiasis admitted to the Touzao Township Hospital of Dong-tai City during the period from November 2012 to November 2013 were treated with STS for 10 days,and the internal diameter of the portal vein,levels of ALT,AST,γ-GT,PIIIP,CIV,HA and LN were measured and compared before and after the adminis-tration of STS. Results The mean levels of all serological parameters except HA were within the normal range before STS treat-ment,while the highest positive rate was detected inγ-GT(26.0%)and HA(54.0%). Following the STS treatment,the mean lev-els of all parameters and the positive rates reduced,with the greatest reduction observed inγ-GT(36.7%)and HA(37.8%);how-ever,the mean HA level was still higher than the normal range. The mean internal diameter of the portal vein reduced from(10.5± 1.7)mm before the STS treatment to(9.8±1.3)mm after the STS administration,with a significant diffrtence(P<0.05). Conclu-sion STS appears effective in the treatment of advanced schistosomiasis.
2.Effect of differential electroacupuncture frequencies on behavior of acute stress depression model of C57BL/6 mice

Objective To explore the effect of different frequencies interference of electoracupuncture on acute stress of C57BL/6 mice. Methods Thirty male C57BL/6 mice were randomly divided into model group,2Hz electroacupunture group and 100 Hz electroacupunture group.After interfering 7 days by electroacupuncture,all the mice were carried forced swimming tests and tail suspension test,and the immobility time in 5 minutes were taken down. Results The immobility time of forced swimming test and tail suspension test of 2 Hz eletroacupuncture group(32.00±14.73)s,(72.50±20.44)s was shorter than those of 100 Hz electoracupuncture group(145.60±34.62)s,(124.22±21.67)s( P <0.05); and the immobility time of 2 Hz eletroacupuncture group was shorter than that of model group(129.50±33.94)s,(142.23±20.05)s( P <0.05); but the immobility time of 100 Hz electroacupuncture group was no difference with that of model group( P >0.05). Conclusions The effect of 2 Hz eletroacupuncture could improve the behaviors of the acute stress of C57BL/6 mice and had the more effectiveness on anti-depression.

6.Methodology and Implementation of Forced Oscillation Technique for Respiratory Mechanics Measurement.

The forced oscillation technique (FOT) is a noninvasive method for respiratory mechanics measurement. For the FOT, external signals (e.g. forced oscillations around 4-40 Hz) are used to drive the respiratory system, and the mechanical characteristic of the respiratory system can be determined with the linear system identification theory. Thus, respiratory mechanical properties and components at different frequency and location of the airway can be explored by specifically developed forcing waveforms. In this paper, the theory, methodology and clinical application of the FOT is reviewed, including measure ment theory, driving signals, models of respiratory system, algorithm for impedance identification, and requirement on apparatus. Finally, the future development of this technique is also discussed.

7.Research on molecular chimeric mice pre-T cells infusion to reduce response to allogeneic T cells stimulation

Objective To research the effect of molecular chimeric mice pre-T cells on proliferation ability of allogeneic mouse T cells.Methods The MHC-Ⅰ gene (H-2Kband H-2Db gene) were extracted and amplified by RT-PCR,the identified pre-T cells were transfected by the constructed eukaryotic expression vector of C57BL/6mouse MHC-I (pIRES-H-2Db and pIRES-H-2Kb),non-transfected group and sham pIRES-transfected control group were set.The molecular chimeric cells were transfused back to BALB/c mouse.After 7 days,T lymphocyte cells of each group were extracted,the ability of molecular chimeric cells inducing spleen T lymphocyte response to allogeneic T cells was observed through mixed lymphocyte culture (MLC).Results Sequencing of the plasmid we have constructed showed that insertion sequence contained C57BL/6 mice H-2Kb and H-2Db series which could be retrieved from GenBank.The result of flow cytometry analysis indicated that H-2Kb and H-2Db protein had an increased expression in pre-T cells,the difference with other two groups was statistically significant (P<0.05).The result of MLC demonstrated that the stimulation index (SI) of T lymphocyte in co-injection transfected pIRES-H-2Kb and pIRES-H-2Db pre-T cells group (0.764±0.074) were significantly decreased compared with non-transfected group(0.983±0.081)and sham pIRES-transfected group(0.994±0.142) (P<0.05).Conclusions The molecular chimeric pre-T cells infusion can reduce spleen T lymphocyte response to allogeneic T cells and it may induce immune tolerance in vivo.
8.Changes of argiuine vaso-pression in blood and cerebral spinal fluid(CSF)in patients with severe brain injury after treated with mild hypothermia

