1.Ki-67 expression in T1 primary breast cancer tissue and its relationship with Her-2 and ER

Objective To study the expression of Ki-67 in T1 primary breast cancer tissue and its relationship with Her-2 and ER,evaluate the prognosis of early breast cancer.Method s Sixty cases of women T1 primary breast cancer tissues of 60 female cases from January 2008 to June 2011 were selected.The expressions of Ki-67 were detected by immunohistochemieal method and the relationships between Ki-67 and clinical-pathological fcatures,Her-2 and ER were analysed.Results The expression of Ki-67 in T1 primary breast cancer was 55% among the 60 patients.The expression of Ki-67 was not related with the clinical-pathological type of breast cancer and menopause (P>0.05).The expression of Ki-67 in group with lymph node metastasis was higer than in group without lymph node metastasis(P =0.047 ,P <0.05).The expression of Ki-67 in Her-2positive group was higher than Her-2 negative group (P= 0.021,P<0.05),and the expression of Ki-67 in ER positive group was lower than that of ER negative group (P = 0.037,P < 0.05).Conclusions The expression of Ki-67 in T1 primary breast cancer indicates the possibility of lymph node metastasis,and its positive correlation with Her-2 positive expression,ER negative expression indicates poor prognosis.

5.Technology of supercritical CO_2 extraction process of Dragon's Blood

Object To study the technology of supercritical CO_2 extraction of effective components from Dragon's Blood which has the inhibiting function on ?-glucosidase. Methods The influence of the extraction pressure, temperature, time and fluid velocity on surplus extraction rate and the improvement of inhibition was mainly discussed. Results The optimal experimental parameters were 15 MPa extraction pressure, 45 ℃ extraction temperature, 30 kg/h fluid velocity and 2 h extraction time. Conclusion The results show that SFE-CO_2 can enormously increase the surplus extraction rate and the inhibitory activities on ?-glucosidase.
6.Regional HPV subtypes distribution characteristics and their relationship with cervical lesions

Objective To understand the infection status and distribution characteristics of human papillomavirus (HPV)in fe-male genital tract in Fengxian district of Shanghai City in order to provide preliminary recommendations for prevention and treat-ment of HPV in this area.Methods The women aged over 1 6 years old receiving HPV testing in the Southern branch Hospital of Shanghai Sixth People′s Hospital From January 2011 to December 2015 were taken as the research subjects and their cervical secre-tions were performed the HPV 21 subtypes detection,among them,1 374 cases underwent colposcopic fixed point biopsy of cervix for determining the pathological grade.Then the data were recorded and analyzed by the SPSS20.0 software.Results Twenty-one HPV subtypes were detected,in which the HPV positive rate was 17.54% (6 313/35 977),and types with high HPV infection rate in these five years were in turn HPV18,HPV52,HPV16,HPVCP8304,HPV58,HPV53,HPV53,HPV53 and HPV58;the HPV positive rate in 1374 cases of cervical lesions was 82.75% (237/1 374),the common types distributions in cervical lesions were HPV16,HPV58,HPV52,HPV18 and HPV31 respectively.Conclusion The HPV infection situation in Fengxian district of Shang-hai City has certain regional characteristics.Then the HPV prevention and treatment strategy suitable for this region may be formu-lated according to the research results.
7.Evaluation of Rabbit Skin Irritation and in vitro Transdermal Absorption of Diclofenac Sodium Microemul-sion

OBJECTIVE:To investigate the property,skin irritation and in vitro transdermal absorption of Diclofenac sodium microemulsion(DS-ME),and to explore the feasibility of local external use of it. METHODS:The content of DS in DS-ME was determined by ultraviolet spectrophotometry. The distribution of particle size was determined by laser particle size analyzer. The ef-fects of DS-ME and blank micro-emulsion on normal skin of single administration,normal skin of multiple administration and dam-aged skin of single administration were investigated by rabbit skin irritation test. The transdermal parameters of DS-ME and commer-cially available DS gel through isolated skin of mice were compared by Franz diffusion cell. RESULTS:The prepared DS-ME was O/W microemulsion with particle size of(30.140±9.020)nm. Compared with blank ME,DS-ME had no significant difference in rabbit skin irritation score. Steady permeation rates of DS-ME and commercially available DS gel were 34.16 and 18.62 μg/(cm2·h), respectively;the permeation coefficient of them were 1.029 and 0.561 cm/h;the delay time were 0.124 2 and 0.367 2 h. CONCLU-SIONS:The particle size of DS-ME is small and not irritant to skin,and can improve transdermal absorption rate of DS.
8.The Effect of Basic Fibroblast Growth Factor (bFGF) on the Grafting Ability of CD34~+Cord Blood Cells

