1.Percutaneous Fixation of Distal Tibia Comminuted Fracture by Using Normal Anatomical Plate

Objective To explore efficacy of minimally invasive percutaneous plate osteosynthesis (MIPPO) for the treatment of distal tibia comminuted fracture by using normal anatomical plate. Methods Between January 2007 and July 2008,18 cases of distal tibia facture were treated by MIPPO using anatomical plate. The clinical data of the patients were reviewed. Results A mean of 6.5 cm incision (5.0-8.5 cm) was made in the patients; the intraoperative blood loss ranged from 60 to 300 ml (mean,145 ml);and the operation time ranged from 30 to 120 min with a mean of 63 min. After the surgery,2 patients developed mild skin necrosis,and was then cured by conventional therapy; no patient had nonunion of the fracture,failure of internal fixation,or delayed wound healing. The 18 cases were followed up for a mean of 8 months (range,4 to 10 months); all of them were healed clinically and could walk without crutch in 4 months postoperatively. According to Johner-Wruhs score system,11 were excellent and 7 were good; the excellent-good rate was 100%. Conclusions MIPPO with anatomical plate is an optimal treatment for distal tibia comminuted fracture with advantages in protecting the soft issues and bony blood supply,promoting the wound-healing process,and reducing the rate of complications.
2.Content Determination of 4 Ingredients in Ligustrum lucidum Decoration Piece by Reference Extract Method

OBJECTIVE:To establish a method for the contents determination of 4 ingredients in Ligustrum lucidum decoration piece by reference extract method. METHODS:HPLC was performed on the column of XBridge C18 with mobile phase of acetoni-trile-0.1% formic acid(gradient elution) at flow rate of 1.0 ml/min,detection wavelength was 254 nm,column temperature was 30℃and volume injection was 10μl. RESULTS:The linear range was 0.037-0.598 mg for neonuezhenide,0.006-0.101 mg for ac-teoside,0.189-3.023 mg for nuezhenide and 0.314-5.027 mg for specnuezhenide(r≥0.999 0);RSDs of precision,reproducibility and stability tests were no more than 3.8%;recoveries were 98.46%-104.83%(RSD=2.43,n=6),95.55%-104.57%(RSD=3.63, n=6),100.09%-104.39%(RSD=1.45,n=6)and 98.84%-104.97%(RSD=2.02,n=6). CONCLUSIONS:The method is simple, good reproducibility,and can be used for the contents determination of 4 ingredients in L. lucidum decoration piece.
3.The apoptotic activity of hydroxycamptothecin against human gastric carcinoma and its mechanism of action

Objective To study the apoptotic activity of hydroxycamptothecin (HCFT) against human gastric carcinoma and its mechanism of action.Methods The apoptotic activity of HCPT against gastric carcinoma SGC-7901 cells was detected by MTT assay.The apoptosis occurrence was determined by Annexin V/PI staining and flow cytometry analysis.The mRNA expression of Bmi-1 was detected by real-time PCR analysis.Western blot analysis was used to detect the protein expression of Bmi-1.Results The IC50 values of HCPT against gastric carcinoma SGC-7901 cells was (4.87 ± 0.35) μ mol/L.After treatment with 0,5,10,20 μ mol/L HCPT for 48 h,the percent of apoptosis was elevated from (1.62 ± 0.37)% to (21.45 ±4.54)%,(36.67 ±5.38)%,(54.26 ±7.14)% and the 2-△Ct values of Bmi-1 mRNA was decreased from 0.614 ±0.022 to 0.445 ±0.018,0.376 ±0.012,0.215 ±0.010,there had significant difference among the different treatment (P < 0.01).Treated with 0,5,10,20 μ mol/L HCPT for 48 h,the protein expression of Bmi-1 was down regulated by HCPT treatment in a dose-dependent manner by Western blot analysis.Conclusion HCPT shows effective activity to induce apoptosis in gastric carcinoma cells,which is mainly related to the down regulation of Bmi-1.
4.Isolation and culture of murine embryonic stem cells of Kunming species

