1.A discussion of informatization management of reagent used in science and research of hospital based on RCA

Objective:To provide scientific references for establishing a whole informatization management system of reagent used in science and research of hospital based on the root cause analysis (RCA). And to find existed problems and analyze main reasons from reagent management, material, time, financial, personnel and other factors through fishbone chart, and to provide relative management measurement of informatization.Methods: The reagent consumptions used in science and research of a tertiary referral center hospital in Fujian province in 2015 year were collected, and then the purchase amount and ratio of them were analyzed.Results: The purchase amount of reagent for science and research of this tertiary referral center hospital in Fujian province was more than 2.55 million RMB in 2015 year, and the ratio of national clinical key special projects was 23.32% and that of provincial clinical key special projects was 6.85%. The two items spent more purchase amount, and the type and ratio of reagent of various department was different depended on their own various developed situation.Conclusion: The informatization management and service mode of reagent used in science and research have very important role in the construction of soft power of hospital. In present, a series of disadvantages existed in various clinical and research department, for example, the use of reagents are scatter, involved professional span is larger and disperse, and accurate trace, dynamic tracking and flexible deploy machines are difficultly achieved. Therefore, how to promote comprehensive informatization management for relative resource, for optimizing structure configuration, for controlling cost and ensuring quality of reagent has became the most important task for informatization management of reagent used in science and research.
2.The game analysis for cost management of medical consumable and stakeholder

Objective:To provide the scientific reference basis in order to establish a perfect cost controlled system for medical consumable. To analyze game factors, such as main resource, the price-off influence and price-off aspiration and so on, from different views, such as medical institutions, patients, manufacturers and other stakeholders and so on, based on game theory and situation of medical consumables management.Methods: To collect the management information of medical consumables cost from a tertiary referral center hospital in Fujian in recent 3 years, and adopt descriptive study to analyze the income situation of medical consumable, medical consumable proportion and medical consumable cost per capita.Results: The medical consumable proportions from 2013-2015 were 14.44%, 16.27% and 18.75%, respectively. Medical consumable cost per patient from 2013-2015 were 3547.95RMB, 4122.3 RMB and 5059.02RMB, respectively. All of these datum contained the drug information.Conclusion: It is obvious that the medical consumable proportions and medical consumable cost per patient were enhancement from 2013 to 2015. The necessity of medical consumable leaded to the direct influence on medical cost. On the other hand, the artificially high price and unreasonable implement of medical consumable are the urgent problems and must be resolved as soon as possible.
3.Effects of Nimesulide on Expression of Matrix Metalloproteinase-2 in Human Colonic Cancer Cell Lines

Objective To investigate the effects of selective cyclooxygenase-2(COX-2) inhibitor nimesulide on the proliferation of colon adenocarcinoma cells in vitro and the expression of matrix metalloproteinase-2(MMP-2).Methods The human colon cancer cell lines HT-29 and HCT-116 were employed in the study,grouped as nimesulide group,DMSO control group and blank control group.After treatment with nimesulide,the inhibitory effect of nimesulide on the proliferation of cancer cells was quantified by MTT assay,and the expression of MMP-2 in the cells was detected by quantitative zymography.Results Nimesulide inhibited the proliferation of HT-29 and HCT-116 cells in time and dose-dependent manners.The inhibitory effect on HT-29 cells was stronger than that on HCT-116 cells.Nimesulide down-regulated the MMP-2 expression in HT-29 cells,whereas the expression in HCT-116 cells remained unchanged.Conclusion Nimesulide can obviously inhibit the growth of colon cancer HT-29 cells with positive COX-2 protein,suggesting that nimesulide may down-regulate the expression of MMP-2 by inhibiting the activity of COX-2.
5.Experimental Study of Testicular Autotransplantation in Dogs

