1.Current status of research on liver innate immunity

Liver innate immunity plays an important role in the defense against pathogen invasion.The mechanisms of liver innate immunity for defending against pathogen invasion are reviewed,including the special anatomical and physiological characteristics of the liver,the fea-tures of innate immune cells in the liver (liver-settled cells and cells raised from blood circulation)and their function and relationship with each other in pathogen invasion,the process of pathogen identification and effect of liver innate immune cells,and the mechanism of liver immune injury and immune tolerance.Given current research on innate immune cells and innate immune molecules in the liver,further stud-ies should be conducted to understand the connection and difference between liver innate immunity and systemic immunity,thus promoting the diagnosis and treatment of liver diseases and deeper research.

4.The clinical features and outcomes in 21 patients with ocular adnexal mucosa-associated lymphoid tissue lymphoma

Objective To evaluate the clinical features,therapies and prognosis in patients with mucosa-assoeiated lymphoid tissue (MALT) lymphoma in ocular adnexal marginal zone (OAML).Methods A retrospective analysis was made upon clinical data from 21 patients with OAML admitted into Beijing Tongren Hospital from June,2008 to December,2011.Results There were 12 (57.1 %) men and 9(42.9%) women,with a median age of 57 (23-79) years old.Majority of patients had localized pathological changes.Among them,16 patients (76.2%) were in stage Ⅰ E,and 5 (23.8%) in stage Ⅳ E.Surgical resection as the sole treatment was performed in 13 patients (61.9%),and positron emission tomography CT(PET-CT) imaging demonstrated normal fluorine 18-fluorodeoxyglucose (FDG) uptake after surgical resection,who were managed with no further therapy.All the 13 patients were followed up for median 14 (5-38) months,and all in complete remission.Combination chemotherapy was given to 8(38.1%) patients.Three patients in stage Ⅰ E treated with COP (cyclophosphamide,vineristine and prednisone) or CHOP (cyclophosphamide,adriamycin,vincristine and prednisone) were all in partial remission.Five patients in stage ⅣE were treated with COP/CHOP in combination with rituximab,and all in complete remission.The 3-year overall survival rate and disease-free survival rate in the total patients were 100.0% and 74.9% respectively.Conclusions The patients with OAML generally have localized disease,show indolent clinical course,and present low lymphoma-related mortality.Surgical resection is a very important treatment in the patients with local disease.Systemic chemotherapy should be considered in patients at advanced stages.Rituximab in combination with chemotherapy can improve the remission rate.
5.Diagnosis and treatment of primary gastric malignant lymphoma

Objective To explore the diagnosis,treatment and prognosis of primary gastric malignant (lymphoma)(PGML).Methods The diagnosis and treatment data of 21 cases of PGML admitted in our(hospital) during 8 years was retrospectively analyzed.The diagnostic methods included barium meal(examination),gastroscopy,B type ultrasonography,and CT scan.All patients underwent operative treatment and most of the patients received adjuvant chemotherapy.Results Upper abdominal pain,digestive tract bleeding,emaciation and abdominal mass were common clinical manifestations of PGML.The resectabilty rate of PGML was 90.5% while the 5-year survival rate was 57.1%.Conclusions The key to a satisfactory prognosis was early diagnosis,radical curative operation and combined treatment.

Objective:To evaluate the inhibitory effect of Herceptin on HeLa,SiHa cells and to investigate the synergistic mechanism of Herceptin and Carboplatin.Methods:HeLa and SiHa cells were treated with Herceptin(at 5,10,20,40,and 80 ?g/ml),Carboplatin(at 0.5,1,2,4,and 8 ?g/ml),and Her+CBP[(10+1),(20+2),and(40+4)?g/ml] separately.The untreated cells were taken as control.SP immunohistochemical method was used to detect the protein expression of HER-2/neu and downstream Ras oncogene.MTT method was used to study the inhibition effect of Herceptin on HeLa,SiHa cells and its synergetic effect with carboplatin.FCM was used to detect the cell apoptosis and cell cycle.The mRNA expression of her-2/neu and ras were assessed by RT-PCR;the protein expression of HER-2/neu and Ras were studied by Western blotting.Results:Herceptin significantly inhibited cervical cancer cells proliferation,and there was a synergistic effect when combined with carboplatin(P
7.The application of super-selective external carotid artery embolization in head and neck diseases

Objective To study the application of super-selective external carotid artery embolization in head and neck diseases. Methods DSA and super-selective external carotid artery embolization were carried out in 41 cases of head and neck diseases including 12 cases of epistaxis,7 nasopharyngeal fibroangioma,1 traumatic arterial bleeding,14 vascular malformation,and 7 malignancies. Results No recurrence of nose bleeding after embolization of epistaxis was seen within 6 - 12-month follow up. The operative bleeding was reduced significantly by preoperative embolization in nasopharyngeal fibroangioma. No recurrence of bleeding was achieved after embolization of traumatic artery. Among the cases of vascular malformation,3 were proven to be significantly effcient,6 effcient,and 5 inefficent in the 6 - 12-month follow up. Among the 7 malignant cases,3 survived more than 2 years. Conclusion Super-selective external carotid artery embolization is safe and effective in the treatment of head and neck diseases. According to the theory of in vitro radioassay, using TSHAb as binder and 125I-staphylococcal protein A (125I-SPA) as tracer agent, a new method for detecting the abnormal immunoglobulin (aIgG) in the sera of patients with Graves disease (GD) was reported. In the initial study of the method, the most appropriate interacting conditions of TSHAb with aIgG were explored and compared with ELISA using ganglioside (GLS) as a binder. The relationship between aIgG and thyroid-stimulating imunoglobulin(TSI) was probed into. The results showed that TSHAb could interact specifically with aIgG in vitro; but it could not interact with IgG from sera of normal subjects, systemic lupus erythematosis (SLE) patients and diabetic patients. With aIgG as an evaluating index, the mean value in 29 normal subjects was 1. 06?0. 17. When the sera of 72 patients with GD in different clinical stages were studied, the aIgG index was positive in 83% of untreated GD patients (n = 24), in 12% of GD patients in clinical remission (n = 25) and in 82% of relapsed GD patients (n = 23). 10.Observation of chimeric hepatitis C virus In infected Huh7.5 cell through transmission electron microscopy

Objective To observe the morphological characteristics of HCV particles and intracel-lular ultrastructure changes in Huh7. 5 cells which was infected with chimeric HCV via transmission electron microscopy. Methods Plasmid J6/JFH encoding the full length HCV chimeric genome was transcribed to HCV RNA in vitro and the RNA was transfected into Huh7.5 cells by electroporation. Quantitative real-time PCR(qRT-PCR) was used to assay HCV copies of the supernatant of transfected cells. Indirect immunofluo-rescence was used to detect the expression of HCV proteins. The cell culture superoatant were used to infect narve Huh7.5 cells, transmission electron microscopy was used to observe morphological characteristics of vi-rus particles and intracellular ultrastructure changes in infected Huh7. 5 cells. Results qRT-PCR showed high level virus copies in supernatant of transfected cells collected in different times, indirect immuno-fluo-rescencc proved high expression of HCV NS5A proteins in the transfected cells. Large numbers of enveloped or unenveloped virus-like particles (VLPs) were observed in infected Huh7. 5 cells via transmission electronmicroscopy. We also found hyperplasia of some membrane-enclosed organelles in the cytoplasm. Several fea-tures characterizing flavivirus infected cells and a cytoplasmic inclusion of unknown origin were observed. Conclusion The chimeric HCV from in vitro cell culture system is proved to be intact virus particles which can efficiently infect Huh7.5 cells.
