2.Effects of mild hypothermia on secondary cerebral vasospusm and levels of endothelin,calcitonin gene related peptide in cerebrospinal fluid and plasma in patients with severe brain injury

Objective To study the effects of mild hypothermia on secondary cerebral vasospasm and levels of endothelin (ET),calcitonin gene related peptide (CGRP) in cerebrospinal fluid (CSF)and plasma in patients with severe brain injury(SBI).Methods Sixty-five patients with SBI were randomly divided into mild hypothermia group(30 cases)and control group(35 cases).Based on the routine treating method,the patients of mild hypothermia group were treated with local mild hypothennia.The levels of ET,CGRP in CSF and plasma were detected before and after treatment.And the incidence rates of secondary cerebral vasospasm cases were compared between the two groups.Results (1)At 7,14 day after treatment,ET levels in CSF and plasma in mild hypothermia group were significantly lower than these in control group (P<0.05or<0.01).The CGRP levels in CSF and plasma were the lowest at 7 day,then gradually increased in two groups.However,the CGRP levels in mild hypothermia group at 7,14 day were higher than those in control group (P<0.05 or<0.01).(2) The rate of secondary cerebral vasospasm in mild hypothermia group was 6.67% which significantly less than that in control group (22.86%) (P<0.05).Conclusion Mild hypothermia can prevent the increasing of ET and the decreasing of CGRP in CSF and plasma,and decrease the incidence rate of secondary cerebral vasespasm in patients with SBI.
3.Cigarette smoke extract regulates expression of heme oxygenase-1 through signaling pathway of protein kinase C-nuclear factor-erythroid 2-related factor 2 in rat airway epithelial cells

[ABSTRACT]AIM:ToobservetheeffectofthesignalingpathwayofproteinkinaseC(PKC)-nuclearfactor-ery-throid 2-related factor 2 (Nrf2) on the expression of heme oxygenase-1 (HO-1) induced by cigarette smoke extract (CSE) in the rat airway epithelial cells .METHODS:After exposed to CSE , the airway epithelial cells of male SD rats were di-vided into control group , CSE3 group, RO318220 group, Nrf2 siRNA group and RO318220+Nrf2 siRNA group.The pro-tein levels of HO-1, Nrf2 and PKC were semi-quantified by Western blotting .The protein expression of HO-1 was assessed by immunocytochemistry .The mRNA expression of HO-1 was detected by reverse transcription-polymerase chain reaction ( RT-PCR) .Nrf2 protein location was observed by immunofluorescence staining .HO-1 activity was also determined .RE-SULTS:After exposure to CSE for 3 h, the Nrf2 protein was mainly located in the nucleus , and the expression level of Nrf2 protein in the nucleus was stronger than that in the control cells .CSE also significantly enhanced the levels of p-PKC protein, and HO-1 mRNA, protein and activity .Pretreatment with RO318220 significantly decreased the levels of PKC pro-tein, Nrf2 protein, and HO-1 mRNA, protein and activity.Meanwhile, Nrf2 protein significantly decreased , and the activ-ity, mRNA and protein levels of HO-1 were also significantly attenuated by pretreatment with siRNA to knock down Nrf2. Pretreatment with RO318220 and Nrf2 siRNA significantly decreased the PKC protein , Nrf2 protein, and HO-1 mRNA, protein and activity .CONCLUSION:CSE up-regulates the HO-1 expression through PKC-induced nuclear translocation of Nrf2 in the rat airway epithelial cells .
4.Effect of Anatoxin-a on Expression of IEGs Genes in Mouse Brain

Objective To explore the role of IEGs in the central neurotoxicity of mice induced by anatoxin-a ANTX-A. Methods 80 Kunming mice were divided into groups and treated with ANTX-A at 200, 50 and 12.5 ?g/kg respectively i.p. for one month, 0.9% sodium chloride solution was used as the control. The expression of c-fos and NGFI-A genes and CREB protein in mouse brain were determined by using RT-PCR and immunocytochemical methods. Results The expression of c-fos gene significantly increased in the females of 50, 200 ?g/kg groups and in the males of 200 ?g/kg group compared with the control group. The expression of C-FOS and CREB-1 protein was significantly higher in 200 ?g/kg group than that in the control group. In all ANTX-A treated groups, the expression of NGFI-A gene did not show a significant difference compared with the control. Conclusion c-fos and CREB-1 genes may play a role in the central neurotoxicity induced by anatoxin-a.
5.In vitro culture and characterization of dental epithelial cells derived from the apical end of rat incisor

Objective:To investigate the biological characteristics of dental epithelial cells derived from cervical-loop epithelium in rat lower incisor. Methods:The apical end of the lower incisor was dissected from two-day-old Sprague-Dawley rats by visual microscopy, and dental epithelial cells were obtained by culture of the apical end explants of the lower incisor. The biological characteristics of dental epithelial cells were determined by morphology and immunocytochemistry fluorescence. Results:Dental epithelial cells emigrated from the cervical loop of rat lower incisor, and epithelium-like morphologies with round-shape or square-shape was observed. Dental epithelial cells had a striking proliferation potential with a high level of BurdU labeling. As a marker of epithelium stem cells, P63 expression was observed in most early cultured cells. However, as cells were subcultured, dental epithelial cells,with the lower level of BurdU labeling,decreasing expression of P63 and increasing expression of alkaline phosphatase, were inclined to terminal differentiation.Conclusion:Stem cells or dental epithelial progenitor cells might reside in the cervical loop of the apical end of the rat lower incisor. The present study may provide an excellent cell source for study on the differentiation of dental epithelial cells and tooth regeneration.

10.Clinical efficacy of sodium tanshinone IIA sulfonate in treatment of advanced schistosomiasis

Objective To evaluate the clinical efficacy of sodium tanshinone IIA sulfonate(STS)in the treatment of ad-vanced schistosomiasis. Methods Fifty cases with advanced schistosomiasis admitted to the Touzao Township Hospital of Dong-tai City during the period from November 2012 to November 2013 were treated with STS for 10 days,and the internal diameter of the portal vein,levels of ALT,AST,γ-GT,PIIIP,CIV,HA and LN were measured and compared before and after the adminis-tration of STS. Results The mean levels of all serological parameters except HA were within the normal range before STS treat-ment,while the highest positive rate was detected inγ-GT(26.0%)and HA(54.0%). Following the STS treatment,the mean lev-els of all parameters and the positive rates reduced,with the greatest reduction observed inγ-GT(36.7%)and HA(37.8%);how-ever,the mean HA level was still higher than the normal range. The mean internal diameter of the portal vein reduced from(10.5± 1.7)mm before the STS treatment to(9.8±1.3)mm after the STS administration,with a significant diffrtence(P<0.05). Conclu-sion STS appears effective in the treatment of advanced schistosomiasis.
