1.Validity and reliability of the Chinese version of Family Management Measure

Objective To assess the psychometric properties of the Chinese version of Family Man-agement Measure (FaMM). Methods FaMM was translated into Chinese and distributed to 330 caregivers whose children had chronic diseases to fill the questionnaire. Results Cronhach's alpha coefficient for the subscales ranged from 0.52 to 0.86, indicating acceptable internal consistency. The content validity index was 0.84. Construct validity was supported by significant correlation between FaMM and FAD and CBCL,Pearson correlation coefficient ranged from 0.11 to 0.38. The relationship among them accorded with the expected trends of Family Management Style Framework. The result of factor analysis indicated 9 factors explaining 60.08% variance. Conclusions The psychometric property of the Chinese version of FaMM was acceptable and it could be regarded as a reliable and valid instrument to measure the family response of Chinese families whose children had chronic diseases.
2.Protoplast fusion in Dendrobium candidum and Gynostemma pentaphyllum

Object To explore the protoplast fusion of Dendrobium candidum Wall ex. Lindl. and Gynostemma pentaphyllum. (Thunb) Makino. Methods The two kinds of protoplasts were fused by PEG method, and then cultured in the modified liquid medium containing 2 mg/L BA and 1 mg/L NAA. Results High yield, viability, and pure mesophyll protoplasts were isolated from D. candidum and G. pentaphyllum. Conclusion The first cell division occurres within three days after fusion. And some of the cells are divided three times.
3.Expression and significance of human defensin-alpha1-3 in breast cancer

Objective To investigate the expression of human defensin-alpha1-3 (HNP1-3) in breast cancer and in normal tissues and determine whether there exists a relationship between HNP1-3 expression and clinicopathologic features.Methods Immunohistochemical staining was used to detect the expression levels of HNP1-3 in 59 breast cancer specimens,and 20 normal tissues (fibroadenoma of breast or tumor-adjacent tissues).The relationship of the expression levels of HNP1-3 with patient’s age,clinical stage,histological grade,lymph node metastases and expression of PR/ER/C-erB-2 was analyzed.Results High level expression of HNP1-3 (95%) was expressed in those normal breast tissues.Different expression of HNP1-3 was found in breast cancer specimens and closely correlated with tumor histological stage and lymph node metastases (P

5.Pharmacologic action of the Magnolia biondii Pamp volatile oil nanometer bangosome

Objective To investigate the pharmacologic action of the Magnolia biondii Pamp volatile oil nanometer bangosome. MethodsThe pharmacologic action of the Magnolia biondii Pamp volatile oil nanometer bangosome was evaluated by observing its effect on Jimpy mice with swelling ear,capillary permeability and rats with allergic symptoms. Results The Magnolia biondii Pamp volatile oil nanometer bangosome could significantly relieve the degree of swelling in Jimpy mice extended by p-xylene,inhibit the increased capillary permeability in Jimpy mice extended by HAC,and combat the symptoms of rhinocnesmus,sneezing,nasal discharge with a better effect than the Magnolia biondii Pamp volatile oil. Conclusion The Magnolia biondii Pamp Volatile oil nanometer bangosome has a good anti-inflammation and anti-hypersensitiveness effect,which upgrades the effect of the Magnolia biondii Pamp volatile oil.
6.Drag resistance diversity of 1731 Pseudomonsd aeruginosa at Xinxiang City

Objective To explore the drug resistance diversity of pseufomonsd aeruginosa.Methods We did sensitivity experiments by 23 antibiotic of 1731 pseudomonad aeruginosa identified from clinic during 1997 to 2008,Results In 1 731 pseudomonad aeruginosa,the samples were main from phlegm,accounted for 58.5%.the second from secretion accounted for 24.7%.The drug resistance of pseudomonad aeruginosa for ampicillin、cefazolin and cefuroxime sodium were all higher than 90%,the second were trimethoprim/sulfamethoxazole 83.2%.The antibacterial medicines of drug resistance higher than 70%were netilmicn(75.2%)、ofloxacin(70.4%)、tetracycline(74.1%)、choramphenicol(72.0%).The antibacterial medicines of drug resistance Iower than 30%were Cefoperazone/Sulbactam、cefepime、imipenem、ceftazidme.In 12 years,the drug resistance diversity had enhanced prevalent trend.The enhanced speed of proportion drug was slow in 2004~2007,but it was fast again in 08 year.Conclusion The pseudomonad aeruginosa have become one of the most clinical routine nosogenetic germs.It showed single and anti muhiresistance of antibioties seriously.In order to improve antibacterial treating effect,we should inspect the drug resistance continuously in our area.We could select rationally antibacterial medicine according to the result of medicine sensitivity experiments.
7.Expression of glycosylphosphatidylinositol-anchored protein in lymphocytes and the subsets of healthy individuals

