1.The Promoting Role of bFGF on the Vascularization of Bone Matrix Gelatin Embedded Vascular Bundles in Rabbit Femoral Anteriomedialis

Objective To investigate the role of basic fibroblast growth factor(bFGF)on the vascularization of bone matrix gelatin(BMG) embedded vascular bundles in femoral anteriomedialis in the rabbits.Methods A longitudinal incision was done at the hyper-knee joint of femoral anteriomedialis in the Japan big ears white rabbit.Thirty six rabbits were divided into group A(n =12,arteria saphena and vena saphena were liberated and embeded to the groove of BMG soaked by 2 μg/ml bFGF0,group B(n =12,arteria saphena and vena saphena were liberated and embeded to the groove of BMG untreated),and group C(n =12,the BMG untreated were directly implanted).The rabbits were sacrificed at 4 weeks,8 weeks and 12 weeks after the surgery respectively by perfusing ink.After the implants were dislodged,transparent specimens were made and Masson stained for histological observations and quantitative analysis.Results After 12 weeks of operation,the neovascularization arranged in an ordered manner in group A,gradually trended to be orderly in group B,and were cluttered mainly on the edge of the implants in group C.The osteogenic and neovascularization areas of group A were the largest on each time point.Conclusion bFGF could promote the vascularization of BMG embedded vascular bundle.There was a positive correlation between osteogenesis and vascularization.
2.Screening of optimal embryonic time for in vitro separation and culture of Kunming mouse embryonic stem cells:Comparison among 2.5,3.5,and 4.5 pregnant days

BACKGROUND:Genotype of Kunming mice was similar to human population,thus an establishment of embryonic stem cell line is beneficial for research of transgenic animal.However,the best time to collect embryo has been less reported yet.OBJECTIVE:To find the best time to collect embryos from Kunming mice.METHODS:The embryos were collected from mother mice of 2.5,3.5,and 4.5 pregnant days.Microscope was used to evaluate the growth condition of embryos,embryo attaching rate(A/C),inner cell mass(ICM)growing rate(I/C),embryonic stem cells (ESCs)clone growing rate(P1/C)and ESCs subclone growing rate(P2/C).The cells were then stained with alkaline phosphatase.RESULTS AND CONCLUSION:Most of the 2.5-pregnant-day embryos were 16-cell-phase embryos.The 3.5-pregnant-day embryos were morulas while the 4.5-pregnant-day embryos were blastulas.There were no significant differences in A/C,I/C,P1/C,P2/C between 2.5 and 3.5 pregnant days(P > 0.05).The 4.5-pregnant-day indicators mentioned above were significantly greater than those two groups;therefore,4.5-pregnant-day embryos were the best source to culture,clone,isolate and passage ESCs.
3.Effects of abstragulus mongholicus bung on apoptosis of neurocytes and the expression of relevant gene in the brain of aging mice

BACKGROUND: The death of aging cells is virtually apoptosis. To a certain extent, it can be interpreted as a series of results of gere activities.Therefore, the inhibition of oncogene's expression can lengthen the life span of cells and delay aging of brain tissues.OBJECTIVE: To study the effect of abstragulus mongholicus bung (AMB) on apoptosis of nerve cells and the expression of relevant gene in aging mice brain.DESIGN: Completely randomized design and controlled experiment.SETTING: Department of Biochemistry, School of Basic Medical Sciences of Jiamusi University.MATERIALS: The experiment was conducted at the Experimental Animal Center and Biochemical Laboratory of Jiamusi University from December 2003 to May 2004. Totally 24 healthy Kunming mice were recruited in this study. There were 8 two-month-old mice (young group) and 16 twelve-month-old mice. All the 16 mice were randomized into abstragulus mongholicus bung group and old control group with 8 in each group.METHODS:① AMB group:Mice in AMB group received gastric gavage was provided by the Department of Traditional Chinese Medicine of the First Hospital Affiliated to Jiamusi University, and evaluated by Jiamusi Drug Inspection Bureau. Water decoction was prepared with 2 kg/L raw materials. Mice in old control group and young group were filled with lukewarm boiled water.② All the animals were treated as above for 30 consecutive days before put to death. Their brains were taken out immediately and the middle parts of the brains were removed to fix with neutral formaldehyde. The remaining brain tissues were made into mitochondria suspension. Content of manganese superoxide dismutase (Mn-SOD) and malondialdehyde (MDA) was determined with xanthosine oxidase method and TBA chemical colorimetry. Apoptotic cells (cells with yellow nuclei were positive ones) were assayed with in situ end-labeling (ISEL) and expression of bcl-2 gene was assayed with immunohistochemical method. The cells stained brown were positive ones. A total of 400 cells were counted under the 400× microscope. We graded the samples according to the percentage of the positive cells: the number of positive cells < 5% -; 5%-10% +; 11%-50% ++; > 51% ().③ Grade and quantitative data were compared with rank sum test and t-test.MAIN OUTCOME MEASURES: Effects of AMB on the rate of neu-ronal apoptosis, the activity of Mn-SOD, the concentration of MDA in mitochondria, and the intensity of the expression of bcl-2 gene.RESULTS: Totally 24 mice entered the final analysis.① Content of MnSOD was higher in young group and AMB group than in old control group (P<0.05).② Concentration of MDA and apoptotic rate in young group and AMB group were lower than those in old control group (P < 0.01).③Expression of bcl-2 gene was significantly different in young group and AMB group from that in old control group (P < 0.01).CONCLUSION: AMB is found to be able to obviously inhibit neuronal apoptosis in aging mice brain by affecting the activity of Mn-SOD, the concentration of MDA and the expression of bcl-2.
4.Age-related biological characteristics of rat adipose-derived stromal cells from different age stages

