1.Effect and mechanism of quercetin from total flavones of Bidens Bipinnata L on the proliferation of hepatic stellate cells induced by TGF-β1

Objective To observe the effect and mechanism of quercetin on the proliferation of rat hepatic stellate cell(HSC)induced by TGF-β1.Methods HSC was isolated by orthotopic hepatic perfusion with collagenase Ⅳ.MTT assay was used to detect the inhibitory effect of quercetin on the proliferation of HSC.The expression of typeⅠcollagen in HSC was detected by ELISA.The effect of quercetin on the expression of typeⅠcollagen,Smad2 and Smad7 mRNA in HSC was detected by real-time quantitative PCR.The expression of type Ⅰ collagen,Smad2 and Smad7 protein in HSC was detected by Western blot.Results Quercetin could inhibit the proliferation of HSC,down-regulated the expression of type Ⅰ collagen,Smad2 mRNA and protein,and Smad7 mRNA and protein expression were significantly up-regulated(all P<0.05).Conclusion The anti-hepatic fibrosis effect of quercetin may be related to its blocking TGF-β1/Smad pathway of HSC and blocking the activation and proliferation of HSC.

Objective To examine the circulating glucose and body weight in the transgenic MKR mouse model who expressed dominant-negative IGF-1 receptor and insulin receptor in skeletal muscle leading to systemic insulin resistance and diabetes. Methods MKR mice were genotyped by PCR analysis of tail DNA.And in these mice we examined the circulating glucose and body weight once a week from 1 to 16 weeks of age, and the circulating insulin and glucose tolerance at age of two-month-old by using C57 mice as controls. 3.Effect of Tanshinone IIA in Preventing and Treating Oxaliplatin Induced Peripheral Neuropathy.

OBJECTIVETo observe preventive and therapeutic effects of Tanshinone IIA (T II A) on oxaliplatin induced peripheral neuropathy (OlPN) and to explore its effects on the expression of calcitonin gene related peptide (CGRP) and never growth factor (NGF).
METHODSTotally 36 phase II - III patients with malignant tumor of digestive tract undergoing chemotherapy program with oxaliplatin, were equally assigned to the T II A group (using THA at 80 mg/day 1 day before oxaliplatin chemotherapy for 3 successive days) and the control group (using chemotherapy program with oxaliplatin alone) by segmented randomization. After 4 cycles of chemotherapy, the incidence degree and incidence of OlPN were evaluated. Sensory nerve conduction velocity (SNCV) and motor nerve conduction velocity ( MNCV) were tested by EMG evoked potential device. Serum levels of CGRP and NGF were also detected in the two groups before and after chemotherapy. The correlation of serum levels of CGRP and NGF to OIPN was assessed using linear correlation analysis.
RESULTSAfter chemotherapy the OlPN incidence was 27.8% (5/18 cases) in the T II A group, obviously lower than that in the control group (55.6%, 10/18 cases; P < 0.05). Compared with before treatment in the same group, SNCV and MNCV of common peroneal nerve were slowed down, serum NGF levels decreased, and serum CGRP levels obviously increased in the two groups (all P < 0.05). Compared with the control group after treatment, SNCV and MNCV of common peroneal nerve were obviously accelerated, serum NGF levels increased, and serum CGRP levels obviously decreased in the THA group (all P < 0.05). Results of linear correlation analysis indicated serum NGF level was negatively correlated with peripheral neuropathy (PN), serum CGRP expression was positively correlated with neurotoxicity (P < 0.05).
CONCLUSIONT II A could reduce the incidence of OlPN, which might be associated with inhibiting the expression of CGRP and up-regulating NGF activities.

