Diagnostics is an important professional medical course concerning basic theory, basic skill and basic approach of diagnosis.Using standardized patients in clinical scene can relieve the shortage of clinical teaching resources and strained doctor-patient relationship as well as improve the clinical skills and doctor-patient communication of students. However, there were many problems in using the standardized patient;therefore, discussions were needed to find a better solution.

AIM:To investigate the feasibility and specificity of gastric carcinoma gene therapy by utilizing RNA interference (RNAi) to inhibit survivin expression in vitro and in vivo. METHODS:Small interference RNA (siRNA) homologous to survivin was designed. pTZU6+1-siRNA-survivin vector was constructed and transfected into BGC-823 cells. The transplanted BGC-823 tumor in nude mice was established to induce RNAi. The changes of survivin gene expression, tumor cell cycle and cell apoptosis were detected by flow cytometry,RT-PCR,Western blotting, immunochemistry and TUNEL. RESULTS:The expression of survivin was obviously inhibited by RNAi in vitro. The phase of cell cycle indicated the reduction of S phase, while G1/G0 phase increased. Cell apoptosis was obvious. Both the mRNA level and the protein expression of survivin decreased obviously. The tumor size reduced after treated with pTZU6+1-siRNA-survivin vector in vivo. The expression of survivin decreased in siRNA treatment group. In contrast, little change in control group in vitro and in vivo was observed. CONCLUSION:RNA interference down-regulates survivin gene expression, inhibits BGC-823 cell proliferation and induces cell apoptosis with good specificity, which may be a possible new approach for neoplasm gene therapy.

Objective To explore the effect of follow-up continuous nursing intervention on the psychological quality and medication compliance in patients with ulcerative colitis after discharge. Methods Ninety-seven patients with ulcerative colitis after discharge from October 2014 to October 2015 were recruited, and 48 cases were selected in the study group and 49 cases in the control group randomly. The control group received routine postdischarge care. The study group received postdischarge follow-up nursing intervention. After 3 months of postdischarge intervention, the psychological quality of patients in two groups was assessed by the emotion application measure scale separately. The Morisky Medication Adherence Scale 8-item version (MMAS 8-item version) was used for the assessment of the medication compliance of the two groups of patients. Results After the intervention, the patients in the study group showed better psychological quality and medication compliance than the patients in the control groups. There were significant differences between two groups. Conclusion Post-discharge follow-up nursing intervention on patients with ulcerative colitis is effective for improving the psychological quality and the medication compliance and may be helpful to improve the life quality of the patients.

Objective To discuss the effect of QGY/CDDP hepatoma lines multi-drug resistance (MDR) reversed by gene transfection of mdr1,mrp-ASODN+ultrasound contrast agent+ultrasound. Methods The QGY/CDDP cells were transfected by mdr1,mrp ASODN+ultrasound contrast agent+ultrasound irradiation respectively and detected of the various indicators:cell adhesion rate, cell sensitivity to cell drug-resistance,the mRNA expression of mdr1 and mrp gene, the expression of P-gp and MRP protein. Results After mdr1-ASODN transfection, the drug sensitivity and expression of P-gp,MRP protein of QGY/CDDP cells were smaller changes(P>0.05),and the rate of cells adherent and expression of resistance gene mRNA were obvious changes(P<0.05).After mrp-ASODN transfection, the cells adherent rate, the drug sensitivity, the expression of resistance gene mRNA and P-gp,MRP protein were obvious changes respectively(P<0.05),the experiment group(group 2')had bigger effects(P<0.05).Conclusions mdr1. mrp-ASODN+ultrasound contrast agent+ultrasound irradiation could safely partly reverse MDR of hepatoma cells, which is a potential new approach for gene therapy.

Proteomics is an emerging discipline and has been widely used in a variety of fields despite of having very short history in comparison with other disciplines. In Chongqing Medical Univer-sity, the course contents were adjusted to fulfill the most effective integration of proteomics research with postgraduate training program for medical university. Diverse teaching was advocated here and af-ter-school communications were greatly encouraged in teaching. Traditional multimedia teaching plat-form remained the main teaching way and students were organized to visit the research platform as supplementing teaching way. The overall quality and effectiveness of teaching were effectively improved by successful implementation of the above initiatives.

AIM: To investigate the effect of huayu xiaoliu fang, a Chinese medicine, on the cell cycle of human lung carcinoma cell line by serologic pharmacological method. METHODS: PGLH7 cells were incubated with rabbit serum containing huayu xiaoliu fang at different doses obtained by serologic pharmacological method. MTT assay was used to calculate the proliferation inhibition rate. The target cells were harvested to analyze the cell cycles by flow cytometry. RESULTS: The Chinese medicine-containing serum inhibited the growth of PGLH7 cells significantly. There was remarkable difference in the proliferation inhibition rate between 10% (high dose) Chinese medicine-containing serum and the control serum (P
