1.Influence of sodium selenite on adiponectin expression in kidney of rats with diabetic nephropathy

Objective:To investigate the influence of sodium selenite on adiponectin expression in kidney of rats with diabetic nephropathy and the relation between sodium selenite and adiponectin,so as to investigate the role of sodium selenite and adiponectin in diabetic nephropathy.Methods: Diabetic nephropathy rat models were established by injecting streptozocin intraperitoneally and high fat diet.The study was divided into 3 groups,namely,a control group,a diabetic nephropathy group and a selenite-treated group.Animals in the selenite-treated group were given selenite solution 1 ?g/kg daily via intragastric administration; those in the other 2 groups were given the same dose of saline in the same way.The animals were sacrificed 10 weeks later and the blood and urine samples were used for biochemical examination.Samples of renal cortex were cut quickly and fixed with 4% glutaral for observation under scanning electron microscope.The renal samples were made into paraffin section for observation of the pathological changes under light microscope and for location of protein expression via immunohistochemistry analysis.The expression of adiponectin mRNA and protein was examined by real-time PCR and Western blotting,respectively.Results: The general condition and biochemical parameters of animals in the selenite-treated group were obviously improved and the pathological changes were obviously releived compared with those in the diabetic nephropathy group.Immunohistochemistry analysis showed that the expression of adiponection in the selenite-treated group was obviously stronger than that in the diabetic nephropathy group,and the expression was found in both glomcrulus and renal tubule.The expression of adiponectin mRNA in the selenite-treated group and diabetic nephropathy group was 2.043 and 1.373 folds that of the control group,respectively.The expression of adiponectin protein in the selenite-treated group was significantly higher than those in the control group and diabetic nephropathy group (P

Objective To investigate the influence of sodium selenite on the expression of adiponectin and nephrin in the kidney of rats with diabetic nephropathy, the relation of sodium selenite with adiponectin and nephrin, and to study the roles played by these factors in diabetic nephropathy. Methods Rat models of diabetic nephropathy were established by injecting intraperitoneally streptozocin combined with high fat diet. Three groups of rats were employed: control group, diabetic nephropathy group, and selenite-treated group. The selenite-treated rats were given selenite solution by intragastric administration every day, and other groups were given same volume of saline. After 10 weeks, all rats were sacrificed, blood and urine samples were collected. The kidneys were used to observe pathological changes with light microscope, and to locate protein expression with immunohistochemistry analysis. The mRNA and protein expressions of adiponectin and nephrin were analyzed via real-time PCR, RT PCR, and Western blot. Results The basic condition, biochemical parameters, and pathological images in selenite-treated group were better than those in diabetic nephropathy group. Immunohistochemistry analysis showed that the expression of adiponectin was in both glomeruli and renal tubules in selenite-treated group, and the coloration was enhanced as compared with diabetic nephropathy group. The mRNA and protein expressions of adiponectin in selenite-treated group were significantly higher than those in diabetic nephropathy group and the control group (all P<0. 05 ). Nephrin expression in selenite-treated group was also higher than that in diabetic nephropathy group, but lower than that in control group. Conclusion Sodium selenite can enhance the mRNA and protein expressions of adiponectin and nephrin in kindey of rats with diabetic nephropathy. Sodium selenite,adiponectin, and nephrin may play important roles in delaying and preventing the development of diabetic nephropathy.
3.A method of plethysmography wave de-noising based on wavelet transform and spline interpolation

Objective: To eliminate the noise of the plethysmography wave and obtain plethysmography wave signal without noise. Methods:Using sym4 and combining the improved wavelet threshold wavelet algorithm to remove high frequency white noise of plethysmography wave, and then using spline differential fitting method to remove baseline drift of pulse wave volume. Results:The experimental results show that the high frequency noise and baseline drift of plethysmography wave have been removed effectively processed by the sym4 wavelet and combining with improved threshold of wavelet algorithm and spline difference after fitting. Conclusion:The method can well remove the noise of the plethysmography wave and retains the effective information of plethysmography wave effectively.
5.Assessing mandibular bone mineral density with morphometric analysis

Objective:To determine the effectiveness and validity of morphometric analysis made from dental panoramic tomograms(DPTs) for predicting mandibular bone mineral density(BMD) while taking the result of dual energy X-ray absorptiometry(DXA) as gold standard.Methods: Twenty healthy subjects(8 male,12 female) with intact dentition,aged from 1126 years old,were included in the present study.The BMD of each patient was measured at the lumbar spine(L1-L4) by DXA;the results were obtained based on the density of surface(g/cm2).Dental panoramic radiography was carried out using dental panoramic tomography(RTG230/ENR,made in Italy).Mandibular cortical thickness(MCT) and panoramic mandibular index(PMI) values were calculated and their relationship with DXA measurements were subjected to correlation analysis.The validity of MCT and PMI measurements were assessed by sensitivity and specificity.Results: We found mandibular BMD was positively correlated with MCT(r=0.268,P

As one of the most important strategies of no drug therapy,biomaterials have drawn extensive application in clinical use.Current animal tests and assays of cell level haven't reached the satisfying depth on the mechanism of biocompatibility and material-host interaction.As the typical method of post-genome era,proteomics contributed to investigation in the biomarker of diseases and the global theory of physiological function and drug effect.Along with the increase of the inquiry of biomaterial,high quality evaluation of bio-effects and the reliability is required.Utilizing proteomic method to study the dynamic variation of the whole proteins after contacting with certain biomaterial is an effective means to explore the direction of its further improvement.This review summarizes most representative studies on function and biocompatibility of biomaterials and focus on the application in dental implant materials,which indicate the significance of the combination of the biomaterials development and the progressive biology assay technique.
7.Eosin Y-water test for sperm function examination.

