1.Study on quality standard for Puhuang Dispensing Granule

Objective: To study the quality standard for Puhuang Dispensing Granule (Pollen Typhae). Methods : Puhuang Dispensing Granule was identified by TLC and isorhamnetin-3-O-neohesperidoside and typhaneoside in Puhuang Dispensing Granule were determined by HPLC. Results : The linear relationship was at the range of 0.4~1.21?g for isorhamnetin-3-O-neohesperidoside and 0.42~1.25?g for typhaneoside, respectively. The average recovery was 98.88% for isorhamnetin-3-O-neohesperidoside and 99.68% for typhaneoside, respectively. Conclusion : The method is available with a good reproducibility and can control the quailty of Puhuang Dispensing Granule.
2.Comparison between methods for preoperative evaluation of cardiac risk for patients scheduled for noncardiovascular surgery

Objective To test the accuracy of American College of Cardiology (ACC)/American Heart Association (AHA) guidelines for preoperative cardiovascular evaluation for noncardiac surgery in Chinese patients and to compare ACC/AHA guidelines with Goldman index and Lee index. Methods From January to December 2003, all patients aged ≥70 yr or patients aged 40-69 yr with a history of cardio- and cerebro-vascular disease, abnormal ECG or diabetes scheduled for noncardiovascular surgery were included in this study. A total of 1 248 patients were eligible. Their clinical data including demographic data, history of cardiovascular disease, routine physical examination and laboratory tests, the scheduled surgery and type of anesthesia were collected. The patients were then evaluated for cardiac risk and classified according to ACC/AHA guidelines (high, moderate, low and no risk), Lee index (class I -IV ) and Goldman index (class I - III ). The cardiac risk of the scheduled surgery was then stratified according to ACC/AHA guidelines. The patients were followed up until discharged from hospital. Cardiac events were defined as cardiac death, myocardial infarct, myocardial ischemia, minor myocardial cell injury, ventricular dysfunction and serious arrhythmia. Likelihood ratio of the 3 methods was calculated. Risk factors for adverse events were identified by univariate analysis and multivariate Logistic regression analysis. Results Of the 1 248 patients 694 were male and 554 female. Their age ranged from 40-102 years (mean age 65.4 yr). 44.7 % of the patients were aged ≥ 70 years. High risk operation accounted for 6.3 % and emergency operation 7.9% . One patients died of cardiac event and ten patients of other causes. Seventy-three perioperative cardiac events occurred in 53 patients. The morbidity rate was 4.2% . 3.THE EFFECT OF RECOMBINANT HUMAN HEPATIC STIMULATOR SUBSTANCE ON GROWTH OF LIVER OVAL CELLS

Objective Hepatic stimulator substance(HSS)is newly defined as a growth mitogen to hepatocytes.This sutdy is aiming to investigate the effect of recombinant human hepatic stimulator substance(rhHSS) on growth of liver oval cells(OVCs). Methods OVCs were prepared from intoxicated rats with 3'-methyl-dimethylaminoazobenzene.After confirmation of OVC morphologically and histochemically,the cell cultures of OVC were exposed to various dosages of rhHSS for 12 and 24?h,respectively.The cellular proliferation was analyzed by MTT and flow cytometry. Results Administration of rhHSS(160-400 mg/L)inhibited the proliferation of OVC,as indicated by MTT and cell cycle analysis,the effect appeared non-dose dependent pattern.The peak of inhibition occurred at 240?mg/L.After incubation with 240?mg/L of HSS for 12 and 24?h,the percentages of S-phase were reduced 47.8% and 35.8% to those of the untreated cells.respectively.Conclusion rhHSS exhibits the inhibitory effect on growth of OVC.
4.OBSERVATION OF CURATIVE EFFECT IN THE NEAR FUTURE OF 124 CASES WITH MULTI DRUG RESISTANT PULMONARY TUBERCULOSIS

The curative effect on 124 cases with multi drug resistant pulmonary tuberculosis from January 1996 to June 1999 was reported.All the cases, with an average history of 5.84 years, were recurrent ones. They included 78 cases of infultative pulmonary tuberculosis,46 cases of chronic fibrocavitative pulmonary tuberculosis. Sputum of all the cases was cultured for Mycobacterium hominis tuberculosis with Kuang`s culture medium and drug susceptibility test was done. As a result, the drug resistant rate to 2 drugs, 3 drugs,4 drugs, 5 drugs and more were 20.16%,25.0%,20.9%,33.72% respectively.The authors added 1~2 kinds of anti TB drugs never used before in each case to KOP (Kanamycin, Ofloxacin, Sodium Aminosalicylate),with a mean 3.3 months treating course. The sputum negative conversion rate was 66.9%,foci absorption rate was 72.6%, cavity close up 50.0% . There wasn′t any serious adverse effect encounted. It suggest that KOP synergetic bacteriocidal to Mycobacterium tuberculosis either inside or outside cells, with fewer side effect, is a good regimen for multi drug tuberculosis treatment.
5.THE CURATIVE EFFECTS OF L-ARGININE ON HIGH ALTITUDE PULMONARY EDEMA