Objective To study the expression of glycosylphosphatidylinositol(GPI) anchored protein in lymphocyte subsets of healthy individuals. Methods Flow cytometry and two/three color McAbs were used to detect CD59 expression in granulocytes, lymphocytes and subsets. Results In 50 samples, granulocytes mainly showed a single population of cells strongly positive for CD59, the percentage of CD59 expression was (98.6?1 5)%. Lymphocytes had a small population of weakly positive and negative for CD59, positive cells being (90 0? 14.9)%. CD3 +T cells had a similar profile as that of total lymphocytes, but there were more weakly positive /negative cells and less strong positive cells. As compared with CD3 +T cells, CD4 +T cells had a larger population of strongly positive for CD59, positive cells being (92.9?8.1)%; while CD8 +T cells had a smaller population of strongly positive, positive cells being (74.6?9.1)%. CD59 expression of activated T cells was between that of CD3 +and CD8 +T cells. CD45RA +and CD45RO +T cells both had a similar profile like that of CD3 +T cells. CD20 +B cells showed a similar profile to that of granulocytes, positive cells being (96.3?0 6)%. Conclusions Population of low or negative expression of CD59 among the lymphocytes of healthy controls was related to T cells, especially CD8 +T cells. B cells had uniform positive expression of CD59, and could be a candidate for detecting PNH.
8.Preparation of long oligonucleotide microarray for detection and sub-typing of human papillomavirus

Objective To design a microarray of ~60mer oligonucleotide for detection and sub-typing of human papillomavirus (HPV). Methods The type-specific oligonucleotide probes of 4 different types of HPV (6, 11, 16, 18) were designed by using biological software Arraydesigner 2.0, which analyzed the whole genome sequences of HPV and selected optimal probes with high specificity, identical length and similar melting temperature (Tm). These probes were synthesized and printed onto the surface of glass slides in order to prepare a low-density microarray. HPV samples were labeled with fluorescence dyes Cy3 using a method of restriction display PCR (RD-PCR). HPV plasmid DNA was restricted with Sau3AⅠ to produce multiple fragments which were ligated to adaptors subsequently and used as PCR template. PCR labeling was performed with the fluorescently labeled universal primer (Cy3-UP) whose sequence is designed according to the adaptor of the RD-PCR approaches. The labeled samples were then hybridized with the oligonucleotide microarray. Results Both single and multiple HPV DNA samples could be detected with oligonucleotides microarray, and the corresponding HPV subtypes were recognized as well. And no signals were detected in all the negative and blank control spots. Conclusion 60mer oligonucleotide microarray designed by appropriate bioinformatics software can be applied to HPV detection and genotyping on gene level.
9.Activity difference of femoral artery preserved by vitrification and freezing in rabbits

0.05).The maximum relaxant forces of vitrification and fresh arteries groups were greater than the freezing group(P 0.05).The arteries of three groups began obvious relaxant responses when sodium nitroprusside was 10-7 mol/L, and nearly reached their maximal relaxant responses when sodium nitroprusside was 10-4 mol/L.CONCLUSION:The vitrification could preserve more active smooth muscle cells of arteries than the freezing method.The norepinephrine has better effect on contraction ability of artery, which preserved in the vitrification group than the freezing group;however, there is no difference in relaxation of sodium nitroferricyanide.
10.Chondrogenic differentiation and osteogenesis gene expression following a short-time induction of adipose-derived stem cells with bone morphogenetic proteins

OBJECTIVE:To investigate osteogenic and chondrogenic differentiation of adipose-derived stem cells following a short time induction with bone morphogenetic protein 2(BMP-2) or BMP-7.METHODS:The subcutaneous adipose tissue was obtained from adult New Zealand rabbits under aseptic condition,and cultured in vitro with collagenase digestion.All cells were divided into 3 groups:in the BMP-2 group,cells were cultured with medium containing 0.1 g/L vitamin C,10 mmol/L?-sodium glycerophosphate and 10?g/L BMP-2 for 10 minutes,followed by 4-14 days inoculation with density of 18?104 cells per pore.In the BMP-7 group,cells were cultured with BMP-7 with the same methods as BMP-2 group.The cells were cultured with simple culture medium in the control group.RESULTS:Compared to the control group,the number of adipose-derived stem cells,protein level and ALalkaline phosphatase(ALP) activity of BMP-2 and BMP-7 groups was up-regulated 1.78-1.79,1.15-1.95,and 32-40 times,respectively.At days 4 after a 15 minutes induction,the runx-2 gene expression,osteopontin gene,and biglycan gene were increased 1.9,2.2 and 1.3-1.7 folds than that of the control group.Meantime,only the biglycan gene expression was increased 1.3-1.7 folds in the BMP-7 group,the runx-2 gene expression,osteopontin gene was not changed.At day 14 after a 15 minutes induction,there was no alter of runx-2,osteopontin,biglycan,as well as aggrecan gene expression in the BMP-2 group;while down-regulated runx-2,osteopontin,biglycan 1.8,5.0 and 1.7 folds,with increased aggrecan gene in the BMP-7 group.CONCLUSION:Following a short time induction,BMP-2 can stimulate runx-2 and osteogenic expression at 4 days after re-culture,whereas BMP-7 down-regulate genes expression at days 14,yet down-regulate aggrecan mRNA expression.
