1.Value of serum APOM concentration for diagnosis of bacterial pneumonia and judgment of disease severity

Objective To investigate the value of serum apolipoprotein M (APOM)concentration in the diagnosis of bacterial pneumonia and its disease severity evaluation.Methods The serum APOM concentration was examined in 250 cases of pneumonia and 50 healthy controls by using the double antibody sandwich ELISA.The ROC curve was used to evaluate the diagnostic value of APOM on bacterial pneumonia and the Bayes discriminant analysis was adopted to construct the discriminant equation based on APOM concentration for evaluating its discriminant significance to the disease severity.Results The serum APOM concentration in the bacterial pneumonia group was (28.9±10.5)μg/mL,which was significantly higher than (19.4±8.93)μg/mL in viral pneu-monia group,(16.4±5.62)μg/mL in the other pneumonia group and(18.1 ±6.15)μg/mL in the healthy controls,the differences had statistical significance(P <0.01).The area under ROC curve for APOM in diagnosing bacterial pneumonia could reach 0.843, showing the good sensitivity and specificity.Moreover,the serum APOM concentration was positively correlated with the disease severity of bacterial pneumonia.The discriminant equation based on APOM concentration as well as sex,age,systolic pressure,body temperature and heart rate could better conduct the classification on the disease severity of bacterial pneumonia and its accuracy could reach to 86.7%.Conclusion The serum APOM concentration has an important clinical value for the diagnosis of bacterial pneumonia and its disease severity evaluation.
2.Construction and verification of recombinant chimeric vesicular stomatitis virus expressing rabies virus G protein

Objective To construct recombinant chimeric vesicular stomatitis virus(VSV)expressing G protein of rabies virus(RV)using VSV as vector.MethodsTo rescue the recombinant virus,G gene of VSV antigenome was replaced with G gene of RV vaccine CTN-1 strain,and co-transfected into 293T cells with helper plasmids coding T7 RNA polymerase and proteins N,P and L of VSV. The expression of RV G gene and G protein was detected by RT-PCR,immunofluorescence assay and Western blot. The recombinant virus was subcultured in Vero cells,the virus titer of different generations was detected and the virus growth curve was drawn.ResultsThe recombinant virus VSV-RVG was successfully rescued. RTPCR results demonstrated that the RV G gene was successfully inserted into the genome of the recombinant virus,and the expression of RVG protein was detected by immunofluorescence assay and Western blot. The recombinant virus was continuously passaged for 5 generations,and the virus titer was stable within 7. 5 ~ 8. 5 lgTCID50/mL.ConclusionThe recombinant chimeric VSV expressing RV G protein was successfully constructed with good genetic stability,which lays a foundation of the construction of reverse genetics technology platform based on VSV vector.
3.Association of TRAF1 genes polymorphism with increased risk of rheumatoid arthritis in Han people

Objective To detect TRAF1 gene single nucleotide polymorphism in Han population ,and analyze its association with increased risk of rheumatoid arthritis (RA) in Han people. Methods The study group was comprised with 100 healthy Han subjects and 100 Han RA patients. Six SNPs was detected by matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOFMS), and the result was analyzed by Haploview software. Fisher's exact test and χ2 test were used for statistical analysis.Results The rs4836834 A/A, A/T, T/T allele frequencies of RA group were 22.22%, 49.49%, 28.28% respectively. The rs4836834 A/A, A/T, T/T allele frequencies of healthy control group were 28.57%, 30.61%,40.82% respectively. There was significant difference (P<0.05) of rs4836834 between Hah patients with RA and healthy controls. For other five sites, including rs10818488, which had been reported to be associated with increased risk of Caucasians RA patients, there was no significant difference (P>0.05) between RA group and the control group. The linkage disequilibrium and haplotype frequency analysis results showed that there was strong linkage disequilibrium between rs7021049 and rs7021880, rs7021049 and rs4836834, rs7021880 and rs4836834, but there was no significant difference between RA group and the control group in the haplo-type frequencies of the three classes of linkage disequilibrium. Conclusion TRAF1 gene may be a susceptible gene associated with Han RA people, but its single nucleotide polymorphisms and performance may be different between Caucasians and Han people.
4.Detection of specific IgE and etiological analysis of Henoch-Schonlein purpura in acute phase in children

