1.Study on design and three-dimension structure of a broad-spectrum inhibitory peptide against staphylococcal enterotoxins superantigen

Objective:Peptides were designed on the basis of high conservative regions of amino acid sequences and structures of the SEs,three-dimension structure of P72 was constructed.Methods: Bioinformatics analysis softwares such as Vector NTI 10.3, InsightII 2000,Discovery Studio 1.7 were used to analyse and predict the space structure of P72.Results:three-dimensional domains of the peptide P72 from SEA, SEB and SEC were quite similar, Peptide P72 was far away from TCRVβchain and MHC class II molecule.Conclusion:The inhibitory activity of peptide P72 may not due to binding to MHCⅡ and TCRVβchain.The exact mechanism of inhibitory activity of P72 should be explored.

3.Photochemical effect of MB on the inactivation of E. coli in blood and biologic functions of blood cells

Objective To observe the photochemical effect of methylene blue (MB) on the inactivation of E. coli in blood and on biological functions of blood cells. Methods The blood was treated with 15 ?mol/L MB, 40 000 Lux halogen lamp irradiation and 0.35 mmol/l rutin for 10~40 min. The photochemical effect of MB on the inactivation of E.coli in blood was observed. The changes of biologic functions of blood cells were measured by means of RBC membrane binding to poly L lysine, reaction promoted by ATP enzyme and neutrophil phagocytosis tests. Results After treatment for 40 min, the E.coli count in blood decreased by 5 log units. The value of T/E of MB and irradiation of halogen lamp was higher than 1. No obvious changes of the morphology of blood cell were found. The negative electrical charges on membrane of RBC decreased by 11.23%~25.49%. After treatment of the blood for 20~30 min, the activity of Na + K +ATP enzyme of RBC membrane increased by 7.60%~20.07%. When the blood had been treated for 10~20 min, the positive rate of neutrophil phagocytosis enhanced by 9.00%. Conclusion MB photochemical treatment can effectively inactivate E. coli in blood. Under morphology specific condition of effectively inactivated blood bacteria, the shapes and functions of blood cells are not altered, but the RBC adhesion may increase by about 11.23%~25.49%.
4.Retrospective analysis on 57 cases of malignant tumor complicating herpes zoster

Objective To analyze the clinical characteristics of malignant tumor complicating herpes zoster.Methods Fifty-seven cases of malignant tumor complicating herpes zoster in our hospital from September 2003 to November 2014 were performed the retrospective analysis on the anti-tumor treatment history and the clinical characteristics of herpes zoster.Results Most of the cases were over 60 years old.Herpes zoster had the high incidence rate within 2 years of operation and radiochemotherapy.The on-set sites of herpes zoster were most commonly in the chest and back.Conclusion The factors of advanced age,surgery,radiochemo-therapy,poor nutritional status,etc.aggravate or promote the occurrence of herpes zoster in the patients with malignant tumor.
5.Effect of chemotherapy on function of isletβcells in patients with breast cancer

Objective To study the effect of chemotherapy on function of islet βcells in patients with breast cancer . Methods One hundred and fifty-six patients with breast cancer who had not received chemotherapy were enrolled from Sep 2012 to Aug 2013 at Chongqing Cancer Institute .One hundred and twenty-three cases completed 2 cycles of chemotherapy;one hundred and twelve cases completed 4 cycles;and, eighty-six cases completed 8 cycles.Height, weight, waist circum-ference(WC), hip circumference(HC) and blood pressure of each patient were measured .Waist-high ratio calculation (WHtR),waist-hip ratio(WHR) and body mass index(BMI) were calculated.All subjects underwent 75 g oral glucose tolerance test(OGTT)and insulin release test before chemotherapy,2 cycles,4 cycles and 8 cycles of chemotherapy, respec-tively.Besides, homeostasis model assessment of insulin resistance index ( HOMA-IR), Matsuda index and pancreatic βcells HOME(HOME-β) were calculated.Results Fasting blood glucose before chemotherapy and after 2 cycles, 4 cycles and 8 cycles of chemotherapy was (4.84 ±0.56), (4.78 ±0.61), (5.02 ±0.56) and (5.96 ±0.52) mmol/L, respec-tively.After therapy, the change in fasting blood glucose after 2 cycles and 4 cycles of chemotherapy was not statistically significant(P>0.05), but was significantly higher after 8 cycles than before therapy(P<0.05).2 h blood glucose before chemotherapy and after 2 cycles, 4 cycles and 8 cycles of chemotherapy was (5.43 ±0.50), (5.66 ±0.58), (8.30 ± 0.66) and (9.65 ±0.80) mmol/L, respectively.With chemotherapy treatment increased , the HOME-IR increased gradu-ally and the sharpest one was after 8 cycles, Matsuda index decreased gradually and the biggest drop was after 4 cycles,and HOME-βdecreased gradually after a transient increase .Conclusion Chemotherapy for breast cancer patients may lead to insulin resistance , pancreatic βcell function decline and the emergence of abnormal glucose tolerance .Four cycles of chemotherapy shoucd be followed by hypoglycemic drugs .
6.Effect of shRNA interfering BAMBI on malignant biological behaviors of human colon cancer SW480 cell

Objective: To explore the effect of shRNA interfering BAMBI (bone morphogenetic protein and activin membrane bound inhibitor) on the proliferation, apoptosis, invasion and migration of human colon cancer SW480 cells and the possible mechanisms. Methods: After successful transfection with sh-BAMBI in SW480 cells, the mRNA and protein epxressions of BAMBI were detected by qRT-PCR and Western blotting, respectively. Cell proliferation was measured by MTT; apoptosis was tested by Hoechst33258 staining; cell invasion was detected by transwell assay; and cell migration was measured by wound healing assay. The expressions of TGF-β/ Smad/2 signaling pathway related proteins were detected by Western blotting. Results: The mRNA and protein levels of BAMBI in shBAMBI group were lower than those of control group (P<0.05). Compared with control group, cell proliferation in sh-BAMBI group was obviously decreased (P<0.05), while apoptosis was obviously increased (P<0.01); in the meanwhile, cell invasion and migration in sh-BAMBI group were significantly reduced (P<0.05). In addition, the protein level of TGF-β and the ratio of p-Smad/2/ Smad/2 in shBAMBI group were significantly higher than those in control group (P<0.05). Conclusion: Interference of BAMBI by shRNA inhibits proliferation, invasion and migration but induces apoptosis of human colon cancer SW480 cells and activates TGF-β/Smad/2 pathway.
