1.Fluorescence in situ hybridization and immunohistochemistry for detecting HER-2 gene amplification and protein expression in breast cancer tissue

Objective To analyze and compare the fluorescence in situ hybridization(FISH) and immunohistochemical(IHC) for detecting HER-2 gene amplification and protein expression in breast cancer tissues .Methods 110 cases of breast cancer from Janu-ary 2008 to May 2012 receiving the modified radical mastectomy were selected .The resected breast cancer tissue was detected by FISH and IHC and the detected results were performed the comparative analysis .Results Among 110 cases of breast cancer tissue , 25 cases(22 .73% ) were the HER-2 protein expression(+ + + ) ,44 cases(40 .00% ) were(+ + ) ,26 cases(23 .64% ) were(+ ) and 15 cases(13 .64% ) were(-) .Among 110 cases ,the gene amplification was in 28 cases(25 .45% ) and no gene amplification was in 82 cases(74 .55% ) .The positive(+ + + ) of the IHC detection was coincident with that of FISH ,and the negative(+ /-) of the IHC detection was also coincident with that of FISH ,there was statistical difference between the suspicious positive of the IHC de-tection and the results of FISH (P<0 .05) .But the total coincidence of the IHC detection results and FISH test results was 89 .29%(25/28) ,and the two detection methods had the positive correlation (χ2 =84 .89 ,P<0 .01) .Conclusion The positive and negative expression of the IHC detection has better consistency with that of the FISH detection .However ,the coincidence of the IHC suspi-cious positive expression and the FISH results is poor ,indicating that the suspicious positive sample of the IHC detection needs to be detected by the FISH detection .
2. Development of printable and online health education material evaluation tools

Objective:
To build a comprehensive evaluation system for printable and online materials, and provide reference for the compilation and selection of patient education materials.
Methods:
Taking Suitability Assessment of Materials (SAM) as frame and combined Patient Education Materials Assessment Tool (PEMAT) and CDC Clear Communication Index, we screened indicator of the evaluation system preliminary and all of them have been adjusted with the language habits and social and cultural characteristics of our country. By Delphi method, 15 experts in related fields were selected to conduct 2 rounds of expert consultation to establish the final indicator system. The weights of the different indicators were also analyzed by Analytic Hierarchy Process (AHP).
Results:
The indicator system of printable and online patient education materials was consisted of 7 first-level indicators,28 second-level indicators,84 third-level indicators. The positive coefficients and the authority coefficients of the experts were 100% and 0.87 of the two rounds. The coordination factors of the experts in second-level and third-level were respectively 0.362 and 0.255 for the second round. The top three indicators of the first-level indicators were content (0.271 5), behavioral suggestion (0.190 4), and language expression (0.190 4); the highest-level index weight was scientific content (0.095 7), and the lowest was the use of commonly numbers, correct interpretation of numbers and units′ meaning (0.007 0).
Conclusion
The comprehensive evaluation system for printable and online materials is scientific, comprehensive and easy to operate. It can provide reference for medical worker to prepare and select printable and online materials.
3.Relationship between expression of Her-2 gene and the transfer number of axillary lymph node and its influence on prognosis

Objective To investigate the relationship between expression of Her-2 gene and the transfer number of axillary lymph node and its influence on prognosis.Methods A total of 351 cases with primary breast cancer from January 2008 to January 2011 were selected.The expression of Her-2 gene was detected by immunohistochemical method,and analyzed the relationship between it and the transfer number of axillary lymph node and its influence on prognosis.Results The expression of Her-2-,+,++ had no correlation with the transfer number of axillary lymph node (x2 =3.757,1.650,2.379,P > 0.05),while the expression of Her-2 +++ had correlation with the transfer number of axillary lymph node (x2 =8.681,P < 0.05).The 2 years survival rates in the expression of Her-2-,+,++,+++ were 77.01% (201/261),85.00% (34/40),29.17%(7/24),1 1.54% (3/26),and which in the expression of Her-2--+ was significantly higher than that in the expression of Her-2 ++-+++ (x2 =61.605,P < 0.01).The transfer number of axillary lymph node was 0 and 1-3,the 2 years survival rate in the expression of Her-2--+ was significantly higher than that in the expression of Her-2 ++-+++,and there was significant difference (x2 =61.605,14.747,P < 0.05).The transfer number of axillary lymph node was 4-9 and ≥ 10,there was no significant difference in the 2 years survival rate between the expression of Her-2--+ and Her-2 ++-+++ (x2 =3.691,3.482,P > 0.05).Conclusions The expression of Her-2-,+,++ has no correlation with axillary lymph node metastasis,and the 2 years survival rate in the expression of Her-2--+ is higher than that in the expression of Her-2 ++-+++,while Her-2-has no difference with Her-2 + in prognosis.While the transfer number of axillary lymph node ≤ 3,the expression of Her-2 gene may be an important prognostic factor.