Objective To explore the allergic status and allergen composition in children with acute Henoch-Schonlein Purpura in Wuxi by allergen detection, thereby to establish appropriate therapeutic strategies. Methods Two hundred and forty-six patients with Henoch-Schonlein Purpura in their acute phase were examined for serum concentrations of total IgE and specific IgE using Germany AllergyScreen system. Results The positive rate of allergen-specific serum IgE was 48.7% (120/246) ,of which,positive reactions to two or more than two allergens were found in 86 cases (34.9%) and that to one kind of allergen were found in 34 cases (13.8%)The positive rate of total serum IgE was 78.9% (194/246). In inhalation allergen group, positive reaction to allergen was found in 134 cases (54.5%). The most common allergen was dust mite/flour mites (19.1%) ,and came the next were house dust(17.1%) and Penicillium notatum/Alternaria alternata(10.2%). In food allergen group,positive reaction to allergen was found in 77 cases (31.3%). The most common allergen was fish (6.5%),and came the next were egg (5.7%) and mutton (5.3%). We found no significantly different allergen positive rates between inhalation allergens group and food allergens group (χ2 = 2.59, P > 0.05). Conclusions The combined allergens attacks were more common in children patients with acute Henoch-Schonlein Purpura by serum allergen detection. Various types of the allergens are associated with the occurrence of acute Henoch-Schonlein Purpura. To reduce the harmness of allergens,it is necessary to avoid touching these allergens.
5.Effect of neuregulins on mtp53 and HIF-1? in MDA-MB-231 cells

AIM:To explore the effect and significance of neuregulins /ErbB2 receptor signal transduction pathway on mtp53 and hypoxia-iducible factor-1?(HIF-1?) in none-overexpression ErbB2 breast cancer cell MDA-MB-231.METHODS:The expression of neuregulin was detected by immunocytochemistry and Western blotting.MDA-MB-231 cells were treated with ErbB2 kinase inhibitor AG825.Proliferation was measured by MTT assay.The cell cycle and apoptosis were determined by flow cytometry.The expressions of mtp53 and HIF-1? were detected by Western blotting.The mRNA expression of HIF-1? was detected by RT-PCR.RESULTS:MDA-MB-231 cells expressed a relative higher level of neuregulin.In the results of Western blotting,the positive reaction band was found in 44 kD which coincides with the molecular weight of neuregulin.When MDA-MB-231 cells were treated with AG825,the proliferation was inhibited in time and dose dependent manners(P
6.Therapeutic Observation of Fire-needle plus Round-sharp-needle Acupuncture for Primary Trigeminal Neuralgia

Objective To observe the clinical efficacy of fire-needle plus round-sharp-needle acupuncture in treating primary trigeminal neuralgia.Method Sixty-two patients with primary trigeminal neuralgia were randomized into a treatment group and a control group, 31 cases in each group. The treatment group was intervened by fire-needle therapy at the trigger points followed by deep needling with round-sharp needle at Xiaguan (ST7), and the distal points Hegu (LI4) and Neiting (ST44) were also treated. The same acupoints were selected in the control group but treated with ordinary needling method. The clinical efficacies were evaluated after 2 treatment courses.Result The total effective rate was 93.5%(29/31) in the treatment group versus 77.4% (24/31) in the control group, and the difference was statistically significant (P<0.05).Conclusion Fire-needle plus round-sharp-needle acupuncture is an effective approach in treating primary trigeminal neuralgia.
7.Preliminary survey on cognitive function of AIDS patients in Guangxi area

