1.Biocompatibility study of acellular bladder matrix patch

Objective To evaluate the biocompatibility of acellular bladder matrix patch as the pelvic floor repair alternative ma-terial .Methods The combination of in vivo and in vitro biological experiments including cytotoxicity ,haemolysis and vaginal im-plant tests were employed to evaluate the biocompatibility of this material .Results In the cytotoxicity test ,the material toxicity was the grade 0-1 .The hemolysis rate of the material extract liquid was 1 .5% .The general and histological observation showed that the material did not cause the host immunologic rejection and led to slight foreign body reaction after implanting ,which was ba-sically degraded in 12 weeks after transplantation .Conclusion Acellular bladder matrix exhibits better biocompatibility ,but needs to adjust its degradation rate in order to adapt to the requirements of pelvic floor repair operation .
2.MMP-1 and TIMP-1 expression in pubocervical fascia from patients with stress urinary incontinence

Objective To investigate collagen metabolism in stress urinary incontinence(SUI) so as to determine the alteration that contributes to SUI.Methods Biopsy specimens of vaginal wall tissue from 24 women with stress urinary incontinence were collected,with matched specimens from 24 female patients without SUI.Immunohistochenical technique were applied to detect the expression of MMP-1 and TIMP-1.Results SUI group demonstrated a significant increase in MMP-1 expression and a decrease in TIMP-1 expression,with significant difference with control group.Conclusion Patients with SUI were in the predominant status of collagen breakdown,which may contribute to the pathogenesis of SUI.

OBJECTIVE: To establish an HPLC method for the determination of plasma concentration of Irbesartan tablets in rats and its pharmacokinetics study.METHODS: The determination was performed on Cosmosil-C18 column with mobile phase consisted of phosphoric acid-methanol-acetonitrile(7 ∶ 80 ∶ 13,V/V/V) at flow rate of 1.0 mL?min-1.The detection wavelength was set at 272 nm.Rats were given Irbesartan solution 30 mg?kg-1 via i.g.gtt.The plasma concentration of irbesartan were determined before medication and at 0 min,5 min,10 min,20 min,30 min,45 min,60 min,120 min,180 min,240 min,360 min,480 min after medication.The pharmacokinetics parameters were estimated.RESULTS: The linear range of irbesartan was 0.5~50 ?g?mL-1(r=0.999 1) with an average recovery of 93.89%(RSD
5.Study on the Protective Effect of Bilobalide on Ischemical Reperfusion Injury in Rats Undergoing Liver Transplantation

OBJECTIVE:To observe the effect of Bilobalide on ischemical reperfusion injury in orthotopic liver transplantation model rats and to discuss its protective effect on ischemical reperfusion injury of rats undergoing liver transplantation.METHODS:The model of orthotopic liver transplantation was established by in rats by setting up the bridge between portal vein and left renal vein and building blood bypass by inlaying of canal in the inferior vena,after modeling,the rats were treated with Bilobalide,then levels of NO(active medium of blood vessel)and ET1 before ischemia and at 10 min,30min and 2h after reperfusion were determined by nitratase reduction method;serum levels of ALT,AST,ALP,LDH enzymology,ATP and MDA in the liver tissues were determined as well.The hepatic tissue sample was taken at 2h and fixed with formalin to be made into specimen for observation of the ultrastructure of liver cells and hepatic lobules under electron microscope.RESULTS:After being treated by Bilobalide,serum NO level was elevated and the pathological elevated levels of ALT,AST and LDH were brought down,the microcirculation in ischemical reperfusion injury rats was improved and the extent of damage in the ultrastructure of liver cells was lessened in rats.CONCLUSION:Bypass of portal vein and inferior vena cava prior liver transplantation is helpful for the prevention of ischemical reperfusion injury of liver.The balance of NO/ET1 is possibly a factor influencing the blood flow in the microcirculation of transplanted liver.Bilobalide was proved to be of protective effect on ischemical reperfusion injury in rats.
6.HPLC Determination of Ferulic Acid in Xuefu Zhuyu Decoction

Objective To establish a HPLC method for the determination of ferulic acid in Xuefu Zhuyu Decoction. Methods A Kromasil C18(250 mm ×4. 6 mm, 5 μm) column was adopted. The mobile phase consisted of acetonitrile-0. 085 % phos-phoric acid(17 : 83)with the flow rate of 1.0 mL/min. The column temperature was at 35 ℃ and detection wavelength was set at 316 nm. Results A good linearity of ferulic acid was in the range of 0. 0252μg to 0. 504μg and r=0. 999 9. The average recovery was 100. 15 % and RSD=1.69 %. Conclusion The method is simple and rapid, and it is suitable for the determi-nation of ferulic acid in Xuefu Zhuyu Decoction.
7.Chronic disease risk index and its predictive effectiveness on chronic diseases

