CHEK2 is an important susceptibility gene of breast cancer. CHEK2 gene mutations are related to pathologic features and prognosis of breast cancer. So the test of CHEK2 gene mutations may be has certain indication role in the treatment of breast cancer. However, the role of CHEK2 gene mutations in Chinese population still needs further research.
2.Measurement of MV CT dose index for Hi-ART helical tomotherapy unit

Objective To evaluate the patient dose from Hi-ART MV helical CT imaging in image-guided radiotherapy.Methods Weighted CT dose index (CTDI_W) was measured with PTW TM30009 CT ion chamber in head and body phantoms,respectively,for slice thicknesses of 2,4,6 mm with scanned range of 5 cm and 15 cm.Dose length products (DLP) were subsequently calculated.The CTDI_W and DLP were compared with XVI kV CBCT and ACQSim simulator CT for routine clinical protocols.Results An inverse relationship between CTDI and the slice thickness was found.The dose distribution was inhomogeneous owing to the attenuation of the couch.CTDI and DLP had close relationship with the slice thickness and the scanned range.Patient dose from MVCT was lower than XVI CBCT for head,but larger for body scan.Results CTDI_W can be used to assess the patient dose in MV helical CT due to its simplicity for measurement and reproducibility.Regular measurement should be performed in QA & QC program.Appropriate slice thickness and scan range should be chosen to reduce the patient dose.
3.Changes of serum S100 proteins and neuron-specific enolase following craniocerebral injury and effect of methylprednisolone intervention

BACKGROUND: In recent researches the concentration of S100 proteins and neuron-specific enolase have been proved positively correlated with the degree of brain injury, and can be used for assessing the degree and prognosis of brain injury. But whether the level of serum S100 proteins and phosphopyruvate hydratase can predict the role of glucocorticosteroid in the treatment of CNS damage is unclear.OBJECTIVE: To observe the changes of serum S100 proteins and phosphopyruvate hydratase level, aiming to explore the role of methylprednisolone in the treatment of craniocerebral inju ryin rats.DESIGN: Randomized controlled study.SETTING: The First Affiliated Hospital of Sun Yat-sen University.MATERIALS: This study was carried out at the Neurological Research Institute of Guangzhou Medical College in September 2003. Totally 72 SD rats were enrolled in this study.METHODS: Totally 72 SD rats were randomly divided into three groups,namely normal group of 8 rats, comparison group and treatment group of 32 rats respectively, the latter two groups were then subdivided into post-trautablishment: Brain contusion and laceration was made in rats according to Feeney's method, rats in normal group were only subjected to craniotomy group, rats were given intraperitoneal injection of methylprednisolone by dosage of 30 mg/kg instantly after injury, which replace by physical saline of the same dosage in comparison group. Rats in normal group were not given intraperitoneal injection, blood specimen was obtained instantly after put to death by cutting off head; in comparison group and treatment group,blood sample was obtained from rat at post-traumatic 1, 6, 12 and 24 hours and centrifuged for collecting superuatant, the level of serum S100 proteins and phosphopyruvate hydratase was detected by using ELISA method.RESULTS: Totally 72 rats were enrolled in this experiment and all regroup was (0.35±0.029) μg/L. The difference of serum S100 proteins was not obvious between normal group and treatment group at post-traumatic 24 hours, but the level of S100 proteins in treatment group and comparison group was significantly higher than normal group (P < 0.01 or P < 0.05);moreover in contrast with comparison group, the level of S100 proteins was found obviously lower in treatment group at post-traumatic 6, 12 hours and hydratase was (8.35±1.01) μg/L in normal group, the difference of serum phosphopyruvate hydratase level between comparison group and treatment group was higher than normal group, displaying statistical significance at all post-traumatic time points (P < 0.01), except for not obvious difference between comparison group at post-traumatic 24 hours and treatment group.While comparing to comparison group, the level of serum phosphopyruvate hydratase was found obviously lower in treatment group at post-traumatic 1,6 and 12 hours time point (P < 0.01 or P < 0.05).CONCLUSION: The level of serum S100 proteins and phosphopyruvate hydratase can be used as predictors for the degree of brain injury in rats, in addition methylprednisolone might exert therapeutic effect on traumatic brain.
4.Clinical efficacy and safety of hysteroscopy surgery combined with methotrexate for treating the patients with cesarean scar pregnancy

