1.The inhibition of intrinsic expression of GFP in nasopharyngeal carcinoma by RNA interferencing

Objective:To construct a nasopharyngeal carcinoma cell strain expressing GFP stably and observe the inhibition effect and cytotoxicity of short hairpin RNA targeting the intrinsic GFP.Methods:HNE1 cells transfected with pEGFP-N1 were followed by the screening with G418 to obtain the stable nasopharyngeal carcinoma cell strain expressing GFP(HNE1-GFP);shGFP,a vector expressing short hairpin RNA targeting GFP,was transfected into HNE1-GFP with various levels.The fluorescence was observed by fluorescent microscope.The inhibition ratios and cytotoxicity of shRNA were analyzed by software and MTT,respectively;The protein and mRNA level of GFP were detected by Western Blot and RT-PCR,respectively.Results:The GFP gene was expressed stably within HNE1-GFP cell line;shGFP suppressed the intrinsic expression of GFP with the ratios of 49.7%,84.8% and 86.7%,respectively,without significant cytotoxicity.Conclusion:A nasopharyngeal carcinoma cell strain expressing GFP stably is established successfully;The inhibition effect of RNA interference is demonstrated in nasopharyngeal carcinoma cells and it is characterized by the significant concentration dependence and rare cytotoxicity.
2.Two methods for air disinfection in neurosurgical intensive care unit

Objective To compare the effects of Chinese herbal fumigation and ultraviolet light exposure on air disinfection in neurosurgical intensive care unit.Methods The floors and the tables in the rooms of ICU were all disinfected using 0.02%84 disinfectant.The air disinfection was performed in the rooms using Chinese herbal(elsholtziae,ramulus cinnamomi,forsythia suspense,flos caryophylli)fumigation as the experiment group and those using ultraviolet light exposure as the control group,both for 30 times.The two disinfection methods were compared before air disinfection,at hours 0,1,2,4,8 and 12 in terms of bacteria tests.Results The air quality on natural bacteria in both groups met the standard of Class Ⅱ Environmental Health Criteria at hour 0,l after air disinfection,without significant difference between the two groups at each time point(P>0.05).In the control group,the bacteria began to grow in number one hour after ultraviolet light exposure and overgrew two hours after air disinfection.In the experiment group,the bacteria grew less,never exceeding the standard level at all time points.The disinfection effect of the experimental group was all significantly better than that of the control group at the time points of 2 h,4 h,8 h,12 h(P<0.05 or P<0.01). Conclusions Chinese herbal fumigation is superior to the UV light irradiation disinfection.It is safe and harmless even in case of medical staff working in the unit.
3.Genotype and Quantitative Expression of Type ⅠComplement Receptor on Erythrocytes from the Patients with Systemic Lupus Erythematosus

Objective Through studing the genotype and quantitatively analysing expression of type Ⅰ complement receptor(CR1) on erythrocytes from the patients with systemic lupus erythematosus(SLE). To investigate the relationship between CR1 and SLE. Methods Polymerase chain reaction(PCR) and Hind Ⅲ restriction enzyme digestion were used to analyse the genotype of CR1, and the quantitative expression of CR1 was demonstrated by FACscan flow cytometry. Results Compared with normal controls, high expression genotype of CR1 was significantly decreased, but low expression genotype was significantly increased in SLE patients. The number of CR1 on erythrocytes in SLE patients was significantly lower than that in normal controls. There was a negative relationship between the expression of CR1 and disease activity index of SLE (SLEDAI). Conclusion The expression of CR1 on erythrocytes might be related to the development of SLE, and it is helpful to detect the CR1 on erythrocytes for the determination of SLE patient′s condition.
4.Functioning and Exercise Rehabilitation Strategies of Obesity Based on ICF-Core Set of Obesity

This paper analyzed the functioning and characteristic of body function, body structure, activities and participation and environments based on the theories and methods of International Classification of Functioning, Disability and Health (ICF). It was concluded that environment was the main reason of the development of obesity, body structure was an objective indicator of determine of obesity and also the degree of obesity's intervention, physical function changes reflected the hazards and their severity, and activity and participation was an out performance of obesity's difficulties or obstacles. Based on these conclusions, several corresponding sports rehabilitation strategies were proposed.
5.Impedance characteristics of ear acupoints in identifying excess or deficiency syndrome of stroke.