4.Motivation alteration influencing factors of internet addiction disorder undergraduates

Objective To explore motivation alteration influencing factors of internet addiction disorder (IAD) undergraduates.Methods The study sampled randomly 793 undergraduates from China University.According to CIAS-R and Young's internet addiction diagnostic criteria,a total of 37 internet addicts were selected.Internet addiction survey was used to study the motivation alteration influencing factors of IAD undergraduates.Results (1) The negative effect of internet using on academy,personal relationship etc.(damage from internet using) had a significant positive correlation with motivation alteration (r =0.672,0.699,0.658,P < 0.01 ; r =0.653,0.673,0.628,P < 0.01 ; r =0.604,0.681,0.703,P < 0.01).(2) Benefit from psychological satisfactory level of internet using had no significant difference with motivation alteration.(3) Self-efficiency of behavioral control on using internet had a significant negative correlation with motivation alteration (r =-0.397,-0.370,P <0.05).Conclusion The motivation alteration of IAD undergraduates is influenced by the factors of damage from internet using,sense of achievement,self-efficiency of behavioral control on using internet and so on.

6.Effect on blockade of MCP 1 in early course of experimental acute necrotizing pancreatitis

Objective To investigate the potential role of MCP-1/CCL2 in experimental acute necrotizing pancreatitis (ANP) and complications. Methods 60 SD male rats were randomly divided into 3 groups: sham operation group ( n = 20 ), ANP group ( n = 20 ) and MCP-1 group ( n = 20 ). ANP model was induced by retrograde infusion of 3.5% sodium taurocholate, MCP-1 group received subcutaneous injection of MCP-1 antibody 0 h and 6 h after ANP induction. The serum levels of amylase, MCP-1, D-lactic acid,histological changes and the expression of MCP-1 mRNA of lung, small intestine and pancreas, the expression of MCP-1 protein in pancreas, MPO levels of small intestine MPO were determined. Results The serum levels of amylase, MCP-1, D-lactic acid in MCP-1 group at 12 h were (4666 ±412)U/L, (39.53 ±8.25)pg/ml and (6.3 ±2.2)mg/L, which were significantly lower than those in ANP group [ (9611 ±363)U/L, (63.42 ±9.32) pg/ml, (9.3 ± 2. 1 ) mg/L, P< 0.05 ) ]; the expression of MCP-1 mRNA in pancreas, small intestine and lung were 0.431 ± 0.009, 0. 211 ± 0.018 and 0.442 ± 0.017, which were significantly lower than those in ANP group [ (0.624 ±0. 010, 0. 523 ±0. 019 and 0. 569 ±0. 024, P <0.05) ]; the expression of MCP-1 protein in pancreas was 2.0 ± 0. 1, which was significantly lower than that in ANP group (4. 0 ± 0. 2, P <0.05). Lung and small intestine MPO were (11.1 ±3.0)U/g and ( 19.2 ±2.0)U/g, which were significantly lower than those in ANP group[(39.2±3.1)U/g and(13.1±2.1)U/g, P<0.05]. Conclusions Early blockade of MCP-1 not only attenuates the severity of ANP, but also decreases the degree of acute lung injury and intestine barrier dysfunction.
7.Application of leukoreduction filter in removal of leukocytes in platelet concentrate

Objective To investigate the application of leukoreduction filter in removal of leukocytes in platelet concentrate . Methods Platelet concentrate was prepared by using platelet‐rich plasma method .35 bags of the same type of prepared platelet concentrate were filtered by using leukoreduction filter .The changes of conventional indicators of platelet before and after filter were measured and recorded .The activating platelets indicators PAC‐1 and CD62p were detected by using flow cytometry .The platelet hypotonic shock was measured by biochemical analyzer .The platelet aggregation was measured by using platelet aggrega‐tion instrument .Results After platelet concentrate was filtered by using leukoreduction filter ,leukocyte removal rate was(98 .28 ± 0 .97)% ,platelet recovery rate was(86 .37 ± 2 .84)% .After filtration ,white blood cell count ,platelets ,red blood cells were reduced than before filtration(P<0 .05) .The differences of capacity of platelets ,mean platelet volume and pH before and after filtration were not statistically significant(P> 0 .05) .Before and after filtration ,the expression of platelet activation markers PAC‐1 and CD62p ,platelet aggregation and platelet hypotonic shock were not statistically different(P>0 .05) .Conclusion Platelet leukore‐duction filter could effectively filter leukocytes in platelet concentrate .It would not change the conventional indicators ,and not affect platelet activation ,aggregation and anti‐hypotonic shock capacity significantly .
8.Effect of Sinomenine on Immune Function of Adjuvant Arthritis Rats

