1.Identification of stem cell characteristics of human diploid cells 2BS

Objective To identify the stem cell characteristics of human diploid cells 2BS and evaluate the potential as biological function positive cells for quality detection of stem cells.Methods Flow cytometry was applied to detect the expression levels of stem cell surface markers in 2BS cells.The staining with alizarin red S,oil red O and alcian blue was used to evaluate the ability of osteogenic,adipogenic and chondrogenic differentiation,respectively.Co-culture with lymphocytes was used to observe the effects on lymphocyte proliferation.Results Human diploid cells 2BS expressed the surface markers CD73,CD 105 and CD90 of stem cells,and possessed the potential of osteogenic differentiation and inhibiting lymphocyte proliferation.Conclusion 2BS cells have several properties of stem cells and could be the positive control for surface markers detection of stem cells by flow cytometry.

IL-8 is a cytokine which belongs to the chemotactic superfamily.It is one of the major mediators in the inflammatory response.In recent years,it was found that it had multiple functions,such as chemoattractant,potent angiogenic,and so on.The cytokine is believed to play an important role in the pathogenesis of inflammation,tumor progress and immune disease.In this review,the properties of IL-8 and highlight its roles on inflammation and tumor progress were surveied.
3.Endoscopic ultrasonography in the diagnosis and endoscopic submucosal dissection in treatment of rectal carcinoid tumors

Objective To evaluate the clinical value of endoscopic ultrasonography (EUS) in diagnosis and treatment of rectal carcinoid tumom, and the safety and efficacy of endoscopic submucosal dissection (ESD) for rectal carcinoid tumors. Methods All the submucous tumor considered rectal carcinoid tumor received EUS examination and according to the result to select appropriate therapeutic regimen. Results All the 45 lesions were verified rectal carcinoid tumors by pathological examination. 42 patients received endoscopic submucosal dissection. Two tumors invaded into the muscularis propria and one tumor invaded beyond serosa and with lymph node metastasis received surgical strategy. Conclusion EUS can be used to determine surgical strategy for rectal carcinoid tumor for it provides precise information of the size, depth, border, muscularis propria or vessel infiltration of the tumor. For rectal carcinoid tumors smaller than 20 mm in diameter, ESD is safe and effective.
4.Effect and safety of methylprednisolone pulse therapy on spinal nerve function in patients with acute spinal cord injury

Objective To investigate the effect of methylprednisolone pulse therapy on spinal nerve functions in patients with acute spinal cord injury (ASCI) and evaluate its safety. Methods 200 patients with ASCI treated in our hospital from January 2010 to December 2016 were selected and were randomly divided into two groups,with 100 cases each group. The patients in control group were treated with routine basic treatment while patients in the study group were treated with methylprednisolone pulse therapy. On the basis of the American spinal cord injury Society scoring criteria (ASIA), the neurological functional recovery scores were performed were scored before and 6 weeks after treatment between the two groups and the adverse reactions were recorded. Results The scores of sensory and motor function of the patients in the two groups were higher than those before the treatment and there was statistical difference (P<0.05), and the scores in the study group was significantly higher than those in the control group, and the difference between the two grouPs was statistically significant (P<0.05). The incidence of adverse reactions in the study group (25.00%) was significantly higher than that in the control group (10.00%), and the difference was statistically significant (P<0.05). The incidence of lung infection and gastrointestinal reaction in the study group were significantly higher than those in the control group and there were statistical difference (P<0.05). Conclusion Methylprednisolone pulse therapy has significant effect on the recovery of neurological function in patients with ASCI, but the incidence of adverse reactions is high, so it is necessary to strengthen the monitoring and intervention in clinic.
5.Bacterial culture and drug susceptibility observation of children with chronic rhinosinusitis in Chongqing city

