1.Study on Quality Standard for Shule Granules

Objective:To establish the quality standard for Shule granules. Methods:By TLC, Angelic asinensis, Panax pseudog-inseng, Fructus liquidam-baris and Leonurus in Shule granules were qualitatively identified;and contents of notoginsenoside R1 and gin-senosides Rg1 in Shule granules were quantitatively determined by HPLC. Results: In the qualitative identification, the clear spots showed good resolution and specificity. The content of notoginsenoside R1 and ginsenosides Rg1 , two main ingredients in Shule gran-ules, was 0. 166 6 and 0. 681 8 mg·g-1 , respectively. Conclusion:The TLC and HPLC-UV determination method is simple, accu-rate and reproducible, and can be used as the quality control method for Shule granules.
2.Determination of Organic Chloride Pesticides and Polycyclic Aromatic Hydrocarbons in Prophyra by Gas Chromatograph-Mass Spectrometry

Objective To develop an analytical method by gas chromatograph-mass spectrometry(GC-MS) for the determination of organic chloride pesticides and polycyclic aromatic hydrocarbons in prophyra Methods The samples were extracted by an acetone-hexane(7∶3,V/V) mixture first,then were purified by C18 solid-phase extraction(SPE),and then were determined with GC-MS.Results The linear range was 0.01-5.0 ?g/ml,the limits of detection were 0.05-0.32 ng/g(dry weight) and 0.25-0.56 ng/g(dry weight)for organic chloride pesticides and polycyclic aromatic hydrocarbons respectively.The recovery rates and relative standard deviation for organic chloride pesticides were 74.27%-121.49% and 3.19 %-17.31% relatively;the recovery rates and relative standard deviation for polycyclic aromatic hydrocarbons were 65.10%-119.26% and 2.75%-14.11% relatively.Organic chloride pesticides were partly detected,but the detection rate of polycyclic aromatic hydrocarbons was higher.Fifteen polycyclic aromatic hydrocarbons were detected in all samples but dibenz(a,h)-anthracene.Conclusion This method is sensitive,accurate and applicable to the determination of organic chloride pesticides and polycyclic aromatic hydrocarbons in prophyra.
3.Dynamic expressions of matrix metalloproteinase-2 and its inhibitor in conjunctival filtering blebs of rats following glaucoma filtering surgery

Background The scarring of conjunctival filtering blebs after glaucomatous surgery is a leading cause of operation failure.Exploring the balance between matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of matrix metalloproteinase-2 (TIMP-2) in conjunctival filtering bleb is very important for the study on pathogenesis of postoperative scarring.Objective This study was to evaluate the role of MMP-2 and TIMP-2 in wounding healing of subconjunctival tissue after filtering surgery in rats.Methods Sixty-three clean male SD rats were divided into normal control group and postoperative 1-day,3-day,5-day,7-day,14-day and 28-day group.The drainage tube was monocularly implanted into the anterior chamber of the rats to establish the filtering surgery models,and the operative response of the eyes was examined under the slit lamp microscope.The animals were sacrificed in corresponding time points,and the frozen sections of eyeballs were prepared,and the expressions of MMP-2 and TIMP-2 in conjunctival and subconjunctival tissues were detected using immunofluorescence technique.Western blot was employed to assay the dynamic expressions of MMP-2 and TIMP-2 proteins in conjunctival and subconjunctival tissues in different groups.The use and care of the rats complied with the Instruction Notions with Respect to Care for Laboratory by State Ministry of Science and Technology.Results Filtering blebs were formed in all the operated eyes 1 day after surgery and remained for 7-17 days,with the average survival time of 12 days.Western blot assay revealed that the expression levels of MMP-2 protein in the filtering blebs of the normal control group were 121.67 ±4.37,and the expressions were gradually elevated from the postoperative 3-day group (183.67±5.61) until the postoperative 7-day group (230.50±8.48) and then gradually declined till the postoperative 28-day group (172.33 ± 8.43),showing a significant difference among the groups (F=280.18,P<0.05).In addition,the expression levels of TIMP-2 protein in filtering blebs of the normal control group were 102.50 ±6.25.The expression levels were gradually raised in postoperative 3-day group (162.67±7.00) and peaked in the postoperative 5-day group (232.00± 11.03),and then the levels gradually reduced till the postoperative 28-day group (150.50±6.41),with a significant difference among the groups (F =145.34,P < 0.05).Immunofluorescent staining showed that MMP-2 and TIMP-2 were weakly expressed in the conjunctival epithelium of the normal rats,while in the operated rats,strong fluorescence for MMP-2 and TIMP-2 was seen in both conjunctival epithelium and subconjunctival tissues of filtering blebs.Conclusions The expression trends of MMP-2 and TIMP-2 in the filtering blebs are consistent to the fibrosis process of conjunctival tissue,indicating that MMP-2 and TIMP-2 participate in the scar formation of conjunctival filtering bleb after glaucoma filtering surgery.
4.A survey of the video training in the application of the nurse self-directed learning readiness

