1.Change of Ethology and Gastronomies of Rats after Spinal Cord Injury

Objective To explore the alteration in ethology and change of muscle fibers of gastronomies of rats following spinal cord injury(SCI). Methods 108 Wistar rats were divided into sham operation group (n=38) and SCI group (n=38) and rehabilitation group (n=32).The ethology was evaluated by Basso, Beattie, and Bresnahan Locomotor Rating Scale (BBB Scale), and the alteration of muscle tissue's reactivitywas observed by immunohistochemistry staining. Results The BBB scores of rehabilitation group were higher than SCI group 3weeks after injury, but all was lower than 10. The cross section areas of gastrocnemius in both groups were getting smaller, but the rehabilitationgroup was slighter. Conclusion Spontaneous recovery occurred in some degree for rats' hind limb after spinal cord injury. Rehabilitationtraining is beneficial for the recovery of rats' motor function, and alleviates the atrophy of hind limb after spinal cord injury.
2.Nano-hydroxyapatite/composite resin:cytototoxicity and application in dental restoration

BACKGROUND:It has been found that the compressive strength and flexural strength of nano-hydroxyapatite combined with light-cured resin matrix can neet the requirements of dental restorative materials. OBJECTIVE:To detect the cytotoxicity of nano-hydroxyapatite/composite resin, and to analyze its effects on the dental restoration. METHODS:In vitro test:passage 5 periodontal ligament fibroblast-like cel s and L-929 cel s suspensions were cultured in the 10%, 50%, 100%of nano-hydroxyapatite/composite resin extracts, respectively, and fresh maintenance medium functioned as negative control group. Absorbance values were detected within 7-day culture. In vivo repair test:57 cases with tooth defect were enrol ed containing 37 males and 20 females aged from 15 to 31 years old, and al were subjected to nano-hydroxyapatite/composite resin treatment. A 3-year fol ow-up was undergone to detect the completeness, marginal adaptation and color matching of the restoration, as wel as the periodontal probing depth, sulcus bleeding index, tooth mobility, dental plaque index, gingival crevicular fluid and aspartate aminotransferase level. RESULTS AND CONCLUSION:In vitro test results showed that the different concentrations of nano-hydroxyapatite/composite resin extracts had no effects on both periodontal ligament fibroblast-like cel s and L-929 cel s. In vivo repair test results found that after 3 years of fol ow-up, the periodontal probing depth, sulcus bleeding index, tooth mobility, dental plaque index, gingival crevicular fluid and aspartate aminotransferase level of patients were significantly decreased than those before repair (P<0.05), but the alkaline phosphatase level revealed no significant change (P>0.05). Unfortunately, three cases with incomplete restoration, four cases with color mismatching, and three cases with the inadaptable edge occurred. To conclude, the nano-hydroxyapatite/composite resin holds good biocompatibility and no cytotoxicity, which can maintain a good periodontal health condition in the dental restoration.
3.Research on Legal Problems of the Transplantation by Taking Foetus as the Donor

Actually in China there has already been much clinical transplantation which takes foetus as the donor,however,there is no regulation or law to restrict it,which is not good for the overall development of national transplantation career.The text discusses the qualification of the transplantation by taking foetus as the donor and the conflicts in different relationships during the process of the transplantation,quotes the correlation of the American legislation and provides the necessity and principles of similar legislation in combination with national circumstances,expecting to give some references for national legislation on fetal tissue transplantation.

5.In vitro expression and in vivo osteogenic capability of pcDNA3-hBMP2-transfected marrow stromal cells in rabbits

BACKGROUND: Whether bone morphogenetic protein 2 (BMP-2) can be transduced into marrow stromal cells (MSCs) and produce osteogenic effects by viral or non-viral vector remains unclear? OBJECTIVE: To observe the expression of cultured rabbit MSCs transfected with pcDNA3-hBMP2 in vitro. Simultaneously, the MSCs were transfected but not screened and then transplanted into autologous muscle to investigate the osteogenic capability by X-ray. DESIGN, TIME AND SETTING: A controlled observation experiment was performed at the Department of Orthopedics, Liaoning Medical University between November 2004 and April 2005. MATERIALS: Six adult New Zealand rabbits, of either gender, weighing 2.0-3.0 kg, were included for this study. BMP2 antibody was the product of Sanaka Company, USA. pcDNA3-hBMP2 was provided by Professor Pu Qin from the Department of Biochemistry, Fourth Military Medical University of Chinese PLA). Restriction enzyme was purchased from Takara biotechnology (Dalian) CO., LTD., China. METHODS: Super-purified plasmid pcDNA3-hBMP2 was extracted from E. coli. Bone marrow was taken from the adult rabbit femur for harvesting MSCs by density gradient separation. The MSCs were divided into the following 4 groups: Group A, cells were transfected and screened by G418; Group B, cells were transfected by pcDNA3-hBMP2; Group C, cells were transfected by empty vector pcDNA3; Group D, only transfection reagent Fugene 6 was added.MAIN OUTCOME MEASURES: Transient BMP2 expression was analyzed by immunohistochemistry. Expression of osteocalcin and collagen I was examined by immunohistochemistry and in situ hybridization, respectively. Two weeks after transfection, MSCs from the group B were autologously transplanted into the muscle. Four weeks later, X-ray assay was used to observe bone formation.RESULTS: pcDNA3-hBMP2 was successfully transduced into MSCs and transiently expressed BMP2 100%. Four weeks after gene transfection, expression levels of osteocalcin and collagen I were significantly higher in the group A than in the groups C and D. X-ray results demonstrated new bone formation four weeks after MSCs transplanted into the muscle.CONCLUSION: pcDNA3-hBMP2 can safely and efficiently transfect MSCs and induce them to differentiate towards osteoblasts by secreting BMP2.
6.Efficacy of Liraglutide for Obesity or Overweight:A Systematic Review