Objective To observe the effect of sinomenine(SINO) on immune function of adjuvant arthritis(AA) rats.Methods SD rats were randomized into normal group,model group,SINO groups(treated with gastric gavage of SINO at the doses of 60,120 and 240 mg/kg respectively),and glucosides of Tripterygium Wilfordii(30 mg/kg) group.Except the normal group,the rats in other groups received subcutaneous injection of the complete Freund's adjuvant 0.1mL into the left hind foot to induce AA.The medication began from 12 days after the modeling and lasted 12 days.The pedal swelling and joint function scores were observed in different time.Radioimmunoassay was used to detect the contents of interleukin-1?(IL-1?) and tumor necrosis factor ?(TNF-?) in synovial cells.Expression of IL-1? and TNF-? mRNA was examined by reverse transcription-polymerase chain reaction(RT-PCR) assay.Results SINO at different concentrations decreased the pedal swelling and arthritis scores to various degrees,inhibited the production of IL-1? and TNF-? in the synovial cells,reduced the expression of IL-1? and TNF-? mRNA,and recovered the normal histological features of synovial cells in AA rats.Conclusion The therapeutic mechanism of SINO for rheumatoid arthritis may be related with the inhibition of secretion of inflammatory mediators in synovial cells,and with the recovery of histological features of synovial cells.
9.Clinical research of lower extremity biomechanical rotation and patellofemoral pain in women

Objective The purpose of this investigation was to discuss the relationship between the rotation of lower extremity biomechanics and patellofemoral pain(PFP) .Methods With normal volunteers as controls ,patients with PFP were scanned by ul‐trathin CT ,keeping the bilateral lower extremity straight neutral .The anatomy parameters were measured and analysed ,such as femoral neck anteversion(FNA) ,femoral neck shaft angle(NSA) ,posterior condylar angle(PCA) ,femoral rotation relative to tibia (FRRT) ,tibial torsion angle(TTA) and patella tilt angle(PTA) .Results Compared with control group ,the PFP group FNA ,FR‐RT ,PCA ,TTA and PTA increase statistically significant (P<0 .05) ,while the NSA is no statistical difference (P>0 .05) .It could be found that PTA had the most obvious influence on the VAS score ,followed by FNA and TTA ;and PTA had the most obvious influence on the WOMAC function score ,followed by TTA ;and significant negative correlation could be found between PTA and age (r= -0 .548 ,P<0 .05) .Conclusion PTA is a sensitive indicator to assess VAS score and WOMAC function score for individ‐uals with PFPS .Increases of PTA ,FNA ,PCA ,FRRT and TTA are associated with PFP for women .
10.Clinical study on tuina for acute cervical radiculopathy

Objective:To observe the clinical efficacy of Ba-pulling and Qian-traction manipulation with neck suspension and movement for acute cervical radiculopathy. Methods: A total of 85 patients who met the inclusion criteria were randomized into an observation group and a control group by random numbers, with 43 cases in the observation group and 42 cases in the control group. The observation group was treated with Ba-pulling and Qian-traction manipulation with neck suspension and movement;while the control group was treated with Bashen-pulling and stretching manipulation in a supine position. The treatment was performed once a day, 10 times as a treatment course. The therapeutic efficacy was evaluated after 1 treatment course, and the changes in the scores of visual analog scale (VAS) and neck disability index (NDI) were observed. Results: The total effective rate was 97.7% in the observation group, and 83.3% in the control group, and the difference between the two groups was statistically significant (P<0.05). After treatment, the VAS and NDI scores of both groups were significantly decreased (both P<0.01), and the differences in the VAS and NDI scores between the two groups were statistically significant (both P<0.01). Conclusion: Both Ba-pulling and Qian-traction manipulation with neck suspension and movement and Bashen-pulling and stretching manipulation in a supine position can relieve pain and improve cervical function in patients with acute cervical radiculopathy, and Ba-pulling and Qian-traction manipulation with neck suspension and movement can produce more significant efficacy than Bashen-pulling and stretching manipulation in a supine position.