BACKGROUND: Hundreds of mouse embryonic stem cell lines are established presently. Most of these cell lines are collected from 129, C57BL/6J and BALB/C mice. Success rate of creating embryonic stem cell lines with Kunming mice is very low. OBJECTIVE: To explore the method of isolation and culture of murine embryonic stem cells of kunming species to increase the efficiency of establishment of embryonic stem cell lines from Kunming species mouse. DESIGN, TIME AND SETTING: The cell experiment was performed at the Chongqing Key Laboratory of Neurology from April 2006 to June 2007. MATERIALS: Blastocysts of 3.5 days and 4 days from Kunming species mouse were collected. METHODS: The blastocysts of 3.5 days and 4 days from Kunming species mouse were cultured on the fibroblast cell feeder layers for 4-5 days, and the cells from inner cell mass were isolated and subsequently cultured in vitro in 2.5 g/L trypase-0.4 g/L ethylenediamine tetraacetic acid (EDTA) and 1 g/L trypase-0.2 g/L EDTA at room temperature for 2-4 minutes. Embryonic stem cell colony was mechanically straggled. MAIN OUTCOME MEASURES: Colony growth was observed and determined by alkaline phosphatase staining, OCT-4 staining and karyotype analysis. Objective To explore the effect of embryonic stem cells(ESCs) differentiating into cardiomyocytes(CM) by cardiomyocytes.Methods The blastocytes of 3.5 d from Kunming mouse,cultured on mouse embryonic fibroblaste cell feeder layers for 4~5 d,the mouse ESCs were isolated and subsequently cultured.Day 2~3 embryoid bodies(EBs) were derived from ESCs of passage 3 to 5 and then incubated with neonatal cardiomyocyte(CM) to induce into cardiomyocytes and the expression of cardiac specific cardiac troponin-T(TnT) and ?-actin were detected by immunofluoresence.Results The rhythmic beating embryoid bodies were observed on day 3 and reached 93% of rhythmic beating embryoid bodies on 12 day in group 2.All the beating cardiomyocytes derived from ESCs expressed cardiac-specific proteins for TnT and ?-actin,and the group induced by direct cell contact acquired the highest differentiating ratio as 56.5%,and was the highest differentiating ratio compared with other groups(P

BACKGROUND:C2 transpedicle screw fixation for Hangman fractures has been paid more attention due to reliability and no loss of physiological function.However,there are lacks of biomechanical evidences for indication treatment because the fixation is single segmental.OBJECTIVE:To investigate the biomechanical stability of C2 transpedicle screw fixation for Hangman fractures.DESIGN,TIME and SEITING:This was a contrast study which was performed at the General Key Laboratory of Biomechanics,First Military Medical University of Chinese PLA from May to August 2004.MATERIALS:AO-universal titanium alloy transpedicle screw of 18 25 mm in length and 3.5 mm in diameter was adopted in this study.Six fresh C1-C4 cervical vetebrae samples were ordiually made into type Ⅰ,ⅡA,and Ⅱ Hangman fracture models.METHODS:After transpedicle screw fixation.Hangman fracture models were measured by non-destroyed style with spinal anterior flexion/posterior extension,left/right lateral curvature,and left/right axial direction.Loading/unlonding circulation was performed three times during each testing.Kinematics indicators were measured on the 3rd circulation.MAIN OUTCOME MEASURES:Spinal motor images at zero load and maximal load were obtained with laser photoscanning(0.1% in precision),and the corresponding systematic software was adopted to calculate 3D range of movement.RESULTS:The relative stability of type Ⅰ Hangman fracture models after C2 transpedicle screw fixation was 100.62%(inflexion),96.91%(posterior extension),99.19%(lateral curvature),and 97.12%(rotation)as compared to control group (P>0.05).The relative stability of type Ⅱ Hangman fracture models after C2 transpedicle screw fixation was 47.84%(inflexion),21.29%(posterior extension),65.98%(lateral curvature),and 41.69%(rotation)as compared to control group (P<0.05).CONCLUSION:Biochemical evaluation suggests that type Ⅰ and Ⅱ A Hangman fractures do fit for C2 transpedicle screw fixation,and the fixation may generate well physiological fixation or stability.However,stability of type Ⅱ Hangman fracture is poor,so it is not suitably adopted single transpedicle screw fixation.
7.The tissue reaction induced by copper ions to the smooth muscle of the terminal rectum

Objective To investigate the tissue stimulation of copper ions to the smooth muscle of murine terminal rectum.Methods In this study copper needle used for electrochemical therapeutic equipment was punctured into submucosal smooth muscle tissue of terminal rectum above the dentate line in 65 male rats.Rats were then divided into 5 groups (10 rats in each group) at random and sacrificed at 1, 3, 4, 6, 8 weeks after respectively, samples were sent for observation of macroscopic and microscopic tissue reaction.Results Rats had no abnormal histological change in the puncture points grossly. Undor microscope, mild edema was detected in the submucosal layer in 1 week group. Remarkable infiltration of inflammatory cells and lymphocytes subsided 3 weeks after, and disappeared five weeks after.Smooth muscle was normal microscopically in all groups.In contrast, platinum and steel needles caused infiltration of plasma cells and neutrophils, ulcers and small abscess formation in around puncture points.Conclusion Inflammatory reaction was induced after puncturing of copper needle into the tissues and the tissue reaction disappeared after 5 Ws.In contrast with platinum and steel needles, copper needle was non-traumatic to the smooth muscle tissue.
8.Efficacy and safety of lamivudine in the patients of hepatitis B virus-associated glomerulonephritis treated by prednisolone with leflunomide