Objective To investigate biological characteristics of rat adipose-derived stromal cells(ADSCs) of different ages.Methods ADSCs were isolated by density gradient centrifugation from different age stages,and cultured in vitro.The adherent cells were preserved to passage,the purity of ADSCs was analyzed by immunocytochemistry method,and cell growth was observed,then proliferation capability and cell cycle were detected.Results All the ADSCs obtained from different age stages grew well and showed good morphology and growth characteristics.The proliferation rate of passage cells was higher than that of primary cells,but the proliferation activity reduced with aging,and cell cycle was prolonged.Conclusion The proliferation capability and activity of ADSCs decreased with aging.However,ADSCs of different age stages can all meet the needs of different patients for tissue engineering seed cells.
5.Effects of ?-lipoic acid on bone metabolism in lead-poisoned juvenile rats

Objective To determine the effects of ?-lipoic acid on bone metabolism in lead-poisoned rats.Methods Totally 31 SD rats were randomly divided into two groups,8 rats in blank control group and 23 in lead group that received gastric lavage with lead acetate(230 mg/L).After 40 days,3 rats from each group were taken for the measurement of lead in the blood and bone.The rats in lead group were then randomly divided into four groups: lead control group and three lead groups with different concentrations of ?-lipoic acid [30,60 and 100 mg/(kg?d)],with 5 rats in each.At the end of the experiment(80 d),the rats were killed and the samples of whole blood,serum and bone were collected to detect osteocalcin content with radioimmunoassay and alkaline phosphatase expression with immunohistochemistry staining.Results ① Blood and bone lead contents in each ?-lipoic treatment group were significantly lower than those in lead control group(P
6.Diagnostic value of CT peritoneography for non-infectious complications of peritoneal dialysis

OBJECTIVE: To assess the diagnostic value of CT peritoneography for peritoneal complications of continuous ambulatory peritoneal dialysis (CAPD). METHODS: CT peritoneography in 11 CAPD patients with clinically suspected dialysis-related complications was prospectively studied. The CAPD patients were all treated in Renji Hospital from 2005 to 2007. CT images were reviewed according to the evidence of peritoneal leaks, hernias, loculate pleural fluid collections, and adhesions. RESULTS: Abnormal findings were found in 9 of 11 CAPD cases including inguinal hernias (3 cases), umbilical hernia (1 case), hydrocele (1 case), leaks in catheter tunnel (2 cases), and peritoneal adhesions (2 cases). Parts of them were confirmed by surgical operation. CONCLUSION: CT peritoneography is useful for the evaluation of complications related to CAPD, and it offers excellent tissue contrast and multiplanar imaging for assessment of the complications.
7.Outcome and risk factors for renal involvement in patients with anti-neutrophil cytoplasmic antibody associated vasculitis

