2.Correspondence analysis on the types of social support and the role of the supporters towards people living with HIV/AIDS in rural areas,Henan province

Objective To explore the relationship between types of social support and roles of supporters,on people living with HIV/AIDS,in rural areas of Henan province.Methods A rural area from Henan province where the main route of HIV transmission was through blood collection was selected as the research site.Survivors registered in that area were randomly selected as subjects.Questionnaire on social support related to social network analysis paradigm was designed and face-to-face interview was used to collect information.Correspondence analysis method was adopted to analyze the relationship between types of social support and roles of social supporters.Results 204 questionnaires were sorted out with 2227 pairs of bind between types of social support and roles of social supporters analyzed.According to scatter plot of row and column points,our data showed that support from the spouses was mainly associated with caring for daily life and companionship for medical treatment on the patients.The research subjects stated that they would primarily discuss over the major issues or chat with their parents and children as they were the ones that they could trust the most.However,they would turn to their brothers,sisters or other relatives to borrow money or asking for other kinds of help.Non-relatives were the resources on social interaction,like going-out together or borrowing life necessities.Conclusion Supporters with different social roles on HIV/AIDS issues,appeared to be corresponded to specific types of social support in rural areas of Henan province.
3.Experimental Study on 99mTc-MIBI Imaging Evaluating Multidrug Resistance of Mice Lewis Lung Cancer

Objectives 99mTc-methoxyisobutylisonitrile (MIBI) SPECT imaging technology was used to observe the condition of tumor cell in-taking imaging agent in the C57BL/6J mice Lewis lung cancer model before and after using Ginsenoside Rg3 (short for Rg3).We aimd so as to discuss the feasibility of using this method to evaluate tumor multidrug resistance (MDR) status.Methods Mice Lewis lung cancer models were randomly divided into Rg3 group and the control group.After applying Rg3,semi-quantitative analysis was made on 99mTc-MIBI SPECT imaging and region of interest (ROI) to observe the multidrug resistance state of tumor and then the results were compared with the detection results of flow cytometry.Results The tumor intake ratio (T/N) difference between the control group and the Rg3 group in imaging,imaging before applying Rg3 and imaging after applying Rg3 were separately 15,60 and 120 min.The differences were statistical significant (P < 0.01).The eliminate indexes (WR) of the control group and Rg3 group were positively related to P-gp protein expression positive cells detected by flow cytometry (P < 0.05).Conclusions 99mTc-MIBI imaging is negatively related to P-glycoprotein (P-gp) expression in mice Lewis lung cancer cells,which can clearly show the multidrug resistance state of tumors and dynamically monitor the effect of Rg3 on multidrug resistance reversion of mice Lewis lung cancer.
4.Analysis of normal eye anterior segments in different ages

Objective To measure and conduct statistic analysis of normal eyes by Pentacam. Methods Two hundred and twenty-one healthy people (442 eyes) underwent examinations of Pentacam, and they were divided into 5 groups by age: <40 years old group (50 people, 100 eyes), 40-49 years old group (50 people, 100 eyes), 50-59 years old group (50 people, 100 eyes), 60-69 years old group (50 people, 100 eyes) and >70 years old group (21 people, 42 eyes). Results With the increase of age, there were significant differences in chamber volume, chamber angle and anterior chamber depth among age groups (P<0.05). In elder age groups, there were significant differences in these parameters between males and females (P<0.01). Conclusion Anterior segment parameters are significantly correlated with age. After 60 years old, the anterior segment parameters become significantly different with gender.

Objective To evaluate the relationship between Multi-slices spiral(MSCT) perfusion and microvessel density (MVD) in maxillofacial tumors. Methods Thirty-one cases of maxillofacial tumors were studied with MSCT perfusion imaging before operation. The time-density curve, perfusion, time to peak(TTP), and Peak enhancement imaging (PEI) of tumors were calculated. MVD of the tumors was measured with immuno-histochemical method by means of detecting factor Ⅷ in all the histologic specimens. Relativity analysis was carried between MSCT perfusion imaging parameters, perfusion curve types and MVD. Results MVD of maxillofacial tumors were higher than normal tissue. MVD remarkably correlated with malignancy of the tumors. Perfusion and time to peak (TTP) correlated well with MVD(t=7.09,4.10, P0.05). 6.Construction of lentiviral vector for shRNA of MACC1 gene and its transfection into MB-231

Objective To construct a lentiviral vector for RNA interference(RNAi)of MACC1 gene and to detect the best trans-fection condition by transfected into MB-231 cells .Methods The siRNA was designed and converted into cDNA of shRNA (small hair pin RNA) of siRNA for MACC1 gene .The cDNA was synthesized and inserted into pMAGic lentiviral plasmid vector which was linearized by enzyme Age Ⅰ and EcoRⅠ .The recombinant plasmid was transformed into competent E .coli DH5α cells .The positive recombinant colony was selected by ampicillin medium agar and identified by DNA sequencing .The recombinant lentivirus was packaged into mature lentivirus by 293FT cells and used to infect MB-231 cells .To detected the transfection condition of high efficiency of infection and low multiplicity of infection .Results PCR and sequencing verified that the recombinant lentivirus plasmid MACC1-shRNA was successfully constructed .The best transfection condition was MOI=40 by transfected into MB-231 .Conclu-sion The lentiviral RNAi expression vector targeting MACC 1 gene is successfully constructed and it can infect MB-231 cells effi-ciently ,which lays the experimental foundation for the research on the changes of malignant biological activity of tumor cell lines and gene therapy .
7.Virological impact of stalk region of neuraminidase in influenza A/Anhui/1/05 (H5N1) and A/Ohio/07/2009 (H1N1) viruses.

