2.Literature study of TCM pattern and medicine-use analysis of systemic lupus erythematosus

Objective To study the rule with the treatment based on the pattern differentiation of systemic lupus erythematosus, by analyzing the literature of traditional Chinese medicine for the treatment of systemic lupus erythematosus from January 1999 to July 2014 included in China National Knowledge Infrastructure(CNKI) and summarizing the frequency of TCM pattern and medicine.Methods The frequency statistics and classified analysis were used to describe the patterns and medicines.Results There were 14 patterns, the common patterns are pyretic toxicity with 23(15.13%), deficiency of both qi and yin with 21(13.82%), qi and blood stasis with 20(13.16%). The disease was involved with liver, spleen and kidney. Pattern factors were mainly sthenia factors, accounting for 65.30% of the total, including hot, blood stasis, toxin, damp-heat, qi stagnation, phlegm; asthenia factors were accounted for 34.70% of the total, including deficiency of yin, deficiency of qi, deficiency of yang and deficiency of blood. There were 201 medicines included, the total frequency was 1 669, which could be into 17 categories. The top three were heat-clearing, tonic and stasis-dissolving medicines. Medicine tropisms are mainly lung tropism, stomach tropism, liver tropism, heart tropism, spleen tropism and kidney tropism, accounting for 88.12% of the total.Conclusions Pyretic toxicity is responsible for the sthenia factors of systemic lupus erythematosus, and deficiency of qi and yin is the main responsibility for asthenia factors, treatment should pay attention to both sthenia and asthenia factors.
3.Correlation of chronic prostatic inflammation with prostate cancer and hyperplasia of prostate in the elderly

Objective To investigate the pathological features of chronic inflammation in prostatic specimens,and study the correlation of chronic prostatic inflammation with prostate cancer and hyperplasia of prostate in the elderly.Methods The histopathologic features of prostatic specimens which were taken during prostatectomy were retrospectively observed.The inflammatory cells in partial cases were labeled by immunohistochemical markers such as CD3,CD20,CD4,CD8,CD117 and CD138,and by inflammation related factors such as nuclear factor(NF)-κB,(COX-2),tumor necrosis factor-α(TNF-α)and inducible nitric oxide synthase (iNOS).Results The chronic prostatic inflammation of different extent were found in 100 patients with prostate cancer and 76 patients with benign prostatic hyperplasia,among them 71 cases (40.3 %) had mild chronic prostatic inflammation,80 cases (45.5 %) had moderate chronic prostatic inflammation,25 cases (14.2 %) had severe chronic prostatic inflammation.Inflammatory cells mainly were CD3-labeled lymphocytes,accompanied by a small amount of mononuclear cells and mast cells.Chronic prostatic inflammation was not correlated with prostatic carcinoma and its differentiation degree,benign prostatic hyperplasia,focal atrophy and its subtypes,and high grade prostatic intraepithelial neoplasia (all P> 0.05).There was correlation between chronic prostatic inflammation and expression of COX-2 (P< 0.05).Conclusions The histological chronic inflammation is common in the prostate specimens in elderly men.There are no direct correlations of prostatic inflammation with prostatic carcinomas and benign prostatic hyperplasia.Prostatic inflammation is positively correlated with the expression of COX 2,which may be associated with the activation of COX-2 pathway induced by oxidative stress.
4.Assessment of two methods in evaluating alveolar bone loss by micro-CT based on periodontitis model in mice

Objective To evaluate two methods measuring alveolar bone loss by micro computed tomography (micro-CT)based on periodontitis model in mice.Methods The silk ligatures were tied around the right maxillary second molars of mice to induce periodontitis model.The right half maxillaries of mice model were harvested for micro-CT analysis.Three dentists were recruited for the measurement with two different methods:Modified tomography (T) method and reconstruction (R) method.Accuracy and consistency of each method were estimated by standard deviation (SD).Results The SDs of R method managed by the same operator (measurement for 3 times) or different operators (3 operators) were 34.87μm and 35.67 μm respectively,while that of T method was 7.82 μm and 14.24 μm respectively.The SDs of T method were significantly lower than those of R method (both P<0.05).Conclusion T method is more accurate and consistent than R method for evaluating alveolar bone loss in mice periodontitis model.
5.Effect of disinfectant with benzethon chloramine and isopropanol as main active ingredients on the accuracy of dental impression.

