1.Application of NMR technique in the discovery and pharmacological studies of active substances from natural products.

The application of HPLC-NMR-MS hyphenated technique in the structural identification of trace substances from complex mixtures and the identification of endogenous and exogenous substances in the establishment process of metabolic profiling have become effective analytical tools in pharmaceutical chemistry, pharmacological and pharmacokinetic studies of active substances from natural products. Metabolomics method based on NMR technology can accurately portray metabolic phenotypes with the characteristics of diseases and a variety of disease-related pathways, and it can greatly enrich and supplement the traditional disease evaluation methods. So it can be used for pharmacological studies of active substances from natural products, such as toxicological studies, the dose optimization, active substances screening and pharmacodynamic evaluation. Hyphenated technique associated with metabolomics method based on NMR technology will accelerate the speed of the discovery of active substances from natural products, and improve the efficiency of their pharmacological evaluation.
2.Research progress in lncRNAs and the diagnosis, prognosis, and drug resistance of gastric cancer

Long non-coding RNAs (lncRNAs) are noncoding RNAs with lengths exceeding 200 bp. These lncRNAs cannot code complete proteins but control gene expression. Recent studies have shown that lncRNAs are closely related to the diagnosis, prognosis, and drug resistance of gastric cancer. This article summarizes the recent progress of studies in China and abroad on lncRNAs relative to the diagnosis, prognosis, and drug resistance of gastric cancer.
3.Significance of circadian gene Period2 expression in epithelial ovarian cancer and the effect of gene overexpression on growth of ovarian cancer xenografts in nude mice

Objective To study the significance of circadian gene Period2 expression in epithelial ovarian cancer tissues and the effect of gene overexpression on the growth of ovarian cancer xenografts in nude mice. Methods Twenty-two cases of ovarian cancer paraffin specimens in the First Hospital of Shanxi Medical University (ovarian cancer group) were chosed during Jau. 2010 to Dec. 2013, including 8 cases of stageⅠ, 8 cases of stageⅡ, and 6 cases of stageⅢ, while 6 cases of benign ovarian epithelial tumor paraffin specimens were selected as control (benign tumor group). Period2 gene were detected by real-time quantitative PCR and western blot methods in different stages of ovarian cancer tumor tissues. Established theovarian cancer xenografts in nude mice with ovarian cancer cell line SKOV3, and they weredivided into 3 groups (n=8), including the recombinant plasmid group, empty plasmid group and control group. Using gene transfection technique to transfer Period2 gene into tumor tissues, tested the expression of Period2 mRNA in tumor tissues by real-time quantitative PCR after transfection into all nude mice, monitoredthetransplant tumor growth and calculating the tumor inhibition rate,detected the antiapoptotic gene BRE, apoptosis related tumor necrosis factor receptor (TNFR1) andtumor suppressor gene NIX in tumor tissues by real-time PCR and western blot in different groups. Results (1)The expression level of Period2 mRNA in tumor tissues amongovarian cancer group stageⅠ,ⅡandⅢwere respectively 2.59±0.50, 0.47± 0.08 and 0.42 ± 0.08, but benign tumor group was 6.59 ± 1.05. The expression level of Period2 protein in ovarian cancer group stage Ⅰ,Ⅱ and Ⅲ were respectively 0.835 ± 0.087, 0.412 ± 0.035 and 0.199 ± 0.031, while benign tumor group was 0.874 ± 0.094. The expression level of Period2 mRNA and protein in benign tumor group was higher than those in ovarian cancer group stageⅠ,ⅡorⅢ(P<0.01). With ovarian cancer stage increased, the expression of Period2 mRNA and protein were decreased or absent (P<0.05).(2)Two weeks after transfection, the expression level of Period2 mRNA in recombinant plasmid group tumor tissue was significantly higher than those in the empty plasmid group or the control group (6.11±0.56 vs 0.50±0.09 vs 0.44 ± 0.08, respectively;P<0.01), the transplanted tumor volume of recombinant plasmid group was significantly less than those in empty plasmid group or the control group[ (486±70) mm3 vs (835±106) mm3 vs (846 ± 110) mm3, respectively;P<0.01], the tumor inhibition rate of the recombinantin plasmid group was as high as 42.9%, that was significantly higher than those in the empty plasmid group and the control group (3.8% and 0, respectively;P<0.05).(3)The expression level of BRE mRNA and protein in transplanted tumor tissues in the recombinant plasmid group were significantly lower than those in empty plasmid group and the control group;the expression level of TNFR1 and NIX were significantly higher than those in the empty plasmid group and the control group (all P<0.05). Conclusions Period2 mRNA and protein expression are absent in ovarian cancer of advanced stage. Transfection and stable expression of Period2 gene could slow down the growth of ovarian cancer, and the tumor inhibition rate could be significantly increased. Period2 gene may promote ovarian cancer cells apoptosis through inhibition of BRE gene expression and promoting TNFR1, NIX gene expressionto exert anti-tumor effect.
4.Vascular Collagen Remodeling, Apoptosis and Bcl-2/Bax of Vascular Smooth Muscle Cells in Carotid Artery on SHR

