Objective To investigate the clinical significance of detection of serum IL -6,IL -17, GM-CSF and TNF -αlevels in patients with rheumatoid arthritis.Methods A retrospective research was made in 79 cases with rheumatoid arthritis,and they were divided into stable period and active period,and another 40 healthy persons were chosen as control group.IL -6,IL -17,GM-CSF and TNF -αlevels were compared,and the relevance was investigated.Results CRP,ESR and DAS28 of active period [39.14 (35.81 )mg/L,41.92 (33.27)mg/L, 4.85(2.74)]were higher than those of the stable period[21.07(16.53)mg/L,29.57(25.14)mg/L,2.88(1.76)] and control group[5.92(2.80)mg/L,3.19(2.24)mg/L],the differences were statistically significant (F =7.013, 6.224,2.136,all P <0.05).Serum levels of GM-CSF,IL -6 and TNF -αof active period[33.81(26.97)pg/mL, 47.23(42.81 )pg/mL,75.35 (41.69 )pg/mL]were higher than those of control group [6.19 (5.11 )pg/mL, 4.68(5.21)pg/mL,33.18 (20.94)pg/mL]and stable period [12.81 (9.76 )pg/mL,8.46 (10.37 )pg/mL, 43.05(38.14)pg/mL],the differences were statistically significant (F =5.328,6.117,8.016,all P <0.05).And there was no significant difference in IL -17 level between the groups (F =0.776,P >0.05).No correlation was observed between IL17 and DAS28,GM-CSF and IL -6 (r =0.213,0.167,0.316,all P >0.05),TNF -αand DAS28,GM-CSF,IL -6 and IL -17 (r =0.761,0.729,0.549,0.421,all P <0.05 ),IL -6 and DAS28 and GM-CSF (r =0.648,0.626,all P <0.05),GM-CSF and DAS28 had good correlation (r =0.563,P <0.05). Conclusion IL -6,GM-CSF and TNF -αlevels were positively correlated with the progression of rheumatoid arthritis,joint detection is of great value in monitoring the disease.

Objectives:To study the distribution status,clinical manifestations and laboratory test characteristics of alpha-thalassaemia gene type in the city of ChongQing to approach the importance of genetic diagnosis for alpha-thalassaemia,and the significance of thia diagnosis for antenatal screening and prenatal diagnosis to promote the survival quality of human being.Methods:This study was performed in the Children's Hospital of Chongqing Medical University.One hundred and three alpha -thalassaemia patients were involved in this study.They had taken the tests of serum iron(SI),total iron binding capacity(TIBC),transferrin saturation(TS),hemoglobin(Hb)electrophoresis and genetic examination.Retrospective study and prospective study were performed with the results.Results:(1)A total of 72 patients underwent SI,TIBC and TS,and 55.56 %(40/72) patients had SI decreased,TIBC increased and TS decreased.(2) 92 serum samples from these patients had Hb electrophoresis,in which 31.52%(29/92) presented fast band.(3) In this study,there were 82.52%(85/103) deletional alpha-thalassaemia and 17.48%(18/103) non-deletional alpha-thalassaemia.(4) In 85 deletional alpha-thalassaemia,alpha0-thalassaemia was more than alpha+-thalassaemia.Conclusion:(1) Deletion form is the main form of ? thalassaemia in ChongQing.(2) alpha0-thalassaemia is less than alpha+-thalassaemia.(3) Genetic diagnosis is the gold standard for this disease.(4) It should be emphasized that antenatal screening and prenatal diagnosis are important for better lives of human beings.(5) Alpha-thalassaemia can affiliate nutritional iron deficiency anemia(NIDA).

Objective:To discuss the different expression of AQP8 in cultured normal human trophocyte and JAR cell line,and the relationship between the changed expression of the channel protein and nosogenesis of chorionic carcinoma.Methods:The human cultured normal trophocyte was primarily cultured,purified and identified.JAR cell line was cultured.The expression of AQP8 was detected in cultured normal human trophocyte and JAR cell line by immunohistochemistry immunofluorescence confocal laser and RT-PCR.Results:(1)In the control group,the limitied expression of AQP8 was observed in cultured normal human trophocyte and JAR cell line,but in the exprimental group,the rate of positive cell of AQP8 in JAR cell line were 93.27%?14.45%,much higher than that in the control group.(2)By immunofluorescence staining confocal laser imaging,it was revealed that,in the same condition and excitation intensity,AQP8 in the JAR choriocarcinoma cell lines showed positive expression of the green,while in normal trophoblast cells,no green light,but when the excitation intensity was strengthened,it may still explore AQP8 in normal human villous trophoblast cell expression(.3)The expression of AQP8mRNA was detected in both cultured normal human trophocyte and JAR cell line.The OD value(0.98?0.35)of AQP8 in the experimental group was significantly higher than that(0.32?0.04)in the control group.Conclusion:Abnormal overexpression of AQP8 will destroy the external water and electrolyte balance of trophoblast cells,which might be one of the reasons for the occurrence of choriocarcinoma.

