5.Expression of brain-derived neurotrophic factor in the brain of old rats after chronic cerebral perfusion deficiency

Objective To explore the roles of expression of brain derived neurotrophic factor (BDNF) in the brain of old rats after chronic cerebral perfusion deficiency in the pathogenesis of vascular dementia. Methods Rat model of vascular dementia was established by modified Pulsinelli 4 vessel occlusion (4VO) and divided into 3d (3 d), 1 week (1 w), 2 weeks (2 w), 4 weeks (4 w) and 2 months (2 m) groups. Computer controlled initiative avoidance response shuttle box system was used to test the learning and memory capacity of rats. Immunohistochemical method was used to examine the expression of BDNF in CA1 region of the hippocampus and cortex of temporal and frontal lobe. Results Compared with those in control group, BDNF positive cells in CA1 region were decreased significantly in 4VO 1 w group ( P 0 05). This paper presents the results of the histochemical study on the activities ofATPase and succinic dehydrogenase and their distribution in mammary gland cancercell of mice by means of electron microscopy.It was found that the activity of ATPase activated by Mg~(++)were mainly locatedon the mitochondrial membrane,the nuclear membrane and the nucleolus,and that ofATPase activated by Ca~(++)were located on the cristae and matrix of mitochondria aswell as the outer nuclear membrane,and also evenly distributed in the chromatin.The activity of the succinic dehydrogenase is mainly located on the cristae andthe inner membrane of mitochondria.An alternitive responses of strong and weakactivities of the enzyme were shown between the outer and inner nuclear membranes.In the mean-while,the activity of the succinic dehydrogenase was also shown in ahigh level in the outer cytoplasmic membrane and the inner nuclear membrane.
7.Treating Metabolic Syndrome from Phlegm and Stasis

[Objective] To review the treatment of metabolic syndrome from phlegm and stasis, so as to provide effective ideas for the treatment of metabol-ic syndrome.[Method] Through searching for literatures related to Chinese medicine treatment of metabolic syndrome in the last decade, we can summarize the role of phlegm and blood stasis in the metabolic syndrome and its relationship with various components of metabolic syndrome. We can also research the application of the resolving the phlegm and removing blood stasis in the metabolic syndrome. [Results] Phlegm, blood stasis consistently run through the metabolic syndrome, and have a close relationship with each component of metabolic syndrome. The prescription of resolving the phlegm and remov-ing blood stasis can not only improve the clinical symptoms, but also can improve obesity-related indicators, reduce blood glucose, increase insulin sensitiv-ity, improve endothelial function and so on. [Conclusion] Treating metabolic syndrome from phlegm and stasis is safe and effective, which can reverse dia-betes, insulin resistance and prevent cardiovascular events through multi-channel, multi-level, multi-target. It has a broad application prospect and is worth further in-depth study.
8.Reduction of postradiation xerostomia in patients with head and neck cancer by superior technology using 3D-RTPS

Objective: To observe the effect of superior technology using three dimension radiation treatment plans (3D-RTPS) in the reduction of postradiation xerostomia in patients with head and neck cancer. Methods: 3D-RTPS of conventional radiation and new superior technique were applied in the patients of 3 kinds of head and neck cancer (tumor in middle, lateral, between middle and lateral area of head and neck). Superior technology including superior ray entrance, multi fields, conformal irradiation and application of wedges. Analysis of 3D dose distributions and dose volume histograms (DVH) was used to evaluate the dose and volume in tumor, parotids and spinal cord in the patients. Tumor dose of 55.6~68.6 Gy (average 58.8 Gy) was given to the patients in conventional radiation group in 8 patients, 53.5~68.5 Gy (average 58.3 Gy) to new radiation technique group in 12 patients. Whole saliva flow rate and subjective xerostomia scores were assessed in patients during radiation course and follow-up. Results: The dose in contralateral parotids in patients with tumor in lateral area, between lateral and middle area in conventional radiation and new radiation technique groups were 51.8~64.3 Gy and 0.4~1.4 Gy, respectively. The dose in parotids in patients with tumor in middle area in conventional radiation and new radiation technique groups were 51.8 and 24.7 Gy respectively. The rate of xerostomia at the end of radiotherapy and in 4~6 months follow-up after radiotherapy in patients of new radiation technique group were 8.3 % (1/12) and 0 respectively. Those in patients in conventional radiation group were 87.5 % (7/8) and 50 % (4/8) respectively. Conclusion: Superior technology using 3D-RTPS can reduce the rate of postradiation xerostomia in patients with head and neck cancer.

Objective] From the Chinese point of view of haze-induced lung injury pathogenesis, diagnosis and treatment, it provides theoretical support. [Method] To exogenous disease TCM theory, the nature of the haze of evils, a sense of evil ways and pathogenic characteristics, etc. based on the analysis of the haze of the etiology and pathogenesis of lung injury, the basic pathogenesis are analyzed and discussed. [Results] For haze pathogenic proposed basic principles should be taken to the prevention, prevention is greater than cure, the treatment should be Quxie, and to righten. Work using the method of elimination of pathogens through purgation and churesis.[Conclusion]For haze urgent treatment, prevention and treatment of traditional Chinese medicine for haze-induced lung injury has very important theoretical and practical value.
10.Isolation and identification of small cell lung cancer stem-like cells

Background and purpose:Small cell lung cancer(SCLC) is a highly aggressive malignancy with a 5-year survival rate of less than 10%.These features suggest the enrichment in cancer stem cells.Our study was aimed to establish a small cell lung cancer cell line from primary cultured from lung cancer tissue,and to identify and isolate cancer stem cells from the early generations of the established cell line.Methods:By using a serum-free medium,lung cancer primary cells were cultured from a small fresh lung cancer cell sample,and a small cell lung cancer cell line was established after passaging the cultured cells.Lung cancer stem cell markers were searched by using flow cytometry analysis to sort through the third or forth generation cells of the established cell line.Biological characteristics of lung cancer stem cells were studied by using the single cell clone formation test,plat colony formation test and cell sphere formation test.Results:A small cell lung cancer cell line was established by primarily culturing a fresh lung cancer sample.The cell line could easily passaged more than 25 generations.When the third or fourth generation cells of the established cell line were checked by flow cytometry,there was a small population of cells that were obviously with CD44 stronger positive(CD44++ cells,5.1%) than the main population cells,which were CD44 weak positive(CD44+ cells).CD44++ cells showed stronger colony formation ability than the CD44+ cells.Furthermore,only the CD44++ cells could form stem cells when a single cell was seeded in a well of 96 well plates.Most importantly,only the CD44++ cells could form cell spheres in ultra low attachment 96 well plates.These results indicated that the CD44++ cells enriched more cancer stem cells in the small cell lung cancer cell line.When CD44 and CD90 antibodies were co-stained,the cells of the established cell line could be separated into 4 populations,i.e.CD44+CD90-,CD44+ CD90+,CD44++ CD90+ and CD44++ CD90-cells.CD44+ CD90+ cells were the smallest population cells in the cell line,with a ratio of about 1.9%.When cultured in ultra low attachment 96 well plates,CD44++ CD90+ cells had the strongest cell sphere formation ability when compared with other population cells,indicating that CD90 might also be a small cell lung cancer stem cell marker.Conclusion:There were cancer stem-like cells in primary cultured small cell lung cancer cell lines,CD44 and CD90,which might mean that they could be lung cancer stem cell markers.