Objective To observe the vascular collagen remodeling, apoptosis and Bcl-2/Bax of vascular smooth muscle cells in carotid artery on SHR . Methods 12 SHR treated with Irbesartan ,12 SHR without treated Irbesartan ,12 wistar rats were normotensive control group. The vascular volume fraction of Collagen were determined by picrosirius red staining .The expression of the proteins Bcl-2 and Bax were determined by Immunhistochemia,VSMC apoptosis was identified by in situ TDT-mediated dUTP nick end labeling(TUNEL). Results Significant Fibrosis exists in carotid artery of SHR. After 20 week's treating, the vascular volume fraction of Collagen and the ratio of wall/cavity were descended, but weren't parallel. The expression of the protein Bcl-2 and APOI were higher than the subjects of normotensive control, its reduced after treating; The expression of the proteins Bax was similar with control, it increased after treating. Conclusions The results suggest that vascular collagen remodeling exist, its maybe related with apoptosis , the proteins Bcl-2 and Bax of VSMC. The Irbesartan can treat it.
5.Epithelial-mesenchymal transition and drug resistance of tumors

Differentiated epithelial cells could be transformed into mesenchymal cells through the epithelial-mesenchymal transi-tion (EMT), which is important for tumorigenesis and metastasis. Recent studies suggest that EMT is closely correlated to the resistance to chemotherapy of tumors. This chemoresistance could enhance tumor malignancy, which includes the induction of the EMT pheno-type. The paper reviews the research progress on EMT and its association with drug resistance of tumors.

Objective To discuss the relationship among hypoxia-inducible factor-1α (HIF-1ct),platelet derived growth factor (PDGF) and vascular endothelial growth factor (VEGF)/VEGFR signaling pathway in breast cancer occurring and metastasis in the early stage.Methods The clinical and pathological features and the expression of HIF-1 α,PDGF-A,VEGF-A and VEGFR-2 were assayed by histomorphology and immunohistochemical staining on tissue specimens from 61 patients of breast cancer and healthy breast tissues healthy breast tissue from 17 patients of benign breast disease.Results VEGF expression was observed in 39 out of 61 malignant breast tissues,more than distribution of positive staining 20 patients (51.3%) of which.VEGFR expression was found in 42 out of 61 lesions,.more than 10% distribution of positive staining 22 patients (52.4%) of which.Simultaneous expression of VEGF and VEGFR-2 was found in 12 cases.HIF-1α was expressed in 50 and PDGF in 39 out of 61 breast cancer patients,and in 36(72.0%) HIF-1α positive specimens and 23 (59.0%) PDGF positive specimens the staining area was more than 10%.The expression of VEGFR was correlated with patient's age (x2 =11.080,P =0.001) and lymph node metastasis (x2 =4.699,P =0.046).VEGF,HIF-1α and PDGF were not correlated with patient's age (x2 =0.880,P =0.415; x2 =1.620,P =0.303 ; x2 =0.150,P =0.786) and lymph node metastasis (x2 =0.677,P =0.437 ; x2 =1.394,P=0.323;x2 =1.841,P=0.192).Conclusion HIF-1α,PDGF,VEGF and VEGFR-2 express in breast tumor tissues at a high level,and VEGFR-2 is correlated with patient's age and lymph node metastasis.Abovementioned proteins can be used as markers for diagnosis,prognosis and targeted therapy.
9.Effect of emulsified isoflurane preconditioning on postsynaptic density protein 95 activation in brain during focal cerebral ischemia-reperfusion injury in rats

