1.Application of fluorescent probes in the diagnosis and treatment of melanoma

Melanoma is a highly aggressive malignant skin tumor characterized by rapid growth and early metastasis. Surgical resection is currently still the main treatment. There are still some challenges in the early diagnosis and thorough surgical resection of melanoma, which affect the prognosis of patients. To date, great progress has been made in the application of fluorescent probes to the diagnosis and surgical resection of, as well as sentinel lymph node localization and resection in melanoma. Fluorescent probes have advantages of good chemical stability, less adverse effects, high specificity, high accuracy and visualization, and may provid new ideas for the diagnosis and treatment of melanoma.
2.Qualitative study of life events how to influence emotional state of heart transplantation patients

ObjectiveTo explore the influence of life events on the emotional state ot heart transplantation patients.Methods All the heart transplantation outpatients were interviewed by a certain psychiatrist in Zhongshan Hospital during Oct.2010 to Oct.2011.The demographic data,life events and the self-reported influenced degree were recorded by the same psychiatrist.A grounded theory-based approach with thematic content analysis of the field notes was used.ResultsEighty-eight patients were enrolled,including 69 males,and19 females.The average age of the patients was 46.5 ±14.3 years old.The average duration after operation was 39.3 ± 39.6 months.The life events were classified into 4 groups,which were medical financial problems,physical condition,family support and future life.The most frequently reported event was physical condition (50.2% ) and the influenced degree was the highest (9.2 ±1.2),while most patients (96.6%) complained high medical and relevant cost and the influenced degree was (6.5 ± 2.1). Family support was reported by13.6% patients and its influenced degree was (7.6 ±1.2).The age of the reporter of future life (6.8%) was under 33 years.ConclusionDifferent events have different influence to patients.Improving the social support of patients is helpful to improve quality of life.
3.Risk Factors,Prevention and Therapy of Hepatic Artery Thrombosis after Liver Transplantation

Objective To investigate the risk factors,prevention and therapy of hepatic artery thrombosis after liver transplantation.Methods The literatures on the risk factors,prevention and therapy of hepatic artery thrombosis after liver transplantation in recent years were collected and reviewed.Results The risk factors include factor Ⅴ Leiden,metabolic liver diseases of recipients,recipient sex,the use of Roux-en-Y biliary reconstructions,virus infection and so on.The measures of prevention and therapy include early diagnosis,detection of activated protein C resistance,postoperative anti-coagulation therapy,liver arteries reconstructions measures,hyperbaric oxygen therapy,continuous transcatheter arterial thrombolysis,liver retransplantation and so on.Conclusion The study of risk factors,prevention and therapy will promote the process of improving the prognosis of patients with liver transplantation.
4.Screening of Proteins Binding to HBsAg from Human Pancreas cDNA Library by Yeast Two-hybrid System

Objective To screen proteins in pancreas cDNA library interacting with HBsAg protein.Methods After being amplified and identified,the human pancreas cDNA library plasmid was transformed into yeast Y187.Thereafter,the bait plasmid(PGBKT7-HBsAg) was transformed into yeast cells AH109.Then,two different transformed yeast cells were mated.The diploid yeast cells were plated on synthetic dropout nutrient medium(SD/-Trp/-Leu/-His/-Ade) and that medium contained X-?-gal for selection two times.After plasmids of true positive blue colonies were extracted and transformed to E.coli,the results were analyzed by DNA sequencing and blast searching in GenBank database.Results Six proteins binding to HBsAg were screened,including aconitase(ACO1),CDK5RAP3,carboxypeptidase,etc.Conclusion Most of these proteins play significant roles in the evolution of 2 type diabetes mellitus,which may supply some new clues for exploring the pathogenesis of abnormality of lipid metabolism and tglycometabolism in the patients with chronic hepatitis virus infection.
5.The expression and significance of serum ghrelin, leptin, IGF-1, and IGFBP3 in children with idiopathic short stature

