1.Total body irradiation before hematopoietic blood stem cells transplantation:clinical analysis of 78 cases

Objective To analyze the efficacy,dose and complications of total body irradiation(TBI) for patients to receive hematopoietic stem cell transplantation(HSCT) in the treatment of leukemia and malignant lymphoma.Methods From May 2002 to February 2007,78 cases with leukemia or malignant lymphoma received the TBI before HSCT in our department.All the patients were irradiated with linear accelerator in 2 consecutive days at a total dose of 7 to 11 Gy,dose rate of 4.5 to 5.0 cGy/min,once per day.Among them,55 cases received traditional high-dose chemotherapy before radiotherapy,and the other 23 cases got TBI first followed by high-dose chemotherapy.Results All the cases used TBI prior to transplantation was successful,and no serious complication such as radiation pneumonitis was observed.Conclusion Our TBI regime is a safe and effective pretreatment for HSCT.TBI used before the large doses of the chemotherapy is recommended because it is proved to be effective,without affecting the survival rate,and of lesser TBI reaction and of lesser preparation work for irradiation to radiotherapist.More attention should be paid when the linear accelerator is employed for TBI.
2.Epidemiological investigation of dental health status of 6-8 years old children in Zhongkai Hi-Tech Zone of Huizhou

Objective:
To investigate the status of tooth caries of 6-8 years old school children in Zhongkai Hi-Tech Zone of Huizhou city, and to provide scientific data for prevention and treatment on caries.
Methods :
Referring to the Guideline for the 3rd National Oral Health Survey, the dental caries status of 1-2 grade students in 32 primary schools were investigated, and the data were analyzed by SPSS 14.0 statistical software.
Results :
The prevalence rate of deciduous teeth caries in 4 390 children was 67.4%, and the DMFT was 3.94; The eruption rate of the first molars was 81.5%, with the prevalence rate of caries was 24.3%, and the DMFT was 0.448. The rate of caries for 8 years old children is higher than in 6-7 years old children (P < 0.001).
Conclusion
There is a high caries rate in children in Zhongkai Hi-Tech Zone of Huizhou, so oral health education, primary teeth caries treatment, and fissure sealant of permanent teeth should be strengthened.
3.Cluster analysis of disaster related topics of national social science foundation and its implication to disaster nursing

Objective:To sort out the situation of disaster related projects in national social science projects in recent 10 years and discuss hot spots and front direction of disaster nursing research.Methods:relevant fund information of disaster research in the national social science foundation and the humanities and social science research foundation of the ministry of education from 2009 to 2019 was analyzed by keyword clustering.Results:A total of 648 fund information was retrieved. Four research hots pots in recent 10 years were obtained: disaster risk prediction mechanism, multi-organization disaster response management, logistics optimization model, disaster public opinion and information dissemination.Conclusion:The development of nursing is inseparable from technology and information. Nursing should cooperate with other disciplines and organizations to form a nursing emergency system.
4.Mast Cells Tryptase Promotes Intestinal Fibrosis in Natural Decellularized Intestinal Scaffolds

BACKGROUND:
Standard two-dimensional (2D) culture has confirmed the mechanism of mast cells (MCs) in the pathogenesis of inflammatory bowel disease (IBD), but the regulation of signaling responses of MCs may well differ in three-dimensional (3D) microenvironments. The aim of the study was to develop a 3D culture model based on decellularized intestinal scaffolds (DIS) and verify how MCs influenced fibroblasts phenotype in the 3D model.
METHODS:
DIS were achieved using the detergent technique and extracellular matrix (ECM) components were verified by histologic analysis, quantification and scanning electron microscope. After human colon fibroblasts recellularized into the scaffolds and activated by MCs tryptase and TGFb1, the changes in genes and signaling pathways during fibroblasts activation in 3D were studied and compared with the changes in 2D cell culture on plastic plates.
RESULTS:
Decellularization process effectively removed native cell debris while retaining natural ECM components and structure. The engrafted fibroblasts could penetrate into the scaffolds and maintain its phenotype. No matter whether fibroblasts were cultured in 2D or 3D, MCs tryptase and transforming growth factor b1 (TGF-b1) could promote the differentiation of fibroblasts into fibrotic-phenotype myofibroblasts through Akt and Smad2/3 signaling pathways. Furthermore, the pro-collagen1a1 and fibronectin synthesis of myofibroblasts in 3D was higher than in 2D culture.
CONCLUSION
Our results demonstrated that the DIS can be used as a bioactive microenvironment for the study of intestinal fibrosis, providing an innovative platform for future intestinal disease modeling and screening of genes and signaling pathways.
5.Remote ischemic conditioning-induced hyperacute and acute responses of plasma proteome in healthy young male adults: a quantitative proteomic analysis.

BACKGROUND:
Long-term remote ischemic conditioning (RIC) has been proven to be beneficial in multiple diseases, such as cerebral and cardiovascular diseases. However, the hyperacute and acute effects of a single RIC stimulus are still not clear. Quantitative proteomic analyses of plasma proteins following RIC application have been conducted in preclinical and clinical studies but exhibit high heterogeneity in results due to wide variations in experimental setups and sampling procedures. Hence, this study aimed to explore the immediate effects of RIC on plasma proteome in healthy young adults to exclude confounding factors of disease entity, such as medications and gender.
METHODS:
Young healthy male participants were enrolled after a systematic physical examination and 6-month lifestyle observation. Individual RIC sessions included five cycles of alternative ischemia and reperfusion, each lasting for 5 min in bilateral forearms. Blood samples were collected at baseline, 5 min after RIC, and 2 h after RIC, and then samples were processed for proteomic analysis using liquid chromatography-tandem mass spectrometry method.
RESULTS:
Proteins related to lipid metabolism (e.g., Apolipoprotein F), coagulation factors (hepatocyte growth factor activator preproprotein), members of complement cascades (mannan-binding lectin serine protease 1 isoform 2 precursor), and inflammatory responses (carboxypeptidase N catalytic chain precursor) were differentially altered at their serum levels following the RIC intervention. The most enriched pathways were protein glycosylation and complement/coagulation cascades.
CONCLUSIONS
One-time RIC stimulus may induce instant cellular responses like anti-inflammation, coagulation, and fibrinolysis balancing, and lipid metabolism regulation which are protective in different perspectives. Protective effects of single RIC in hyperacute and acute phases may be exploited in clinical emergency settings due to apparently beneficial alterations in plasma proteome profile. Furthermore, the beneficial effects of long-term (repeated) RIC interventions in preventing chronic cardiovascular diseases among general populations can also be expected based on our study findings.
