Objective To study the changes of Caspase8 and Caspase3 protein in HL-60 cells during apoptosis induced by psoralen (PSO) plus ultraviolet A (UVA). Methods HL-60 cells were treated with PSO, extracted from Chinese medicine psoralea fruits in different concentrations, plus UVA of wave length 360 nm in different irradiation time. The apoptosis ratios, ultrastructure changes and the expression of Caspase8 and Caspase3 protein were detected. The factorial design and analysis of variance were used to analyze the interaction among the factors. Results PSO, UVA and PUVA all increased the apoptosis ratios and up-regulated the expression of Caspase8 and Caspase3 protein, but the effects of PUVA were the strongest. There were obvious changes about apoptosis under the electron microscope after treatment with PUVA. Conclusions PUVA can induce the apoptosis of HL-60 cells and activate Caspase8 and Caspase3 gene.

With high incident of cancer and chronic disease, it is more and more important to develop palliative care; as developing later, there is neither unified palliative care mode and related policy, nor mature assessment collaboration in China. It pointed out that Australia as an advanced country with stable development of palliative care and assessment collaboration, called palliative care outcomes collaboration (PCOC). It expatiated the PCOC on structure, function and operation. China should learn from the mode of Australia palliative care assessment and management collaboration, find references to improve and further develop Chinese palliative care assessment collaboration.

Objective:To investigate the effect of all-trans retinoic acid (ATRA) on the differentiation of marrow stromal stem cells(MSCs)into neurons. Methods: Rat MSCs were expanded as undifferentiated cells in culture for 5-7 passages and pretreated with ATRA for 24 h, then induced by modified neuronal medium (MNM) for 18 h. The control cells were directly cultured in MNM without ATRA pretreatment. Immunocytochemistry was used to detect the expression of nestin, neuron-specific enolase (NSE), neuron-specific nuclear protein (NeuN) and microtuble-associated protein (MAP-2) in the experimental groups and the control cells. The change of cell proliferative cycles and the expression of retinoic acid receptor beta (RAR?) were detected before and after ATRA treatment. Results: 1. The expression of nestin, NSE, NeuN and MAP-2 was much higher than that of the control group. 2. No changes were found in the cell proliferative cycles before and after ATRA treatment. 3. No expression of RAR? was found in MSCs; but positive expression of RAR? was detected afer 24 h of ATRA treatment and it was (20.3?4.2)%. Conclusion: ATRA can activate gene transcription via nuclear receptor RAR? and promote MSCs transdifferentiation into neurons.

Objective:To study the association of RAC1 gene polymorphisms with protein expression levels of Rac1-GTP. Methods:A total of 182 healthy Hans population in Hubei were recruited. The 4 tag-SNPs in RAC1 gene were genotyped by Real time TaqMan-MGB genotyping assay. And the Rac1-GTP protein levels in plasma samples from all participants were determined by enzyme linked immunosorbent assays ( ELISA ) . The distribution characteristics of Rac1-GTP expression levels were also analyzed. Furthermore, the expression levels of Rac1-GTP were compared among different genotypes of the 4 tag-SNPs in RAC1 gene. Results:The distribution of Rac1-GTP expression levels was positive skewed in healthy Chinese Hans population. The expression levels were significantly higher in females than in males (P<0. 05),and appeared in decreased trend with age,but without significant differences (P>0. 05). Different expression levels of Rac1-GTP were observed in different genotypes for rs702482 and rs10951982 (P<0. 05). However,no significant difference was found for rs702483 and rs6954996 (P>0. 05). Conclusion:RAC1 genetic polymorphisms can potentially affect the expression levels of Rac1-GTP protein in healthy Chinese Hans population.

This study aims to construct the recombinant lentivirus vector containing specific small interfering RNA (siRNA) targeting rat CREB binding protein(CBP)gene and to identify its function of inhibiting the expressions of acetylated histone in primarily cultured hippocampal neurons. Firstly, we constructed four kinds of recombinant lentivirus siCBP. And then we used them to infect the primarily cultured hippocampal neurons, and performed real-time PCR, western blot respectively to detect the expressions of CBP. Afterwards, the most effective lentivirus siCBP was used to infect the primarily cultured hippocampal neurons, and then the HAT activity and protein expressions of acetylated histone Ac-H3, Ac-H4 of the neurons were examined. By using PCR, endonuclease cutting and gene sequencing, we confirmed that the target genes were correctly cloned in lentivirus vector. Besides, CBP mRNA and protein expressions in neurons were found to be with varying degrees of decreases after infections of the four kinds of lentivirus siCBP. Furthermore, the representative and most effective lentivirus GR806 could effectively inhibit the HAT activity and the protein expressions of Ac-H3, Ac-H4 in neurons. It provides the experimental basis for the subsequent application of siCBP to clarify the effects and corresponding molecular mechanism of the CBP-dependent histone acetylation on learning and memory function in hippocampus.

