1.The influences of maternal thyroid dysfunction on children

A series of changes of maternal thyroid hormones occur during pregnancy.The growth and development of offspring,especially the growth of the thyroid gland,depends a lot on maternal normal thyroid function.If the thyroid dysfunction of pregnant women is not diagnosed and treated properly,the children may suffer with serious consequences,including fetal loss (abortion,stillbirth),premature birth,congenital malformation,hyperthyroidism or hypothyroidism,delayed development of the nervous system,etc.Maternal thyroid dysfunction even influences the long-term growth of offspring.Therefore,we should detect and control thyroid dysfunction of pregnant women and their children as early as possible,in order to improve the survival rate and life quality of the offspring.
2.Construction,identification and expression of recombinant Bb-MPT64 vaccine of mycobacterium tuberculosis

Objective:To construct and identify recombinant Bb-MPT64 vaccine of mycobacterium tuberculosis. Methods: MPT64 antigen gene were amplified by PCR from mycobacterium tuberculosis H37Rv,then the gene was cloned into Escherichia coli-Bifidobacteria shuttle plasmid pGEX-1?T to construct pGEX-MPT64.The recombinant plasmid was electroporated into Bifidobacteria bifidum(Bb) to construct rBb-MPT64 vaccine.The expression of pGEX-MPT64 was induced with isopropyl-?-D- thiogalactopyranosid(IPTG) and protein MPT64/GST was examined by SDS-PAGE and Western blotting techniques. Results:688bp MPT64 gene was successfully amplified by PCR and cloned into pGEX-1?T by restriction endonuclease analysis.rBb-MPT64 vaccine was successfully constructed by PCR.SDS-PAGE and Western blotting demonstrated that the protein MPT64/GST were expressed in E.coli,BL21. Conclusion:rBb-MPT64 vaccine of mycobacterium tuberculosis is successfully constructed,which may provide basis for the development of new anti-tuberculosis vaccine.
3.Application and analysis of PDCA in medical equipment maintenance

Objective:To explore effective ways to improve the medical quality and safety management level of medical equipment.Methods: Using the PDCA management tool in the daily maintenance of medical equipment, the management of medical equipment is more reasonable. Results:During 2 years’ PDCA plan implementation, fault total cases in 32 sets of hemodialysis equipment were 461, and annual average cases were 230.5. In comparison with the total number of 441 fault cases of 28 units in the previous 1 years, the failure rate is greatly decreased. The mis-operation caused by fault is also greatly reduced. The circuit and water failures were annually 16.5 times and 197 times, and they were greatly reduced compared to 25 times and 324 times in the previous year.Conclusion: It can effectively improve the medical quality and safety management level of medical equipment by using the PDCA management tool in the daily maintenance of medical equipment, and also in the tertiary general hospital accreditation standards provisions to clear quality and safety indicators and records of continuous improvement effect of the department of medical equipment. Therefore, In the medical equipment maintenance work, it can carry out the new trend of PDCA and medical equipment management.
4.Positive Association between Macrophage Infiltration and Senile Calcific Aortic Stenosis

Objective To investigate the relation between macrophage infiltration and degree of calcific aortic stenosis (CAVS)in elder patients.Methods The aortic valve specimens were collected from 2012 September to 2014 March in Depart‐ment of Cardiac Surgery of Hainan General Hospital ,for operation removal ,from 80 cases[40 males and 40 females ,age:(58 ± 6)years old];normal aortic valve specimens were collected from 7 patients undergoing surgical removal of the normal aortic valve because of aneurysm[4 males ,3 female patients ,aged(41 ± 8)years] ,and 80 healthy adults served as control group.The clinical data of patients were collected ,and CAVS valve and normal valve were observed by HE staining.Expression feature of macrophages was studied by immunohistochemical(IHC)staining.IPP(Image‐Pro Plus)software was used to measure the densi‐ty of macrophages ,and Pearson relation analysis was employed to study the correlation between the density of macrophages and the degree of valve stenosis.Results As compared with healthy adults ,cholesterol and C reactive protein in patients with CAVS were significantly elevated.There were infiltration of inflammatory cells ,new blood capillaries and calcification in the valves of CAVS lesions observed by HE staining.No positive expression of macrophage marker CD68 was found in normal aortic valves , but CD68 positive expression was observed in the valves of CAVS by IHC staining ,and mainly concentrated in the peripheral calcification and osteoid tissues.Simple linear regression analysis showed the density of macrophages was positively correlated with the large pressure gradient(r=0.75 ,P<0.05) ,the average differential pressure(r=0.75 ,P<0.05) ,and the maximum aortic jet velocity(r=0.72 ,P<0.05) ,however ,was negatively correlated(r= -0.71 ,P<0.05)with the valve orifice area .Con‐clusion Compared with healthy adult volunteers ,the concentration of cholesterol and C reactive protein were elevated in blood of the patients with CAVS.Compared with the normal valve ,the macrophages were infiltrated in CAVS valves ;the density of macrophage was positively correlated with the degree of CAVS.
5.Therapeutic effect of rehabilitation intervention on patients with diabetes mellitus complicated tubercu-losis