ObjectiveTo investigate the outcome and risk factors for kidney involvement by analyzing 64 patients with anti-neutrophil cytoplasmic antibody(ANCA)-associated vasculitis.MethodsData analyzed including the demographic information,survival status,renal survival status and laboratory parameters such asserum albumin level,serum creatinine level,urinary protein excretion level,hematuria,high sensitivity C-reactive protein(CRP),ANCA titer,and the Birmingham vasculitis activity score (BVAS).Logistic regression analysis,Cox regression analysis and ROC curve were used to evaluate the risk factors of patients with renal involvement and all-event survival.ResultsTotally 64 patients were enrolled [24 females with the average age of (59.9±2.0) years] and followed up for a median of (38±16) months.The morality rate was 14%,and the prevalence of end stage renal disease was 39%.Compared with those who had better outcomes,patients who died or with end stage renal disease had higher serum creatinine level [ (624±246),(245±127 ) μ mol/L,respectively,t=7.17,P=0.005 ] and erythrocyte sediment rate [ (112±24),(76±48) mm/1 h,respectively,t=3.74,P<0.01 ],but lower serum albumin level [(294±31 ),(316±42) g/L,respectively,t=-2.27,P=0.01 ] and hemoglobin level [ (79±13),(99±33) g/L,respectively,t=-3.23,P<0.01 ] at baseline.Logistic regression analysis found that serum creatinine level and erythrocyte sediment rate at baseline were associated with poor outcome and Cox regression analysis further confirmed this result[Scrβ=1.004,95%CI1.002~1.006,P<0.01; ESR β=l.018,95%CI 1.000~1.037,P=0.046].ROC curve analysis showed that serum creatinine and erythrocyte sediment rate were predictors for AAV patients' prognosis and their AUC were 0.95 and 0.80,the sensitivity of these parameters was both 94%,and the specificity was 93% and 70%respectively.ConclusionThe intensity of initial treatment should be based on disease severity and activity in order to improve the prognosis of those with ANCA-associated vasculitis with renal involvement.Increased serum creatinine and erythrocyte sediment rate may serve as predictors for poor prognosis in this patient cohort.

Objective To analyze the status of job satisfaction of human organ donation coordinators in Fujian province during the normalization period of novel coronavirus pneumonia (COVID-19) epidemic prevention and control and put forward corresponding suggestions. Methods The job satisfaction of human organ donation coordinators in Fujian province was investigated by online questionnaires. The correlation analysis of all dimensions of the overall job satisfaction of human organ donation coordinators was carried out, and the influencing factors of the overall job satisfaction were analyzed. Results The overall job satisfaction of human organ donation coordinators was (2.9±0.8), which was a relatively low score. The job promotion satisfaction was (4.7±1.3), (2.0±1.4) for the job pay and benefits satisfaction, and (2.0±1.3) for the job communication satisfaction. Objective To label Flk-1+CD31-CD34-human mesenchymal stem cells (hMSCs) with superparamagnetic iron oxide (SPIO) and to evaluate the effect of SPIO on proliferation and neural differentiation of labeled cells. Methods hMSCs were incubated with SPIO (50 mg/L) and PLL (1.5 mg/L) overnight(12~18 hours). Both labeled and unlabeled cells went through growth curve test,Trypan blue staining and flow cytometer to evaluate the effects of SPIO on cell proliferation,cell viability and surface markers. Immunofluorescence assay was conducted for neuron and neuroglia specific cell surface markers after neural induction protocols were used. Results Cell viability of the two groups were both more than 90% for 7 days. There was no significant difference in cell viability and growth curve test between two groups. The results of flow cytometer showed that both labeled and unlabeled cells expressed CD44, CD105 and Flk-1 markers, while CD31 and CD34 were negative. After neural induction, the statistical analysis of A value for all the markers showed no significant difference between the two groups.Conclusion SPIO, as MRI cellular contrast, is safe and efficient.
10.Effect of osteoporosis on the prognosis of implant dentures

BACKGROUND:Due to good function and aesthetic effect, implant dentures become prevalent in osteoporotic patients with missing teeth. However, whether osteoporosis is a contraindication of dentures as wel as effects of osteoporosis on the bone-implant integration have been not ful y understood.
OBJECTIVE:To compare the levels of alkaline phosphatase, bone mineral density and implant stability in patients with osteoporosis and non-osteoporosis, and to explore whether the osteoporotic patients can achieve good prognosis. METHODS:Forty patients undergoing implant dentures in the Department of Prosthodontics, the First Affiliated Hospital of Xinjiang Medical University since February 2015 were enrol ed, and al otted to experimental (osteoporosis) and control (non-osteoporosis) groups (n=20 per group).
RESULTS AND CONCLUSION:The levels of salivary alkaline phosphatase, bone mineral density and initial implant stability in the experiment and control groups both were lower than those before surgery. There were significant
differences in the implant stability quotient at 1 and 3 months and immediately after implantation between groups. These results indicate that osteoporotic patients undergoing implant surgery can achieve a good prognosis that is similar with non-osteoporosis ones.