OBJECTIVE:
To assess the effect of disinfectant (Cavicide) with benzethon chloramine and isopropanol as main active ingredients disinfectant on dental impression accuracy.
METHODS:
The effect of Cavicide on three impression materials (alginate, polyether and vinylpolysiloxane) were assessed using a standard model. The standard model was digitized by an extraoral scanner (IScan D103i, Imetric). For each kind of impression materials, thirty impressions were taken following the manufactures' instruction in the same conditions. Subsequently, the impressions were randomly divided into three groups, with ten impressions in each group. After the impression taking was completed, the three groups underwent pure water rinse for 1 min (blank control, BC), 2% glutaraldehyde solution immersion disinfection for 30 min (glutaraldehyde, GD), and Cavicide solution spray disinfection for 5 min (Cavicide, CC), respectively. All the impressions were digitized by the extraoral scanner (IScan D103i, Imetric) after disinfection and exported to a dedicated three-dimensional analysis software (Geomagic Qualify 2014, Geomagic, USA). In the software, the digital models of the impressions were trimmed to teeth and then superimposed with the digitized standard model via best-fit alignment. Root mean square (RMS) was used to evaluate the deviations between the impression and the standard model. The deviation in the anterior and posterior regions was evaluated respectively. One-way ANOVA test and the LSD post-hoc test were used to compare the deviations between the three groups (P < 0.05). The color map of each superimposition was saved for visual analysis.
RESULTS:
For the polyether and vinylpolysiloxane materials, the difference between the three groups was not statistically significant (P=0.933, P=0.827). For the alginate material, the difference in posterior region between group GD and group BC, as well as group GD and group CC were statistically significant (GD vs. BC, P=0.001; GD vs. CC, P=0.002), while the difference between group BC and group CC was not statistically significant (P=0.854). The visual analysis showed an obvious deviation in the buccal-lingual direction in group GD.
CONCLUSION
Disinfectant (Cavicide) with benzethon chloramine and isopropanol as main active ingredients using spray disinfection has no effect on the accuracy of the alginate, polyether and vinylpolysiloxane impressions.

6.Cloning of two different IL-15 genes and their effect on the humoral immune response elicited by HBV vaccine

Objective:To study the effect of IL-15 eukaryotic expression plasmid on the humoral immune responses to HBV surface antigen protein vaccine.Methods:Had constructed two of IL-15 eukaryotic expression plasmides.One is the eukaryotic expression plasmid of the IL-15 whole sequence,the other is the chimeric eukaryotic expression plasmid of the IL-2 signal peptide and IL-15 mature peptide sequence(IL-2s-15).The bioactivities of the expression products of the two plasmids were identified by CTLL-2 proliferation assay.Results:After IL-15 plasmid co-immunized with HBsAg,the titer of anti-HBsIgG was much higher than that of control( P 0.05),however,the anti-HBsIgG2a/IgG1 ratio was higher than that of control.Conclusion:IL-15 expression plasmids effect differently on the immune responses induced by protein vaccine.
7.The Mutasynthetic Production of CHC-B1 from a Streptomyces avermitilis Mutant in the Presence of Cyclohexanecarboxylic Acid

Gene deletion vector pLJ04(pKC1139∷bkdF+bkdH)was used to disrupt bkdFGH in Streptomyces avermitilis 76-02-e,an industrial producer of anthelmintic avermectin.The disruptants were confirmed by PCR.Shake flask experiment and HPLC analysis showed that the mutant lost the ability to produce avermectins.As it is expected,the mutant,named S.avermitilis bkd76-3,could restore the ability of producing avermectins when the feeding of methylbutyric acid and isobutyric acid to its fermentations was carried out.The addition of cyclohexanecarboxylic acid(CHC)into fermentations of the S.avermitilis bkd76-3 allowed for production of four components,two of which was confirmed as CHC-B1 and CHC-A2 by LC/MS analysis,respectively.
8.Identification of Viticis Fructus and Its Adulterants by ITS2 Sequence

ITS2 sequence was used as a barcode to identify Viticis Fructus and its adulterants. The study collected 46 samples include Viticis Fructus and its adulterants. The sequences were obtained by extracting DNA, amplification, sequencing bi-direstionlly, and then assembling with CodonCode Aligner. To identify Viticis Fructus and its adulterants, the study used the methods of computing the genetic distances by Kimura 2-Parameter (K2P) model and constructing the Neighbor-joining (NJ) phylogenetic trees using MEGA5.0. The results showed that the maximum inter-specific K2P distance of Viticis Fructus was less than the minimum inter-specific K2P distance of Viticis Fructus and its adulterants and the NJ tree indicated that the Viticis Fructus and its adulterants could be distinguished clearly. Therefore, using ITS2 barcode can distinguished Viticis Fructus and its adulterants accurately.
9.Detection rate of post-traumatic stress disorder symptoms among military rescuers in Wenchuan earthquake and its interventional factors