Objective:To explore therapeutic effect of rehabilitation intervention on patients with diabetes mellitus (DM)complicated tuberculosis (TB).Methods:A total of 108 DM + TB patients treated in our hospital from Jan 2008 to Aug 2013 were enrolled.According to random number method,they were randomly and equally divided into rehabilitation intervention group and routine treatment group.The intervention course was eight weeks,and then rehabilitation effect was compared between two groups.Results:Compared with before treatment,after treatment, there were significant reductions in fasting blood glucose (FBG)and 2h postprandial blood glucose (2hPBG)in both groups,P <0.01 all;compared with routine treatment group,after treatment,there were significant reductions in levels of FBG [(6.77±1.82)mmol/L vs.(5.54±1.46)mmol/L]and 2hPBG [(7.94±1.83)mmol/L vs.(6.87 ±1.42)mmol/L],P <0.01 both,and significant rise in percentages of significant reduced or disappeared pleural effusion (51.85% vs.83.33%)and significant reduced or closed TB focus (44.44% vs.72.22%)in rehabilitation intervention group,P <0.01 both.Conclusion:Rehabilitation intervention can make patients get better control of blood glucose,promote absorption of pleural effusion and pulmonary tuberculosis focus.
6.Studies on serum proteomics of psychological stressed rats

Objective To explore the changes in serum protein profiles in rats after psychological stress. Methods Twenty-four male Wistar rats were randomly divided into psychological stress group,control group and stressor group. Rats in stressor group received intermittent foot-shock. Instead of foot-shock,the rats in psychological stress group received the stimulating information from stressor group by watching,listening and smelling. Rats in control group were kept in individual cages and received no stimulation. Experiments were performed once every morning for two weeks. Serum samples of rats in both psychological stress group and control group were obtained and the changes in protein profiles were determined and statistically analyzed by Surface Enhanced Laser Desorption Ionization-Time of Flight-Mass Spectrometry (SELDI-TOF-MS) after the experiments. 7.Correlation between tear osmolarity and the dry eye severity

Background There still is no a uniform diagnostic criteria for increasing population of dry eye.Studies showed that high tear osmotic pressure is one of causes of a variety of ocular surface discomfort,and the change of tear osmolality has been included in the definition of dry eye syndrome by Dry Eye World Study Group (DEWS) in 2007.It is very clinically significant to study the relationship of tear osmolality with dry eye.Objective This clinical trial was to study the correlation between tear osmolarity and dry eye severity based on Preferred Practice Pattern(PPP)of DEWS.Methods A descriptive study was performed.Fifty-four eyes of 54 patients with dry eye disease were asked to complete an evaluation of dry eye signs and symptoms questionnaire based on PPP.The ocular anterior segment was examined under the slit lamp biomicroscope.Cornealfluorescein staining,tear-film breakup time (BUT),Schirmer test Ⅰ (S Ⅰ t)without anesthesia were performed,and tear sample was collected for tear imageology and tear osmolarity admeasurement.The correlation between dry eye score with tear osmolarity was evaluated using Pearson linear correlation analysis.Written informed consent as obtained prior to any relevant medical procedure.Results The ratio of male and female patients was 1: 2 in this study.The negative correlations were found between tear osmolarity with S Ⅰ t or BUT(r =-0.456,-0.699,P<0.01),and significantly positive correlations were found between tear osmolarity with tear imaging examination,corneal fluorescein staining,conjunctival hyperemia or dry eye symptom score(r=0.545,0.686,0.691,0.803,P<0.01).Conclusions As a diagnostic method,tear osmolarity can objectively reflects dry eye severity.It can be a biomarker in dry eye severity grade when combined with clinical symptom.
8.Study on Bactericidal Activity of Thymopentin Peptide and Its Derived Peptides