Objective To investigate the nurse self-directed learning readiness in some 3A hospital in Shandong province and discuss the strategies how to improve it. Methods Self-designed questionnaire and The Nurses Self-Directed Learning Readiness Scale were used to investigate 210 nurses from 23 departments in Shandong provincial hospital. Results Before and after the implementation of the video training, a total of 230 and 210 self-directed learning readiness scales were distributed, 210 and 210 questionnaires were returned, the effective recovery rate was 91.30%and 100%respectively. The data of questionnaires were statistically analyzed by the method of paired t test. The results showed that after the implementation of the project, the total score of the after test (156.63±19.48) was higher than the before (137.91 ± 13.71). The difference was statistically significant (t=-9.54, P<0.05). The score in the three dimensions of self-management, self-control, and desire for study (48.03 ± 7.25), (48.71 ± 6.44), (59.89 ± 7.61) points, were higher than (40.11±5.48), (43.40±5.34), (54.40±6.26) pointsbefore implementation. The difference was statistically significant (t=-10.22,-7.67,-6.83,P<0.05). Conclusions The video training would provide the convenient and quick way and could inspire the nurses′ learning motivation even more which is of great significance to improve the nurses ability of autonomic learning.
5.Correlation of diastolic blood pressure and isolated systolic hypertension with coronary heart disease in the elderly

Objective To investigate the correlation of diastolic blood pressure (DBP) <70mm Hg and isolated systolic hypertension (ISH) with coronary atherosclerosis (CAS) and coronary heart disease (CHD) in elderly patients and to analyze the independent risk factors for low DBP.Methods A total of 246 elderly patients with untreated ISH who underwent coronary angiography were divided into the low DBP group (n=102) and the non-low DBP group (n=144) according to DBP level.All clinical and angiographic data were collected and the retrospective analysis was performed to assess the risk of CAS and CHD in patients with low DBP and to analyze the risk factors for low DBP.Results There were no significant differences in systolic pressure level between the low DBP group and the non low DBP group [(156.2±15.6) mmHgvs.(154.4±14.2) mmHg,t=0.93,P>0.05],while pulse pressure level was significantly higher in the low DBP group than in the non-lowDBPgroup [(91.3±±17.7) mmHgvs.(72.9±15.1) mm Hg,t=8.54,P<0.01].Stepwise logistic analysis showed that age,diabetes and smoking were independent risk factors for low DBP.After adjustment for age,gender and other common risk factors,low DBP was independently associated with CAS and CHD (OR =1.72 and 1.44,95% CI:1.082.72 and 1.04-1.99,respectively,both P< 0.05).Conclusions Low DBP is independently associated with CAS and CHD in elderly patients with untreated ISH.Age,diabetes and smoking are the independent risk factors for low DBP.
6.Relation of Isolated Systolic High-Normal Blood Pressure and Coronary Heart Disease in Middle-Aged and Elderly Persons

Objective To investigate the relationship between isolated systolic high-normal blood pressure and cor-onary heart disease (CHD) in the middle-aged and elderly population. Methods A total of 236 patients over 50 years old and undergone coronary angiography were enrolled and divided into isolated systolic high-normal blood pressure (ISHNBP) group (n=135) and non-ISHNBP (NISHNBP) group (n=101) according to their systolic and diastolic blood pressure levels. All clinical data and angiographic data were collected. Results There was no significant difference in systolic blood pres-sure between NISHNBP group and ISHNBP group (P>0.05). There were significantly lower levels of diastolic blood pres-sure and higher pulse pressure in ISHNBP group (P<0.01). Results of multivariate logistic analysis showed that isolated sys-tolic high-normal blood pressure was the independent risk factor for CHD (OR=2.67,95%CI:1.50-4.75, P<0.01). And the distribution of diseased coronary vessel numbers was more extensive in the ISHNBP group (P<0.01). Conclusion The iso-lated systolic blood pressure in the middle-aged and elderly population with high risk of coronary heart disease should be paid attention to, and make appropriate interventions, which may help reduce the incidence of cardiovascular disease.
7.A survey of the practice status of the new nursing practice standards for intravenous therapy in Shandong province

Objective To investigate the practice status of the newNursing practice standards for intravenous therapy in secondary and above hospitals in Shandong province. Methods Self-designed questionnaire was used to investigate 3051 nurses from 129 hospitals in 17 cities in Shandong province. Results Nurses′ awareness rate of the new Nursing practice standards for intravenous therapy was 92.69%(2828/3051);92.13%(2811/3051) nurses reported that their hospitals used the new standard, 65.78%(2007/3051) nurses reported that their hospitals had applied the standard since the second half of 2014; 92.92% (2835/3051) nurses reported that their hospitals had revised their practice rules according to the new standard;93.54%(2854/3051) nurses reported that their hospitals had revised the related nursing process; 50.08% (1528/3051) nurses thought the new standard helpful to clinical practice;22.98%(701/3051) nurses knew well about the new standard;53.85%(1643/3051) nurses thought the standard applicable to clinical practice. Conclusions As nurses do not master the Nursing practice standards for intravenous therapy well, they need strengthened training. Hospitals should fresh their rules, protocolsand supply training program to improve nurses′ knowledge and skill. Meanwhile, Nursing practice standards for intravenous therapyalso needs to be revised and improved according to nurses feedback.

