1.Effects of the EL-1 Receptor Antagonist on Matrix Metalloproteinase 1 Expression in UV-irradiated Fibroblasts

Objective To study the effects of the IL-1 receptor antagonist (IL-1Ra) on matrix metalloproteinase 1 (MMP-1) expression in UV-irradiated fibroblasts. Methods Simulating the impact of environmental ultraviolet (UV) light on human skin, UVA-irradiated human fibroblasts were cultured in medium obtained from UVB-irradiated HaCaT cells. MMP-1 was detected by ELISA in the culture medium of fibroblasts. After treatment with IL-1Ra, the mRNA expression levels of C-Jun, C-Fos and GAPDH (internal control) of fibroblasts were measured by real-time fluorescent quantitative RT-PCR. Results Production of MMP-1 by UVA (10 J/cm2)-irradiated fibroblasts was increased in culture medium from UVB-irradiated HaCaT cells. The fibroblasts produced significantly higher levels of MMP-1 in culture medium from HaCaT cells treated without UVB than those with 15 mJ/cm2 UVB (t = 8.413,P= 0.014). However, IL-1Ra inhibited MMP-1 production of fibroblasts in a dose-dependent manner. Standard curves of real-time fluorescent quantitative RT-PCR showed a linear correlation between the copy number and the threshold cycle (Tc). Melting curves confirmed the specificity of PCR products. The original copy numbers of C-Jun and C-Fos as well as the ratios of the numbers to the GAPDH copy number showed that IL-1Ra inhibited the C-Jun mRNA expression of fibroblasts in a dose-dependent manner but had no significant effects on C-Fos mRNA expression. Conclusions The culture medium from UVB-irradiated HaCaT cells can promote MMP-1 production by UVA-irradiated fibroblasts. IL-1Ra reduces MMP-1 production via inhibition of C-Jun mRNA expression.
2.Effect of puerarin on glucogen synthase Kinase-3 bate in adipocyte of rats with insulin resistance

AIM: To investigate the effect of puerarin on expression of the glucogen synthase kinase-3 bate (GSK-3?) in adipocyte of rats with insulin resistance induced by diets. METHODS: 30 Wister rats were randomized into two groups: control group (n=10) and model group (n=20).Control group were fed with general diets, while model group were fed with the high fat diets to induce insulin resistance. After 4 weeks, the models have formed then were randomized into two groups: model group (n=10) and puerarin group (n=10). Puerarin group were treated with puerarin (100 mg?kg -1 ?d -1 , ip) for 6 weeks.GSK-3? protein was detected by Western-blot analysis. Body weight, serum triglyceride and cholesterol, fasting plasma glucose and serum insulin concentration were detected termly and the insulin sensitivity index was calculated. RESULTS: The model group showed hyperglycemia, hyperinsulinism, obviously obesity and the insulin resistance index (ISI) decrease by high fat diets, insulin resistance was induced by this way. 3.Ultrasonographic diagnosis of mesenteric vascular diseases

Objective To investigate the diagnosis of mesenteric vascular diseases by two-dimensional ultrasonography and color Doppler flow imaging (CDFI) and to summarize ultrasonographic imaging features.Methods Eighteen patients were examined by ultrasonography in emergency.The thickness,scope and echo of the wall of the small intestine,the condition of intestinal peristalsis,the blood flow of the wall of the bowel and the mesentery were measured by ultrasonography.Results The diagnosis of all the cases was confirmed by operation.The accuracy rate of ultrasonographic examination was 83.3% (15/18).Two patients were pseudo-positive( 11.1% ) and one patient was pseudo-negative( 5.6% ).Conclusions The ultrasonographic examination with directimaging features can determine the diagnosis of the disease and assess the classification and scope of mesenteric vascular diseases,hence is the first choice and an indispensable method.

7.Significance of the mutation of c-kit gene in the clinical prognosis of gastrointestinal stromal tumor

Objective To investigate the significance of the mutation of c-kit gene in the development,(clinical) pathology and prognosis of gastrointestinal stromal tumors(GISTs).Methods The technique of(PCR-SSCP) was used to detect ckit gene mutation in 106 cases of GIST,and 57 of the cases were(followed-up).Results The c-kit gene mutation rate in the whole group was 45.3%(48/106),and(3.4)%(2/41) in benign GIST,and 70.8%(46/65) in malignant GIST.The 1-,3 and 5-year survival rates and median survival time of the mutation positive group were all markedlly lower than those of the(mutation) negative group.A comparison of the parameters that can differentiate the benign GISTs from malignant ones such as size,histological necrosis,aggressive grade,nucleus mitotic count and nuclear discrepancy,showed significant difference between the of postive and negative gene mutation(P