AIM To observe the protective effect of Ganduqing Granule(GDQ) on acute hepatic injury. Objective:To detect the expression of has_circ_0001459 in peripheral blood mononuclear cells (PBMCs) of Epstein-Barr virus (EBV)-infected individuals and evaluate its diagnostic value for EBV infection.Methods:The expression profiles of circRNAs in PBMCs of patients with EBV infection were obtained by whole-transcriptome sequencing. Differentially expressed circRNAs with statistical significance were selected based on the criteria of ∣log 2Fold Change∣≥1 and P<0.05. Based on the composition, length and primer specificity, hsa_circ_0001459 with high expression was selected for further research. The relative expression of hsa_circ_0001459 in PBMCs of 60 patients with EBV infection and 45 healthy people was detected by real-time fluorescent quantitative PCR. Receiver operating characteristic (ROC) curve and Kappa analysis were used to analyze the diagnostic value of hsa_circ_0001459 expression for EBV infection. Results:The expression of hsa_circ_0001459 showed an up-regulation trend in patients with EBV infection as compared with that in the healthy people, and the results were consistent with the sequencing results. The area under the ROC curve for screening EBV-infected individuals was 0.83(95%CI: 0.75-0.91) with the sensitivity of 0.80(95%CI: 0.66-0.89) and the specificity of 0.77(95%CI: 0.65-0.86). Kappa analysis indicated that hsa_circ_0001459 was moderately consistent with EBV DNA in the diagnosis of EBV infection (κ=0.51).Conclusions:Hsa_circ_0001459 tended to be highly expressed in PBMCs of EBV-infected individuals, which might be uses as a diagnostic marker for EBV infection.
6.Study on reconstruction of rabbit alveolar bone defect with strontium-containing brushite bone ce-ments

Objective To investigate whether 5% strontium-containing brushite bone cements(DCPD) has repair effect on alveolar bone defects in osteoporotic rabbits. Methods Eighteen healthy adult female rabbits were used to establish osteoporosis models and were randomly divided into three groups:the blank control group, DCPD group,doped 5%DCPD group(5%strontium),with 6 rats in each group.In addition to the blank group, rats the other groups were filled with the corresponding bone cements in the bilateral alveolar bone defects. At 4 weeks and 8 weeks after operation,3 rats in each group were killed and given shooting the defect area X-ray.The expression of b-FGF was detected by immunohistochemistry. Results X-ray results showed that the defected of doped 5% DCPD group have been nearly completedat the 8 week,DCPD group partially completed repair,but blank group was not fully repaired. The results of immunohistochemistry showed that the expression of b-FGF was the highest at 4 weeks and decreased at 8 weeks after operation.Expression of b-FGF was significantly different at 4 weeks and 8 weeks among the three groups.Conclusion The 5%strontium-containing brushite bone cements can repair bone defect in osteoporotic rabbits.
7.Remineralization effect of nanohydroxyapatite on adjacent glazed surfaces

Objective:
To investigate the effect of a nanohydroxyapatite suspension with a 10% mass fraction on enamel demineralization after adjacent enamel removal and to provide a reference for the clinical use of nanohydroxyapatite to prevent enamel demineralization after adjacent enamel removal.
Methods :
Forty fresh extracted premolars were incised from the coronal surface of the crown after glazing and polishing with sand strips. Each tooth was made into two specimens. Eighty teeth were randomly divided into four groups: a blank group, a dolofluoride group, a dental mousse group and a 10% nanohydroxyapatite group. After 30 days of pH cycling in vitro, a microhardness tester was used to detect the enamel surface elements before and after deglazing treatment. Scanning electron microscopy (SEM) and X-ray energy dispersive spectrometry (EDS) were used to analyze the enamel surface elements.
Results:
After the experiment, the microhardness values of the adjacent glazed specimens of the four groups were (128.18 ± 6.89) kg/mm2 in the blank group, (216.75 ±9.87) kg/mm2 in the dolefluoride group, (198.88 ± 4.76) kg/mm2 in the dental care group and (218.44 ± 7.88) kg/mm2 in the 10% nanohydroxyapatite group, which were significantly different (F=189.14, P< 0.001). The LSD-t test showed that there was no significant difference between the dolefluoride group and the 10% nanohydroxyapatite group (P< 0.05), but the differences between the other groups were statistically significant (P > 0.05). There were significant differences in the Ca/P ratio among the blank group (1.39 ± 0.08), the dolefluoride group (1.70 ± 0.11), the dental care group (1.54 ± 0.12) and the 10% nanohydroxyapatite group (1.71 ± 0.14) (F=10.539, P< 0.001). There was no significant difference between the dolefluoride group and the nanohydroxyapatite group after the experiment (P > 0.05), but there were significant differences among the other groups (P< 0.05).
Conclusion
Dolerofluoride, dental protector and 10% nanohydroxyapatite can promote remineralization of enamel after enamel removal. The remineralization induced by 10% nanohydroxyapatite is similar to that induced by dolerofluoride.
