BACKGROUND:Hip prosthesis needs to meet the good structural compatibility with femoral bone marrow cavity and the biomechanical properties of the original bone tissue. However, the difference of individual anatomical characteristics and the stress concentration in the local area after replacement directly affect postoperative combination of femoral prosthesis stem, force flow transfer and femoral reconstruction, and finaly result in aseptic loosening of hip joint in some patients.
OBJECTIVE:To analyze the biomechanical characteristics of modified biotype self-locking hip prosthesis by the method of finite element analysis, and to provide the reference for the application of self-locking hip prosthesis in hip replacement.
METHODS:Experimental design contained traditional biotype hip joint prosthesis group and self-locking hip joint prosthesis group. In accordance with the finite element models of self-locking hip prosthesis, 28 kinds of screw fixation were established. Three-dimensional models were generated in the Minics system. After the optimization of prosthesis, prosthesis was designed. In the Solidworks software, the femur was re-positioned and set for contacting set in Ansys to analyze the stress distribution and displacement distribution of the femoral-prosthesis-screw by Ansys. We compared the peak of stress and displacement of the femur and prosthesis, and analyzed the biomechanical stability of prosthesis.
RESULTS AND CONCLUSION:(1) By using the Ansys finite element method, we analyzed the stress and displacement distribution of the femoral-prosthesis-screw. The minimum stress peak value of the femur was 15.698 MPa. The minimum stress peak value of prosthesis was 45.491 MPa. The minimum stress peak value of screw was 8.359 MPa. The minimum displacement peak value of femur was 1.125 3 mm. The minimum displacement peak value of prosthesis was 1.039 6 mm. The minimum displacement peak value of screw was 0.566 4 mm. (2) Compared with the traditional biotype hip joint prosthesis group, in 28 kinds of self-locking hip joint prostheses, the stress was minimum in the group E. The displacement was minimum in groups A-F. Regarding comprehensive stress and displacement, groups A-F were the best combination. The maximum stress of femoral-prosthesis-screw was 18.936, 59.494 and 12.382 MPa. The maximum displacement of femoral-prosthesis-screw was 1.125 3, 1.039 6 and 0.626 3 mm. (3) These findings indicated that the stress and displacement parameters of groups A-F was the best parameter combination in 28 kinds of self-locking hip joint prostheses, and it is the recommended index of researching and developing self-locking hip prosthesis.

Objective To assess dietary glycemic load (GL) in young adult males with acne vulgaris and to evaluate its association with severity of skin lesions,in hope to provide evidences for comprehensive treatment of acne vulgaris. Methods The Global Acne Grading System (GAGS) was used to evaluate the severity of acne, and food frequency questionnaire (FFQ) and 24-hour dietary recall (24-HDR) to assess GL.Differences in GL and body mass index (BMI) were assessed by t test and analysis of variance, and Logistic regression analysis was conducted to analyze the factors influencing BMI and GL.Results This study included 99 young adult males with acne vulgaris who were 19-24 years old.Logistic regression analysis revealed that GL was the risk factor of BMI,and higher GL was always associated with increased BMI (β3 =1.119,P < 0.05,OR =3.062,95% confidence intervals:1.044-8.986).No factors were found to have influences on GL.The patients with a disease duration of > 6 months showed a higher GL (t =2.947,P < 0.01),but similar BMI (t =0.798,P >0.05) compared with those with a disease duration of ≤6 months.Significant differences were observed in GL (F =76.296,P < 0.01),but not in BMI (F =3.142,P > 0.05),among the three groups of patients with different GAGS score.GL was higher in patients with GAGS score of ≥39 than those with GAGS score of 1-18 (P <0.01) and GAGS score of 19-30 (P < 0.01),and higher in patients with GAGS score of 19-30 than in those with GAGS score of 1-18 (P < 0.01).Conclusion GL is positively correlated with the severity of lesions and disease duration in young adult males with acne vulgaris.

Objective To explore the role of ERK1/2 in the central pathogenesis of migraine. Methods Healthy adult male SD rats were randomly divided into five groups:normal group (group C),sham operation group(group C),migraine model group(group M),DMSO group (group D)and PD-98059group (PD group),with 12 rats in each group. The extracellular discharge frequency in the spinal trigeminal nucleus was recorded and ERK1/2 phosphorylation was tested. Results (1) The percentage of extracellular discharge frequency change:Two hours after treatment, the percentage of discharge frequency change was (325.9±47.32)%.The percentage of extracellula discharge frequency change in group M (325.9±47.3)% was higher than that in group N (100.0± 0.0) % and group C(107.3± 16.4)%.There was no significant difference in the percentage of discharge frequency change between group D(319.3±42.5) % and group M (325.9±47.3) %.The percentage of discharge frequency change in group PD(218.5±31.7)% was lower than that in group M(325.9±47.3)% and group D(319.3± 42.5)%.(2) ERK1/2 phosphorylation:the ERK1/2 phosphorylation in group M and group D was higher than that in group N and group C.There was no significant difference in ERK1/2 phosphorylation between group D and group M.The ERK1/2 phosphorylation in group PD was lower than the other four groups. Conclusion During the process of central sensitization to migraine, neuronal excitability and ERK1/2 phosphorylation were increased.ERK1/2 inhibitor (PD98059) reduced ERK1/2 phosphorylation and neuronal excitability. These indicated that ERK1/2 may play a role in central sensitization of migraine in rats.

Background and purpose:Previous studies have suggested Na+-Ca2+ exchanger isoform 1 (NCX1) as a key component of calcium homeostasis was involved in the tumorigenesis. However, the role of NCX1 and calcium signal in tumorigenesis of hepatocellular carcinoma (HCC) has not been explored. This study aimed to investigate the effect of NCX1 on cell proliferation and migration of HCC HepG2 cells in vitro and the possible mechanism. Methods:Both the real-time fluorescent quantitative polymerase chain reaction (RTFQ-PCR) and Western blot were applied to assess the expression of NCX1 mRNA and protein in normal hepatic cells (LO2), HCC cell line (HepG2), human normal hepatic tissues and hepatocellular carcinoma tissues. The change of intracellular calcium signal in LO2 and HepG2 cells via acti-vated NCX1 channel in the presence or absence of Na+ was examined by a confocal laser scanning microscope. The effects of NCX1 special inhibitor KB-R7943 on cell proliferation and migration of HepG2 cells were measured by MTT and cellscratch test. Results:Both mRNA and protein expression of NCX1 were higher in HCC tissues and cell line HepG2 than in the normal tissues and cell line LO2 (P<0.05). The activation of NCX1 channel induced a slight rise in cytoplasmic Ca2+concentration ([Ca2+]cyt) in normal cells, but caused a marked increase in cancer cells. And the NCX1 activation induced intracellular calcium increase was significantly reversed by NCX1 inhibitor KB-R7943 (P<0.05). Both NCX1-mediated proliferation and migration of HepG2 were also significantly attenuated by the KB-R7943 (P<0.05).Conclusion:NCX1 is up-regulated in HCC cells and tissues. The activation of NCX1 mediates intracellular calcium homeostasis. The inhibition of NCX1 activity can suppress the proliferation and migration of HepG2 cells. It is suggested that NCX1 may be involved in the development and progression of HCC.
