1.Clinical Efficacy of Fasudil Hydrochloride Injections (Administered at Different Time) for Acute Cerebral Infarction

OBJECTIVE:To evaluate the efficacy and safety of hydrochloric fasudil(administered at different time after onset)for acute cerebral infarction(ACI).METHODS:90 patients developed ACI within 72 h were randomly divided into trial group and control group of 45 each.The trial group was subdivided into trial group 1(developed ACI within 24 h)and trial group 2(developed ACI for more than 24 h).The control group received Compound Radix Salviae Miltiorrhizae injection for 15 days while the trial groups received Fasudil Hydrochloride for 15 days.Improvement of neurological function was evaluated using China Stroke Scales(CSS).RESULTS:The CSS of trial group 1 and trial group 2 was significantly decreased as compared with that of control group,and the CSS in trial group 1 was markedly decreased than in trial group 2,all showing significant differences(P

4.Ultrastructural changes in platelets with experimental severe decompression sickness

The present study describes systematically the ultrastructural changes of blood and some organs of rabbits, goats and rats with severe decompression sickness. In our experiment, the animals were exposed to pressure of 0.6 MPa and 1.0 MPa for 60 minutes respectively, to induce severe decompression sickness. The results are as follows: 1. Abnormality of platelets. The granules of platelets decreased and the platelet pseudopods were formed with adhesion and aggregation of platelets. 2. Deformation of erythrocytes. Triangle-like or shell-like cells, dacryocytes, twisted cells, and Ⅰ—Ⅲ type echinocytes were found. 3. Changes of endotheliocytes of the vascular wall became flattened. The space between these cells significantly widened (up to 4-18?m), and blood cells could be found to have wedged into it. 4. Formation of air-blood surface by the intravascular blood. 5. Alteration of organelies. There occured the expansion of rough endoplasmic reticulum, the degranulation and rupture of membranes, as well as the swelling and dissolution of mitochondria.
5.Laboratory Accreditation as an Important Means for the Enhancement of Drug Analysis Capability

OBJECTIVE: To provide references for domestic drug manufacturing enterprises' participation in laboratory accreditation as well as the enhancing of drug analysis capability. METHODS: Both international and domestic laboratory-ratified standards, the importance of laboratory accreditation and drug manufacturing enterprises' participation in laboratory accreditation were expounded. RESULTS & CONCLUSIONS: Domestic drug manufacturing enterprises' participation in laboratory accreditation is conducive to the enhancing of their drug analysis capability.
6.Commissioning Manufacture of Overseas Drugs in China in view of the Amendment of the Pharmaceutical Affairs Law in Japan

OBJECTIVE:To provide references for Chinese pharmaceutical enterprises in developing commissioning manufacture of overseas drugs.METHODS:The registration system of medicine commissioning manufacture in China,Japan and other countries were compared.The current status of Chinese drugs commissioning manufacture and the feasibility for Chinese pharmaceutical enterprises to gain commissioning manufacture of overseas drugs were analyzed.RESULTS & CONCLUSIONS:Both the government and pharmaceutical enterprises in China should take advantage of the amendment of the pharmaceutical affairs Law in Japan to contribute to the development of commissioning manufacture of overseas drugs.
7.The Value of Diagnosis and Prognosis of Chest Imaging Feature for Severe Acute Pancreatitis

Objective To study mechanism and relationship of acute lung injury that be complicated with severe acute pancreatitis(SAP).To analyze the chest imaging feature of X-ray and CT of SAP and improve the recognition of clinical value of diagnosis and prognosis.Methods Thirtynine SAP patients(17 male,22 female)age ranged from 26 to 64 of SAP that be diagnosed by clinic and CT were analysed retrospectively.All cases were checked by chest X-ray film and rechecked at least three times,of them chest CT were taken in 11 cases.Results Ascites was found in 34 cases,pleural effusion was found in 19 cases.Pulmonary edema sign of X-ray and CT was found in 16(41%) cases when SAP was diagnosed by pancreas CT.The sign was found in 24(61.5%) cases in 24 hours and in 37(94.9%)cases in 48 hours and all cases in 72 hours.In the 16 early pulmonary edema cases,There were pleural effusion in 11 and pericardiac effusion in one case,10 cases of them developed to acute respiratory distress syndrome(ARDS)and 7 cases died.Conclusion The chest imaging feature of SAP has important significance for early diagnosis of ARDS and prognosis of SAP,and can be served as a clinical indication of severe degree of SAP.
8.ERCC1 and Ki67 Expression in Patients with Breast Cancer and Relationship Between Its Expression and Sensitivity of Platinum Chemotherapy