Ventilatory volumes and respiratory flow rate during quiet and rapid respiration were measured in 480 healthy pilots of our Air Force. The mean values were: VE, 9.58?1.74 1/min; MVV, 154.69?19.48 l/min; PIFR, 23.56?4.60 l/min; and FPIFR, 182.388?29.95 l/min.Regression analysis showed that they correlated well with each other as well as with body surface area (BSA), age, sitting height etc. The regression. formulae are Y = 2.97398 + 3.74173X2 for VE, Y = 103.921-0.839956X1 + 44.4811X2 for MVV, Y = 20.2761+0.735782X4 for PIFR, and Y = 99.9757-0.69924X1+ 59.7506X2 for FPIFR, where X1 stands for age, X2 for BSA, and X4 for vital capacity.The above results should provide a valuable physiological basis for designing aircraft oxygen equipment,for laying down the standard of on-board oxygen supply, and for pulmonary functional tests in pilots.
9.Correlation of matrix metalloproteinases and Kashin-Beck disease

Objective To investigate the expressions of matrix metalloproteinases(MMPs) in Kashin-Beck disease(KBD) cartilage as well as in a KBD rat model of T-2 toxin poisoning under selenium deficient conditions, and to investigate the effect of T-2 toxin on MMP-13 expression in human chondrocytes in vitro in order to determine a possible mechanism underlying KBD. Methods Samples of articular cartilage were divided into 2 groups:controls(samples from 5 normal children, traffic accident or operation), and KBD(samples from 5 children with KBD, auctopsy). Thirty-two Sprague-Dawley rats were divided into two groups by body weight using random number table: normal diet group(n = 16) and selenium-deficient diet group(n=16). The selenium level in normal diet was 101.500μg/kg, and in selenium-deficient diet was 1.118μg/kg. Rats were fed for 4 weeks with selenium-deficient or normal diet, respectively. After successful build up of the low selenium rat model, normal diet group was then subdivided into 2 sub-groups: normal group(n = 8) and normal diet plus low T-2 toxin group(n = 8);and selenium-deficient diet group was also subdivided into 2 sub-groups: selenium-deficient group ( n = 8 ) and selenium-deficient diet plus T-2 toxin group ( n = 8 ) . T-2 toxin of 100 μg·kg-1·d-1 was administered by intragastric administration for 30 days. Then the rats were sacrificed, and their knee joints were processed for histopathological evaluation. MMP-1 and MMP-13 locations in cartilages were performed by inmmunohistochemistry. Human chondrocytes C28/I2 were cultured in vitro. The experiment was divided into 4 groups: empty vector plasmid group, MMP-13 promoter plasmid group, MMP-13 promoter plasmid plus 20 μg/L T-2 toxin group and MMP-13 promoter plasmid plus 40 μg/L T-2 toxin group. MMP-13-luciferase reporter plasmid and vector plasmid were transiently transfected into C28/I2 cells for 24 hours, and then treated with 20 - 40 μg/L T-2 toxin for 24 hours. Transactivation of human MMP-13 promoter was analyzed using luciferase reporter constructs containing sequences spanning-1602 to+20 bp in C28/I2 chondrocytes. Results The percentages of chondrocytes staining for MMP-1 in the superficial and middle zones of KBD samples [(29.73 ± 10.12)%, (28.27 ± 0.91)%] were significantly higher than those of controls[(2.47 ± 0.11)%, (0.00 ± 0.00)%, all P < 0.05]. The percentages of chondrocytes staining for MMP-13 in the superficial and middle zones of KBD samples [(13.21 ± 4.32)%, (41.85 ± 6.32)%] were significantly higher than those of controls[(5.72 ± 0.31)%, (0.00 ± 0.00)%, all P<0.05]. The percentages of chondrocytes staining for MMP-13 in the superficial and middle zones of rats fed with selenium-deficient diet plus T-2 toxin group[(13.21 ± 4.32)%, (61.85 ± 8.68)%] were significantly higher than those of the normal and selenium-deficient groups[(2.43 ± 0.22)%, (5.89 ± 0.69)%, (3.03 ± 0.29)%, (25.99 ± 0.57)%, all P < 0.05]. Moreover, T-2 toxin activated the MMP-13 promoter detected with luciferase reporter assays in C28/I2 cells. The luciferase activities in MMP-13 promoter plasmid plus 20 μg/L T-2 toxin group and MMP-13 promoter plasmid plus 40μg/L T-2 toxin group(0.082 78 ± 0.008 40, 0.103 35 ± 0.013 19) were significantly higher than those in empty vector plasmid group and MMP-13 promoter plasmid group(0.024 19 ± 0.000 96, 0.040 32 ± 0.003 56, all P < 0.05). Conclusions These data suggest that T-2 toxin induces cartilage matrix degradation through up-regulation of MMP-13 promoter expression. Increased MMPs staining intensity in KBD cartilage and the rat KBD model of T-2 toxin poisoning under selenium deficient conditions suggest that matrix degradation appear to be driven by MMPs activity.