DSA and color Doppler ultrasound examination showed that autografts had good blood supply. Conclusion The modified testicular autotransplantation operation could improve the successful rate of transplantation, and had not obvious influence on testicular function. It can be further applied in homologous testicular transplantation experimental and clinical research.
6.Preliminary Study of Nocturnal Breathing Disturbance in Patients of Ischemic Cerebral Stroke with Dysphagia

Objective To analyze the role of dysphagia during the course of sleep apnea in patients who experienced a first-ever ischemic stroke.Methods According to the video-fluoroscopy,50 patients were divided into two groups:dysphagia and non-dysphagia groups.The effect of different sites and severity of infarction on the breathing disturbance was analyzed,and the possible role of dysphagia was discussed.Results Patients with dysphagia showed more number of OAIs at the acute phase(AHI,20.9;OAI,19.1)than that of non-dysphagic patients(AHI,9.5;OAI,8.6).Multiple correlation analysis found the extent of dysphagia had relationship with the AHI,especially OAI(AHI,P=0.018;OAI,P=0.024).Conclusion The number and type of obstructive apneic events may occur at the acute phase of a first-ever cerebral infarction in patients with pharyngeal muscle alterations secondary to the neurological lesion.
7.Study on in vitro biomineralization of enamel-binding peptide.

We present the binding ability of a new peptide (CMPQVMPMC-) with dental enamel after being evaluated in the present study. Under a standard procedure, the recovery of M13 filamentous phage was greatly enhanced by displaying the peptide in phage coat protein p III. Then the cyclic peptide was synthesized using a solid method. The effect of the cyclic peptide in vitro biomineralization was tested in a single-diffusion microtiter plate gel system. Absorbance at 405 nm of each sample was recorded for 24 h at every 6 h intervals. The relatively increased values of each sample were expressed as percentages relative to the blank group (100%). The cyclic peptide resulted in a concentration-dependent delayed nucleation. In addition, the overall values of peptide groups at the end of 24 h were lower than those in the control group but much higher than those in the BSA control group.

8.Effect of morphine dependence on nucleic metabolism in hippocampal neurons in mice

Objective To study the molecular mechanism about injurious effect of morphine dependence on the construction and function of hippocampal neurons. Method Mice were given (sc) increasing doses of morphine to form morphine-dependence model ,and the DNA and RNA changes stained with acridine orange (AO) fluorescence probe technique were investigated in hippocampal neurons of morphine-dependence group,naloxone-precipitated withdrawal group in morphine-dependence mice and control group. Results Compared with control group , the staining changes of DNA,RNA decreased obviously in hippocampal neurons of both morphine-dependence group and naloxone- precipitated withdrawal group in morphine-dependence mice,especially the later. Conclusion Both morphine-dependence and naloxone-precipitated withdrawal in morphine-dependence mice injured nucleic acid metabolism in hippocampal neurons ,especially the later.Those changes may be some reasons of decreased brain function ,especially in learning and memory deficits.
9.Effects of Celecoxib on Proliferation of Human Colonic Cancer Cells and on The Hepatic Metastasis of Animal Model

10.Relationship between MDR1 exon 26 genotype and dosage of FK506 and acute rejection following renal transplantation

Objective To evaluate whether the MDR1 exon 26 genotype is associated with the dosage of FK506 and acute rejection in renal transplant patients. Methods One hundred and six patients who administrated FK506 after renal transplantation were retrospectively analyzed. The patients were investigated for MDR1 genotype determined by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) assays. According to the genotype, the patients were divided into 3 subgroups, wild homozygous-CC, mutant homozygous-TT and heterozygous-CT. At 3rd, 6th and 12th month after renal transplantation, enzyme-linked immunosorbent assay (ELISA) was used to measure the FK506 whole blood concentration. Whole blood FK506 concentration per dose/kg/day and acute rejection within one month postoperation were compared among 3 subgroups. Results Of the 106 patients, 32 (30.2 %), 44 (41.5 %) and 30 (28.3 %) were carriers of CC, CT, TT in MDR1 exon 26, respectively.