Objective: To explore the impedance characteristics of ear acupoints in stroke patients with excess or deficiency syndrome, and to provide basis data for objective study of the syndromes of stroke. Methods: The data of electrical characteristics of ear acupoints in stroke patients and healthy people were collected, and excess syndrome and deficiency syndrome of stroke were identified by quantifying the syndromes of stroke using scales. The differences in impedance characteristics of ear acupoints between stroke patients and healthy people were analyzed, and the differences in impedance characteristics of ear acupoints between stroke patients with excess syndrome and stroke patients with deficiency syndrome were analyzed too. The correlation among impedance characteristics of ear acupoints, stroke and the syndromes was also analyzed. Results: There were significant differences in impedance characteristics of ear acupoints between stroke patients and healthy people (P<0.05,P<0.01). The ear acupoints CO12 (Gan) and CO13 (Pi) had a significant role in diagnosing stroke as compared with CO18 (Neifenmi), AT3.4.AH12i (Naogan), CO10 (Shen), TG2p (Shenshangxian), AH6a (Jiaogan), AT4 (Pizhixia), and CO15 (Xin). There were significant differences in impedance characteristics of ear acupoints between stroke patients with excess syndrome and stroke patients with deficiency syndrome (P<0.05, P<0.01). The ear acupoints AH6a (Jiaogan) and CO10 (Shen) played an important role in differentiation diagnosis of excess syndrome and deficiency syndrome of stroke, followed by CO18 (Neifenmi), TF4 (Shenmen) and TG2p (Shenshangxian). Conclusion: Some ear acupoints with diagnostic value for stroke may provide basis of objective research for stroke diagnosis as well as identifying excess syndrome and deficiency syndrome of stroke.
6.Treatment and outcome of polypropylene mesh or tape related pain after reconstructive pelvic surgery

Objective To investigate clinical treatment and outcome of mesh-related pain after pelvic floor reconstruction. Methods Twelve patients were referred to Peking University People′s Hospital for post-operation pain related to polypropylene mesh or tape used in pelvic floor reconstruction and received reoperation from January 2007 to December 2014. The demographic characteristics, clinical manifestation, operation method and follow-up outcome were retrospectively analyzed, and current literature was also reviewed. Results (1)General information:the median age was 54 years old(range 43-74 years old). The median vaginal delivery times was 2 (range 1-5). Initial pelvic floor operation procedures included 4 cases vaginal mesh, 5 cases vaginal mesh plus tension-free vaginal tape (TVT), 1 case rectal surface mesh implantation, and 2 cases sacrocolpopexy. (2) Related complications:all 12 patients had pain-related post-operation symptoms including dyspareunia, perineal pain, urination and defecation difficulty. Seven patients also complicated with other symptoms including discharge and bleeding. Mean time from first operation to medical consultation was 30 months (range 1-72 months). (3) Reoperation results:all 12 patients received reoperation on mesh revision or tape, 10 cases of them reoperated by vaginal approach, while the other 2 cases by laparoscopy. The range of the mesh or tape depended on tender or firm tissue and scar by palpation. The visual analogue scale (VAS) was significantly improved, which was 7.1 ± 2.2 pre-operation, and 0.9 ± 1.9 post-operation (95%CI: 4.48-7.86, P<0.01). Conclusions Pain is one of the reason why patients was removal of the mesh or tape after pelvic floor reconstruction operation. The surgery procedure may be considered as partial or entire mesh or tape excision by vaginal or abdominal way. Mesh or tape removal surgery could relieve related pain and other complications, and the VAS score was significantly improved after surgical management.
7.Application and Prospect of Multivariate Statistical Analysis in TCM Syndrome Classification

Standardization, objectification, and normalization of TCM syndromes are emphasis, difficulty, and hot issue of inheritance and development of TCM.With the technological development of computer and extensive application of multi-disciplinary integration, research methods for the identification of TCM syndrome classification emerge in endlessly. This article analyzed the application status of multivariate statistical analysis in the identification of TCM symdrome classification, summarized application status and existing problems of cluster analysis, PCA and factor analysis, discriminant analysis and Logistic regression analysis, and structure equation model in the identification of TCM symdrome classification, which provided references for further studies on identification methods of TCM symdrome classification.
8.Comparative Study on Cun-Kou Six-pulse Diagram between Chronic Gastritis and Bronchial Asthma