Objective To analyze the effect of Smad7 on the expression of TGF-?R,R Smads and STRAP in the process of malignant transformation of immortalized human bronchial epithelial cells BEP2D.Methods Cells were cultured and protein were extracted from BEP2D,BERP35T2,BS7 and RS7 cells,then Western blotting was performed to examine the expression level of TGF-?RⅠ,TGF-?RⅡ,Smad2/3,Smad4 and STRAP.(Results) Stable transfection of Smad7 gene led to decreased expression of TGF-?RⅡ and increased expression of Smad2 and STRAP in both BEP2D and BERP35T2 cells.There were no changes in the expression of Smad3 and Smad4.Conclusion Overexpression of Smad7 leads to decreased expression of TGF-?R Ⅱ and increased expression of STRAP in BEP2D and BERP35T2 cells,which could be one of the mechanisms of the malignant transformation of BEP2D cells.

?AIM: To observe the application of phacoemulsification combined with laser peripheral iridoplasty ( LPIP ) for acute angle-closure glaucoma with cataract which could not be controlled well by drugs.?METHODS:From January 2011 to June 2015, 49 eyes in 47 patients with acute angle - closure glaucoma and cataract were recruited while their intraocular pressure remained higher than 40mmHg 12h after drug treatment. LPIP were performed and phacoemulsification and intraocular lens implantation were carried out 3d after. Their clinical data and follow-up results were reviewed.?RESULTS: The intraocular pressure before treatment, 1d after LPIP, 1d after phacoemulsification, 1mo after phacoemulsification and 3mo after phacoemulsification were 62. 35+10. 31mmHg, 17. 96 ± 4. 64 mmHg, 16. 58 ± 3. 19mmHg, 13. 50 ± 2. 74 mmHg and 13. 46 ± 2. 48 mmHg respectively (F=10. 02,P<0. 05); the intraocular pressure of 1d (t=4. 35), 1mo (t=6. 43) and 3mo (t=6. 97) after phacoemulsification were all lower than the initial pressure (P<0. 05). The visual acuity showed the same trendy, while the visual acuity before treatment, 1d after LPIP, 1d, 1 and 3mo after phacoemulsification were 0. 06± 0. 02, 0. 20 ± 0. 18, 0. 45 ± 0. 19, 0. 60 ± 0. 11 , 0. 65 ± 0. 09 respectively (F=8. 36,P<0. 05). The best corrected visual acuity at 1d, 1 and 3mo after phacoemulsification were better than that before laser treatment ( t= 3. 97, 5. 12, 5. 89,P<0. 05). At 1d and 3mo after phacoemulsification, the anterior chamber depth, angle opening distance, trabecular/iris angle were all better than the initial ones (P<0. 05). At 1 and 3mo after phacoemulsification, the goniosynechia got better as well (P<0. 05).?CONCLUSION:Phacoemulsification combined with LPIP can reduce intraocular pressure, improve the visual acuity and make the anterior chamber depth, angle opening distance, trabecular/ iris angle, goniosynechia better for patients with acute angle-closure glaucoma and cataract when drug cannot control intraocular pressure well.

A variety of genetic mutations are closely related to the occurrence and development of hematological tumors. Apolipoprotein B mRNA-editing catalytic polypeptide 3B(APOBEC3B),a member of the cytosine deaminase family,can induce mutations in target DNA or RNA,which is one of the causes of tumor genome damage.APOBEC3B is highly expressed in many hematological tumors. This paper mainly reviewed the research progress on the role of APOBEC3B in hematological tumors,so as to provide potential targets for targeted therapy and accurate personalized therapy of hematological tumors.

OBJECTIVE:To establish an HPLC method for determination of Emodin in Yanyan granules.METHODS:The stationary phase was Diamonsil C18(150mm? 4.6mm,5? m)column and the mobile phase was methanol-0.1% phosphoric acid(68∶ 32),with detection wavelength set at 254nm.RESULTS:Within the range of 0.26～ 3.90? g Emodin had a good linearity,and the average recovery was 96.73%(RSD=1.49%).CONCLUSION:The method is accurate,sensitive,reproducible,simple and can be used for the quality control of Yanyan granules.
