1.Silent information regulator 1 regulates the human alveolar epithelial A549 cell apoptosis induced by cigarette smoke extract

Objective To investigate the effect of silent information regulator 1 (SIRT1)on cigarette smoke extract (CSE)-induced apoptosis of human alveolar epithelial cell (AECs).Methods The expression levels of SIRT1 protein were examined by Western Blotting in A549 cells that were treated with CSE at different concentrations.The impairment models of A549 cells induced by CSE were established.The concentration of CSE was 20.0% and the treatment time of CSE was 24 hours.A549 cells were pretreated with 20 μmol/L resveratrol(Res)or 5 mmol/L nicotinamide (NAM) for 2 h before CSE treatment.The protein levels of SIRT1,Bax and Bcl2 were further explored by Western blotting.The proportion of apoptotic A549 cells was measured using MTT.Results The expression of SIRT1 was reduced after treatment with CSE in a dose-dependent manner(P<0.05).Compared with the control group,the group of CSE treatment with 2.5%,5.0%,10.0%,20.0%and 40.0% CSE for 24h showed that the expression of SIRT1 in A549 cells was decreased by 25.6%(P<0.05),35.2%(P<0.05),38.7% (P<0.05),57.9% (P<0.05)and 64.0%(all P<0.05)separately; that A549 cell viability was decreased in a dose-dependent manner; and that A549 cell viability was decreased by 10.2%(t=2.035,P<0.05),18.4%(t=4.269,P<0.05),27.7% (t=5.963,P<0.05),59.0%(t=21.140,P<0.05)and 88.1%(t=58.827,P<0.05)separately.CSE plus 20 μmol/L Res pretreatment reversed the expression levels of SIRT1,Bax and Bcl2 in A549 cells (all P<0.05)and reduced the apoptosis of A549 cells.The effects of CSE on inhibiting SIRT1 pathways were aggravated by NAM (an inhibitor of SIRT1) in the A549 cells (P< 0.05).Conclusions SIRT1 plays important role in regulating the apoptosis of human alveolar epithelial A549cell induced by CSE.SIRT1 may inhibit apoptosis by up-regulating Bcl2 expression and downregulating Bax expression,which has a protective effect on A549 cells apoptosis induced by CSE.

3.Relationship of A20 mutation with clinicopathologic features and prognosis of diffuse large B cell lymphoma

Objective:To detect A20 mutation and to investigate its relationship with clinicopathologic features, prognosis, and re-sistance to therapy of diffuse large B cell lymphoma (DLBCL). Methods:A total of 104 cases with DLBCL and their clinical data were collected;follow-up was performed on a few of DLBCL patients. The expression of P-gP and Ki-67 protein was detected with immuno-histochemical staining. The A20 gene mutation in exons 3, 6, and 7 were examined by polymerase chain reaction and DNA sequencing. Finally, the correlation of genetic abnormality of A20 with clinicopathologic features was analyzed. Results:Missense mutation in ex-on 3 of A20 gene was identified in 18 of 104 patients (17.3%). The A20 gene mutation at site 73 of exon 3 was greater in the cases with activated B cell-like-DLBCL than with germinal center B-cell-like-DLBCL (18.5%vs. 2.5%, P=0.010). In contrast to the advanced clin-ical stage and high International Prognostic Index (IPI) cases, the rate of A20 mutation was superior to the opposite (P<0.05). The ex-pression for P-gP was higher in the cases with mutation than that of those with wild-type A20 gene (16/18 vs. 52/86, P=0.021). The dif-ference in A20 mutation between the groups of low and high positive expression for Ki-67 (1/17 vs. 26/60, P=0.030) was significant. DLBCL with A20 mutation had an increasing recurrence tendency (P=0.06) and a worse survival (P=0.016) compared with those with wild-type A20 gene. Conclusion:The A20 mutation has a certain influence on the clinicopathologic characteristics and survival condi-tions of BLBCL patients. Probably, A20 mutation is a factor associated with a poor prognosis of DLBCL.
4.A Method for Pretreatment and Character Extraction of Epicardial Signals Based on Template Matching

Objective To explore a method for real-time pretreatment and character extraction of epicardial signals and promote the research on the electrophysiological mechanism of atrial fibrillation (AF).Methods Firstly,an analysis was made to get the properties of epicardial signals. Then,a method based on template matching was used to extract atrial activities from epicardial signals. At last,the feature points of atrial activities could be recognized.Results The correctness of recognizing feature points of sinus rhythm and AF were 99.7% and 97.0% respectively. Furthermore,only 1~2 s were cost to compute a 10-second data record with this method.Conclusion The proposed method can extract crucial information about atrium depolarization from epicardial signals and is adequate for online analysis.
5.Inhibitory effect of CIK cells on growth and liver metastasis of human primary gastric malignant lymphoma xenograft