This study aims to investigate the virological impact of the stalk region and cysteine (C) in neuraminidase (NA) of influenza A/Anhui/1/05 (H5N1) and A/Ohio/07/2009 (H1N1) viruses. The NA of A/ Anhui/1/05 (H5N1), defined as AH N1, lacked 20 amino acids (including C, defined as s20) as compared with NA of A/Ohio/07/2009 (H1N1) (defined as 09N1). We deleted s20 of 09N1 to construct 09N1-s20, and inserted s20 into AH N1 to construct AH N1+s20. To investigate the impact of C on the biological function of NA, we deleted C in 09N1 to construct 09N1-C and inserted C into AH N1 to construct AH N1-C. The pseudo-type viral particle (pp) system was used to evaluate the impact of these mutants on virology. The combination of 09N1-C and 09H1 (defined as 09H1::09N1-C) showed an infectivity 8 times that of the wild type 09H1::09N1, while the infectivity of the combination of AH N1+C and AH H5 (defined as AH H5::AH N1+C) was much lower than that of the wild type AH H5::AH N1. The infectivity of the combination of 09N1-s20 and 09H1 (defined as 09H1::09N1-s20) was 4 times that of the wild type 09H1::09N1; the infectivity of the combination of AH N1+s20 and AH H5 (defined as AH H5:: AH N1+s20) was 1/7 that of the wild type AH H5::AH N1. The co-existence of 09N1-C and AH H5 displayed 6 times the infectivity of AH H5::09N1, while the infectivity of 09H1::AH N1+C was very low. Multimer analysis showed that in the wild type 09N1, the forms of NA were dimer > tetramer > monomer; the major component of NA in 09N1-C was monomer; in 09N1-s20, the forms of NA were monomer > dimer. AH N1 was mainly composed of monomer; in AH N1+s20, the forms of NA were dimer > monomer > tetramer; in AH N1+C, the forms of NA were dimer > tetramer. Deletion of C or s20 from 09N1 did not change the expression of NA. The study suggested that deletion of C from the stalk region of NA in A/Ohio/07/2009 (H1N1) increases infectivity. Insertion of C into NA's stalk region of A/ Anhui/1/05 (H5N1) significantly decreases infectivity. Cysteine deletion in the stalk region is important for the infectivity of A/Anhui/1/05 (H5N1) and A/Ohio/07/2009 (H1N1). It may interfere with the infectivity via changes in NA polymerization.

8.A study on“one-stop”premarital healthcare services based on SWOT analysis

Based on SWOT analysis and relevant information about the premarital healthcare services through on-site survey and literature research , this paper systematically analyzed the Strengths , Weaknesses , Opportunities and Threats of “one-stop” premarital healthcare services .It also puts forward some strategic suggestions on premarital healthcare services in China including enhancing communication and cooperation between the related national minis -tries and commissions , building a government-led multi-sectoral coordination mechanism , increasing the resource in-vestment , promoting the premarital health care service and pre-pregnancy physical examination integration , and strengthening publicity and mobilization of “one stop” service for premarital healthcare service policy recommenda-tions to provide a basis for decision-making in the promotion of the policy-related works .
9.Meta analysis of clinical and pathological features of breast cancer in young women in China

Objective To compare the clinical and pathological features of Chinese young breast cancer(age ≤ 35)with elder patients(> 35)using Meta analysis .Methods Published studies concerning clinical and pathological features of young breast cancer in China were searched systemically and assessed .Stata12 .0 software was used for data analyzing and calculating OR and its 95%CI .Results Totally 31 studies were selected for Meta analysis ,and most of them were classified as 6 - 7 scores ,which showed the quality of articles was high .The risk factors of breast cancer and its pooled odds ratio values with statistical significance were as fol‐lows 6 .42(95% CI :4 .22 - 9 .79) ,0 .61(95% CI :0 .50 - 0 .74)when clinical staging of 0 - Ⅱ phase or Ⅰ - Ⅱ phase ,2 .25(95% CI :1 .69 - 2 .99)when histological type of Invasive carcinoma ,1 .73(95% CI :1 .23 - 2 .43)when histological grade of III grade ,1 .80 (95% CI :1 .23 - 2 .43)when positive of lymph node metastasis .Conclusion Compared with elder breast cancer ,the clinical and pathological characteristics of young breast cancer were mainly for the high misdiagnosis rate ,the late clinical stage ,the high pro‐portion invasive carcinoma ,the poor histological differentiation and the lymph node metastasising easily ,the hint of young breast cancer screening and treatment may be different principles and measures should be adopted .
