1.Effect of Electroacupuncture on Hand Dysfunction after Stroke

Objective To observe the effect of electroacupuncture on hand dysfunction after stroke. Methods Forty hemiplegic patients after stroke were selected from a multi-center large-scale trail, belonged to rehabilitation group (n=20) and electroacupuncture group (n=20), who accepted routine rehabilitation and additional electroacupuncture, respectively, for six weeks. They were assessed with Brunnstrom Grade, Fugl-Meyer Assessment (FMA), Wolf Motor Function Test (WMFT) and modified Barthel index (MBI) before treatment, two, four and six weeks during treatment, and twelve weeks after treatment. Results There were differences between two groups in scores of FMA, WMFT and total time to complete WMFT twelve weeks after treatment (t>1.900, P<0.05). There was no difference between two groups in scores of MBI and Brunnstrom Grade (t=0.532, Z<1.79, P>0.05), but electroacupuncture group seemed to improve more. Conclusion Elec-troacupuncture may improve the muscle strength for patients in flaccid paralysis, and promote the recovery of hand function for patients af-ter stroke.

Oral tuberculosis is one of oral infectious diseases and there are some difficulties in its diagnosis.This article describes a case of oral gingival tuberculosis for that the diagnosis had not been confirmed by biopsy.Local injection with streptomycin was undertaken.1 month after treatment the gingival lesion was improved.
4.Analysis of nutrition and health and mental situation on puerperal women in Chongqing

Objective To investigate nutrition and health behavior situation and mental situation on puerperal women of Chongqing,in order to provide scientific basis on their pertinent health education. Methods 424 puerperal women were chosen to take health care questionnaire and anxiety self-test table,and investigated by interviewers who had been through professional training,and then doing statistical analysis. Results Women in puerperal in Chongqing had some unreasonable nutrition and health knowledge and mental situation,diet behavior inhibition and so on. Among them,only 49.51% needed calcium supplementation,only 79.37% held that they could have fruit during confinement in childbirth,as high as 66.02% went out after confinement,only 33.49% washed during puerperal,20.39% had over light anxiety. Conclusion It is necessary to reinforce health education and puerperal health guide to puerperal women in Chongqing to improve nutrition and health level and mental situation of puerperal women.
5.Effects of LPS and PMA on proliferation of human len epithelial cells and expression of epidermal growth factor receptor in human len epithelial cells

Objective To observe the effects of LPS and PMA on proliferation of human len epithelial( HLE )cells and expression of epidermal growth factor receptor(EGFR) in HLE cells.Methods The expressions of EGFR protein of HLE cells from felus,adult lens age-related cataract and cultured HLE cells were detected by immunohistochemical staining.The expression of EGFR mRNA was detected by RT-PCR.The effects of LPS (0.5,1.0,2.0 mg?L-1 ) and PMA(25,50,100 nmol?L-1 )on proliferation of HLE cells were detected by MTT colorimetry method,and the EGFR mRNA expression in HLE cells was determined by RT-PCR. Results The expressions of EGFR protein and mRNA were positive in HLE cells from felus,adult lens age-related cataract and cultured HLE cells.The proliferation rates of HLE cells treated with 0.5,1.0,2.0 mg?L-1 LPS were (3.21?0.42)%,(12.25?1.34)% and (36.67?3.65)%,respectively.The proliferation rate of HLE cells in 2.0 mg?L-1 LPS group was higher than those in 0.5 and 1.0 mg?L-1 LPS groups(F=7.709,P0.05).PMA(25,50,100 nmol?L-1 )could not effect the expression of EGFR mRNA in HLE cells .Conclusion Inflammation stimulant factor such as LPS can promote the proliferation of HLE cells by increasing the expression of EGFR and result in occurrence of posterior capsular opacifition(PCO).
6.Effects of vitamin A deficiency and its supplementation on tibial growth in rats

Objective To study the effects of vitamin A deficiency(VAD)and its supplementation on the skeletal growth in rats.Methods Eighteen female Wistar rats were randomly divided into VAD(Vit A 400 IU/kg diet,n=3),supplement(n=9)and control groups(normal diet Vit A 6 500 IU/kg,n=6).The supplement groups were randomly divided into 3 sub-groups,and their diet was changed to Vit A 6 500 IU/kg diet from 14 d after pregnancy,0 d and 4 weeks after birth respectively until they were 7 weeks age old.When they were 0 day,4 and 7 weeks old,their body weight,the length of the tibia,serum Vit A level were measured,and the histology of growth plate was also observed.Results At every measuring times,the serum Vit A level was significantly lower,and the tibial length was shorter in VAD group than in the control(P
7.Effect of different transfection reagents and injection methods in mice testicular injection on the expression of exogenous gene.

The purpose of this study was to study the effect of three different transfection reagents (Lipofectamine™ LTX & PLUS™, Lipofectamine 2000 and Nano-PAMAM-D) and three different testicular injection methods (rete testicular injection, seminiferous tubules injection and testicular interstitial injection) on the efficiency of production transgenic mice. After the mixtures of plasmid DNA (pEFP-C1) and transfection reagent were injected with different testicular injection methods, the sperm density, vitality, positive sperm rates and PCR positive transgenic mice rate were examined 30 days after injection. The results showed that the damage degree from slight to serious of three transfection reagents was Lipofectamine™ LTX & PLUS™, Lipofectamine 2000, and PAMAM-D. The sperm positive rates with green fluorescence of these three groups were 35.65%±0.69%, 12.86%±0.35% and 10.04%±0.20%, respectively. The PCR positive rates of transgenic newborn mice were 29.17%, 13.70% and 5.88%, respectively. Among the groups of different testicular injection methods, the damage degree from slight to serious was rete testicular injection, seminiferous tubules injection, and testicular interstitial injection, whereas the sperm positive rates with green fluorescence were 35.13%, 15.13%, and 0%, respectively. The PCR positive rates of transgenic newborn mice among different testicular injection groups were 33.3%, 12.5%, and 0.0%. The combination of rete testicular injection and Lipofectamine™ LTX & PLUS™ had the lowest toxicity and highest transgenic efficiency in the production of transgenic mice.