Objective To investigate the effect of bFGF on the grafting ability (grafing efficiency)of CD34 + cord blood cells seeded in 5 FU injured human bone marrow stromal cells.Methods Using combined methods of bone marrow stromal cell culture system,light microscopy and electron microscopy,flow cytometry,we investigated the toxic effects of 5 FU on human bone marrow stromal cells (hBMSC) and the benefit of bFGF on dynamics of hBMSC as well as the ability of the cord blood stem cell to bind hBMSC.Results After 3 hours incubation with 5 FU,significant changes in hBMSC cytosolic and nuclear morphology were observed.Nuclear splitting and abnormal shapes of the pseudopodia were found under the electron microscope.Flow cytometry revealed that apoptosis appeared before G 0/G 1,and that cells in G 0/G 1and G 2/M phases were remarkably lower,while the cells in S phase were remarkably higher than the controls.Two hours after adding the cord blood hematopoietic progenitor cells,the group treated with bFGF(injury with intervention of bFGF group,IBG)showed higher proprtion of the adherence layer compared to the group without adding bFGF(injury group,IG)(29% versus 12%) Conclusion 5 FU causes DNA damage in hBMSC.It also impairs the binding ability of cord blood stem cell to hBMSC.bFGF is able to repair hBMSC damage and improve the binding of CD34 + cord blood hematopoietic progenitor cells onto hBMSC.
9.Effect of Different Methods on Sterilizing Hands:A Randomized and Controlled Experiment Study

OBJECTIVE To explore the method for sterilizing hands which is effective,convenient and without stimulation for medical personnel.METHODS A total of 150 nurses on duty were divided into three groups randomly.In the test groups 1and 2,towels with 0.2% peracetic acid and BANENG disinfectant fluid were used separately,but in the control group only soap with flowing water was used for hands sterilization.RESULTS The rate of bacterial removal was 98.82%,72.63% and 68.64%,respectively,among the 3 groups.The efficacy of BANENG disinfectant fluid was similar to 0.2% peracetic acid.Both of them showed significant differences compared with control group.CONCLUSIONS Both 0.2% Peracetic acid and BANENG disinfectant fluid have better disinfection efficacy,and BANENG disinfectant fluid is more easier in use.
10.Clinical effect of amiodarone combined with bisoprolol in patients with congestive heart failure and ventricular arrhythmia

Objective To explore clinical effect and safety of amiodarone combined with bisoprolol in patients with congestive heart failure and ventricular arrhythmia. Methods Selected 100 cases with congestive heart failure and ventricular arrhythmia in our hospital from January 2011 to April 2017 as research objectives and divided them into two groups randomly with 50 cases in each group. Provided amiodarone to control group and provided amiodarone combined with bisoprolol to observation group. Compared two groups' arrhythmia and cardiac function, heart rate, ejection fraction, QT time (QTc) corrected for heart rate before and after treatment as well as adverse events. Results Observation group's effective rate of arrhythmia treatment and the effective rate of cardiac function were 94.00%, 96.00%, those were significant higher than control group's 70.00%, 78.00% (P<0.05). Two groups' heart rate and ejection fraction score after 3 month treatment were significant higher than those before treatment (P<0.05), and observation group's improvement was more significant (P<0.05). Observation group's QTc was significantly prolonged after 3 months of treatment (P<0.05), but control group's QTc did not change significantly. There was no significant difference in the incidence of adverse reactions between the two groups. Results Amiodarone combined with bisoprolol has significant clinical effect on patients with congestive heart failure and ventricular arrhythmia. It is safe and worthy to be promoted clinically.