Objective To evaluate the effectiveness and safety of liraglutide for obesity or overweight. Methods Random controlled trials of liraglutide for obesity or overweight were gathered from MEDLINE, EMbase, CENTRAL, CNKI, VIP and WanFang. We screened the retrieved studies according to the inclusion and exclusion criteria, evaluated the quality of included studies, and then performed meta-analyses with The Cochrane Collaboration's Revman 5. 3. 0 software. Results Twelve randomized controlled trials of liraglutide for obesity or overweight were included, in which 11 trials were written in English and one trial in Chinese. The results of meta-analyses showed that the body weight was significantly reduced in the liraglutide group than in placebo, insulin, exenatide and glimepiride groups [RR=-0. 91,95%CI(-1. 01,-0. 81),P<0. 000 01;RR=-2. 88,95%CI(-3. 37,-2. 39),P<0. 000 01; RR=-1. 12,95%CI(-1. 32,-0. 92),P<0. 000 01; RR=0. 45,95%CI(-0. 62,-0. 27),P<0. 000 01]. Moreover, liraglutide had significant effect in decreasing HbA1c and systolic blood pressure of patients with obesity or overweight. Conclusion Liraglutide is effective for controlling body weight of patients with obesity or overweight. But its long-term efficacy still needs to be confirmed by performing more RCTs with high quality, large sample and long term follow-up.
7.Evaluation of application effect of clinical nursing pathway in patients with laparoscopic cholecystectomy

Objective To discuss the application effect of clinical nursing pathway (CNP) in patients with laparoscopic cholecystectomy(LC). Methods 168 patients with laparoscopic cholecystectomy were analyzed from January 2009 to March 2011.They were randomly divided into the experimental group and the control group with 84 patients in each group.The control group adopted traditional nursing mode,and the experimental group adopted clinical nursing pathway.Hospitalization time,hospital expenses,untoward effect,mastering of health knowledge,satisfaction degree of patients were analyzed. Results Hospitalization time,hospital expenses,untoward effect,mastering of health knowledge and satisfaction degree were better in the experimental group than those of the control group. Conclusions Application of clinical nursing pathway in nursing quality management can shorten hospitalization time,and reduce hospital expenses,promote mastering of health knowledge,and improve satisfaction degree of patients and their families with nursing service.
8.Relationship between utrophin and dystrophin in muscle of patients with several nerve and muscle diseases

Objective To investigate relationship between utrophin and dystrophin in muscle of patients suffered from several neurological muscular diseases.Methods Muscle biopsies of 26 cases of patients suffered from 8 categories neurological muscular diseases and 2 cases of control were analysed for utrophin and dystrophin by immunofluorescence experiments.Results In a majority of Duchenne muscular dystrophy (DMD) patients,their sarcolemma revealed absent,weak or discontinuous fluorescence for dystrophin.In non-DMD muscular dystrophies,lipid storage myopathy,myotonic dystrophy,inflammatory myopathies, neurogenic amyotrophy, polymyositis, mitochondrial encephalomyopathy, myogenic amyotrophy,immunofluorescence reactivity for dystrophin were strongly exhibited in entire sarcolemma.In normal biopsy sample,strong immunofluorescence reactivity for dystrophin was identified in entire sarcolemma,while weak and discontinuous fluorescence was identified on a minority of sarcolemma of DMD patients with severely reduced dystrophin.There was no immunofluorescence reactivity for utrophin in sarcolemma of DMD patients with moderate decreased dystrophin,non-DMD muscular dystrophies and other 6 categories of neurological muscular diseases,nor in sarcolemma of normal biopsies.Conclusions utrophin is expressed in sarcolemma of DMD patients,who have severely reduced dystrophin simultaneously. utrophin is absent in sarcolemma of other categories of neurological muscular diseases including non-DMD muscular dystrophies with normal dystrophin expression and DMD patients with moderately decreased dystrophin.