Objective:
To explore the relationship between parents’ unstable marital status and offspring’s suicide attempts and the possible mediating factors, and to provide reference for suicide prevention among off spring grom unst able families.
Methods:
From 2017 to 2018, 2 766 valid questionnaires were collected from freshmen in a university in Hunan province. Students completed questionnaires of childhood trauma and depression traits, and reported their parents’ marital status and suicide attempts etc.
Results:
Compared to college students with parents of stable marital status, students with parents of unstable marital status showed higher reported rate of suicide attempts and higher scores of depression trait and childhood trauma (t/χ2=2.78, 5.83, 20.30, P<0.05). Controlling possible confounding factors, unconditional Logistic regression showed that childhood trauma, unstable marital status of parents and depressive traits were positively correlated with suicide attempt(OR=1.05, 1.45, 1.08, P<0.05). The 95% confidence interval of the Bootstrapping method with four mediation pathways (parental marital status→childhood trauma→depressive trait, childhood trauma→depressive trait→suicide attempt, parental marital status→childhood trauma→suicide attempt and parental marital status→childhood trauma→depressive trait→suicide attempt) does not include zero.
Conclusion
Childhood trauma and depressive traits mediate the relationship between parental instable marriage and suicide attempts in off spring.

Chinese Critical Care Medicine 2018;30(8):760-763
Objective To analyze the main characteristics of air pollution in Nanchang City, and discuss the correlation between air pollution exposure (especially PM2.5) and the development of pneumonia related intensive care unit (ICU) patients and their lag effect. Methods 2454 patients who lived in Nanchang City admitted to ICU of Jiangxi Provincial People's Hospital from January 1st, 2014 to December 31st, 2016 were enrolled. According to the diagnosis, the patients were divided into pneumonia group (156 cases) and non-pneumonia group (2298 cases). The general clinical characteristics of patients and air pollution concentration in Nanchang in the year between 2014-2016 were collected. Multiple regression model was used to analyze the influence of meteorological factors on the condition of ICU patients associated with pneumonia. Using odds ratio (OR), the correlation intensity of air pollution exposure and pneumonia related ICU patients' development was reflected, and the confidence level of association intensity was reflected by the 95% confidence interval (95%CI). The distribution lag nonlinear model (DLNM) was established to evaluate the effect of air mass parameters on the time lag effect. Results The results of air pollution analysis in Nanchang City in the year between 2014-2016 showed that the annual average concentration of carbon monoxide (CO), sulfur dioxide (SO2) and nitrogen dioxide (NO2) was low and maintained at the same level in the year between 2014-2016. The annual average concentration of CO and NO2 increased in the year between 2014-2016, but the average annual concentration of SO2 decreased rapidly in the year between 2014-2016, and the average annual concentration of PM2.5 tended to slow down after the year between 2014-2016. The annual average concentration of PM10 decreased in the year between 2014-2016, but continued to rise in the year between 2014-2016. The annual mean concentration of O3 showed a trend of continuous increase from the year between 2014-2016. The age of pneumonia group was generally higher than that of non-pneumonia group, most of them were male, and had higher expected mortality and acute physiology and chronic health evaluation Ⅱ(APACHEⅡ) score. The average air temperature in the pneumonia group was lower than that in the non-pneumonia group on the day of entering the group, and the air pollutants such as PM2.5 and PM10 were significantly higher than those in the non-pneumonia group. The analysis of multiple regression models showed that PM2.5 and air temperature were significantly related to patients with ICU pneumonia on the day of entry (PM2.5:OR = 1.02, 95%CI =1.00-1.03,P < 0.05; air temperature:OR = 0.96, 95%CI = 0.92-0.98,P < 0.05), and the effect of PM2.5 on patients with ICU pneumonia could last for at least 5 days (OR = 1.04, 95%CI = 1.00-1.09,P < 0.05), and the effect disappeared until the 7th day. According to the analysis of the influence of different concentrations of PM2.5 on ICU pneumonia patients, when the PM2.5 concentration reached 200μg/m3, its effect on ICU pneumonia patients would last for 5 days (OR = 1.45, 95%CI = 1.07-1.76,P < 0.01).Conclusion PM2.5 and air temperature are significantly related to the condition of ICU patients with pneumonia, and the influence of high concentration of PM2.5 on ICU patients with pneumonia has a lag effect.