Objective Liver metastasis model of human primary gastric lymphoma in nude mice was reproduced for an experiment to evaluate the inhibitory effect of cytokine-induced killer (CIK) cells on the growth and liver metastasis of primary gastric lymphoma. Methods Surgical orthotopic implantation of a histologically intact liver metastasis fragment derived from a surgical specimen of a patient with metastatic gastric lymphoma was initally implanted, in order to reprodueing a liver metastasis model of human primary gastric lymphoma. Peripheral blood mononuclear cells (PBMC), isolated by blood cell separator from healthy donors and patients with primary gastric lymphoma, were incubated in vitro. rhIFN-?, rhIL-2 and anti-CD3 McAb were added to PBMC in order to prepare CIK cells as well as lymphokine-activated killer (LAK) cells. CIK and LAK cells from different donors were used in treating gastric malignant lymphoma, so as to investigate the inhibitory effect of 2 kinds of effector cells on the growth of the tumor and liver metastasis. Results The liver metastasis model of human primary gastric malignant lymphoma in nude mice was successfully reproduced. After administration of different agents continuously for 20 days (0.3ml/d), the inhibitory rates of the following 4 groups, healthy donors LAK group (2?1010/ml), healthy donors CIK group (2?1010/ml), patients CIK group (1?1010/ml) and patient CIK group (2?1010/ml), were 39.28%, 53.57%, 40.38% and 56.42%, respectively. The liver metastasis rates in control group, healthy donors LAK group, healthy donors CIK group, patients CIK group (1?1010/ml) and patient CIK group (2?1010/ml), were 100.0%, 62.5%, 50.0%, 62.5% and 37.5%, respectively. 6.Research advances on late intraocular lens dislocation after phacoemulsification

Late intraocular lens dislocation is one of the most severe late complications after phacoemulsification.It often occurs 3mo after the surgery.Different from early intraocular lens dislocation, late intraocular lens dislocation is caused by zonular dehiscence and contraction of the capsular bag many years after phacoemulsification.In recent years, the incidence of late intraocular lens dislocation gradually increases, having a risk of 0.1% after 10a and 1.7% after 25a.In the long-term follow-up patients who underwent cataract surgery, 90% had zonular insufficiency and capsular contraction.Among the multiple factors which may contribute to zonular weakness and capsular contraction, pseudoexfoliation is the most common cause, accounting for 50% of all the cases.Other risk factors include aging, high myopia, uveitis, trauma, previous vitreoretinal surgery, retinitis pigmentosa, diabetes mellitus, atopic dermatitis, previous acute angle-closure glaucoma attack, and connective tissue disorders.The understanding of these predisposing factors will suggest necessary preventions for high-risk patients in the future.
7.Effect of compound trabeculectomy for primary angle - closure glaucoma with persistent high intraocular pressure

AIM: To evaluate the clinical effects of compound trabeculectomy for primary angle-closure glaucoma with persistent high intraocular pressure.
●METHODS:Thirty-three cases ( 34 eyes ) with primary angle - closure glaucoma were executed compound trabeculectomy. All patients were divided into two groups: group A: 18 cases ( 18 eyes ) were executed compound trabeculectomy under high intraocular pressure higher than 35mmHg after using lOP-lowering medicine for 48-72h; group B: 15 cases (16 eyes) were executed compound trabeculectomy, the intraocular pressure was normal after using lOP-lowering medicine.
●RESULTS:Thirty-four eyes were successfully completed surgery with follow-up of 6 to 18mo and there were no serious complications, including suprachoroidal hemorrhage. The visual acuity before operation in group A and group B were 0. 02 ± 0. 01, 0. 04 ± 0. 02, respectively. And the postoperative visual acuity in group A and group B increased 0.2±0. 06, 0. 3±0. 07, respectively (P<0. 01). All of them, intraocular pressure with 30 eyes were controlled in 9-23 mmHg, 3 eye was controlled by normal through adding in several kinds different lOP-lowering medicine. One eye was failure. Compared to preoperation, postoperative intraocular pressure of two groups were decreased by normal, which were 17. 9 ± 9. 1 mmHg and 15. 4±8. 4mmHg, respectively (P<0. 01). There was no significant difference between two groups for postoperative intraocular pressure.
●CONCLUSION: The primary angle-closure glaucoma with persistent high intraocular pressure should be executed compound trabeculectomy decisively, even under high intraocular pressure, which prevent further damage and loss of visual function. As long as to fully consideration the preoperation, intraoperative careful operation, careful nursing, it is safety and effective to have compound trabeculectomy under the continuous high intraocular pressure for primary angle-closure glaucoma.
9.Establishment of lung metastasis model of human primary malignant melanoma in the small intestine in nude mice