Objective To investigate the clinical efficacy and safety of hysteroscopy surgery combined with methotrexate (MTX) for treating the patients with cesarean scar pregnancy(CSP).Methods The clinical data of 64 patients with CSP were retrospectively analyzed.The patients were divided into observation group (46 cases) and control group (18 cases) according to treatment methods,the control group was given the therapy of local pregnancy intracapsular MTX combined with uterine curettage,and the observation group was given the therapy of local pregnancy intracapsular MTX combined with lesions removed surgery by hysteroscopy.The intraoperative blood Ioss,epistrophy time of β-human chorionic gonadotrophin (hCG),hospitalization time,clinical efficacy of the two groups were observed.Follow-up for 6 months the menstruation of the two groups was observed.Results The total effective rate in observation group was significantly higher than that in control group [100.0% (46/46) vs.66.7% (12/18),x2 =16.920,P < 0.01].The intraoperative blood loss,epistrophy time of β-hCG and hospitalization time in observation group were significantly lower than those in control group [(35.46 ± 11.63) ml vs.(176.78 ± 57.96) ml,(21.04 ± 6.79)d vs.(39.65 ± 12.79) d,(13.89 ±4.63) d vs.(26.82 ±8.94) d,P<0.01].The observation group had a successful surgery and no complications.In the control group,4 cases occurred uterine curettage intraoperative bleeding,2 cases underwent open lesion resection and then were forced to line hysterectomy.The rate of menstruation in observation group after operation 30-39 d was significantly higher than that in control group [84.8% (39/46) vs.25.0% (4/16),P < 0.01].Conclusions Hysteroscopy surgery combined with MTX for treating the patients with CSP has exact clinical effect.It is safe and reliable,and is worthy of promotion and application.
5.Effects of methylprednisolone on serum S-100B protein and neuron specific enolase in rats with brain injuries

Objective To observe the role of methylprednisolone in treatment of brain injuries through comparing the changes of serum S 100B protein and neuron specific enolase (NSE) levels. Methods Seventy two rats with brain contusion made by frontoparietal bone windows plasty with extradural hitting were divided into three groups: control group ( n =32), normal group ( n =8) and treatment group ( n =32) that were subdivided into groups 1, 6, 12 and 24 hours after injury. The levels of S 100B protein and NSE were measured at different time points after hitting by ELISA. Results ①The levels of serum S 100B protein and NSE was (0.35?0.03) ?g/L and (8.35?1.01) ?g/L, respectively in normal group. 7.Correlation between microalbuminuria and cardiovascular events in patients with coronary heart disease

Objective To investigate the correlation between microalbuminuria and cardiovascular events in patients with coronary heart disease.Methods A total of 120 CHD patients were divided into experiment group (30 mg/g< UACR<300 mg/g, n=80)and control group(UACR≤30 mg/g,n=40)The relationship between MAU and cardiovascular events rate was compared after 1 5 months.Results Microalbuminuria level in experiment group was significantly higher than control group(P <0.05).The incidence of cardiovascular event in experiment group was significantly higher(t = 3.71 7,P <0.05 ).MAU and cardiovascular e-vents were positively related by Spearman′s analysis(r=0.721,P <0.01).The effective factors were MAU and CRP(MAU:P =0.001;CRP:P =0.037).Conclusion MAU concentration had a great impact on the incidence of cardiovascular event.The incidence of cardiovascular might be predicted by the MAU.

10.Treatment of HBV/HCV co-infected patients in DAA era

Asian-pacific area,especially China,is Hepatitis B high epidemic area.Since 2011,the first generation of oral direct anti-HCV agents (DAAs) came to clinical use,the treatment of chronic hepatitis C has switched from interferon-based regimen era to DAA era.There is an increased awareness of hepatitis B (HBV) reactivation in chronic hepatitis C (CHC) patients coinfected with HBV treated with pan-oral direct-acting antivirals(DAAs).Compared with interferon-based regimen,HBV reactivation occurred earlier and more severe among patients received DAA regimen,and even fetal cases or case end up with liver transplantation was reported.Thus,association of liver diseases called to alert the occurrence of HBV reactivation among CHC patients who received DAAs regimen.It is hence important to have HBV serology screened in all CHC patients before initiation of pan-oral DAAs therapy and the usefulness of preemptive administration of effective anti-HBV nucleos(t) ide analogues in coinfected patients need to be further studied.
