1.Effect of phosphodiesterase type 5 inhibitors on prostate cancer

Erectile dysfunction was one of the most frequently postoperative complications after radical prostatectomy,which has been deeply concentrated by patients and urologists,along with more and more people were diagnosed as localized prostate cancer.Widely being used for erectile dysfunction after radical prostatectomy,the efficacy and safety of phosphodiesterase type 5 inhibitors have been confirmed by many clinical studies.However,the influences of phosphodiesterase type 5 inhibitors on the occurrence and progression of cancer are still not completely revealed at present.Additionally,the efficiency of phosphodiesterase type 5 inhibitors on biochemical recurrence after radical prostatectomny is still controversial,though it has been demonstrated that phosphodiesterase type 5 inhibitors is not associated with the occunrence of prostate cancer based on recent clinical studies.We will review the possibly mechanisms of phosphodiesterase type 5 inhibitors in oncologic occurrence,especially in the occurrence of prostate cancer and its biochemical recurrence after radical prostatectomy.
2.Polyhydroxybutyrate Synthesis in Recombinant Zymonomas mobilis Affected Ethanol Production

Zymonomas mobilis was transformed with a polyhydroxybutyrate synthesis operon phbCAB equipped with a pdc promoter from Z. mobilis. For the first time,PHB was produced in recombinant Z.mobilis. Shake flask studies indicated that accumulation of PHB in Zymomonas mobilis increased approximately 10% ethanol productivity for the first 48h of anaerobic fermentation. After that,the PHB effect was observed as insignificant probably due to the exhaustion of the sugar.
3.Effect of Professor Deng Tietao’s Brain-strengthening Recipe 1 on Learning and Memory Abilities as well as N-methyl-D-aspartate Receptor Subunit 2B mRNA Expression in Rats with Vascular Dementia

Objective To observe the effect of Professor Deng Tietao’s Brain-strengthening Recipe 1 on learning and memory abilities of rats with vascular dementia, and to explore its molecular biology mechanism. Methods The vascular dementia rat model was established by permanent occlusion of the bilateral arteria carotis communis. Sixty male SD rats were randomly divided into 5 groups, namely sham-operation group, model group, nimodipine (5.4 mg·kg-1·d-1) group, and high-and low-dose Chinese medicine groups (57.6 and 14.4 g·kg-1·d-1, respecitvely) . After medication for 30 days, the learning and memory abilities were evaluated by Morris water maze test. The histological changes of the rat hippocampal tissues were observed after HE staining, and the mRNA expression of N-methyl-D-aspartate receptor subunit 2B ( NR2B) in rat hippocampus was measured by real-time fluorescence quantitative polymerase chain reaction (RT-PCR) . Results The water maze test results showed that the escape latency of the rats in the sham-operation group was significantly decreased on the third day, and was significantly decreased in the high-dose Chinese medicine group on the fourth day ( P<0.05 compared with the model group). The results of spatial probe test showed that the times of rats passing through the platform within 60 seconds were significantly higher in high-dose Chinese medicine group ( P<0.01compared with the model group) , but the time for rat staying at the quadrant of the platform in the other four groups did not differ from that in the model group ( P>0.05). HE staining results for the model group were presented as the absence of neural cells in hippocampus CA1 region, karyorrhexis of nerve cells, indistinctness of nuclear membrane, disappearance of nucleolus, shrinkage of nerve cells, increase of cytoplasm acidophily, and reactive hyperplasia of astrocytes. And in the hippocampal CA1 region of high- and low-dose Chinese medicine groups, the injured neural cells were found, and the lesions were slighter than those of the model group. Compared with the model group, the mRNA expression of NR2B was increased in nimodipine group and high-dose Chinese medicine group ( P<0.05) . The mRNA expression of NR2B showed a increasing trend in low-dose Chinese medicine group, but the differences were insignificant compared with that of the model group ( P>0.05). Conclusion Brain-strengthening Recipe 1 has obvious effects on improving the learning and memory abilities of vascular dementia rats, and the therapeutic mechanism is associated with reduction of the degree of neuronal damage in hippocampus and with the upregulation of the mRNA expression of NR2B.
5.Current situation of HIV/AIDS voluntary counseling and testing in Jiangxi province

Objective To analyze the current situation of voluntary counseling and testing(VCT) and improve HIV/AIDS prevention and control in Jiangxi province.Method VCT services in the province were analyzed using descriptive methodology.Result The province is in shortage of full time VCT workers.Of the individuals receiving pre-testing counseling,those receiving post-testing counseling and receiving referral counseling accounted for 5.22% and 1.39%,respectively.The individuals receiving tests mostly were aged 20-29 years,and a little less were aged 30-39 years.Higher positive rates of HIV screening tests were found in Pingxiang and Nanchang cities,accounting for 39.13% and 26.09%,respectively.Conclusion To improve VCT service and contain the spread of HIV/AIDS in this province,it is necessary to establish VCT networks,upgrade quality of counseling service and publicize VCT and its outreach activities among target population.
6.Effects of Electroacupuncture at Ouch Point on Muscle Regeneration and Basic Fibroblast Growth Factor Expression in Skeletal Muscle Damaged Rats

