1.Adrenomedullin and the endothelial cell functions in vitro

Objective To explore the effects of adrenomedullin(AM) on human umbilical vein endothelial cells (HUVECs) and possible mechanisms involved. Methods The HUVECs were selected to be a model and the following aspects were studied: (1) Effects of high glucose (30 mmol/L) on the endothelial dysfunction induced. (2) The role of protein kinase C (PKC)? and PKC? in the endothelial dysfunction induced by high glucose and the effects of AM. The translocation of PKC? or PKC? in a single HUVEC was observed by laser-scanning confocal microscope and the expressions analysis was conducted quantificationally by Western blotting. HUVECs were cultured and divided into 3 groups: (1) Matched control group; (2) High glucose group (30 mmol/L glucose); (3) AM (10~(-9), 10~(-8), 10~(-7) mol/L) + highglucosegroupHUVECswereincubated with AM for 48 h. Results (1) High glucose could induce HUVECs dysfunction: increased apoptosis, decreased NO concentration and increased sICAM level of HUVECs. AM reversed the above changes of HUVECs induced by high glucose. (2) It was observed that AM inhibited the translocation of PKC? from plasma to nucleus in HUVECs induced by high glucose and the translocation of PKC? from nucleus to plasma and membrane in HUVECs. (3) AM inhibited the increasing expressions of PKC? in HUVECs induced by high glucose. The expressions of PKC? in high glucose group were not different from that of the control. Conclusion AM appears to correct the endothelial cell dysfunctions induced by high glucose. The inhibition of PKC? and PKC? seems to play some roles during the process.

Objective To investigate the alteration of energy metabolism and oxidative injury in the myocardia suffering from lethal ventricular tachyarrhythmia (LVTA). Methods Two LVTA-SCD SD rat models, induced by aconitine injection or coronary artery ligation (CAL), respectively, were developed. Rats that died from over-anaesthesia or CAL-induced heart failure were served as their controls, respectively. Mitochondrial membrane potential (MMP), reactive oxygen species (ROS), malonaldehyde (MDA), phosphocreatine (PCr) in the ventricular myocardia, and serum troponin I (cTnI) were detected, and compared between LVTA rats and their controls. Results Fourteen ACO-LVTA and six CAL-LVTA rats were successfully developed. As compared to their controls, ACO-LVTA and CAL-LVTA rats had higher ROS and MDA content, and lower concentration of PCr in the ventricular myocardia. MDA content in ACO-LVTA group is signiifcantly higher than that of its control (P<0.05). MMP in myocardia of ACO-LVTA is lower than that of its control, but is higher than those of two CAL groups. Serum cTnI in rats of both LVTA models is higher than those of their controls and pre-treated control. Specially, serum cTnI in CAL-LVTA was signiifcantly higher than that of ACO-LVTA and its control (P<0.01). The myocardial ROS content is correlated with the duration of VT and VF (P<0.05), with correlation coefifcients being 0.44 and 0.46, respectively. Conclusions After LVTA, the ventricular myocardia had lower MMP and PCr content, higher concentration of ROS, MDA, as well as higher serum cTnI than their controls, indicative of oxidative injury and alteration of energy metabolism under LVTA-SCD.
3.Homological analysis of a foodborne disease outbreak caused by Salmonella typhimurium and Salmonella choleraesuis

Objective The homological analysis of a foodborne disease outbreak event that occurred on July 15, 2022, in Sanya was carried out to provide scientific basis for clinical treatment, incident tracing and prevention and control, and to explore the feasibility of pulsed field gel electrophoresis (PFGE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) in the homology analysis of Salmonella bacteria. Methods The traceability analysis of food poisoning incidents was accurately analyzed through the combination of traditional bacterial detection methods, PFGE, MALDI-TOF MS and the clinical symptom, epidemiological characteristics. Results In this outbreak, 14 cases of poisoning symptoms such as fever, nausea, and vomiting appeared, all of which had a history of dining in the same restaurant, with similar onset and clinical manifestations. Among them, one was the chef who made the fried rice in the restaurant. The pathogen culture and identification showed that it was caused by Salmonella typhimurium and Salmonella choleraesuis. Among the 14 cases of patients, 5 cases of Salmonella choleraesuis, 3 cases of Salmonella typhimurium, with the positive rate of 57.14% (8/14). One case of environment and two cases of food were detected. The result of PFGE showed nine PFGE types, 95.00% homology for 7 strains, and 94.00% homology for 4 strains. Conclusions The food poisoning was caused by eating egg fried rice containing Salmonella choleraesuis and pork intestine containing Salmonella typhimurium. In the future, , it is necessary to strengthen the risk monitoring of food contamination and harmful factors, to ensure food safety from the source of food supply, and to prevent the occurrence of foodborne infection events. At the same time, it is also crucial to standardize the health check of related personnel, and to conduct multi-channel education on the harm and prevention of foodborne diseases.

Osteoporotic thoracolumbar fracture in the elderly will seriously reduce their quality of life and life expectancy. For osteoporotic thoracolumbar fracture in the elderly, spinal reconstruction is necessary, which should comprehensively consider factors such as the physical condition, fracture type, clinical characteristics and osteoporosis degree. While there lacks relevant clinical norms or guidelines on selection of spinal reconstruction strategies. In order to standardize the concept of spinal reconstruction for osteoporotic thoracolumbar fracture in the elderly, based on the principles of scientificity, practicality and progressiveness, the authors formulated the Clinical guideline for spinal reconstruction of osteoporotic thoracolumbar fracture in elderly patients ( version 2022), in which suggestions based on evidence of evidence-based medicine were put forward upon 10 important issues related to the fracture classification, non-operative treatment strategies and surgical treatment strategies in spinal reconstruction after osteoporosis thoracolumbar fracture in the elderly, hoping to provide a reference for clinical treatment.