Objective To examine the expressions of serum growth hormone (ghrelin), leptin (LP), in sulin-like growth factor-1 (IGF-1) and insulin-like growth factor binding protein-3 (IGFBP3) in children with id-iopathic short stature (ISS), and their significance. Methods A total of 40 patients with ISS were selected from May. 2012 to Oct. 2014 in Fuyang Maternal and Child Health Hospital, while a total of 40 children in good health were selected as the control group. Serum ghrelin and LP levels were measured by ELISA. Serum IGF-1 and IGF-BP3 levels were detected by chemiluminescence immune assay. Results were analyzed statistically. Results The height, weight, BMI, GH, serum LP, IGF-1, and IGFBP3 levels in the observation group were significantly lower than those in the control group, while serum ghrelin expression level was significantly higher than that in the control group(P<0.05). The correlation analysis showed that ghrelin and LP were negatively correlated(r=-0.611, P<0.01), ghrelin and IGF-1 levels were negatively correlated(r=-0.520, P<0.05), Ghrelin and IGFBP3 were pos-itively correlated (r=0.586, P<0.01), IGF-1 and IGFBP3 was negatively correlated (r=-0.576, P<0.01), and LP and IGFBP3 were negatively correlated (r=-0.609, P<0.01). Conclusions It shows that ghrelin, LP, IGF-1, and IGFBP3 levels in children with ISS are related to growth hormone secretion status. The interactions between ghre-lin and insulin-like growth factor axis regulate growth and development of children.
6.Aerobic exercise relieves oxidative stress, inflammation and apoptosis due to colitis

Objective:To explore the effect of regular aerobic exercise on oxidative stress, inflammatory reaction and apoptosis with a rat model of colitis.Methods:Forty-five C57BL/6 mice were randomly divided into a control group, a model group and a model exercise group, each of 15. Colitis was induced in the rats of the model and model exercise groups through administration of 3.5% sodium dextran sulfate. The control group and the model group were kept quiet in their cages, while the model exercise group performed voluntary wheel running for 8 weeks after the successful modelling. Any histopathological changes in the colon tissue were observed after hematoxylin and eosin staining. Oxidative stress, inflammatory reaction and apoptosis were measured from colon tissue homogenates.Results:Compared with the control group, the average expression of the tumor necrosis factor α (TNF-α), interleukin (IL)-1β, IL-6 and prostaglandin E2 (PGE2) as well as levels of reactive oxygen species (ROS), malondialdehyde (MDA) and nitric oxide (NO) and inducible nitric oxide synthase (iNOS) and Caspase-3 activity and the protein expression of cyclooxygenase 2 (COX2), nuclear factor kappa B (NF-κB) p65, apoptosis inducing factor (AIF), cytochrome c (Cyt C) and Bax had all increased significantly in the other two groups, while the levels of IL-10 and glutathione (GSH), the activity of superoxide dismutase (SOD) and glutathione peroxidase (GPx) and the protein expression of Bcl-2 protein had decreased significantly in the model group. Compared with the model group, the average content of TNF-α, IL-1 β, IL-6, PGE2, ROS, MDA and NO, the activity of iNOS and Caspase-3 and the protein expression of COX2, NF-κB p65, AIF, CytC and Bax had decreased significantly, while that of IL-10 and GSH, the activity of SOD and GPx and the protein expression of Bcl-2 protein had increased significantly in the model exercise group.Conclusion:Regular aerobic exercise has a protective effect against colitis through improving oxidative stress, inflammation and apoptosis, at least in rats.
7.The effect of RNA interference induced by inhibition of hTERT and Bi-1 gene expression

