2.Analysis of stability and healing on minimally invasive osteotomy for the treatment of hallux valgus.

To treat hallux valgus, minimally invasive osteotomy on 1st metatarsal neck,immobilization with "8-shape" bandagev was performed, and the effect was definited. The principles of osteotomy stability were analyzed from the osteotomy position, methods, direction and angle. The grinding drill was used to increase friction coefficient between the ends of osteotomy. Correct direction of osteotomy and suitable angle were the key point of stability. The immobilization with "8-shape" bandage complied with the principle of elastic fixation created the conditions for the slight movement of the osteotomy ends. Compared with internal fixation ,it was better on osteotomy healing,and the osteotomy ends were stable and healed with cartilage

3.Preparation and clinical application of cDNA microarray for combined detection of hepatitis virus

Objective To develop the cDNA microarray for the combined detection of hepatitis virus, and to study the feasibility of applying the microarray in clinical setting. Methods For the small and simple genome of HBV and HDV, the specific primers of PCR were designed with Primer Premier 5.0 program according to the conserved region of HBV and HDV, and 10 and 4 gene fragments were obtained respectively, which could be used as the probes of gene chip. As for the complex genome of HCV, the technique of restriction display PCR (RD-PCR) was employed. Some of gene fragments were selected which were comparatively more specific and sensitive as microarray probes. In order to explore the experimental conditions of microarray in vitro detection, three types of gene chip were prepared successively including HBV and HDV simultaneous detection, HCV detection and modified HCV detection. Results The hybridized signals on the gene chip showed that the effect in detection was satisfactory. Through the prepared gene chips mentioned above, some probes with good quality were selected and the microarray was prepared for HBV, HCV and HDV simultaneous detection. The diagnostic capability of the microarray was evaluated following the washing and scanning steps. Linearity: Serial dilutions of the target DNA or cDNA showed that a strong linear relationship existed between the various concentrations of target DNA or cDNA and the fluorescence intensities obtained from microarray assay (r=0.990 2, r=0.992 1, r=0.981 9), and that the detection range for the microarray was from 104 to 1011 copies/ml. Specificity: Samples from other viruses such as YFV, JET and DV were also subjected to the test and the results were all negative. Reproducibility: The reproducibility of this assay system was evaluated by repeated measurements, and the within-run coefficient of validation of HBV, HCV and HDV were 7.1%, 7.2% and 6.6%, respectively, while the between-run coefficient of validation was 7.9%, 8.2% and 7.6%, respectively. Accuracy: By using the BLAST and the GenBank database, the identity of the obtained sequences including 16 fragments of PCR, 24 RD fragments of HCV and some positive serum samples were verified, each sequenced product was confirmed to be a genome fragment of expected size. In order to fulfill the request of clinical diagnosis, a modified protocol for microarray detection was established. This new protocol consisted of two hours of hybridization, omitting the steps of prehybridization and purification of samples, and the hybridization temperature was elevated from 42℃ to 52℃, et al. The whole protocol could be completed in less than 8 hours. 98, 42 and 5 serum samples from hepatitis B, C and D patients and 130 samples from healthy people were analyzed, respectively, by microarray assay and real-time PCR (Taqmam method). 5.Application effect of Fufukangspray in post partum women

Objective To explore the application effect of Fufukangspray in postpartum women in the treatment of vaginitis . Methods From July 2013 to June 2012 in our hospital ,80 cases of postpartum vaginitis patients as the research object .The control group was treated with Metronidazole Suppositories treatment ,the experimental group were treated with Fufukangspray treatment . Comparison of the two groups in the therapeutic effect and adverse reaction of differences .Results Compared with the control group ,we found that the experimental group total effective rate was significantly higher ,the incidence of adverse reaction was low‐er ,the differences between groups by chi square test analysis that was statistically significant(P<0 .05) .Conclusion Fufukang‐spray has good application effect in the treatment of postpartum vaginitis ,curative effect ,less adverse reactions ,it is worthy of clini‐cal application .
6.Content Determination of 4 Index Components in Uygur Medicine Capparis spinosa by HPLC