Objective To investigate the effect of emulsified isoflurane preconditioning on postsynaptic density protein 95 (PSD95) activation in brain during focal cerebral ischemia-reperfusion (I/R)injury in rats.Methods Thirty-two pathogen-free male SD rats weighing 280-300 g were randomly divided into 4 groups (n =8each): sham operation group(group S),group I/R,emulsified isoflurane group(group EI) and lipid emulsion group(group LE).Focal cerebral I/R was induced by middle cerebral artery occlusion for 2 h followed by reperfusion.8% emulsified isoflurane 10.5 ml/mg or 30% lipid emulsion 10.5 ml/mg was injected intraperitoneally at 24h before I/R in groups EI and LE respectively.The neurologic deficit score (NDS) was evaluated at 6 h of reperfusion and then 4 rats were sacrificed,and brains were removed for determination of phosphorylatied PSD95 (pPSD95) expression in ischemia cortex and hippocampus by Western blot.Four rats were sacrificed at 24 h of reperfusion and then their brains were removed for determination of infarct volume percentage.Results NDS,infarct volume percentage and pPSD95 expression in ischemia cortex and hippocampus were higher in groups I/R,EI and LE than in group S( P < 0.01 ).NDS,infarct volume percentage and pPSD95 expression in ischemia cortex and hippocampus were lower in group EI than in group I/R( P < 0.05).There was no significant difference in NDS,infarct volume percentage and pPSD95 expression between groups I/R and LE(P > 0.05).Conclusion Emulsified isoflurane preconditioning can attenuate focal cerebral I/R injury in rats by inhibiting PSD95 activation in brain.
10.Effects of emulsified isoflurane preconditioning on expression of platelet-activating factor and its receptor in brain during focal cerebral ischemia-reperfusion in rats

ObjectiveTo investigate the effects of emulsified isoflurane preconditioning on the expression of platelet-activating factor (PAF) and PAF receptor during focal cerebral ischemia-reperfusion (I/R) in rats.MethodsThirty-two healthy adult male SD rats weighing 250-300 g were randomly divided into 4 groups ( n =8each):group sham operation (group S); group I/R; group emulsified isoflurane preconditioning( group EI) and group lipid emulsion (group LE).Focal cerebral I/R was induced by 2 h occlusion of middle cerebral artery followed by reperfusion in groups I/R,EI and LE.8% emulsified isoflurane 10.5 ml/kg and 30% lipid emulsion 10.5 ml/kg were injected intraperitoneally at 24 h before cerebral ischemia in groups EI and LE respectively.The neurologic deficit score (NDS) (0 =no deficit,4 =unable to control) was evaluated at 12 h of reperfusion.Venous blood samples were collected for measurement of plasma PAF concentration.The animals were then sacrificed and their brains removed for determination of infarct size (by TTC staining) and PAF receptor expression in hippocampus and cerebral cortex (by Western blot).ResultsFocal cerebral I/R significantly increased NDS,the infarct size,plasma PAF concentration and PAF receptor expression in cerebral cortex and hippocampus in group I/R as compared with group S.Emulsified isoflurane preconditioning significantly attenuated the focal cerebral I/R induced above changes in group EI as compared with group I/R,but there was no significant difference between groups I/R and LE.ConclusionEmulsified isoflurane preconditioning can attenuate focal cerebral I/R injury by inhibiting PAF and PAF receptor expression.