Objective To investigate the changes in TNF? content of sciatic nerve induced by chronic constriction injury (CCI) of sciatic nerve to determine the role of TNFa in the development of neuropathic pain. Methods Eight-four female health SD rats weighing 250-300g were anesthetized with sodium barbiturate. Unilateral sciatic nerve was exposed and ligated at the middle of thigh. Three ligatures (chromic catgut 4.0) were placed around the sciatic nerve and tied. The distance between the two ligatures was about 1 mm. Sham operation was performed on the contralateral thigh. The sciatic nerve was exposed and mobilized but not ligated. The thermal nociceptive threshold was determined by measuring the withdrawal latency of hindpaw placed on a 58℃ hot plate on days 0.5, 1, 3, 5, 7, 9, 11, 13 and 14 after surgery. Animals were sacrificed on days 0.5, 1, 3, 7, 10 and 14 after surgery. Sciatic nerves were removed from both thighs and frozen at - 80℃ for determination of TNF? content. Sciatic nerve from healthy animals was used as control. The percentage of maximal possible response (% MPR) , was determined for each group (CCI, sham operation, control) % MPR= (new withdrawal latency- average baseline latency)/( 15-average baseline latency) . The distribution of TNF? between supernatant and sediment was also determined. Results The average baseline nociceptive threshold (withdrawal latency) was (7.9?0.2)s. There was significant different in %MPR between the two hindpaws on days 1, 3, 5, 7, 9 and 11 after surgery. The TNF? content of sciatic nerve from healthy rats was (40.62? 0.24) pg/mg protein. The TNF? content of the ligated sciatic nerve was elevated abruptly in 12h after ligation, then abruptly declined to a plateau but was still significantly higher than that of sham-operated side on days 1 and 3. There was no significant difference in TNF? content of sciatic nerve between control group and sham-operation group. The relative content of TNF? content in the sediment of ligated sciatic nerve gradually increased and reached the peak on day 7 and then gradually decreased. Conclusion The TNF? content of peripheral nerves plays an important role in the development of neuropathic pain. Membrane-combined TNF? is involved in the process of nerve repairing.
9.Inhibitory effects of intensive insulin on myocardium expression of tumor necrosis factor alpha in severely burnt rats

Objective To investigate the inhibitory effects of intensive insulin on myocardium expression of tumor necrosis factor alpha(TNF-?)in severely burnt rats and its correlative mechanism.Methods In eighteen SD rats the right jugular vein and the left ventricle were cathetetized,and were divided into three groups randomly:(1)the sham burn group,the rats were treated by sham burn;(2)the burn group,the rats were inflicted with 30%TBSA Ⅲ degree burn,and then given fluid resuscitation routinely;(3)the intensive insulin group,the rats received intensive insulin treatment after burn,and the liquid volume given was equal to the burn group.The level of plasma glucose contents,the left ventricular systolic pressure(LVSP)and left ventricular end-diastolic pressure(LVEDP)were recorded at 1,2,3,4,5,6h after burn,and animals were sacrificed at 6 hours after burn.The level of TNF-? in myocardium tissue was assayed by ELISA,and mRNA expression was determined by real-time PCR.Western blot was used to detect I?B? phosphorylation and degradation in left ventricle tissue.Meanwhile,burn serum-challenged cardiomyocytes were also used to detect the levels of insulin and LY294002,a specific insulin activation inhibitor,on the phosphorylation of I?B?.Results The animals showed stress hyperglycemia after burns,the function of heart was damaged(LVSP degraded and LVEDP increased),the level of TNF and the expression of mRNA in the myocardium increased apparently,I?B? phosphorylation and degradation increased in left ventricle tissue and burn serum-challenged cardiomyocytes.Given the intensive insulin treatment to keep the blood glucose level between 4.5 to 5.2 mmol/L,the function of heart was significantly improved,the level of TNF-? was apparently lowered,and the I?B? phosphorylation and degradation were significantly decreased.LY294002 could abolish the inhibitory effect of insulin,and it acted on the I?B? phosphorylation on burn serum-challenged cardiomyocytes model.Conclusion Intensive insulin may decrease the activation of NF-?B through diminishing the I?B? phosphorylation and degradation,which inhibit the transcription and expression of TNF-? in myocardial cell,and improve the heart function accordingly.
10.EFFECT OF LOW FAT DIET ON EXPRESSTION OF PTOTEIN KINASE B IN ADIPOSE TISSUE OF RATS WITH INSULIN RESISTANCE

Objective: To study the effect of low fat diet on expression of protein kinase B (PKB) in adipose tissue of rats with insulin resistance. Method: Thirty male Wistar rats were randomly divided into control group ( C, n=10),given low fat diet; model group (M, n=20), given high fat diet. After 4 w, group M was randomly divided into 2 subgroups accepting diet intervention of 6 w.(1)High fat group (HF) was continually given high fat diet;(2)Low fat group (LF) accepted low fat diet. The expression of PKB stimulated by insulin in adipose tissue was determined with Western blotting at the end of the experiment. Results: (1) Group M showed hyperglycemia, hyperinsulinism, hypertriglyceride, hypercholesterolemia and elevated HOMA insulin resistance index (HOMA-IR) while the insulin sensitivity index (ISI) was obviously reduced compared to group C, indicating that the insulin resistant model was induced ; (2) Low fat diet treatment decreased FBG,TG,TC,which were accompanied by lower HOMA-IR and elevated ISI; (3) Because of long term high fat diet, expression of PKB in adipose tissue of group HF decreased by 23.5% (P