Objective To investigate the positive detectable rate of post-traumatic stress disorder ( PTSD ) and its influencing factors among the military rescuers , and explore preventive measures for rescuers .Method At three points , 2600 soldiers from Wenchuan military rescuers were enrolled , whose PTSD symptoms were evaluated by PCL-S.Self-edited questionnaires on the general state , Alerting Test Questionnaires for Military Group Stress ( ATQMGS ) , and State-Trait Anxiety Inventory(STAI)were used to evaluate the influencing factors of rescuers′PTSD symptoms.Results Seventy-one rescuers were tested positive for PTSD and the positive detectable rate was 2.9%.On the basis of demographic data , the difference test suggested that rescuers different in terms of positions , duration of rescue, family structure, trauma history, mental problem history or trauma history during the rescue were significantly differenct in PTSD symptoms , which were strongly and positively correlated with anxiety , morale, and self-efficacy, but were strongly and negatively correlated with leadership support , control feeling , promotion motivation , cognition of tasks , sense of belonging and work value .Multiple linear regression showed that the influencing factors of PTSD symptoms included the trait of anxiety , leadership support , trauma history, control feeling, mental problem history, trauma history during the rescue, position and duration of rescue. Conclusion The PTSD positive detectable rates among the military rescuers are lower than in the previous research.PTSD symptoms of military rescuers are related to such factors as the position , duration of rescue , family structure , trauma histo ry, mental problem history, trauma history during the rescue, trait of anxiety, morale, self -efficacy, leadership support, control feeling , promotion motivation , cognition of tasks , sense of belonging and work value .The influencing factors of PTSD symptoms are the trait of anxiety , leadership support, trauma history, control feeling, mental problem history, trau-ma history during the rescue , position level and rescue duration .
10.Efficacy Observation for Treating Ankylosing Spondylitis by Chinese Herbs and Recombinant Hu- man Tumor Necrosis Factor Receptor II-Antibody Fusion Protein.

OBJECTIVETo observe the clinical effect of Chinese medical (CM) syndrome differentiation based Chinese herbs and recombinant human tumor necrosis factor receptor II-antibody fusion protein (etanercept) for treating ankylosing spondylitis (AS) patients.
METHODSTotally 35 AS patients were treated with syndrome differentiation based Chinese herbs and etanercept. Reinforcing Shen and strengthening Du channel, activating meridians to stop pain was principle used in syndrome differentiation based treatment. Etanercept was subcutaneously injected, 25 mg each time; twice per week for the first three months and once a week for the latter three months. The clinical efficacy was evaluated after 3 and 6 months of treatment. Meanwhile, ASAS20 and ASAS50 standards arriving rates were also observed. Bath Ankylosing Spondylitis Disease Activity Index (BASDAI), Bath Ankylosing Spondylitis Functional Index (BASFI), visual analog score (VAS) for spine pain, VAS for night pain, patient global assessment (PGA), VAS for physician global assessment, CM syndrome score, finger-ground distance, thoracic activity, tragus-wall distance, lumbar scoliosis, cervical rotation, Schober improved test, erythrocyte sedimentation rate (ESR), and C-reactive protein (CRP) were observed before treatment, 3 and 6 months after treatment.
RESULTSCompared with before treatment, BASDAI, BASFI, VAS for spine pain, night pain, physician global assessment, PGA, CM syndrome score, finger-ground distance, thoracic activity, tragus-wall distance, lumbar scoliosis, Schober improved test, ESR, and CRP all decreased after 3 and 6 months of treatment, with statistical difference (P < 0.05). Cervical rotation also decreased after 6 months of treatment, with statistical difference (P < 0.05). Compared with 3 months of treatment, total effective rate of CM syndrome, ASAS20 and ASAS50 standards arriving rates increased after 6 months of treatment, with statistical difference (P < 0.05). There were statistical differences in all indices mentioned above between after 3 months of treatment and after 6 months of treatment (P < 0.05).
CONCLUSIONSyndrome differentiation based Chinese herbs combined etanercept could alleviate inflammatory reaction favorably, control the progression of active AS, and improve joint functions.
