1.Protective effect of pyrrolidine dithiocarbamate pretreatment on lung against LPS-induced injury in rats

Objective To investigate the protective effect of pyrrolidine dithiocarbamate (PDTC) pretreatment on lung agaillst LPS-indueed injury in rats.Methods Fifty-four Wistar rats of both sexes weighing 180-220 g were randomly idivided into 3 groups:group I normal control(group C,n=6);group Ⅱ LPS (group L,n=24)and group Ⅲ PDTC(group P,n=24).Endotoxemia was induced by intraperitoneal LPS 8 mg/kg in group Ⅱ and Ⅲ.In group Ⅲ PDTC 120 mg/kg was injected iv at 30 min before IP LPS.Venous blood samples were collected at 1,3,6 and 12 h after LPS administration in group L and P and after normal saline administration in group C for determination of serum IL-6 concentration by ELISA.Six animals were killed at each time point after blood sample collection.The lungs were removed for microscopic examination and determination of W/D lung weight ratio,HSP70 expression and NF-κB activity.Results Intraperitoneal LPS significantly increased serum IL-6 concentration and W/D lung weight ratio,HSP70 expression and NF-κB activity in group Ⅱ.PDTC pretreatment significantly attenuated LPS-induced increase in serum IL-6 concentration,W/D lung weight ratio andNF-κB activity.The lung histologic dalnage induced by LPS Wag also ameliorated by pretreatment with PDTC.Conclusion Pyrrolidine dithiocarbamate pretreatment Can protect lung against LPS-induced injury through inhibition of activation of lung tissue NF-κB and up-regulation of expression of HSP70.
2.The reference range of copper,iron,zinc,calcium and magnesium in peripheral blood of preschool children in Suzhou

Objective Accodring to the guideline of Reference Intervals in the Clinical Laboratory(ISO15189) ,to establish the reference range of Cu ,Fe ,Zn ,Ca and Mg in the peripheral blood of preschool children in Suzhou .Methods Using atomic absorp-tion spectrometry to detect the levels of Cu ,Fe ,Zn ,Ca and Mg in peripheral blood of 3 992 Kindergarden children in Suzhou from Apr to May 2014 ,according the age ,the children were divided into three groups ,1 511 cases in 3 to 4 years old group including 750 boys ,761 girls ,1 169 cases in 4 to 5 years old group including 570 boys ,599 girls ,1 312 cases in 5 to 6 years old group inclu-ding 604 boys 708 girls .According to NCCLS's guideline C28 -A2 to define the reference range .Results The concentration of copper ,iron ,zinc ,calcium ,magnesium had no significant differences in different gender and age groups(P>0 .05) ,the reference ranges in peripheral blood of the preschool children in Suzhou were Cu 11 .47-32 .53 umol/L ,Fe 6 .86-9 .80 mmol/L ,Zn 64 .46-133 .02 umol/L ,Ca 1 .47-2 .09 mmol/L ,Mg 1 .21 -1 .75 mmol/L .Conclusion There are regional difference on the levels of Cu ,Fe ,Zn ,Ca ,Mg in children's peripheral blood ,each region should to establish the local reference range follow the guideline .
3.NUTRITIONAL EVALUATION OF POWDERED GOAT'S MILK PROTEIN

This study is conducted to evaluate the protein quality of the spray-dried goat's milk powder made from milk of Qingdao's Laoshan dairy goats. Chemical analysis and rat experiment are included in the study with spray-dried powdered cow's milk and isolated soy protein as comparative substances and chemical pure casein as reference protein.The content of crude protein in the powdered goat's milk is 26.01% and the protein's limitting amino acid is sulphur containing amino acids with the AAS and CS being 0.91 and 0.52 respectively, similar to those of cow's milk protein. The percentage of EAA and the EAA index are 49.1% and 96.96%, as high as those of cow's milk. The results of the rat experiment designed according to AOAC showed that the PER, NPR, AD, TD, BV, NPU of the protein are 3.01, 4.62, 90.49%, 96.75%, 86.02 and 83.22% respectively. Except that the AD of goat's milk protein is slightly higher than that of cow's milk protein, there is no significant difference between two milk proteins on other nutritional parameters. The present study gives the conclusion that the protein of powdered goat's milk is at least as nutritious as that of powdered cow's milk, but more nutritious than chemical pure casein and far more nutritious than isolated soy protein.
4.Ghrelin expression in the tissues of different thyroid diseases