Objective To study the changes of blood arginine vaso-pression(AVP)levels in patients with severe brain injury after treated with mild hypotherima.Methods Seventy-eight patients with severe brain injury were divided into mild hypothermia(33~35℃)group and normothermia group.The blood AVP levels and CSF AVP levels were determined at the third and the seventh day after brain injury.Results The AVP levels in blood and CSF of mild hypothermia group were lower than those of the normothermia group at the third and seventh day after brain injury(P<0.05).According to GOS,prognosis of the mild hypothermia group was better than that of the nonnotbcrmia group(P<0.05).Conclusion The mild hypothermia treatment may have inhibitive effects on the production of blood AVP,CSF AVP,and brain edema.Mild hypothermia is an effective method in the treatment of acute severe brain iniury in reducing the mortality and in increasing the survival rate.
9.Preparation and immunogenicity of influenza vaccine lyophilized liposomes

Aim: To prepare the influenza vaccine lyophilized liposomes and to characterize its particle distribution, encapsulation efficiency and immunogenicity. Methods: Flu vaccine liposome based on the method of thin-film evaporation was prepared using phospholipids , cholesterol and the purified influenza virus split vaccine, and was further subjected to frozen-drying. The polymorph was observed by microscope; the particle distribution and the average size were analysed by transmission electron microscope; its encapsulation efficiency was determined by Lowry method and the antibody titers were assessed by hemagglutination-inhibition after pulmonary delivery to mice. Results: The reconstitated influenza vaccine liposome under electronic microscope were round or elliptic particles evenly distributed at a mean size of 2. 14 祄, with the encapsulation efficiency of more than 80%. The antibody titer through pulmonary delivery was higher than that through intraperitoneal injection. Conclusion: The prepared influenza vaccine lyophilized liposomes possess high encapsulation efficiency, better particle distribution and marked immunogenicity through pulmonary delivery to mice. Pulmonary delivery of influenza vaccine liposomes is a potential immunization approach worthy of further exploitation.
10.Effects of propofol versus sevoflurane combined with remifentanil anesthesia on activity of nuclear factor kappa B in neutrophil granulocyte during one-lung ventilation in patients undergoing radical resection of esophagus cancer

Objective To compare the effects of propofol versus sevoflurane combined with remifentanil anesthesia on the activity of nuclear factor-kappa B (NF-κB) in neutrophil granulocyte during one-lung ventilation (OLV) in patients undergoing radical resection of esophagus cancer.Methods Forty ASA physical status Ⅰ or Ⅱpatients of both sexes,aged 40-64 yr,with body mass index 18-25 kg/m2,undergoing radical resection of esophagus cancer,were randomized into 2 groups (n =20 each):propofol-remifentanil group (group P) and sevofluraneremifentanil group (group S).Anesthesia was induced with midazolam,sufentanil,etomidate and cisatracurium besylate in both groups.The patients were tracheal intubated and mechanically ventilated.Anesthesia was maintained with iv infusion of propofol 4-8 mg· kg-1 · min-1 in group P,or with inhalation of 1%-3% sevoflurane in group S,and with iv infusion of remifentanil 0.2μg· kg-1 · min-1 and intermittent iv boluses of cisatracurium besylate 7 mg in both groups.The Narcotrend index value was maintained between 40 and 50.Blood samples were taken from the radial artery at 5 min after intubation,the beginning of OLV,30,60 and 90 min of OLV,0 and 10 min after re-expansion of the collapsed lung,and the end of surgery for blood gas analysis and for determination of plasma concentrations of IL-1.The oxygenation index and respiratory index were calculated.The nuclear protein of neutrophil granulocytes was extracted for detection of NF-κB-DNA binding activity.Results Compared with group P,the respiratory index,plasma concentrations of IL-1 and NF-κB-DNA binding activity were significantly increased (P < 0.05),and no significant change was found in the oxygenation index in group S (P > 0.05).Conclusion Compared with sevoflurane-remifentanil anesthesia,propofol-remifentanil anesthesia can inhibit activation of NF-κB in neutrophil granulocyte and is helpful in attenuating the inflammatory response in lung tissues during OLV in patients undergoing radical resection of esophagus cancer.