This study was aimed to discuss the difference of Cun-Kou six-pulse (CKSP) diagram of different diseases. The ZBOX-I Type Pulse Digital Analyzer was used to collect CKSP diagram of 30 bronchial asthma cases and 33 chronic gastritis cases. Time-domain parameter was extracted for analysis. Comparison was made by statistical methods. The results showed that there was statistical difference on time-domain parameter of pulse diagram for different diseases (P<0.05). There was statistical difference on time-domain parameter of pulse diagram between the left side and right side (P<0.05). There were statistical differences on t4 of left Cun, t4 and h5 of right Cun, t1\t4 of left Chi between bronchial asthma and chronic gastritis cases (P<0.05). It was concluded that the single-cycle pulse diagram of chronic gastritis was wider but no higher than that of bronchial asthma. The systole phase of single-cycle pulse diagram of bronchial asthma occupied greater proportion than that of chronic gastritis. There was difference on Cun of time-domain parameter in pulse diagram between bronchial asthma and chronic gastritis. It was consistent with the theory of Cun-Kou sub-designated zang-fu organs, which was “left Cun designates heart, right Cun designates lung.” However, there was no further explanation on functional differences between the spleen and stomach. The mechanism of pulse difference on the left and right side still required further illustration.
9.Immunohistochemical analysis of OTUB1 in 90 cases of ovarian mucinous tumors

Background and purpose:The ovarian mucinous tumor is one of the major subtypes of the ovari-an epithelial cancer. Ubiquitination is one of the main post-translational modifications, which has proven to be involved in tumorigenicity. Deubiquitinase is the protein enzyme that could reverse the process of ubiquitination to affect the initiation and progression of malignancies. This study aimed to analyze the expression and clinical application of deubiquitinase OTUB1 (OTU deubiquitinase, ubiquitin aldehyde binding 1) in ovarian primary mucinous tumors. Methods:This study collected 90 cases of ovarian primary mucinous tumors during 2010-2015 in Obstetrics and Gynecology Hospital of Fudan University, and then collected the clinicopathological information and performed the immunochemistry.Results:Fourteen out of 90 cases were ovarian primary mucinous cystadenoma, 17 were borderline mucinous tumor (intestinal type and intraepithelial carcinoma), and 59 were ovarian primary mucinous carcinoma. The expression rate and intensity of OTUB1 were much higher in malignant cases than those in benign ones (P<0.05). The expression rate and intensity of OTUB1 were much higher in cases with mucinous intraepithelial carcinoma than those in cases with intestinal type borderline mucinous tumor (P<0.05). The expression rate and intensity of OTUB1 increased with the advance of FIGO staging (P<0.05). The expression rate and intensity of OTUB1 were much higher in cases with involved fallopian tubes than those in cases without involved fallopian tubes (P<0.05). The expression rate and intensity of OTUB1were much higher in cases with involved uterus and omentum than those in cases with-out involvement (P<0.05). The expression rate and intensity of OTUB1were much higher in cases with lymph node metastasis than those in cases without involvement (P<0.05).Conclusion:There is significant difference in OTUB1 expression between ovarian primary mucinous carcinoma and benign mucinous cystadenoma. It is highly correlated to FIGO staging and invasion and metastasis of tumor. OTUB1 could be used in differential diagnosis and in monitoring the tumor initiation and progression in ovarian mucinous carcinoma.
10.Efficacy of gemcetabine and LY294002 on the growth of human pancreatic cancer cell lines BxPc-3 and MiaPaCa-2

Objective To investigate the effects of gemcetabine and LY294002 monotherapy or combination on the proliferation and poptosis of pancreatic cancer cell lines BxPc-3 and MiaPaCa-2.Methods Cell proliferation and poptosis were detected by MTT and Annexin V-FTTC,respectively.Results Both gemcetabine and LY294002 could inhabit the proliferation of the two cell lines.Their inhibitory effects were increased accompanied with increased drug concentrations and the cell survival rates was negatively correlated with logarithmic of the drug concentrations (r<-0.95,P<0.01).The inhibitory effects of gemcetabine and LY294002 to the BxPc-3 proliferation were significantly stronger than to the MiaPaCa-2(P<0.05).For BxPc-3 and MiaPaCa-2,the IC50 of gemcetabine were(10.07±1.83),(36.45±2.71)μmol/L(P<0.05),and the IC50 of LY294002 were(7.84±1.48),(17.89±1.98)μmol/L(P<0.05),respectively.Gemcetabine and LY294002 could induce cell apoptosis(P<0.01).Though both the concurrent or consecutive use of these two drugs could promote cell apoptosis,the effect of the concurrent group was significantly stronger(P<0.05).The order of these two drugs in the concurrent group had no significant influence on their effects(P>0.05).Conclusion Both gemcetabine and LY294002 could inhibit the proliferation of pancreatic cancer cell lines.Their concurrent application shows a significant inhibitory effect on the cell apoptosis.