Objective To investigate the expressions of excision repair cross complementation group 1 ( ERCC1) and Ki67 in patients with breast cancer, and the relationships between their expressions and sensitivity of platinum-based chemotherapy. Methods Totally, 129 cases were pathologically diagnosed as breast cancer.Paclitaxel and carboplatin were used simultaneously. Chemotherapy regimen was as follows:Gemcitabine 1 000 mg??( m2 )-1 , IV drop on day 1 and 8;cisplatin 25 mg??( m2 )-1 , IV drop on day 1-3, for six cycles ( 21 days a cycle ) . ERCC1 and Ki67 expression in tumor tissue was observed by immunohistochemical analysis.Platinum-based chemotherapy sensitivity and survival of patients with different levels of ERCC1 and Ki67 expression were analyzed. Results In 129 patients, 18 cases were ERCC1 and Ki67 double-negative ( ERCC1-Ki67-) , and the clinical effective rate and 3-year cumulative survival rate were 88.89%and 83.33%, respectively.Twenty-four cases were ERCC1 positive but Ki67 negative ( ERCC1+Ki67-) , and the clinical effective rate and 3-year cumulative survival rate were 50. 00% and 62.50%, respectively.Thirty-three cases were ERCC1 negative but Ki67 positive (ERCC1-Ki67+), and the clinical effective rate and 3-year cumulative survival rate were 54. 55% and 60. 60%, respectively. Fifty-four patients were ERCC1 and Ki67 double-positive ( ERCC1+Ki67+) , and the clinical effective rate and 3-year cumulative survival rate were 22.78% and 31. 48%, respectively.Compared with ERCC1-Ki67- group, the clinical treatment efficiencies of cisplatin-based chemotherapy in ERCC1+Ki67- group, ERCC1-Ki67+ group, and ERCC1+Ki67+ group were significantly decreased ( P<0. 05 ) . The clinical treatment efficiency in patients of ERCC1+Ki67+ group with cisplatin-based chemotherapy was significantly decreased as compared with ERCC1+Ki67- group and ERCC1-Ki67+ group (P<0.05).Compared with ERCC1- Ki67- group, three-year cumulative survival rate in patients of ERCC1+ Ki67- group and ERCC1- Ki67+ group, ERCC1+Ki67+ group was significantly decreased ( P<0. 05 ) . Compared with ERCC1+Ki67-group and ERCC1-Ki67+group, three-year cumulative survival rate in patients of the ERCC1+Ki67+group was significantly decreased ( P<0.05) . Conclusion The expression levels of ERCC1 and Ki67 in breast cancer were high. Their expression levels are closely related with clinical efficiency of platinum-based chemotherapy.
9.Relationship between polymorphism of cytochrome P450 2C19 and clinical response to clopidogrel

Objective To discuss the relationships of the genotype of cytochrome P450 2C19 (CYP2C19) gene and and clopidogrel responsiveness.Methods The study enrolled 110 patients with acute myocardial infarction who were treated with clopidogrel in Tiantan Hospital of Beijing from November 2012 to May 2014.Patients were treated by aspirin and clopidogrel.CYP2C19 polymorphisms were detected by genotype testing kits.Platelet inhibition rates were measured by thrombelastography to represent the antiplatelet effect of clopidogrel.The platelet inhibition rates of clopidogrel were compared among different genotypes.Results Compared with carriers of CYP2C19 * 2 or * 3 reduced-function allele,CYP2C19 * 1 wild type had higher platelet inhibition rate of clopidogrel ((35.73 ± 19.29)% vs.(48.30± 20.84)%),and the difference was significant(t =3.264,P<0.05).There was no difference between intermediate metabolizer((35.72± 19.27)%) and poor metabolizer((35.75±19.89)%;P>0.05).The frequency of wild-type homozygotes CYP2C19 * 1/* 1 was higher in responders than in low responders(frequency of low reaction group CYP2C19 * 1/* 1 14 cases (40.0%),* 1/.2 10 cases(28.6%),* 1/* 3 4 cases(11.4%),* 2/* 2 5 cases(14.3%),* 2/* 3 2 cases (5.7%),frequency of reaction group were 45 cases (60.0%),15 cases (20.0%),4 cases (5.3 %),7 cases (9.3%),4 cases(5.3%);x2 =3.838,P =0.05).Conclusion Polymorphism of gene CYP2C19 is associated with different responses to clopidogrel.CYP2C19 * 2/ * 3 reduced-function allele is associated with low response to clopidogrel.
10.Effect of caveolin-1 on renal injury and the expression of tight junction protein in MRL/lpr mice kidney

Objective To study the effect of caveolin-1 on renal injury and the expression of tight junction proteins in MRL/lpr mice kidney.Methods The mice were divided into 4 groups:5 mice in the normal control group (BALB/c mice);the MRL/lpr lupus mice (n=18) were randomly divided into the MRL/lpr group in which 6 mice were included;the negative control group in which 6 mice were included;the caveolin1 transfection group in which 6 mice were included.The changes of urine protein,the levels of urea (BUN) and creatinine (Cr) were detected.The expressions of claudin-5,occludin,ZO-1 and caveolin-1 protein were determined by western bloting.Analysis of variance was used to determine statistical significant differences between the two groups.A significance level of 0.05 was considered as signigicant.Results Compared with the control group,24 h urine protein [(2 894±437) mg,(412±72) mg],BUN [(8.7±1.5) mmol/L,(6.9±0.4) mmol/L],Cr [(106±22) μmol/L,(85±4) μmol/L] were significantly increased,level of caveolin-1 protein increased (265±17,61±6),the level of occludin (114±12,190±12),claudin-5 (60±5,80±6) and ZO-1 (98±11,206±15) protein decreased in the MRL/lpr group (P<0.05).After caveolin-1 transfection,the levels of urinary protein [(1 253±249) mg,(2 894±437) mg],BUN [(6.5±1.3) mmol/L,(8.7±1.5) mmol/L],Cr [(78±17) μmol/L,(106±22)μmol/L] were significantly decreased,and the levels of occludin (218±16,114±12),claudin-5 (87±6,60±5)ZO-1 (313±17,98±11) were increased (P<0.05).Conclusion The expression of caveolin-1 protein in the renal tissues of lupus nephritis increases.Caveolin-1 can reduce the expression of tight junction proteins and contribute to progres-sion of lupus nephritis.