Objective To provide an ideal animal model for exploring the pathogenesis and experimental treatment of malignant melanoma in the small intestine.Methods Fresh tissue of lung metastatic lesions from patients with malignant melanoma of the smallintestine were transplanted into mucosa of the small intestine in nude mice.After 4 times of screening.the tissue of the lung metastatic lesions from the nude mice were transplanted into the small intestine of additionat nude mice.Tumorgenecity and metastasis of transplanted tumors were observed,and were analyzed by morphology,karyotype and flow cytometry.Results A lung metastatic model of human primary malignant melanoma of the small intestine in nude mice was successfully constructed and named HSIM-0601.Massive melanin granules and melanin complex were seen in cytoplasm of tumor cells.Immunohistochemical straining of S-100 and HMB-45 were positive.The number of chromosome was between 57 and 59.DNA index was 1.49.HSIM-0601 was passed for 26 generations.A total of 173 nude mice were used for tumor transplantation.The growth rate of the transplanted tumors and resuscitation rate of liquid nitrogen cryopreservation were both 100%.In HSIM-0601.lung metastasis rate was 100%(173/173)and lymph node metastasis rate was 61.3%(106/173).Conclusions The HSIM-0601 successfully mimics the natural clinicopathologic course of patients with primary small intestinal melanoma,and provides an ideal animal model for research on pathogenesis,metastasis and experimentM therapy of malignant lymphoma in the small intestine.
10.Modified design of biotype self-locking hip joint prosthesis

BACKGROUND:Hip prosthesis needs to meet the good structural compatibility with femoral bone marrow cavity and the biomechanical properties of the original bone tissue. However, the difference of individual anatomical characteristics and the stress concentration in the local area after replacement directly affect postoperative combination of femoral prosthesis stem, force flow transfer and femoral reconstruction, and finaly result in aseptic loosening of hip joint in some patients.
OBJECTIVE:To analyze the biomechanical characteristics of modified biotype self-locking hip prosthesis by the method of finite element analysis, and to provide the reference for the application of self-locking hip prosthesis in hip replacement.
METHODS:Experimental design contained traditional biotype hip joint prosthesis group and self-locking hip joint prosthesis group. In accordance with the finite element models of self-locking hip prosthesis, 28 kinds of screw fixation were established. Three-dimensional models were generated in the Minics system. After the optimization of prosthesis, prosthesis was designed. In the Solidworks software, the femur was re-positioned and set for contacting set in Ansys to analyze the stress distribution and displacement distribution of the femoral-prosthesis-screw by Ansys. We compared the peak of stress and displacement of the femur and prosthesis, and analyzed the biomechanical stability of prosthesis.
RESULTS AND CONCLUSION:(1) By using the Ansys finite element method, we analyzed the stress and displacement distribution of the femoral-prosthesis-screw. The minimum stress peak value of the femur was 15.698 MPa. The minimum stress peak value of prosthesis was 45.491 MPa. The minimum stress peak value of screw was 8.359 MPa. The minimum displacement peak value of femur was 1.125 3 mm. The minimum displacement peak value of prosthesis was 1.039 6 mm. The minimum displacement peak value of screw was 0.566 4 mm. (2) Compared with the traditional biotype hip joint prosthesis group, in 28 kinds of self-locking hip joint prostheses, the stress was minimum in the group E. The displacement was minimum in groups A-F. Regarding comprehensive stress and displacement, groups A-F were the best combination. The maximum stress of femoral-prosthesis-screw was 18.936, 59.494 and 12.382 MPa. The maximum displacement of femoral-prosthesis-screw was 1.125 3, 1.039 6 and 0.626 3 mm. (3) These findings indicated that the stress and displacement parameters of groups A-F was the best parameter combination in 28 kinds of self-locking hip joint prostheses, and it is the recommended index of researching and developing self-locking hip prosthesis.