Objective To observe the effects of electroacupuncture at Ouch Point on proliferation cell nuclear antigen (PCNA), Desmin and basic fibroblast growth factor (bFGF) expression in gastrocnemius muscle, and explore the possible mechanisms of the effect. Methods 78 male Sprague-Dawley rats were randomly divided into model group (n=6), blank group (n=24), self-repair group (n=24) and electroacupuncture group (n=24). Each of the later 3 groups was divided into 1 d, 4 d, 7 d and 14 d subgroups, 6 rats in each subgroup. The model group, self-repair group and electroacupuncture group were modeled by gastrocnemius muscle crush injury combined with eccentric exercise-induced injury. After that, the model group given HE stain, and the other groups were given correspondent approach according to time point and immunohistochemical technology was used to test the expression of PCNA, Desmin and bFGF. Results PCNA expression was higher in the self-repair group than in the blank group at each time point (P<0.01). Desmin expression was lower in the self-repair group than in the blank group 1 d and 14 d after injury (P<0.05), but higher 7 d after injury (P>0.05). bFGF expression was higher in the self-repair group than in the blank group 7 d after injury (P<0.01). PCNA, bFGF and Desmin expression were higher in the electroacupuncture group than in the self-repair group 1 d and 4 d after injury (P<0.05). Conclusion Electroacupuncture at Ouch Point can promote the proliferation of muscle satellite cells, accelerate the myogenic differentiation and shorten the repair process, which may be related to the increased and advanced expression of bFGF.
7.Influence Factors and Absorbing Mechanics of Copper Ion by Yeast Strain Y17

Using the high efficient copper-adsorbing yeast strain Y17 as absorbing material, the major affect factors including pH, original concentration of Cu2+, cell biomass, adsorption time and temperature were examined, and then the absorbing sites of the Y17 was determined. The results showed that the solution pH was the most dominate factor which affected the biosorption of Cu2+, the other affecting factors were the ini- tial concentration of Cu2+, the cell biomass added, and adsorption time, respectively; the temperature had lit- tle effect on the rate of biosorption. The orthogonal experiment showed that the optimal absorption condition was as follow: the solution pH was 5.0, the absorption time was 40 min, the cell biomass of Y17 added was 5.0 g/L, and the concentration of Cu2+ was 8 mmol/L; the highest adsorbing rate was up to 82.7% at this condition. Based on the results of different pretreatments and the desorption of Cu2+, the cell wall of Y17 was identified as the main place occurring boisorption process, and the -NH2 group, -COOH group on the surface of the yeast cells played an important role on the boisorption process.
8.Clinical application of percutaneous vertebroplasty

Objective To evaluate the efficacy of percutaneous vertebroplasty for treating the vertebral compression fractures and vertebral metastatic neoplasms, and to assess its clinical curative effects Methods Among 81 cases (47 men and 34 women; ages 35~84, mean age 61 2 years), 110 vertebrae with metastatic neoplasms (61 cases) or vertebral osteoporosis (20 cases) were infused with methacrylate guided by DSA Effect of the operations was observed closely after the procedures Results The procedures were successful in all the involved patients and no significant complications were noted clinically Among the 61 patients treated for malignancies, 58 showed marked pain relief and 3 moderate pain relief 20 patients with vertebral osteoporosis demonstrated complete pain relief The cement leakage observed on radiographs included slight leakage to the adjacent disc (6 of 110, 5 5%), the epidural fat (8 of 110, 7 3%), the perivertebral venous plexus (2 of 110, 1 8%), and the paravertebral soft tissues (2 of 110, 1 8%) Conclusion Percutaneous vertebroplasty can significantly relieve the pain in patients with osteoporotic fractures caused by the malignancies and the vertebral osteoporosis We suggest that slight PMM leaks, when not symptomatic, should not be considered as complications
9.Quality standard of Zhike Pingchuan Capsules

AIM: To establish the quality standard for Zhike Pingchuan Capsule (Radix Panacis Quinquefolii, Semen Pruni Armeniacae, Radix Scutellariae, Radix Glycyrrhizae, Bulbus Fritillariae Cirrhosae, etc.). METHODS: Radix Panacis Quinquefolii, Radix Glycyrrhizae, Bulbus Fritillariae Cirrhosae in Zhike Pingchuan Capsule were identified by TLC and ginsenoside Rb_1 was determined by HPLC. The analysis was carried out on C_~18 column by HPLC. The mobile phase was CH_3CN-H_2O(34∶66). The flow rate was 1.2 mL?min~-1 and the detection wavelength was at 203 nm. The column temperature was at 40.0 ℃ and sensitivity was 0.02 AUFS. RESULTS:The average recovery was 97.20% and RSD was 2.25% and the linear range of ginsenoside Rb_1 was in 0.612-~6.120 ?g. CONCLUSION:This method is simple, rapid with a good reproducibility. This method can be used for the quality control of Zhike Pingchuan Capsule.
10.Construction of bioartificial renal tubule assist device In Vitro and its function of transporting sodium and glucose.

To explore a new way of constructing bioartificial renal tubule assist device (RAD) in vitro and its function of transporting sodium (Na(+)) and glucose and to evaluate the application of atomic force microscope in the RAD construction, rat renal tubular epithelial cell line NRK-52E was cultured in vitro, seeded onto the outer surfaces of hollow fibers in a bioreactor, and then cultured for two weeks to construct RAD. Bioreactor hollow fibers without NRK-52E cells were used as control. The morphologies of attached cells were observed with scanning electron microscope, and the junctions of cells and polysulfone membrane were observed with atomic force microscope. Transportation of Na(+) and glucose was measured. Oubaine and phlorizin were used to inhibit the transporting property. The results showed that NRK-52E cells and polysulfone membrane were closely linked, as observed under atomic force microscope. After exposure to oubaine and phlorizin, transporting rates of Na(+) and glucose were decreased significantly in the RAD group as compared with that in the control group (P<0.01). Furthermore, when the inhibitors were removed, transportation of Na(+) and glucose was restored. It is concluded that a new RAD was constructed successfully in vitro, and it is able to selectively transport Na(+) and glucose.