Objective To investigate the efficacy and safety of lamivudine in the patients of hepatitis B virus-associated glomerulonephritis (HBV-GN) treated by prednisolone with leflunomide. Methods 41 HBV-GN patients treated by prednisolone and leflunomide were enrolled in this study. According to the presence of the indications of antiviral treatment, the patients were divided into the prevention group and the treatment group received lamivudine treatment, and another 15 patients with chronic hepatitis B were treated with lamivudine as the control group. The biochemical, virological and serological responses during the process of the treatment and the serum levels of IFN-γ and IL-4 were determined. Results No significant differences of biochemical, virological and serological responses in the prevention and treatment groups compared with the control group at 48 weeks after treatment. Levels of the IFN-γ and IFN-γ / IL-4 decreased significantly in the prevention group and treatment groups compared with the control group (P < 0.05, respectively). No serious adverse reactions occurred in all patients. Conclusion The antiviral treatment of lamivudine lead to good curative effect and security in HBV-GN patients received hormones combined with leflunomide treatments, but leflunomide with prednisolone inhibited the production of IFN-γ and removal of hepatitis B virus.
9.A sensitive LC-MS/MS method for simultaneous assay of three iridiods in Gardenia jasminoides Ellis

Objective To develop a simple, rapid and sensitive method for the determination of major compounds from Gardenia jasminoides Ellis using liquid chromatography tandem mass spectrometry (LC-MS/MS).Methods The analysis was performed on Dikma Diamonsil?C18 (100mm×4.6mm, 5μm) column with acetonitrile-0.1%acetic acid and 0.1%acetic acidwater as mobile phase at a rate of 0.4ml/min.The column temperature was set at 40℃and the injection volume was 2μl.Quantification of these compounds was performed by LC-MS/MS with positive or negative ion mode electrospray ionization (ESI) in the multiple reaction monitoring (MRM) mode.Nebulizer gas, 3L/h;drying gas, 15L/h;desolvation line (DL) temperature, 240℃;heat block temperature, 300℃;CID, 230kPa.The mass transition of the precursor/product ions was monitored at m/z 391.10→149.30for shanzhiside, 573.40→365.05for genipin-1-gentiobioside, 447.30→225.15for geniposide.Results The regress equation of shanzhiside, genipin-1-gentiobioside and geniposide were Y=243.810 X-289.957, r=0.999 9;Y=137.125 X+2 092.76, r=0.999 6;Y=2 030.32 X+823 213, r=0.999 8in the range of 10.76-215.2ng/ml;516-4 128ng/ml;2 000-20 000ng/ml respectively.This validated method has good repeatability, precision, recovery and stability.The results meet the requirements by regulation.Conclusion This method shortened the analysis time and improved efficiency.It assayed the three iridoid glycosides in Gardenia jasminoides Ellis sensitively and precisely.This method can be used for the quality control of Gardenia jasminoides Ellis.
10. Reflections on the teaching of infectious diseases based on the One Health concept

Infectious disease is a major public health problem threatening human health and social development, and infectious disease teaching is an important part of public health education. However, traditional infectious disease teaching faces challenges such as overly theoretical and fragmented content, overly traditional and monotonous methods, and a narrow and isolated vision, which makes it difficult to adapt to the current complex and changing situation of infectious disease prevention and control. The “One Health” concept emphasizes interdisciplinary, cross-sectoral, and cross-regional communication and cooperation to achieve the harmonious unity of health for humans, animals, and the environment, which is significant in preventing and controlling infectious diseases. This paper proposes some ideas for reforming infectious disease teaching based on the One Health concept, which aims to improve the quality and effectiveness of infectious disease teaching by recognizing the close connection between human health, animal health, and environmental health. Specifically, it is suggested to reform the teaching in four aspects: introducing the basic concepts, principles, and practices of One Health, increasing the content of infectious diseases related to One Health, adopting diversified and interactive teaching methods, and establishing cross-disciplinary teaching cooperation. Concrete recommendations are provided for each aspect. This paper argues that reforming infectious disease teaching based on the One Health concept is conducive to cultivating public health talents with global vision, systems thinking, and cross-disciplinary collaboration capabilities, providing robust talent support for coping with emerging, re-emerging, and endemic infectious disease threats.
