1.Expression and localization of Kuzbanian-DN mRNA in trans-gene NOD mouse pancreas cells

Objective Kuzbanian (KUZ) is the functional homologue of the vertebrate ADAM10. ADAMs are a protein family of cell surface-anchored membrane, whose members have extensive biological activities. This paper was to explore KUZ-DN (the dominant negative construct of KUZ) mRNA expression and localization in the trans-gene NOD mouse pancreas tissue and to provide molecular basis for the mechanism of the gene intervened stem cell differentiation to treat diabetes. Methods Pancreas was frozen and paraffin sections of pancreas in eight trans-gene NOD mice were made. Non-radioactivity in situ hybridization experiment was conducted on the tissue sections by the cRNA probes of KUZ-DN (360 bp) sense and antisense with DIG labeled. Results There was the strong positive KUZ-DN mRNA signal report of in situ hybridization in the trans-gene NOD mouse pancreas cells. Sense chain probes and blank comparison sections were negative. Conclusion The positive expression and localization of KUZ-DN mRNA in the trans-gene NOD mouse pancreas cells provides a vital data for the gene therapy research project of the KUZ-DN intervened Notch/Delta signaling pathway by the stem cell differentiation to treat diabetes
2.RCR AND ADP/O RATIO CHANGES OF RAT LIVER MITOCHONDRIA DURING EARLY PHASE OF BURN INJURY

Using succinate as a substrate,the respiratory control ratio (RCR) of liver mito-chondria of 20 burned Sprague-Dawley rats (male, 20%of burned body surface area, III degree burn), and 32 burned Wistar rats (male), increased 30 minutes and 2 hours after burn, and ADP/O ratio increased 39 minutes after burn. But ADP/O ratio decreased and RCR was nearly normal 4 hours after burn.The observations suggest that there is an increasing mitochondria coupling during the early phase of burn injury, which has not been reported.
3.FUNCTIONAL CHANGES IN RAT LIVER MITOCHONDRIA DURING THE EARLY PHASE OF BURN INJURY

Using succinate as a substrate, the respiratory control rates (RCR's) of liver mitochondria of burned Sprague-Dawley rats (male, 20 percent of burned body surface area,III degree burn) were increased 15,30, 45, and 60 min after burn, the peak being at 30 min after burn (the sham group as control).In comparison with the sham group, the specific activity of succinic dehydro-genase was slightly decreased and the activities of cytochrome b and c + c1 were decreased significantly 30 min after burn.
4.Binding of Purine Nucleotides to Brown Adipose Tissue Uncoupling Protein from Different Kinds of Animals

Using brown adipose tissue submitochondrial particles from different kinds of animals, binding of uncoupling protein to different purine nucleotides was estimated. The results indicate that the dissociate constants (Kds) of uncoupling protein from hamsters to purine nucleotides are more than the Kds of uncoupling protein from guinea pigs, which are more than the Kds of that from rats, presumably because of different amino acid residue sequences of uncoupling protein polypeptide from different kinds of animals.
5.Effect of pH on Binding of Purine Nudeotides to Uncoupling Protein in Subnitochondrial Parti-cles from Rat Brown Adipose Tissue

Using submitochondrial particles, from rat brown adipose tissue mitochondria treated by Lubrol and 3 mol/L urea, as a model, the relationship between different pH and pKd of binding of purine nucleotides to uncoupling protein was estimated. When it was shown as plots of pKd against pH, it was found that the pKd peak of binding of GDP or ADP to uncoupling protein was about pH 5.5. The greater pH deviated, the less pKd became. The highest pKd of binding of GTP or ATP to uncoupling protein was also near pH 5.5, below which pKd remained constant, but above which pKd dropped rapidly. At a certain pH, pKd of binding of GDP or GTP to uncoupling protein was higher than that of binding of ADP or ATP, respectively.
6.Research of Tongue Image Analysis Instrument Based on Image Analysis