8.Effect of dietary vitamin A intake on plasma vitamin A concentration in preschool children of Banan district, Chongqing, China

Objective:To study the relationship between dietary vitamin A intake and plasma vitamin A concentration , and establish the theoretical basis for dietary intake predicting vitamin A nutritional status . Methods:By using cluster sampling , 492 children aged 2-7 years in kindergartens in Banan district of Chongqing were selected .A cross-sectional nutrition and health survey was conducted , including the clinical examination , anthropometry , laboratory test and dietary survey .Results: Among the children surveyed, 229 were boys, and 263 girls, the mean age was (4.54 ±0.87) years, height (107.50 ± 7.20) cm, and weight (18.42 ±3.41) kg, the mean value of plasma vitamin A was (1.04 ±0.30)μmol/L.The prevalence of marginal vitamin A deficiency ( MVAD) was 43.5%.No cases of severe clinical vitamin A deficiency were found (plasma vitamin A≤0.35μmol/L).Clinical examination found no conjunctiva, corneaor skin abnormalities, and no Bitot’s spots.Prevalence of the last two weeks colds were 27.4% (135/492), no diarrhea and other gastrointestinal or digestive diseases were found .The proportion of insufficient dietary vitamin A intake ( <600 μg RE/d) was as high as 50.0%.By using correlation analysis, plasma retinol concentrations were related to dietary vitamin A intake (r=0.162, P<0.001), and to dietary energy intake (r=0.107, P=0.017).After adjustment for the effects of other non-dietary factors on vitamin A deficiency , the multivariate logistic regression showed that vitamin A-rich foods of liver intake=0 g/d (OR=1.95, 95% CI:1.05-3.61, P=0.034), vitamin A-rich fruits intake=0 g/d (OR =1.55, 95% CI: 1.03 -2.33, P =0.034), vitamin A-rich vegetables intake<200 g/d (OR=3.47, 95%CI:1.37-8.75, P=0.009) were important risk factors of vitamin A deficiency .But we had not found the correlation between the intake of meat , eggs and milk and vita-min A deficiency .Conclusion:Dietary factors may be the major risk factor of vitamin A deficiency in the three kindergartens .The dietary vitamin A intakes are significantly related to plasma retinol concentra-tions, and the vitamin A-rich foods intakes can predict the body ’ s vitamin A nutritional status .
9.Protective effect of quercetin on L-02 cells by inhibiting DNA damage of INH-induced mitochondrial oxidative stress

AIM: To investigate the role of reactive oxygen species ( ROS)-mediated mitochondrial oxidative injury in isonicotinyl hydrazide ( INH)-induced DNA damage and the protective effect of quercetin on L-02 cells.ME-THODS:The injury model of hepatocyte L-02cells in vitro induced by INH was established .The cells were divided into control group, INH group, low-dose quercetin group and high-dose quercetin group.The DNA damage of L-02 cells was evaluated by the comet test .The mitochondrion was prepared , and the level of mitochondrial ROS and the value of mitochondrial membrane potential (ΔΨm ) were detected by fluorescent probes DCFH-DA and rhodamine 123.The content of MDA was measured by TBA method .The activity of SOD was assessed with the xanthine oxidase method .The protein expression of Bcl-2 and Bax was determined by Western blotting , and the value of Bax/Bcl-2 was calculated .RESULTS:INH induced obvious DNA damage , increased the level of mitochondrial ROS , the content of MDA and the value of Bax/Bcl-2, and markedly reduced the value of ΔΨm and the activity of SOD in the L-02 cells.Quercetin attenuated DNA dam-age, reduced the level of mitochondrial ROS , elevated the value of ΔΨm , declined the content of MDA , increased the ac-tivity of SOD and decreased the value of Bax/Bcl-2 in the L-02 cells.CONCLUSION:INH induces DNA damage in L-02 cells by generation of mitochondrial oxidative stress .Quercetin has a protective effect on L-02 cells to attenuate the INH-in-duced DNA damage by inhibiting ROS-mediated mitochondrial oxidative damage .
10.MRI characteristics and evaluation of cryptococcus meningoencephalitis in HIV-negative patients

Objective To search MRI features of patients with HIV-negative cryptococcus meningoencephalitis (CM)and evaluate the value in judging the prognosis of CM.Methods The findings of cranial MRI and prognosis were retrospectively analyzed in 19 cases with India-ink capsule staining-proved HIV-negative CM in our department.The prognosis was evaluated by Glasgow Outcome Scale:score 1=death,score 2=persistent vegetative state,score 3=severe disability,score 4=moderate disability,score 5=good recovery. The patients with score 1-3 were classified as the group of poor prognosis,while those with score 4,5 as good prognosis.Results Abnormal manifestations on cranial MRI were observed in 84.2%(1 6/1 9)cases,meninges enhanced in 84.2% (1 6/1 9),brain edema in 31.6%(6/1 9),brain parenchyma lesion in 47.4% (9/1 9 ),vasculitis in 5.2% (1/1 9 ).The mean duration of follow-up was 3.5 years,The prognosis of 5/9(44.4%)cases with brain parenchyma lesion confirmed by cranial MR was poor.Conclusion The MRI manifestation of HIV-negative CM is diverse,and brain parenchyma lesion confirmed by cranial MRI may associated with poor prognosis.
