1.Research progress of fluorometholone on dry eye

?Dry eye is a common eye disease around the world, which has highly incidence. Although there are many treatment measures for dry eye, the clinical effect is not satisfactory. There is a close relationship between inflammation and dry eye, so the researchers pay more attention on anti -inflammatory therapy, such as fluorometholone. To explore the effect of anti -inflammatory medicine on dry eye, put forward the problems and shortcomings as well, lay the foundation for further study, and provide a strong theoretical basis for the clinical use of anti-inflammatory drugs in the treatment of dry eye, the author summarizes the clinical and experimental research progress of fluorometholone on dry eye in China and abroad.
2.Discussion on construction of medical equipment in grass-roots unit

This paper aims to find out the use status of medical and sanitary equipment in primary unit. By means of investigating grass-roots health institutions and summarizing practical works, such problems in equipment were detected as standard vacancy, datedness and laggardness, partly absence, nonstandard management, untimely maintenance and low rate of utilization, etc. This paper puts forward some suggestions corresponding to the above disadvantages.
3.Literature analysis on perimenopausal syndrome in TCM by intelligent statistics

Objective:To collect and evaluate the information of literature on perimenopausal syndrome.Methods:The computer retrieval method supplemented by manual inspection of literature.Results:590 relevant documents were retrieved and among which 129 were comprehensively studied.Conclusion:The quality of the literature should be improved though there was an annual quantitive increase of the literature on perimenopausal syndrome in TCM.Great efforts should be made in this aspect.
4.Isolation and Identification of Promoter Sequence of Interferon Regulatory Factor-3 and Detection of Its Promotor Activity in Human Embryonic Kidney-293 Cells

Objective To construct a luciferase reporter plasmid containing interferon regulatory factor 3(IRF-3)human gene promoter and to evaluate promoter activity in human embryonic kidney(HEK)-293 cells.Methods The 1 000 bp fragment was amplified by PCR with human genomic DNA as a template and was directionally cloned into pGL3-basic multiple cloning sites to construct the luciferase repor-ter plasmid pGL3-pIRF-3.Transfection of HEK-293 cells with the promoter-driven lucife-rase construct was performed to induce lucife-rase gene expression and calculate the relative luciferase activity unit(RLU).Promoter sequence of 1 000 bp upstream of transcription initiation site of IRF-3 was analyzed by using Promoter 2.0 Prediction software.Results DNA sequencing and restriction endonuclease analysis verified the successful construction of the plasmid pGL3-pIRF-3.This IRF-3 promoter exhibited a strong promoter activity with an increase of 42.2-fold of RLU in HEK-293 cells when compared with pGL-3 basic vector.The transfection experiment confirmed that the levels of its activation were significantly higher than that in controls in HEK-293 cells.Function analysis of IRF-3 promoter disclosed seve-ral GATA-1 and specific protein 1(Sp1) sites and E2F in minimal promoter region.Conclusion The plasmid pGL3-pIRF-3 promoter is successfully constructed and has a strong basal promoter activity in HEK-293 cells.
5.Prediction of multifocal visual evoked potential for postoperative visual recovery in cataract patients

AIM: To determine multifocal visual evoked potential (mVEP) in the prediction of postoperative visual acuity in cataract. METHODS: We examined 30 normal eyes as control and 60 eyes of 52 cataract patients, senile cataract in 27 cases 30 eyes, cataract with glaucoma in 25 cases 30 eyes by mVEP examination. All patients underwent phacoemulsifi-cation (Phaco) and intraocular lens (IOL) implantation. After surgery,best corrected visual acuity (BCVA) was examined at 1 week, 1 month, and 3 months respectively.RESULTS: The mean amplitude and latency in mVEP responses between normal control group were 183±11nV, 95±8ms, and in senile cataract group were 177±10nV, 96± 8ms respectively, there were no significant difference between two groups (P>0.05). The mean amplitude and latency of cataract with glaucoma 138±7nV, 99±6ms were significantly different comparing to both control and senile cataract group (P<0.05). After surgery, the am-plitude and latency were 276±11nV and 93±8ms respec-tively, which did not change significantly comparing to the normal eyes (P<0.05), their visual function got no obvious damage and visual recovery were better (BCVA≥0.8). While those with central amplitude 221±6nV and latency 105±7ms that were significantly deviated from control group (P<0.05), and their visual function were seriously damaged and visual recovery were much poorer (BCVA<0.3).CONCLUSION: mVEP waveform might enable us to evaluate objective visual function detection before cataract surgery. A subject with visual function damage, their mVEP responses to stimulation were severely changed when it compared to controls.

By reviewing the status of domestic intellectual property right and patent in particular the authors pointed out that the key of increasing the number of patents and patent quality is to promote the trans formation of research achievements and the management of research achievements.Based on the framework scheme of the transformation chain of research achievements.four major stages of patent management were described,which contained ①the planning and design of patent index in the initial management of research achievement,②the forming and outputting of patents in the management of the formation of research a chievement and patent forming,③implementing and industrialization of patents in the stage of industrializing of research achievement and patent,④realizing the maximum efficiency of patents and establishing patent net in the stage of research extension and patent implementing.
7.Effect of rAAV2-TGFβ3 on collagen degradation of liver fibrosis in rats

