1.Effect of estrogen receptor ? on the formation of mice endometriosis

Objective To investigate the effect of estrogen receptor ?(ER-?)on the formation of mice endometriosis.Methods Endometriosis animal model was established with 30 ER-? knockout(?ERKO)mice and 22 ER-? non-knockout(?ER)mice.All the mice were divided into three groups:intact group(?ERKO 10 and ?ER 8),ovariectomy group(?ERKO 10 and ?ER 7)and ovariectomy + estradiol treatment group(?ERKO 10 and ?ER 7).The endometriotic grafts were taken out from all the mice 2 weeks after operation,and the weight,histological structure and the expression of MMP-9 in the grafts were analyzed and compared between ?ERKO and ?ER mice.Results The endometriotic grafts were significantly heavier 2 weeks after transplantation than that before transplantation in intact group(P0.05).In addition,no significant differences were found between the ?ERKO and the ?ER mice in endometriotic grafts' weight,the histopathologic structure and the expression of MMP-9.Conclusion ER-? may throw few influence on the formation of endometriosis.
2.Relationship between childhood trauma and depressive traits in college students and the influence of coping style and automatic thinking

Objective:
To explore the moderated and mediating mechanism between childhood trauma and depressive traits, and to provide a reference for conducting the related research and intervention.
Methods:
From 2017 to 2018, a cluster sampling method was adopted to select 2 786 freshman. Childhood trauma questionnaire, state-trait depression Scale, the automatic thought questionnaire and trait coping style questionnaire was administered.
Results:
Regression analysis showed that childhood trauma, automatic thinking and negative coping was positively associated with depressive traits, and positive coping was negatively associated with depressive traits (β=0.12, 0.40, 0.14, -0.33, P<0.05). The mediation analysis showed that automatic thinking mediated the relationship between childhood trauma and depressive traits (95%CI=0.03-0.06). The results of moderation effect showed that negative coping moderated the relationship between childhood trauma and automatic thinking and the relationship between childhood trauma and depressive traits, respectively (P<0.05).
Conclusion
Childhood trauma affects depressive traits,and its relationship is influenced by boping styles and automatic thinking.
3.Unstable parental marriage and suicide attempts in adult offspring and possible mediating factors

Objective:
To explore the relationship between parents’ unstable marital status and offspring’s suicide attempts and the possible mediating factors, and to provide reference for suicide prevention among off spring grom unst able families.
Methods:
From 2017 to 2018, 2 766 valid questionnaires were collected from freshmen in a university in Hunan province. Students completed questionnaires of childhood trauma and depression traits,and reported their parents’ marital status and suicide attempts etc.
Results:
Compared to college students with parents of stable marital status,students with parents of unstable marital status showed higher reported rate of suicide attempts and higher scores of depression trait and childhood trauma (t/χ2=2.78, 5.83, 20.30, P<0.05). Controlling possible confounding factors,unconditional Logistic regression showed that childhood trauma,unstable marital status of parents and depressive traits were positively correlated with suicide attempt(OR=1.05, 1.45, 1.08, P<0.05). The 95% confidence interval of the Bootstrapping method with four mediation pathways (parental marital status→childhood trauma→depressive trait,childhood trauma→depressive trait→suicide attempt,parental marital status→childhood trauma→suicide attempt and parental marital status→childhood trauma→depressive trait→suicide attempt) does not include zero.
Conclusion
Childhood trauma and depressive traits mediate the relationship between parental instable marriage and suicide attempts in off spring.
4.Recombinant adenovirus expressing siRNA is generated to inhibit the expression of RARbeta in rat mesenchymal stem cells treated by all-trans retinoic acid.

To construct the recombinant adenovirus vector expressing specific siRNA for rat retinoic acid receptor-beta (RARbeta) gene, and to detect its effect on RARbeta expression and neuronal differentiation of all-trans retinoic acid (ATRA) treated mesenchymal stem cells (MSCs). First, we designed four pairs of siRNA sequence for rat RARbeta gene and annealed complementary oligonucleotides in vitro, then cloned double-stranded DNA in pSES-HUS vector containing U6/H1 double-promoter and recombinated with the backbone vector to construct pAd-siRARbeta plasmid. We infected MSCs by using adenovirus Ad-siRARbeta which was packaged in HEK293 cell line, then performed Real-time, Western blotting and immunoflourencence to detect the expression of RARbeta. We used combination of ATRA and MNM to induce MSCs into neural-like cells, then performed Real-time PCR and immunoflourencence to detect neuronal specific markers of induced neural cells. By using PCR, endonuclease cutting and gene sequencing, we confirmed that the target genes were correctly cloned in adenovirus vector. We could observe more than 60% RFP-positive MSCs at 24 h after adenovirus infection. The expression of RARbeta was significantly increased to 16.5 +/- 2.34 fold in ATRA treated MSCs (P < 0.05) and located in nucleus. Three of four pairs siRNA could effectively inhibit the expression of RARbeta with inhibition efficacy of (66.26 +/- 9.12)%, (48.70 +/- 5.78)%, (64.09 +/- 0.53)% (P < 0.05), especially siRNA-pool group with inhibition efficacy of (78.09 +/- 4.24)% (P < 0.01). Combination of ATRA and MNM induced MSCs into neural-like cells which expressed neuronal specific markers, Nestin, NSE, MAP-2, and Tau. Immunoflourencence result showed that about 50-88 present of cells were positive for Nestin, NSE, Tjul, however, adenovirus medicated expression of siRARbeta could effectively inhibit the expression level of neural specific proteins and the ratio of positive stained cells (P < 0.05). Therefore, we successfully constructed the recombinant adenovirus vector containing siRNA for rat RARP gene, adenovirus could effectively infect MSCs and inhibit the expression of induced RARbeta in ATRA treated MSCs, then inhibit neuronal differentiation of MSCs.