Objective To investigate the distribution of pathogenic bacteria in children with chronic rhinosinusitis (CRS) and their susceptibility to commonly used antibacterial drugs .Methods 95 cases of CRS complicating adenoidal hypertrophy were per-formed adenoidectomy .The secretion of maxillary sinus ostium and posterior ethmoid sinus ostium was collected during endoscopic adenoidectomy by the disinfected cotton swab for conducting the bacterial culture and the drug sensitivity test .Results 89 speci-mens showed the bacterial growth ,the positive rate of bacterial culture in all culture specimens was 93 .7% ,86 strains of aerobic bacteria ,28 strains of anaerobic bacteria .Among 95 patients ,81 cases of aerobic infection ,52 cases of anaerobic infection ,44 cases of mixed infection .The drugs susceptibility test in vitro displayed that amoxicillin and clavulanate potassium had the higher antibacte-rial activity to aerobic bacteria and metronidazole had the higher antibacterial activity to anaerobic bacteria .Conclusion Children CRS is dominated by the aerobic bacterial infection .The bacterial infection plays a larger role in children CRS .Antibacterial drugs have better effect for treating CRS .
6.Effects of bone marrow mesenchymal stem cell intravenous transplantation combined with autologous bone marrow stem cell mobilization on cardiac function in rats with cardiomyopathy-induced heart failure

BACKGROUND: Bone marrow mesenchymal stem cells (BMSCs) possess the potential in multi-directional differentiation and can differentiate into cardiomyocytes, which had self-discipline jumpiness. There are few reports on intravenous transplantation or autologous mobilization of BMSCs for treating dilated cardiomyopathy-induced heart failure. OBJECTIVE: To study the effect of BMSC intravenous transplantation combined with bone marrow stem cell autologous mobilization on cardiac function in rats with cardiomyopathy-induced heart failure. DESIGN, TIME AND SETTING: This control experiment was conducted at the National Physiological Laboratory, Molecule Laboratory, Animal Experimental Center and Central Laboratory of Shanxi Medical University from January 10th, 2007 to January 10 th, 2008. MATERIALS: Five Wistar rats served as BMSC donor. Fifty rat models of cardiomyopathy-induced heart failure were created by intraperitoneally injected with 15 mg/kg adriamycin for six weeks. Recombinant human granulocyte stimulating factor (GSF) was bought from Qilu Pharmacy Co., Ltd. CD44, CD34 and CD45 were given by Flow Cytometric Analysis Laboratory, Experimental Center, Shanxi Medical University. METHODS: BMSCs were isolated, purified and cultured in vitro. Surface marker CD45, CDD44 and CD34 were detected with f1ow cytometer. Wistar rat models were randomized into 4 groups. BMSCs and recombinant human GSF (50 ?g/kg) were injected into models by tail vein and hypodermoclysis infusion respectively in the combination treatment group (MG group). Recombinant human GSF (50 ?g/kg) was subcutaneously injected into models in the bone marrow stem cell autologous mobilization group (G group). BMSCs were injected into rat models by tail vein in the BMSC group. Rat models in the heart failure group were intact. MAIN OUTCOME MEASURES: ①Peripheral blood leukocyte and mononuclear cell counts and CD34 expression were obtained before treatment and 6 days after treatment. ②At 4 weeks, cardiac function parameters were evaluated by echocardiography and hemodynamic measurement. Diabetic peripheral neuropathy(DPN)is the most common complications of DM.It is considered to be progressive and irreversible.DPN occurred in a stocking and glove distribution,the mainly symptoms are pain,numbness and paresthesias.It is also the main cause of ulceration,infection and amputation.Usually,there is no special treatment for DPN besides blood-glucose control and pain control.Recently,Surgical decompression of peripheral nerves is used to restore sensation and relieve pian among DPN patients by Prof.Dellon and other doctors who have proved its efficacy in treatment of DPN.

10.Photochemical tissue bonding in tissue repair

Suture was a traditional method for tissue repair. However, sutures used in the closure of surgical wounds can induce inflammation and lead to scarring that appears as crosshatch marks. Threads used for sutures may cause immune response and surgical infection. In this review, we introduce a novel sutureless technique for wound repair, called photochemical tissue bonding (PTB). Absorption of visible light by a photosensitizing dye initiates photochemical reactions leading to covalent protein-protein crosslinks that bridge the wound surfaces, thus producing an immediate seal between the wound surfaces. This technique has been applied in various tissues including skin, cornea, nerve, blood vessels, et al. PTB reduces inflammation resulting in less scarring in skin. PTB dramatically improves the incidences of vascular anastomotic leakage and stricture. In addition, PTB enhances the recovery of nerve function after nerve repair.