Objective To investigate effect of transforming growth factor β3(TGFβ3)on expression of MMP-9,MMP-2,TIMP-1 and collagen I in rats with liver fibrosis through transduction by recombinant adeno-associated virus 2(rAAV2).Methods Rats were randomly divided into 4 groups:normal control group,model group,negative control group and TGFβ3 group.Liver fibrosis model was induced by hypodermic injection of 40% CCl4.rAAV2-TGFβ3,was injected via vena caudalis of the rats one week before CCl4 Was given.All rats were sacrificed and the liver tissues were taken 8 weeks afler injection of CCl4.The histopathological changes were observed on HE sections;the expressions of MMP-9,MMP-2,TIMP-1 and collagen I were examined by histochemistry and the positive area rates were semi-quantitatively analyzed.Results Compared with the model group and negative control group,the decreases of inflammatory infiltration and collagen fibers hyperplasia were observed in the TGFβ3 group,and the expression of MMP-9 increased(q=23.664,27.746,P<0.01),the expression of collagen I(q=5.503,5.251,P<0.01)and TIMP-1(q=5.800,8.608,P<0.01)decreased,but that of MMP-2 was not changed(q=2.1 08,0.996,P>0.05).Conclusion rAAV2-TGFβ3 can reduce the histological damage and degree of liver fibrosis in rats by inhibiting expression of TIMP-1 and collagen I,and upregulating expression of MMP-9.
8.A comparison of preliminary bond strength between oxide ceramics and three resin luting cements

Objective:To compare the shear bond strength between oxide ceramics and three resin cements. Methods: Blocks of zirconium dioxide and aluminium sesquioxide ceramics were treated with sandblasting and bonded with three luting cements respectively. After preserved in 37 ℃ water for 24 h, the shear bond strengths of these specimens were tested and the data were analyzed by SAS9.12 software package. Results: There were significant differences among alumina ceramics which manifested that shear bond strength of Superbond C&B was the highest (37.60±2.06) MPa. However, in zirconium dioxide groups, RelyX~(TM) Unicem showed the lowest bond strength(13.29±3.05) MPa,but there was no difference between Superbond C&B and Panavia~(TM) F. Conclusion: Both Superbond C&B and Panavia~(TM) F have reliable shear bond strength with oxide ceramics, while RelyX~(TM) Unicem is not so perfect.
9.Astilbin inhibits proliferation of rat aortic smooth muscle cells induced by angiotensin II and down-regulates expression of protooncogene.

This study examined the effect of astilbin on the proliferation of rat aortic smooth muscle cells (RASMCs) induced by angiotensin II (AngII) and explored the possible mechanisms. Cell proliferation model of RASMCs was induced by treatmente with AngII. Cells were randomly divided to 8 groups. Normally cultured VSMCs serves as blank control group; in AngII model group, cells were treated with AngII at 10(-7) mol/L; in three astilbin groups, cells were treated with 10, 15, 30 mg/L of astilbin; in three AngII+astilbin groups, cells were treated with AngII (at 10(-7) mol/L) and astilbin at 10, 15, 30 mg/L. Cell proliferation ability was detected by MTT method and the cell cycles and proliferation index were flow cytometrically determined. The expression of c-myc mRNA was assessed by using reverse transcription polymerase chain reaction (RT-PCR), and the expression of NF-κB in RASMCs was immunocytochemically observed. Our results showed that MTT metabolism in RASMCs in the basic and AngII stimulated situation was inhibited by astilbin, and the cells numbers of G(0)/G(1) phase were increased and that of G(2)/S phase were decreased markedly. Not only highly expression of c-myc gene stimulated by AngII could be inhibited by Astilbin significantly, but also the expression of NF-κB protein can be down regulated by Astilbin. We are led to conclude that astilbin astilbin can inhibit the AngII-mediated proliferation of RASMCs by blocking the transition of RASMCs from G(0)/G(1) phase to S phase and by down-regulating the expression of NF-κB, c-myc gene.
10.Evaluation of clinical efficacy of BAY 38-4766 for cytomegalovirus rentinitis

Background At present,researches about retinitis and acquired immunodeficiency syndrome (AIDS) mainly focus on observation and analysis of the cytomegalovirus (CMV)-induced ocular complication.To screen the effective drugs is very important for the treatment of AIDS-related CMV-induced retinitis.Objective This study was to describe the clinical features,diagnosis and treatment outcome of CMV-induced retinitis with BAY 38-4766 and evaluate the relationship between CMV retinitis and AIDS.Methods This was a case observational study.A total of 154 eyes of 84 patients with CMV retinitis and AIDS were enrolled in this study.Before the definitive diagnosis of CMV retinitis,the AIDS course of these patients were 4-26 months.In the initial examination,the visual acuity ranged from finger counting to 0.4,and the number of CD4+ T-lymphocyte was 0-30 cells/μl.The survival time in the patients ranged from 3 weeks to 18 months.BAY 38-4766 was used in 117 eyes of 62 patients,and 102 eyes of 53 patients showed the srinked of retinal lesion and improvement of vision (0.1-0.7).BAY 38-4766 was used to treat the CMV retinitis in 117 eyes of 62 patients with the initial intravenous dose of 5 mg/kg,twice per day for consecutive 2 weeks and followed by oral dose 1 gram per day.The follow-up duration was 2 weeks to 18 months.The fundus feature,visual acuity and CD4+ T-lymphocyte counts were analyzed.This study proposal was approved by Ethic Committee of the 474th Hospital of PLA,and written informed consent was obtained from each patient prior to entering this study.Results The numbers of CD4+T-lymphocytes increased to 12-402 eells/μl after administration of BAY 38-4766.The CMV retinitis aggravated and the vision decreased in the untreated 22 patients with the CD4+T-lymphocytes 0-30 cells/μl.Conclusions CMV retinitis is the most common intraocular complication in patients with AIDS.Diagnosis of CMV retinitis is based on the characteristics of necrotizing retinitis,which is typically associated with retinal hemorrhage and vasculitis.BAY 38-4766 is an effective drug for the treatment of CMV retinitis.