Based on the principles of the in vitro staining technique, hypotonic swelling test, and water test, the Eosin Y-water test method was developed to simultaneously detect the integrity of the sperm head and tail and sperm membrane structure and function. As a widely used method in clinical laboratories in China, the Eosin Y-water test is methodologically characterized by three advantages. Firstly, both the sperm head and tail can be detected at the same time, which allows easy and comprehensive assessment of membrane damage in different parts of sperm. Secondly, distilled water is used instead of the usual formula solution to simplify and standardize the test by eliminating any potential effects on the water molecules through the sperm membrane due to different osmotic pressure or different sugar proportions and electrolyte solutions. Thirdly, the test takes less time and thus can be repeated before and after treatment. This article focuses on the fundamental principles and modification of the Eosin Y-water test and its application in sperm function examination and routine semen analysis for male infertility, assessment of the quality of sperm retrieved by testicular fine needle aspiration, semen cryopreservation program development, and evaluation of sperm membrane integrity after microwave radiation.

8.Evaluation of the anterior transsphincteric removal of rectal villous adenoma

Objective To evaluate the value and safety of the anterior transsphincteric operation for treating rectal villous adenoma.Methods The clinical data of 17 cases with rectal villous adenoma,including 6 cases with malignant change,operated by anterior transsphincteric approach at Changzheng hospital,from February 2000 to August 2006,were analyzed retrospectively.The distance between the anal verge and the lower margin of the tumor was 5~ 8 cm(average 7 cm).The tumors were 2~ 5 cm in diamater.Results The rectal villous adenoma was completely removed with negative resection margins in all the 17 patients.Fecal continence,urinary function and sexual function were preserved successfully in all the patients.No postoperative complications developed.The postoperative pathological stage of rectal villous adenoma with malignant change was pTisN0M0 in 3 cases,pT1N0M0 in 2,and pT2N0M0 in 1.At a median follow-up of 46 months,no patients had recurrence.Conclusions Anterior transsphincteric approach is useful and safe for operation of rectal villous adenomas and those with early mmalignant change.It is particularly valuable for large midrectal villous adenoma.
9.The effects of titanium surface modified by micro-arc oxidation on the collagen secretion and the expres-sion of osteogenesis-related genes of MC3T3-E1 cells

Objective:To investigate the effects of titanium surface modified by micro-arc oxidation (MAO)on the collagen secre-tion and the expression of osteogenesis-related genes of MC3T3-E1 cells.Methods:Pure titanium discs were divided into two groups:Titanium with modified surface by MAO(MAO group)and titanium with polished surface(PT group).Tissue culture polysty-rene plates (TC)were used as controls.Surface morphology of the samples was examined by field-emission scanning electron micros-copy(FESEM).Surface roughness of samples was measured by surface roughness meter.MC3T3-E1 cells were cultured on the sur-face of the samples and the collagen secretion on the samples was measured by sirius red-based colorimetric microassay after 12 days of culture.On day 16 of culture,the expression of osteogenesis-related genes was examined by RT-PCR.Results:A porous oxide layer was observed on the surface of MAO treated samples and the surface roughness was more than that of PT group(P<0.01 ). Cells in MAO group showed higher collagen secretion than in PT or TC groups(P<0.05).MAO treated surface induced higher OSX, COL-Iα1 and OPN expression than PT.Conclusion:MAO treated titanium surface may induce more collagen secretion of MC3T3-E1 cells than PT.
10.Lumbar disc degeneration:tissue engineering repair and biomechanical evaluation

BACKGROUND:At present, the clinical treatment of lumbar disc degeneration mainly includes conservative treatment, traditional surgery and minimal y invasive surgery. The therapeutic purpose is to relieve symptoms, but the long-term effect is not very satisfactory. Therapeutic methods focusing on biological functional recovery have been concerned gradual y, but the clinical application is far in sight. OBJECTIVE:To review the advances in the treatment of lumbar disc degeneration regarding tissue-engineered repair and biomechanics. METHODS:PubMed database was searched by the first author for relevant articles published before December 2014 using the keywords of“intervertebral disc degeneration, clinical treatment, biological treatment, tissue engineering, biomechanics, repair, progress”in English. A total of 100 articles were searched initial y and final y, 40 articles were included in result analysis. RESULTS AND CONCLUSION:Although the therapeutic schemes are varied, the treatment of intervertebral disc degeneration is a great chal enge for clinicians and basic researchers. Currently there is no perfect clinical treatment, and indications corresponding to various therapies should be paid attention as wel as long-term fol ow-up evaluation. For various reasons, the biological treatment for intervertebral disc degenerative disease is becoming more and more popular, providing a promising prospect for the treatment of intervertebral disc degeneration. So far, large amounts of data have been obtained from animal experiments, but there are stil many problems to be solved. Other chal enges also involve the al aspects of general tissue engineering methods, such as cel s, cytokines and scaffolds. In these studies, the nucleus pulposus tissue engineering based on the combination of heparin-functionalized chitosan hydrogel, cytokines and stem cel s exhibits a promising prospect.
