1.Effects of Jinmaitong serumon the expression of β-catenin, GSK-3β and myelin protein zeroin Schwann cells of rats cultured in high glucose medium

Objective To investigate the effect of serum containing different concentrations of traditional Chinese medicine Jinmaitong on β-catenin, GSK-3β and P0 in Schwann cells cultured in high glucose medium.MethodsTwenty-five male Sprague-Dawley rats were randomly treated with 5, 10, 15 and 20 times of Jinmaitong and distilled water.Schwann cells were divided into six groups, which are control group, high glucose group, 5 times group, 10 times group, 15 times group, 20 times group.72 hours later, the proliferative activity of SCs cells were detected by CCK, the mRNA and protein expression of β-catenin, P0 and GSK-3β was detectived by rt-PCR and Western blot.Results High glucose medium could inhibit the proliferation of Schwann cells, down-regulate the expression of β-catenin and P0(P<0.01), and up-regulate the expression of GSK-3β(P<0.05) mRNA significantly.But Jinmaitong can invert the results (P<0.01, P<0.05).Conclusions High glucose medium will injure the proliferation of Schwann cells, but Jinmaitongcan increase the proliferation activity of Schwann cells, and promotes the secretion of P0 partially dependent on up-regulating the activity of β-catenin and down-regulating the activity of GSK-3β.
2.Contrast ultrasound guided 915 MHz microwave percutaneous coagulation therapy of the hepatic artery and portal vein injury in vivo experiment

Objective To evaluate the hemostasis effect of 915 MHz microwave in the hepatic artery and portal vein injury in vivo dog liver.Methods After manufactured model of the hepatic artery and portal vein injury,915MHz microwave percutaneous coagulation therapy and drug injections were performed under the guidance of contrast-enhanced ultrasound (CEUS),the hemostatic effect,amount of bleeding and histologic findings were compared between the two groups.Results 915 MHz microwave coagulation had better hemostatic effect than drug injection,the amount of bleeding was significantly less than the injection method,and the pathological results showed that the vessel wall necrosis and surrounding tissue coagulation were more obvious than the injection.Conclusions CEUS guided 915 MHz microwave has a significant role in hemostasis,so the 915 MHz microwave percutaneous coagulation therapy of the hepatic artery and portal vein injury in vivo dog liver has important clinical auxiliary significance.
3.Contrast-enhanced ultrasound guided 915 MHz percutaneous microwave coagulation therapy of the splenic artery injury:an in vivo experiment

Objective To evaluate the effect of 915 MHz microwave percutaneous coagulation of splenic artery injury in vivo experimental.Methods The manufacture model of the splenic artery injury were made and divided into two groups:group A (peripheral splenic artery injury) and group B (central type of splenic artery injury),915 MHz,2450 MHz microwave coagulation therapy and drug injections were performed under the guidance of ultrasound contrast.The hemostatic effect,operating time,total therapy time,amount of bleeding,amount of fluid infusion and histologic findings were compared.Results The treatment time of 915 MHz microwave[group A(4.65 ± 5.45),group B (9.38 ± 5.62) s] was less than those of 2450 MHz microwave[group A(86.36 ± 6.34) s,group B(127.72 ± 5.34) s] and drug injection [group A(128 ± 4.85)s,group B(182 ± 7.28)s],on the other hand,the amount of bleeding and fluid volume was less than that of rest methods.Conclusions Contrast-enhanced ultrasound guided 915 MHz microwave has significant hemostatic effect compared with the other two methods,915 MHz microwave percutaneous coagulation therapy has important significance to splenic artery injury in clinical.
4.Mechanism of protective effects of low dosage of ultrashortwave diathermy on cerebral ischemia-reperfusion injury

Objective To observe the effect of low dosage of uhrashortwave(USW) on infarction volume, B cell lymphocytoma-xl (Bcl-xl) and tumor necrosis factor alpha (TNF-?) after cerebral ischemia-reperfusion in rats and discuss its acting mechanisms. Methods Focal ischemia-reperfusion model was established in 25 rats by re- versible right middle cerebral artery occlusion with filament. The right side cerebral ischemia was lasted for 2 hours and then followed with 24 hours of reperfusion. The content of neurological deficits were evaluated by the Zea-Longa 5-degree scoring system to select rats. After surgery, the rats were divided into 3 groups: blank control group, control group and USW treatment group. The brain of all rats was taken at 24 hours after reperfusion. The cerebral infarction volume, the expression of Bcl-xl and TNF-?were measured and analyzed. Results Twenty-five rats were used in the analysis of results. 6.Analysis of the necessity of extraperitonealization in ileal conduit surgery after radical cystectomy