OBJECTIVE:To establish the method for determining the contents of protocatechuic acid,rutin,gallic acid and kaempferol in Uygur medicine Capparis spinosa. METHODS:HPLC was performed on the column of Agilent C18 with the mobile phase of acetonitrile-0.1% phosphoric acid solution(7∶93,V/V)at the flow rate of 1.0 ml/min,the detection wavelength was 327 nm,column temperature was 30 ℃ and volume was 10 μl. RESULTS:The linear range was 0.260 0-50.0 μg for protocatechuic ac-id(r=0.995 6),3.109 0-102.0 μg for rutin(r=0.999 9),1.018 0-40.0 μg for gallic acid (r=0.998 9) and 0.063 0-36.0 μg for kaempferol(r=0.998 8);RSDs of precision,stability and reproducibility tests were all no more than 1.20%;average recoveries were respectively 101.51%(RSD=1.85%,n=6),99.70%(RSD=1.23%,n=6),98.28%(RSD=1.86%,n=6) and 100.97%(RSD=1.74%,n=6). CONCLUSIONS:The method is specific,and can fast and accurately determine the contents of protocate-chuic acid,rutin,gallic acid and kaempferol in Uygur medicine C. spinosa.
7.Anticancer Effects of Xiaotansanjie Recipe on Microsatellite Instability in MKN-45 Human Gastric Cancer Orthotopic Transplantation Tumor

Objective To explore the mechanism of Xiaotansanjie recipe on MKN-45 human gastric cancer orthotopic transplantation tumor with microsatellite instability. Methods MKN-45 human gastric cancer cell line was used to establish subcutaneous tumor model in nude mice as an experimental model which passed three generations of tumor source. Then OB glue was used to establish human gastric cancer orthotopic xenograft model. Twenty model nude were divided into model group and Xiaotansanjie recipe group (treated with Xiaotansanjie recipe). After 6 weeks of treatment intervention, tumor weight was measured, inhibitory rate was calculated and microsatellite instability (D17S250, D2S123, D5S346, Bat-25 and Bat-26) was detected. Results The tumor weight in Xiaotansanjie recipe group was significantly lower than that in model group (P<0.01). The tumor-inhibition rate was 40.84%. Five microsatellite markers (D17S250, D2S123, D5S346, Bat-25 and Bat-26) were amplified in the tumor tissue of model group, and microsatellite markers D2S123, D5S346 appeared some prolong and reduction. Microsatellite markers (D2S123, D5S346) mutation was stabilized after 6 weeks treatment of Xiaotansanjie recipe. Conclusion Xiaotansanjie recipe can inhibit the MKN-45 human gastric cancer orthotopic transplantation tumor by down-regulating the microsatellite instability.
8.Self-evaluation of dentofacial esthetics in skeletal malocclusion patients

Objective To investigate the difference between the self-evaluation of patients with dental and facial esthetics in skeletal malocclusion and normal persons.Methods The subjects were divided into three groups:62 patients with skeletal Class Ⅱ,84 patients with skeletal Class Ⅲ,as well as 130 persons in control group.There was no obvious difference in gender and age among the three groups.Happiness with dental and facial appearance was assessed using questionnaires.Analysis of variance,linear regression and logistic regression were used for statistical analysis.Results Skeletal malocclusion patients were less happy with their dental appearance than control group.Class Ⅱ patients and women had lower happiness scores for their dental appearance.Among skeletal malocclusion patients,the shape (44.5 %) and prominence of their teeth (17.8 %) were the most frequent causes of concern.Older subjects,women,and skeletal malocclusion patients were less happy with their facial appearance.Most of them were dissatisfied with the facial appearance focused on the chin (53 %),facial contours (37 %),and the position of the teeth when smiling (33 %).A greater proportion of Class Ⅲ subjects than Class Ⅱ subjects wished to change their appearance.Conclusions The findings indicate that women and skeletal malocclusion patients have lower levels of happiness with their dentofacial appearance.Although Class Ⅱ patients exhibit the lowest levels of happiness with their dental appearance,there is some evidence that concerns and awareness about their facial profile are more pronounced among the Class Ⅲ patients.
9.Interpretation of standards on training and evaluation of urine analyzers in grass-roots medical unit

Objective To analyze the training and evaluation standards on urine analyzers in the grass-roots medical unit.Methods The applicability,requirements,subject setup and items of the standards as well as the requirements of evaluation scoring chart were expounded in detail,and the related elementary knowledge was analyzed including the structure,working principle,installation and adjustment,operation,application,maintenance and etc of the analyzer.Combined personnel with the equipment was suggested to ensure the accuracy and high efficiency of the test result.Results The interpretation of the standards contributed to enhancing the quality of medical equipment training and the ability of practical support,and facilitated the examiner to perform annual evaluation with the scoring chart.Conclusion It is necessary to grasp accurately the connotation of the standards,to improve the quality of medical equipment training and to enhance combat support capability for the grass-roots medical unit and examiner.
