2.Gene modified dendritic cell vaccine in anti-tumor immunity

Dendritic cells(DCs) are the most potent antigen presenting cells,which play a vital role in the initiation of immune response by presenting antigens to T cells and followed by induction of T-cell response.This established function of dendritic cells has attracted much attention in efforts to develop useful vaccines for the treatment of cancer.In several studies,DCs were genetically engineered by tumor-associated antigens or by immune molecules such as costimulatory molecules,cytokines,and chemokines.These new DC vaccines are more powerful in stimulating anti-tumor immunity.This review focuses on DC gene modifications for enhancing the multiple effector functions of DC,a variety of transferred genes,and recent clinical trials.
3.Clinical observation on trimetazidine in treatment of chronic heart failure

Objective To explore the clinical effect of chlorpromazine(TMZ)in treatment of chronic heart failure(CHF).Methods 70 patients with CHF were randomly divided into control group and treatment group 35 cases in each gronp,control group was given conventional treatment,treatment group on the basis of additional application TMZ,20mg/times,3 times/d.8 weeks after the observation and comparison of two groups of cardiac function in improving the situation.Results Treatment group before the treatment of LVEF(38.0 ± 2.4)% After treatment,LVEF(48.0 ±2.3)% ;control group before treatment LVEF(38.2 ±2.2)% after treatment,LVEF(42.0 t2.5)% ;Twosets of treatment LVEFdifference statistically significant(t =6.9419,2.8482,allP < 0.05); Treatment group compared with control groups after treatment improve even more obvious(t =4.3147,P <0.05)o Control groups and then hospitalized 10 cases(28.5%),the treatment group hospitalization 3 cases(8.5%),both groups rate difference statistically significant(x2 =4.629,P <0.05).Both groups were no deaths,no serious adverse effects.Conclusion General against heart failure therapy on the basis of the combined with TMZ could significantly improve the cardiac function of patients with chronic heart failure,reducing hospitalization rates.
4.Clinical effect of different doses of anthracyclines on breast cancer patients before chemotherapy

Objective To explore the effect of different doses of anthracycline on preoperative chemotherapy of breast cancer.Methods In 40 cases of breast cancer patients using small doses of anthracycline therapy, and classified as the control group, the other 40 patients using high-dose anthracycline therapy, and classified as observation group.Then observe two groups of patients were compared;the patients were in our hospital from January to December 2016 were treated.Results The total effective rate was 82.5% in the observation group and 62.5% in the control group, the difference between the two groups was statistically significant (P<0.05).The incidence of adverse reactions in the observation group was 0, the control group was 27.5%, and the incidence of adverse reactions in the two groups was statistically significant (P<0.05).Conclusion Different doses of anthracyclines on the clinical effect of chemotherapy before surgery for breast cancer research found that different the clinical effect of different doses produced by E100C and E75C, E100C can improve the treatment effect of the patients, and reduce the occurrence of toxicity, so it is worthy of reference.

6.Design on Intelligent Sickbed Calling System Based on PLC

Objective To enrich the control ways of Sickbed Calling System. Except for real time communication between sickrooms and the nursing station, situations in every sickroom are excepted to be learned conveniently, thus realizing scientific sickroom management of the hospital. Methods The Calling System of Sickbed was designed by using programmable controller instead of singlechip control system. Results The experiment demonstrated that the Sickbed Calling System designed by programmable controller was simpler and more popular. Conclusion Calling System of Sickbed designed by programmable controller can realize the management of Sickbed Calling System promptly and accurately. It has a bright application future.
7. β-amyloid and Alzheimer disease: Recent progress

Alzheimer disease (AD) is the most common form of dementia in aged population; it is a neurodegenerative disorder with unknown etiology and pathogenesis. The major pathological findings of AD include senile plaques, neurofibrillay tangles, and apoptosis-related regional synaptic and neuronal degeneration. The brain senile plaques are mainly produced by β-amyloid, a peptide with neurotoxicity and vasculotoxicity. Due to the polygenic and heterogeneous characteristics of AD, Amyloid Cascaded Hypothesis is currently drawing increasingly more attention. This paper reviews the relation between Aβ and AD published recently.
9.Retinal lesions and potential confounders in mouse models

During the application of spontaneous or genetically engineered mouse models for retinal diseases,some phenotypes of the models might not be related to the gene (s) of interest or intentional manipulation.This problem often arises from founder effect of inbreed mouse strains.The most impact experiences in vision research community are mouse models incorporated with mutations in Crb1 (rd8),phosphodiesterase 6 beta (Pde6b) (rd1),Gnat2 and RPE65 genes.This paper reviewed the most relevant articles on this matter.We encourage that vision researchers who apply or develop mouse lines to study retinal diseases should carefully track and check the genetic background of their stains and/or choose proper controls in the study design.
10.Inhibitory effect of dendritic cells combined with cytokine-induced killer cells on the growth and liver metastasis of human primary malignant melanoma of the small intestine

Objective To investigate the inhibitory effect of dendritic cells (DCs) and homologous cytokine-induced killer cells (CIKs) on the growth and liver metastasis of human primary malignant melanoma of the small intestine. Methods A histologically intact liver metastasis fragment derived from a surgical specimen of patients with primary malignant melanoma of the small intestine was implanted in the mucous layer of small intestine in nude mice to construct the liver metastasis model of the malignant melanoma. Peripheral blood mononuclear cells from healthy donors and patients with malignant melanoma of the small intestine were isolated. DCs and CIKs were separately produced according to corresponding culture method, and the DC-CIKs were harvested after coculorthotopically implanted in 56 nude mice, and 72 hours later, all the mice were equally divided into normal saline group (0.3 ml), mitomycin treatment group (400 μg/0.3 ml), healthy donor CIK group (6 × 107/0.3 ml),healthy donor DC-CIK group (3 × 107/0.3 ml), autologous CIK treatment group (6 × 107/0.3 ml), autologous DC-CIK group (6 × 107/0. 3 ml) and DC-CIK (6 × 107/0.3 ml) + mitomycin (400 μg/0. 3 ml) treatment group. Mice in the experimental groups were administered the agents once daily by vena caudalis injection for 28 days. Mice were sacrificed 72 hours after treatment, and the orthotopic xenografts were removed for weighing,and the liver metastases were simultaneously evaluated by macroscopy and microscopy. All data were analyzed using the analysis of variance, LSD test and chi-square test. Results A liver metastatic model of human primary malignant melanoma of the small intestine ( HSIM-0603 ) was established. Of the control group, healthy donor CIK group, healthy donor DC-CIK group, autologous CIK treatment group, autologous DC-CIK group and DC-CIK +mitomycin group, the tumor weights were ( 1.18 ± 0. 17), (0.71 ± 0.06), (0.68 ± 0. 15 ), (0. 43 ± 0.08 ),(0.67 ± 0.07 ), (0.37 ± 0.08 ) and (0.20 ± 0.05 ) g, respectively; the tumor inhibition rates were 0, 39.82%,42.37%, 65.56%, 43.22%, 68.64% and 83.05%, respectively; liver metastases were detected in 8, 8, 5,4, 4, 3, 2 rats, respectively, in the above mentioned groups. There were significant differences in the tumor weight, tumor inhibition rate and liver metastasis rate among the seven groups (F = 152. 300, x2= 200. 538,94. 325, P < 0.05 ). Conclusions The liver metastatic model could be applied to the studies on the pathogenesis,invasion, metastasis and treatment of human primary malignant melanoma of the small intestine. DC-CIK has the potential in inhibiting the growth and liver metastasis of the malignant melanoma of the small intestine.