Objective In this study ,we constructed a series of recombinant plasmids carriers expressing shRNA targeting hTERT and Bi‐1 gene .These recombinant plasmids carriers were transfected into CNE‐2Z cell lines using Lip and continuously in‐duced the expression of shRNAs .Furthermore ,the shRNAs caused the degradation of mRNAs homologous in sequence with the target genes ,which lead to a sequence‐specific gene silencing .Methods The CNE‐2Z cells was divided into untreated group ,pEG‐FP‐N1 group and pEGFP‐N1/Lip group .Flow cytometry(FCM ) was applied to determine the transfection efficiency .The changes of hTERT and Bi‐1 gene expression were detected by Real‐time RT‐PCR and Western blotting .Results The best transfection effi‐ciency between plasmid and Lip was 2 .5 μg plasmid and 6 .25μL Lip .Conclusion We constructed several shRNA recombinant eu‐karyotic expression plasmids successfully .The recombinant plasmid can inhibit the expression of hTERT and Bi‐1 gene specifically and effectively .
8.The depression and anxiety state of heart transplantation patients

ObjectiveTo investigate the depression and anxiety state of heart transplantation patients. MethodsFifty-weight heart transplantation outpatients were assessed by Hamilton Depression Rating Scale (HAMD) and Hamilton Anxiety Rating Scale (HAMA) in Zhongshan Hospital during October 2010 to April 2011.Those diagnosed with depression disorder should meet the criteria of depression in Chinese Classification of Mental Disorder-3nd version (CCMD-3) and the score of HAMD≥14,and those diagnosed with anxiety disorder should meet the criteria of anxiety disorder CCMD-3 and the score of HAMA≥ 14.The diagnoses of concurrent depression and anxiety were made.Results The prevalence of depression disorder and anxiety disorder was respectively 15.5% (9/58) and 22.5% (13/58).The prevalence of depression disorder and anxiety disorder within 12 months was respectively 16.7% (2/12) and 33.3% (4/12).The score of HAMD was positively related to HAMA score (P<0.01 ).ConclusionThe prevalence of depression disorder and anxiety disorder is very high and the co-existence of depression and anxiety is very common.
9.Expression of ErbB3 in pancreatic cancer cell line regulated by microRNA-148a

Objective To look for and confirm the downstream regulated gene of microRNA (miRNA)-148a in pancreatic cancer cell line.Methods The target gene regulated by miRNA-148a was predicted through bio-informatics analysis.The plasmid containing desired gene and with luciferase 3'-untranslated region (3'-UTR) reporter was constructed.Pancreatic cancer cells BXPC-3 were transfected with analogs and inhibitors of miRNA-148a by liposomes.The activity of luciferase was measured to determine whether miRNA-148a directly connected with desired gene.The expression level of miRNA-148a was changed in BXPC-3 cells,and the changes of target gene v-verb-b2 erythroblastic leukemia viral oncogene homolog 3 (awian) (ErbB3) expression were detected by Western blot at protein level.The data were analyzed by one way ANOVA.Results There was a conservative binding site of ErbB3 with miRNA-148a detected by bio-informatics analysis,miRNA-148a directly combined with ErbB3 and the activity of luciferase decreased to (25.00+47.00) % of the negative control (F=4.66,P< 0.01).After miRNA-148a overexpression,the gray value of ErbB3 expression in BXPC-3 cells decreased to (26.16±4.69)% of control group (F=6.563,P<0.05).Conclusion miRNA-148a directly targeted and regulated the expression of ErbB3 in pancreatic cancer cell line BXPC-3.
10.Clinical characteristic and surgical management of adult presacral tumors

Objective To investigate the clinical characteristics and surgical management of adult presacral tumors.Methods The clinical data of 24 patients with adult presacral tumors from 2007 to 2012 were retrospectively analysed.All patients were diagnosed by digital rectal examination and imaging examination before operation.Seventeen patients with via sacrococcygeal approach,4 patients with via abdominal approach,and 3 patients with via combined abdominal and sacrococcygeal approach.Results The incision of 22 patients primary healing.There were 2 patients with postoperative incision infections,after dressing change cure.No patients died during perioperative period.Twenty patients were followed up for 3-17 months,1 patient with via abdominal approach recurrenced,and was healed after a second surgical resection.Conclusions Digital rectal examination and imaging examination are the main methods for diagnosis of adult presacral tumors.Most adult presacral tumors can be resected through sacrococcygeal route.