OBJECTIVE:To study the bactericidal activity of thymopentin and its derived peptides. METHODS:Agar plate count was adopted to determine the bactericidal activity of thymopentin [arginine(R)-lysine(K)-aspartic acid(D)-valine(V)-tyro-sine(Y),RKDVY],its derived peptide 1 [RKN(agedoite,N)VY] and derived peptide 2(RKKVY)to Gram negative bacterial (Proteusbacillus vulgaris,Escherichia coli) and Gram positive bacterial (Staphylococcus aureus,Enterococcus faecium). There were 15.625-1 000 μg/ml for peptides,102 CFU for bacteria. RESULTS:Three pentapeptides possessed bactericidal activity against Gram negative bacteria. The activities of RKKVY and RKNVY were stronger than RKDVY(P<0.01),there was no significant dif-ference between RKKVY and RKNVY(P>0.05). They also possessed bactericidal activity against Gram positive bacteria,and the activity from strong to weak was RKKVY>RKNVY>RKDVY(P<0.01). CONCLUSIONS:Thymopentin and its derived peptides possess bactericidal activity against Gram negative and positive bacteria,with dose-effect relationship.
9. Determination of lobetyolin, scopolin, scopoletin, quercetin, kaempferol and isorhamnetin in Shenqi Jiu by HPLC with gradient elution method

Objective To develop an HPLC method with gradient elution for determination of lobetyolin, scopolin, scopoletin, quercetin, kaempferol and isorhamnetin in Shenqi Jiu. Methods A Shiseido C18 column (4.6 mm×250 mm, 5μm) was adopted, the mobile phase was acetonitrile (A)-0.5% acetic acid solution (B) with gradient elution at a flow rate of 1.0 ml/min, the column temperature was set at 30 ℃, the sample quantity was 20 μl, and the detection wavelengths were 269 (lobetyolin), 346 (scopolin and scopoletin) and 360 nm (quercetin, kaempferol and isorhamnetin). Results The linear ranges of above mentioned 6 ingredients in order fell within the range of 4.59-91.80 (r=0.9999), 2.62-52.40 (r=0.9996), 1.95-39.00 (r=0.9998), 3.34-66.80 (r=0.9995), 2.30-46.00 (r=0.9991), and 2.86-57.20 μg/ml (r=0.9999), respectively. The average recoveries and RSD (n=9) were 98.27% (1.33%), 97.62% (1.48%), 99.17% (0.99%), 98.50% (1.37%), 97.35% (1.35%), and 98.86% (1.70%), respectively. Conclusion The established HPLC gradient elution method can simultaneously determine the above mentioned 6 ingredients Shenqi Jiu. The method is simple, accurate, with good reproducibility. The method is helpful for the quality control of Shenqi Jiu.
10.Progress on association between COMT gene and violence behavior in patients with schizophrenia.

The prevalence of violence behavior in patients with schizophrenia is higher than that in common population. Data suggest that genetic factors may play a substantial role for the etiology of the behavior. Among the particular gene polymorphisms that have been considered to be involved in violence behavior, the catechol-O-methyltransferase (COMT) gene had been the focus of recent research. This article reviews the association research between COMT gene and violence behavior in patients with schizophrenia in several aspects: SNP polymorphism of COMT Val158Met and COMT Ala72Ser, haplotype of COMT gene and DNA methylation of promoter region of COMT gene. The genetic research direction is presented for patients with schizophrenia.
