Aims: Microbial induced calcite precipitation (MICP) is a natural occurring biological process that employs the usage of
ureolytic bacteria for a wide range of applications such as improving the mechanical properties of soils. The aim of this
study was to isolate and identify local urease-producing bacteria from the limestone caves of Sarawak and characterise
their specific urease activities.
Methodology and results: Enrichment culture technique was used to isolate urease-producing bacteria. These local
isolates were identified using phenotypic and molecular characterisation. Conductivity method and biomass (OD600)
measurements were conducted to analyze and determine the specific urease activities of the local isolates. 16S rRNA
gene sequence analysis identified the bacterial isolates as Sporosarcina pasteurii, Sporosarcina luteola and Bacillus
lentus.
Conclusion, significance and impact of study: This is the first study reporting the isolation and identification of
urease-producing bacteria from Fairy and Wind Caves situated in Bau, Sarawak, Malaysia. The findings in this study
suggest the bacterial isolates are capable of inducing calcite precipitation and serve as alternative microbial ureolytic
agents.

4.Advances in microbial enzymatic elimination of ethyl carbamate in Chinese rice wine.

Ethyl carbamate (EC), a carcinogenic and teratogenic chemical that is widely distributed in various alcoholic beverages, has attracted much attention. Microbial enzymatic degradation of EC in rice wine is always efficient and attractive. In this review, we summarize the research progress and problems of microbial enzymatic elimination of EC in rice wine from three aspects: the mechanisms of EC formation in rice wine, the research progress of acid urease, and the research progress of urethanase. Then, we propose the corresponding strategies to solve the problems: screening new urethanase with satisfied enzyme properties, food-grade expression and directed evolution of the bifunctional Fe³⁺-dependent acid urease and acid urease used in combination with urethanase to eliminate both urea and EC in rice wine.

Urease decomposes urea to ammonia, and has application potential in agriculture and medical treatment. Urease proteins include structural proteins (UreA, UreB and UreC) and accessory proteins (UreD/UreH, UreE, UreF and UreG), each of them has its own unique role in urease maturation. The structural proteins form the active center of urease, and the accessory proteins are responsible for the delivery of nickel. We review here the structure and function of bacterial urease complexes, and how each protein interacts to complete the activation process. We hope to provide theoretical basis for the regulation of urease activity and the development of urease inhibitors.

6.Enhancing the value of biposy urease test by two biposy specimen collection in the diagnosis of H. pylori infection on peptic ulcer disease patient

200 patients over 18 years old were diagnosed with peptic ulcer. Two specimens were taken on each patient: from the corpus and 1/3 andral, which were put into each urease test hole. The results were diagnosed with H. pylori infection with urease test by reading biopsyspecimen after 1 hour and 24 hours. With 83.5% patient having used drugs before endoscopy, the percentage of H. pylori (+) from the corpus and the antral specimen is significantly different (65% vs 68.5%) but using specimens from both sites increases significantly the H. pylori positivity from 65% to 78%

8.Mycological findings of Trichophyton tonsurans isolated in New Orleans area.

There had been no reports of trichophyton tonsurans infection in Korea yet. We have much chances to import the organism through international travels and exchanges. But we, Korean, have no experiences to observe T. tonsurans. Author tried mycological studies with T. tonsurans insolated in New Orleans, USA. The results are as follows: Gross findings of T. tonsurans showed that fine granular surface with light yellow hue or white color and grooving in the central area on the front side and mahogany brown color on the reverse side. On the urease test, various reddish discoloration was noted. Microscopic findings showed that septated hyphae, macroconidia, chlamydoconidia and microconidia. Macroconidia were 305 septated, smooth-surfaced, and were found more frequently in the whitish colony. Microconidia were characteristic in their arrangement and shape; round or oval shaped microconidia laterally to hyphae, some were match-head like terminal swelling. Hair perforation test showed positive results in 16 strains among 19 tested strains. All these findings are similar to T. rubrum and T. menatgrophytes, the most frequent isolates of dermatophytes in Korea, and we have to pay an attention to differentiate carefully.

9.Advances of enzymes related to microbial cement.

Microbial induced calcium carbonate precipitation (MICP) refers to the natural biological process of calcium carbonate precipitation induced by microbial metabolism in its surrounding environment. Based on the principles of MICP, microbial cement has been developed and has received widespread attention in the field of biology, civil engineering, and environment owing to the merits of environmental friendliness and economic competence. Urease and carbonic anhydrase are the key enzymes closely related to microbial cement. This review summarizes the genes, protein structures, regulatory mechanisms, engineering strains and mutual synergistic relationship of these two enzymes. The application of bioinformatics and synthetic biology is expected to develop biocement with a wide range of environmental adaptability and high performance, and will bring the MICP research to a new height.

Enzyme activities of Cenococcum geophilum isolates were examined on enzyme-specific solid media. Deoxyribonuclease, phosphatase, and urease were detected in all isolates, whereas cellulase was not detected in any of the isolates. Variations in enzyme activities of amylase, caseinolysis, gelatinase, lipase, and ribonuclease were observed among isolates.