In order to study the curative effects of L arginine (L Arg) on high altitude pulmonary edema (HAPE),we had treated 10 patients with HAPE by intravenous L Arg at 3700m, and the result was compared with 11 patients who inhaled low concentration nitric oxide (NO) and 24 subjects with routine treatment.The findings were as follows:compared with the routine group, inhaling NO or intravenous L Arg could hasten the disappearance of moist rales and X ray shadow, and shorten the illness time( P 0.05). It suggested that intravenous L Arg is a handy way to treat HAPE, and there is no toxicity. It is worthy of further trial.
6.THE RESEARCH OF FIVE DRUG-RESISTANCE GENES OF MYCOBACTERIUM TUBERCULOSIS IN 134 PATIENTS

M.tuberculosis strains isolated strains from sputum specimens of 134 patients were analyzed by PCR SSCP and traditional drug susceptibility tests. The results sbowed that the gene mutation rate of PZA, SM, RFP,INH and EMB resistance in those clinically isolated strains was 42 7%,71 7%,78 9%,68 6% and 43 9%, respectively. The gene mutation was in relation with drug resistance level of M. tuberculosis. The gene mutation rate was higher in high concentration resistance strains than in low concentration resistance strains.

8.Effect of Iodine Deficiency and Hypothyroidism on the Protein Expressions of Calcineurin in the Hippocampus of Pups

Objective To observe the effect of iodine deficiency and hypothyroidism on the protein expressions of calcineurin in the hip-pocampus of pups. Methods Female Wistar rats (n=28) after pregenancy were randomly divided into control group,hypothyroid group and iodine deficient group. According to the dose of propylthiouracil (PTU) in the fed water, hypothyroid group was divided into 5 ppm group and 15 ppm group (7 rats in each group). Totally 5 pups from each group were sacrificed and perfused intracardially in postnatal day (PN) 7,PN14 and PN21. Brains were removed,fixed and sectioned coronally. All sections were observed and analysed for the protein exression of calcineurin by immunohistochemistry in the hippocampus CA1,CA3 and DG regions. Results In PN14 and PN21,protein levels of cal-cineurin in GA1 and CA3 regions of the hippocampus in iodine-deficient and 15 ppm treatment groups were significantly higher than those of the controls (P< 0.05) and in DG region,the contrary was true. In PN7,the positive products were scarely observated in each region and the protein expression was no significantly different in all four groups. Conclusion Iodine deficiency and hypothyroidism may increase the protein expression of calcineurin.
9.Mutations in the D-loop region of mitochondrial DNA in gastric cancer

Objective:To investigate the variations o f mitochondrial D-loop region in gastric cancers. Methods:The D-loop region of 12 gastric cancers together with th e adjacent normal tissues were amplified by PCR and sequenced. Results: Among the 12 cases,16 mutations and 28 polymorphisms were identified with the mutations rate of 50%.There were 3 new polymophisms which we re not reported previously in the GeneBank among the 28 polymorphisms. Conclusion:We think that the mitochondrial DNA D-loop region is a highly polymorphric and mutable region and the mutation rate is relatively high in patients with gastric cancer.
10.Injury of hepatic mitochondria and its pathogenesis in rats with endotoxemia

AIM: To investigate the effect of endotoxin on rat hepatic mitochondria. METHODS: Rats were randomly divided into two groups:endotoxin group and the control. 8 cases of animals were included in each group. The effect of electron leak on the production of endogenous oxygen free radicals and the changes of mitochondria function were studied.RESULTS: Treated with endotoxin, a significant increase in O 2 and the rate of state 3,4 were observed in liver mitochondria; The rate of electron transfer to proton pump of mitochondria respiratory chain complex Ⅱ+Ⅲ( H +/2e -), respiratory control rate and ADP/O decreased significantly. CONCLUSION: A increase in production of endogenous oxygen free radicals induced by endotoxin plays an important role in the injury of rat liver mitochondria.