Objectives To develop a measurement for obesity assessment which could be used as a simple tool of risk screening. Methods The data of National Nutrition and Health Survey ( 2002 ) were used to analyze the relationship between body mass index ( BMI ), waist circumference (WC) and chronic diseases, based on which chronic disease index ( CRI ) was established. Receiver operating characteristic curve (ROC) was used to determine the cut-off of CRI and to compare the predictive effectiveness of CRI,BMI and WC on chronic diseases. The kappa test was chosen to estimate the consistency of different cut-off of CRI with BMI and WC. The odds ratios of chronic diseases in different cut-offs of CRI were calculated by multiple Logistic regression analysis. Results CRI was calculated as CRI = BMI + 3.5 WC. In ROC curve,the cut-off of CRI was 300. The predictive effectiveness of CRI was higher than that of BMI and WC. CRI at 300 showed the best consistency with 24 kg/m2 BMI and 85 cm WC (P <0. 05 ). In comparison with CRI <300,the risk of chronic diseases was significantly increased with an increase of 20 U CRI. Conclusion CRI shows good predietive effectiveness and could be used to identify those with higher risk of chronic diseases.
8.Effects of muscular traumatic fluid on biological properties of muscle-derived stem cells in rats

Objective To study the effects of muscular trumatic fluid on the biological properties of muscle-derived stem cells (MDSCs) in rats.Methods MDSCs were isolated and purified by the preplate technique,muscle injury was made for the extraction of muscular traumatic fluid.Bradford colorimetric was used to detect the protein content in the muscular traumatic fluid,and the fluid with the highest protein was used to co-culture with MDSCs.Methyl thiazolyl tetrazolium (MTT) assay and wound model of monolayer of cells were used to observe the effects of wound fluids on the proliferation and mobility of MDSCs.The expressions of α-SMA and Vimentin were tested by immunohistochemistry and Western blot technique.Results MTT assay showed that wound fluid with concentration of 10% could most significantly increase the proliferation of MDSCs.The increased expressions of α-SMA and Vimentin were found in MDSCs after cultrue with muscular traumatic fluid in vitro,and the time-dependent relation exists.Conclusions Wound environment can directly participate in the muscle regeneration by inducing the proliferation,mobility of MDSCs;on the other hand,MDSCs can differentiate into fibrotic cells under stimulation of muscular traumatic fluid.
9.Endoglin in assessing angiogenesis of epithelial ovarian tumor:report of 70 cases

Objective To investigate the significance of endoglin in the angiogenesis of ovarian carcinoma.Methods Immunohistochemical assay was used to investigate 70 tissue samples of pathologically identified epithelial ovarian tumor(including 31 specimen of benign and 39 of malignant)from December 2000 to December 2003 in our department using monoclonal antibodies against CD34 and endoglin.Microvessels density(MVD)was counted by experienced pathologists.Results No significant difference of MVD for CD34 was observed in the ovarian cystadenoma and ovarian carcinoma.In contrast,an increment of MVD for endoglin from low-grade to high-grade dysplasia and that from earlier stage to later stage was statistically significant.Conclusion Endoglin may be a valuable marker for assessing neovascularization in the process of ovarian carcinoma development and for predicting the risk of ovarian carcinoma development.
10.Cell-cell contact induces mesenchymal stem cells differentiation to smooth muscle cells

Objective To establish a co-culture system between mesenchymal stem cells (MSCs) and smooth muscle cells (SMCs) in vitro, and study the influence of SMCs on the differentiation of bone marrow MSCs (BMSCs) into SMCs in the co-culture system. Methods Density gradient centrifugation was used to separate and culture BMSCs, and collagenase digestion was employed to separate and culture bladder smooth muscle cells(BSMCs). Then the 2 types of cells were identified respectively. Then the cells were divided into 4 groups, BMSCs culture group (positive control), BSMCs cells culture group (negative control), BMSCs and BSMCs co-culture groups with contact or non-contact. The expression of ?-smooth muscle actin (?-SMA) and calponin were detected by immunofluorescence staining. Western blot analysis was employed to determine protein expression of calponin. Results In contact co-culture group, the positive rate of CD90 abd CD44 were 99.78% and 60.27% respectively in BMSCs, while only 0.21% for CD45. Western blotting showed that BMSCs expressed little calponin, but those contacting with BSMCs had an increasingly stronger expression of calponin with the elapse of contact culture time, while there was no significant change in its expression in BMSCs with contact. Immunofluorescence staining indicated that part of BMSCs were calponin positive at the 8th day after culture, but no positive cell was observed in non-contact culture group and BMSCs culture group. Conclusion The contact between BSMCs and BMSCs can promote the differentiation of BMSCs into SMCs.