Objective To discuss the necessity of closing the peritoneum during the operation of ileal conduit after the radical cystectomy.Methods We retrospectively analyzed the clinical data of 395 patients with bladder cancer who received radical cystectomy from Jan.2014 to Sep.2016.The amount of male was 327,female was 68.The mean age was (65.8 ± 9.7) years old.Patients were divided into four groups according to the surgical method of cystectomy and urinary diversion.In group A,patients,including 78 males and 9 females,were received open radical cysectomy (ORC) with extraperitoneal ileal conduit.The mean age was (67.8 ± 9.2) years old.In the preoperative clinical staging,66 cases were less than T2 and 21 cases were more than T2.Preoperative pathological grade in 83 cases and low grade in 4 cases.In group B,patients,including 31 males and 2 females,were accepted ORC with ileal conduit without peritoneum closure.The mean age was (67.3 ± 8.7) years old.Preoperative clinical staging showed less than T2 in 25 cases,more than T2 in 8 cases,The preoperative pathological grade showed high grade in 33 cases.In group C,patients,including 112 males and 27 females,were accpeted LRC with ileal conduit without peritoneum closure.The mean age was (64.3 ± 10.5)years old.The preoperative clinical staging showed less than T2 in 107 cases and more than T2 in 32 cases.The preoperative pathological grade showed high grade in 135 cases and low grade in 4 cases.In group D,patients,including 106 males and 30 females,were accepted RARC with ileal conduit without peritoneum closure.The mean age was (65.9 ±10.0)years old.Preoperative clinical staging showed less than T2 in 103 cases and more than T2 in 33 cases.The preoperative pathological grade showed high grade in 132 cases and low grade in 4 cases.Ileal conduit without peritoneum closure means completely open the peritoneum after anastomosis of the ureter and intestine in the urinary diversion surgery without shutting down the peritoneum,which is different from the extraperitoneal ileal conduit.The operating time,blood loss,blood transfusion rate,recovery time of intestinal function and perioperative complications and rate of hydronephrosis were analyzed.Results The 395 cases completed operation successfully,no LRC or RARC had been converted to ORC.The operative time was (280.1 ± 92.3) min,(233.6 ± 99.4) min,(304.8 ± 108.9) min,(364.6 ± 86.4) min in four groups,respectively (P < 0.05).The blood loss in four groups were (489.1 ± 285.6) ml,(431.8 ± 233.1) ml,(373.0 ±213.7) ml,(205.6 ± 137.8) ml,respectively (P <0.05).The transfusion rate in four groups were 18 (20.7%),16 (48.0%),15 (10.8%),14 (10.3%),respectively (P < 0.05).The mean time to flatus in four groups were (3.7 ±1.8)d,(3.6±1.0)d,(3.5±1.2)d,(2.2±1.7)d,respectively (P < 0.05).While ileal obstruction rate had no statistical difference in four groups [group A 17 cases(19.5%),group B 6 cases(18.2%),group C 27 cases(19.4%),group D 19 cases(14.0%),P =0.678].Urine leakage,intestinal leakage,lymphocyst were only occurred in group A [7 cases (8.0%),2 cases (2.3%),2 cases (2.3%)].Pyelonephritis was noticed in each group,including 14 cases(16.1%)in group A,2 cases(6.1%)in group B,9 cases (6.5%)in group C,6 cases(4.4%)in group D (P < 0.05).Hydronephrosis 6 months after surgery was observed in four groups,including 15 cases(17.2%)in group A,3 cases(9.1%)in group B,7 cases(5.0%)in group C,5 cases(3.7%)in group D (P < 0.05).Conclusions Ileal conduit without peritoneum closure would not increase the incidence of complications,on the contrary,it would relieve the tension of anastomosis,and reduce the occurrence of complications such as urine leakage.

PCR method was used to introduce the code sequence of Factor Xa cleavage site to the 5′ end of cecropin CMIV mutant gene X, then the gene was cloned into the expression vector pGEX-KG, and was highly expressed in E. coli BL21 by IPTG induction. The fusion protein was purified by affinity-chromatography and was cleaved by Factor Xa. Cecropin X with antibacterial activity was obtained after purified by ion-exchange chromatography.
9.Case control study of hook needle knife for the treatment of stenosing tenovaginitis of flexor digitorum.

OBJECTIVETo investigate the effect and safety of the hook needle knife for the treatment of stenosing tenovaginitis of flexor digitorum.
METHODSFrom September 2007 to September 2008, 60 outpatients with stenosing tenovaginitis of flexor digitorum were randomized divided into the treatment group and the control group, 30 cases in each group. Among the patients, 44 patients were female and 16 patients were male, aged from 34 to 69 years, averaged 56 years, the duration of disease ranged from 1 month to 1 year, averaged 3 months. All the patients were treated with hook needle knife and local-blocking respectively. The patients were followed up for 6 months, and the relief of moving-pain, tender-pain, stretching-pain and resist-ing--pain were observed respectively. All the patients were evaluated by the symptoms with numerical rating scale.
RESULTSThe relief of moving-pain, tender-pain, stretching-pain and resisting-pain in the treatment group were significantly better than those of the control group; and the therapeutic effects of treatment group were better than those of the control group.
CONCLUSIONThe method for treating stenosing tenovaginitis of flexor digitorum with hook needle knife has advantages of definite effects, micro-invasion and safety.

10.A cyclotide against influenza A H1N1 virus from Viola yedoensis.

Three cyclotides were isolated from the whole plant of Viola yedoensis in this study. The two, vary peptide E and cycloviolacin Y5, were previously reported, and a novel cycloviolacin VY1 was characterized according to the interpretation of MS/MS fragmentation of peptides which were produced from the reduced and alkylated parent peptide with the digestion of Endo Lys-C, trypsin and chymotrypsin, separately. The stability of remarkable resistance to proteolytic degradation by trypsin and chymotrypsin, and that of thermal denaturation was confirmed again. Besides, the IC50 value of cycloviolacin VY1 against influenza A H1N1 virus was (2.27 +/- 0.20) microg x mL(-1). It is the first cyclotide reported with anti-influenza A H1N1 virus activity in vitro assay.