Objective:To investigate whether ghrelin, a novel endogenous ligand of growth hormone secretagogue receptor ( GHS-R) , was expressed in the thyroid tissues of different thyroid diseases, and its implication. Methods:The paraffin-embedded specimens of thyroid tissues from 2000 to 2004 were obtained from 57 patients with different thyroid diseases, including 5 subacute thyroiditis, 8 Hashimoto' s thyroiditis, 7 hyperthyroidism (Graves disease) , 8 nodular goiter, 5 thyroid adenoma, 3 thyroid lympho-ma, 8 papillary carcinoma, 3 follicular carcinoma, 5 medullary carcinoma and 5 undifferentiated carcinoma cases. The specimens of normal peri-adenoma thyroid tissues served as controls. Immunohistochemi-cal staining was used to detect ghrelin expression. Results:(1) ghrelin expression was undetectable in the thyroid tissues of normal control, subacute thyroiditis, Hashimoto' s thyroiditis and Graves disease. (2) ghrelin expression was also undetected in the tissues of nodular goiter, thyroid adenoma and thyroid lymphoma. (3 ) ghrelin-positive staining was found in the tumor cells of different types of thyroid carcin-moma. Five cases were positive within 8 cases in papillary carcinoma, 2 cases were positive within 3 cases in follicular carcinoma, 3 cases were positive within 5 cases in medullary carcinoma, 3 cases were positive within 5 cases in undifferentiated carcinoma. Conclusion:Ghrelin is expressed in malignant epithelial thyroid neoplasmas, but not in autoimmune or inflammatory thyroid diseases and benign nodular thyroid diseases. The results indicate that ghrelin expression may play an important role in the occurrence and development of thyroid carcinoma.

6.Experimental Study on Preventive and Therapeutic Effect of Chinese Traditional Compound Medicine to Subclinical Hepatic Encephalopathy in Liver Cirrhotic Rats

Objective To study the preventive and therapeutic effects and the mechanism of Chinese traditional compound medicine to subcilincal hepatic encephalopathy(SHE)induced by endotoxin in liver cirrhotic rats.Methods The liver cirrhosis model of rats were made in subcutaneous injection of 40% carbon tetrachloride,the SHE model was made by infused with endotoxin into the tail vein.Treatment group was given low dose or moderate dose or high dose of Chinese traditional compound medicine,Control group was treated with lactulose.The morbidity of SHE,hepatic function,blood ammonium(BA),endotoxin,liver tissue pathology were observed.Results Above indexes were improved by every group of Chinese traditional compound medicine.The curative effect of moderate and high group of Chinese traditional compound medicine to the hepatic function,BA,endotoxin,liver tissue pathology were better than that of lactulose.Conclusions The effects of the Chinese traditional compound medicines(compose of Polygonum cuspidatum,raw hawthornfruit,seed of Zizyphus jujuba Mill,Mongolian Dandelion Herb and Rhizoma Acoori Graminei)in SHE prevention and treatment is significant.Its mechanism might be as follows:① recovering liver function,decreasing inflammatory changes of liver tissue;② preventing the absorption of BA,lowering the content of BA;③ reducing the sensitivity of the body to endotoxin.
7.Other Effects of Cimetidine in the Decompensated Stage of Chronic Cardiopulmonary Disease

OBJECTIVE:To observe the other effects of cimetidine in the decompensated stage of chronic cardiopulmonary disease. METHODS: 72 patients with uncompensated chronic cardiopulmonary disease were assigned into therapy group and control group (n=36). Both groups were given routine treatment. Therapy group were additionally treated with cimetidine intravenously for 48 h. The changes of respiration frequency, arterial partial pressure of oxygen (PaO2), arterial partial pressure of carbon dioxide (PaCO2), intake of staple food and electrolytes in blood serum of 2 groups before and after treatment were determined. RESULTS:In therapy group, the respiration frequency, the level of PaCO2 and disorder of electrolytes in blood serum were decreased and the level of PaO2 and intake of staple food were increased. 9.Diagnosis and treatment of accessory breast cancer in 16 cases

Objective To investigate the clinicopathological characteristics,diagnosis and therapy,as well as the prognosis of accessory breast cancer. Methods Twenty-two cases were diagnosed as accessory breast cancer from Jan 1,1984 to Dec 31,2008, their clinicopatholgical data were analyzed retrospectively.Results Up to Mar. 1, 2010,2 cases had local recurrence;7 cases had long-distance metastasis;6 cases died. In the current study,the 5-year survival rate of accessory breast cancer was 43. 7%. Conclusions Accessory breast cancer is aggressive. The diagnosis was mainly based on clinical characteristics and postoperative pathology. The combined therapies may improve the survival rate.
10.An experimental study on distribution of musk into the brain through blood brain barrier

OBJECTIVE: To study the possible pathway of the effect of musk on brain disorder, distributing into the brain through blood brain barrier. METHODS: We used the musk ketone (muscone), a main composition of musk, to inject through the tail vein of the rats into the blood and took the brain and other organs at different times to make samples. Then gas chromatography was used to measure the distribution of muscone in the brain and other organs. RESULTS: Muscone could pass through the normal rat's blood brain barrier into the brain and soon reached the highest peak and remained in higher concentration, and more slowly metabolized as compared with other organs. CONCLUSION: Musk distributing into the brain through blood brain barrier provides the basis for its effect in treating brain disorders. Chromatography is an effective method to study the active composition of Chinese herbal medicine distributing through the blood brain barrier into the brain.