Objective To develop a kind of tongue image analysis instrument which is used to deal with the deviation problem on the process of tongue image analysis currently. Methods The system included two parts:hardware and software. The hardware was made up of special tongue image acquisition device and computer. With the application of digital image analysis technology, the instrument implements functions such as tongue image collection, image preprocess, image segmentation and feature analysis. Results After a certain amount of sample study process, the accuracy of tongue shape recognition was about 90%. Conclusion This instrument can achieve the aim of tongue information objectification and are widely used for clinical medicine and teaching.
7.Detection of extended-spectrum β-lactamases and antimicrobial resistance of Escherichia coli isolated from different clinical specimens of a county hospital in 2012-2014

Objective To analyze the distribution and antimicrobial resistance of Escherichia coli (E.coli)causing infection in a county hospital,and provide basis for clinical treatment of E.coli infection.Methods From 2012 to 2014,all kinds of clinical specimens in a hospital were performed bacterial culture,identification,and antimicrobial susceptibility testing;extended-spectrum β-lactamases (ESBLs )were initially screened by broth microdilution method,and confirmed by disc diffusion method;data were statistically analyzed by WHONET 5.6 and SPSS 19.0 software.Results 271 isolates of E.coli were detected,and mainly isolated from midstream urine (26.94%).The detection rate of ESBLs-producing E.coli was 49.82%,most were isolated from sputum specimens (56.52%), but there was no significant difference in detection rates of ESBLs-producing E.coli isolated from different speci-mens (P >0.05).Antimicrobial susceptibility testing results showed that the resistance rate of E.coli to penicillins was the highest (> 90%),to cefalotin and cefuroxime were both > 75%,susceptibility rates to amikacin and piperacillin/ tazobactam were both high (resistance rates < 10%),carbapenem-resistant strain was not found, resistance rates of ESBLs-producing strains to most antimicrobial agents were significantly higher than non-ESBLs-producing strains.Conclusion E.coli is mainly isolated from midstream urine specimens,antimicrobial resistance of ESBL-producing strains is more serious.
8.Comparison of three methods for evaluation of wrinkles

Objective Wriokle is the sign of skin aging. In order to provide a method for the further study of skin aging, we established some methods for the evaluation of wrinkles. Methods Three methods were used to evaluate wrinkles. 481 Chinese women with wrinkles on their faces were assessed and analyzed by clinical grading, Photoshop and Visionscan meter. We tried to check out the correlation between them. 481 pictures were divided into nine ranks. By using the softwares, we analyzed pictures again. And we calculated the correlation of all the parameters. Results There was good linear relationship between clinical grade and median, and also between values of LEVEL and SD by Photoshop histognam analysis. By Visionscan meter, there was good linear relationship between clinical grade and texture parameters, and also between SELS parameters. Conclusions All the three methods can reflect the skin aging in some degree. In clinical practice, different method or parameter will be chosen according to the different instance.

10.An analysis on epidemic trend of HIV/AIDS and sexual behavior characteristics among MSM from 2008 to 2012 in Yinchuan city

Objective To analyzes the epidemic trend and sexual behavior changes among men who have sex with men (MSM ) in Yinchuan city so as to provide scientific evidence for the future development of prevention and control strategies .Methods A cross-sectional survey was conducted and data about the sexual behavior as well as trend of HIV epidemic among MSM from 2008 to 2012 were collected and analyzed .Results A total of 1 644 MSM were recruited from 2008 to 2012 .The MSM condom use rate in last time homosexual sex fluctuated between 73 .5% -80 .2% ,and their condom use rate in every time was increasing significant-ly(P<0 .05) from 54 .3% in 2012 to 44 .0% in 2008 .The HIV prevalence and 90% CI was 2 .6% (1 .1-4 .0) ,4 .5% (2 .5-7 .5) , 5 .7% (6 .6-7 .8) ,3 .8% (2 .2-5 .3) and 7 .0% (4 .9 -9 .1) respectively .The HIV prevalence was increasing significantly (P<0 . 01) from 7 .0% in 2012 to 2 .6% in 2008 .Syphilis prevalence was increasing significantly (P<0 .01) from 9 .0% in 2009 to 2 .9%in 2008 ,then decreasing significantly (P<0 .01) from 6 .3% in 2012 to 13 .6% in 2010 .Conclusion The HIV infection rate have a rising trend among MSM in Yinchuan city .More effective intervention measures should be taken to promote safe sex and curb the AIDS epidemic spread among MSM .