Objective To understand the cognitive function of the patientwith acquired immune deficiency syndrome (AIDS) in Guangxi are.MethodEighty caseof AIDconfirmed by laboratory in the Nanning Fourth People′Hospital were selected athe patientgroup ;athe same time 88 individualwithouthe dangeroubehaviorwithin 3 monthwhich could cause human im-munodeficiency virus(HIV) infection and with negative anti-HIV antibody detection were selected from the accompanying family memberathe control group .The Chinese translation version of the Montreal cognitive assessmenscale (MoCA) waadopted to conducthe investigation and evaluation on the cognition function in the two group.ResultThe total score span in the patientgroup wa2-29 pointwith the average of (20 .04 ± 6 .13) point,of normal cognitive function and 48 cases(60 .00% ) of cognitive impairmen;the total score span in the control group wa17-29 pointwith the average of (25 .02 ± 2 .98) point,of normal cogni-tive function and 6 cases(6 .82% ) of cognitive impairmen.The scoreof MoCtotal score ,visual and execution ,naming ,delayed recall ,attention ,abstraction and orientation in the patientgroup were lowethan those in the control group ,the differencebetween the two groupwere statistically significan(P<0 .01) .The personnel distribution and proportion in the variouscore segmenta-tionhad statistical differencebetween the two group(P<0 .05) .The factorof gende,age and educational level were performed the Logistiregression analysi,the resultshowed thaonly the educational level wathe influencing facto.Conclusion Differendegreeof cognitive impairmenexisin the AIDpatientand the educational level may be factoaffecting the scale score .

Replenishing qi, nourishing yin and activating blood circulation therapy is an effective treatment for diabetic nephropathy (DN) in traditional Chinese medicine (TCM). In this article, mechanisms of this therapy in DN were summarized, which included improvement of glucose and lipid metabolism, reducing inflammation and relieving renal fibrosis. And it provided theoretical supports for DN treatment with TCM.
9.Analysis of related risk factors of unstable plaques in extracraniai carotid arteries in patients with ischemic cerebrovascular diseases

[Objective]To investigate the related risk factors of unstable plaques in extracranial carotid arteries in patients with ischemic cerebrovascular diseases(ICVD).[Methods] Two hundred and eight ICVD patients were enrolled and plaques in extracranial carotid arteries were detected by c(a)rotid color ultrasound.All the patients were divided into unstable plaque group(75 cases)and stable plaque group (133 cases)according to the ultrasound performance.Related disease history and the traditional risk factors of ICVD were recorded.The risk factors with statistical significance were screened out by univariate nonconditional Logistic regression analysis,and then the independent risk factors of unstable plaques in extracnmial carotid arteries were analyzed by multivariate non-conditional Logistic regression analysis.[Results]Univariate non-conditional Logistic regression analysis showed that male,history of hypertension,history of diabetes,smoking were positively correlated with the formation of unstable plaques in extracranial carotid arteries(P<0.01 or <0.05).High density lipoprotein cholesterol(HDL-C)was negatively correlated with the formation of unstable plaques in extracranial carotid arteries(P<0.01).There were no statistical significance in history of coronary heart disease,law of life,family history of hypertension,drinking,family history of stroke and low density lipoprotein cholesterol between two groups(p >0.05).Multivariate non-conditional Logistic regression analysis showed that male,history of hypertension,history of diabetes and smoking were the independent risk factors of the formation of unstable plaques in ex tracranial carotid arteries (OR =2.33,7.16,6.15,2.28,respectively,P<0.01).And HDL-C was the protection factor of the formation of unstable plaques in extracranial carotid arteries(OR=0.84,P < 0.01).[Conclusions] The independent risk factors of the formation of unstable plaques in extracranial carotid arteries in patients with ICVD are male,history of hypertension,history of diabetes and smoking.And HDL-C is the protection factor of the formation of unstable plaques in extracranial carotid arteries in patients with ICVD.It has important significance to take out necessary intervention for patients with risk factors.
10.A preliminary study of anti-aging and wound healing of recombination cytoglobin.

In this paper, the preliminary study on antioxidant, enhancement of antioxidant enzymes activity, reducing the content of oxygen free radicals, delaying skin aging of the recombination cytoglobin (rCygb) purified by our lab were investigated through human keratinocyte cell line (HaCAT) H2O2 oxidative stress model, mouse skin aging model caused by continuous subcutaneous injection D-gal, rat acute liver injury model induced by CCl4 and rat skin wound healing model. The results showed that rCygb improved the activities of total superoxide dismutase (T-SOD), glutathione peroxidase (GSH-Px) and catalase (CAT), reduced the activities of lactate dehydrogenase (LDH) and alanine aminotransferase (ALT) as well as decreased the content of malondialdehyde (MDA). Skin biopsy showed that rCygb promoted angiogenesis, increased expression of collagen and improved the anti-inflammatory ability. All results displayed that rCygb improved the oxygen free radical scavenging ability, delayed skin aging and promoted wound healing.
