2.Rapid N-glycosylation profiling of glycoprotein based on ultrafiltration-devices.

With development of bio-technique, more and more proteins were applied as clinical approaches. However, the protein homogeneity, especially the N-glycosylation limited the further research and application of these protein drugs. The analysis method for N-glycans is believed to be critical in protein drugs development. To enhance the N-glycans isolation efficiency and accelerate the pretreatment, a new strategy was built on ultrafiltration-devices. New methods increased the isolation efficiency of N-glycans containing N-acetylglucosa mine with 10%-20%. The degrading of N-glycans containing sialic acids was also minimized with this method. 20%-100% more N-glycans with sialic acids were isolated. The pretreatment was finished within 30 min. Coupled with HPLC-HRMS, an effective and reliable strategy designed for protein drugs N-glycans analysis were developed.

3.Clinical significance of the changes in serum NT-proBNP level in aged patients with severe brain dysfunction in acute phase

Objective To explore the changes and influencing factors of serum NT-proBNP level in aged patients with severe brain dysfunction in acute phase, and the corresponding clinical significance. Methods The serum NT-proBNP and cTn-I levels of patients with severe brain dysfunction at day 1, 3, 5 and 7 were measured respectively. The correlation between acute neurological dysfunction caused by brain dysfunction and serum NT-proBNP and cTn-I levels were analyzed, and the impact on prognosis was explored. Results The serum NT-proBNP levels were significantly higher in death group at day 3[(759±341)ng/L], 5[(1980±839)ng/L] and 7[(2490±1862)ng/L] than in survival group[(594±612)ng/L,(733±424)ng/L,(315±346)ng/L]. Serum NT-proBNP and cTnI levels were associated with progressive cerebral edema in both groups. Location of early intracranial lesions was significantly different between two groups. Death group had higher ratio of intracranial lesions in basal ganglia and brainstem than did survival group. High serum NT-proBNP level after day 7 suggested poor prognosis. Serum NT-proBNP level was not associated with serum cTn-I level. Conclusions Progressively increased serum NT-proBNP level in aged patients with severe brain dysfunction in acute phase suggests poor prognosis. The increased degree of serum NT-proBNP in aged patients with severe brain dysfunction in acute phase is associated with the location of intracranial lesions.
4.Effect of meridians massage on the growth and development of premature children

Objective To investigate the effect of meridian massage on the growth and development of premature infants.Methods 60 preterm infants were randomly divided into the observation group and the control group with 30 cases in each group,the control group adopted modified domestic simple meridian massage method,and the observation group increase meridian massage based on the control group,2 times a day and 15 minutes one time.Body weight,length,intake of milk,sleep time,time to regain birth weight,and meconium drained time were eomparel between two groups.Results The observation group was significantly better than the control group in body weight,length,intake of milk,sleep time,time to regain birth weight,and meconium drained time.Conclusions Meridian massage based on the domestic simple touch method plays a significant role in promoting the growth and development of premature infants.
5.The Distribution of Body Weight Related Index and Relationship with Cardiovascular Diseases in Rural Residents of Yunnan Province

Objective The aim of this study was to investigate the distribution of body weight related index and to explore the relationship between weight related index and cardiovascular diseases in rural residents in Yunnan Province. Methods By random cluster sampling method,rural residents were selected in Yunnan Province. The data were collected by face-to-face questionnaire and medical examination. Results 4070 rural residents in Yunnan participated in this study. The survey results showed that gender,age,education level and ethnicity affected both the distribution of body weight related index and low body weight,overweight,obesity,and central obesity(P < 0.05). Compared with low body weight,non overweight,non obesity and non central obesity group,non low body weight,overweight,obesity and central obesity group had higher prevalence rate of hypertension(P < 0.01). Among central obesity group,the prevalence rate of coronary heart disease was higher than non central obesity group(P < 0.01). Conclusion The distribution of body weight related index in rural residents of Yunnan province showed some special characteristic. The overweight,obesity,and central obesity were related to cardiovascular diseases. Change unhealthy living habits or behavior as well as improving their cultural quality have effects on controlling body weight related index and help to reduce the prevalence rates of overweight,obesity and central obesity which can further reduce the prevalence rates of cardiovascular diseases.

8.Study on cytomegalovirus gene mutations conferring resistance to ganciclovir in heart and lung transplant patients

Objective To establish a rapid and convenient assay to screen for emerging ganciclovir(GCV) resistant mutations in the UL97 gene of human cytomegalovirus (HCMV) from transplant recipients. Methods Thirteen spefcific oligonucleotide probes were designed according to the target sequence encoding HCMV-UL97 gene. A nested polymerase chain reaction (PCR) amplifying UL97 gene was employed. Line probes assay (LiPA) was configured to detect relevant non-synonymous mutations at codon 460,520,594,595,603, and 607. In parallel, nested-PCR amplicons were subsequently sequenced directly. Results Four of 16 heart and lung transplant recipients had been infected with HCMV resistant strains during GCV therapy. The mutant codons of UL97 gene were at codon 520、595 and 603. All amplicons detected by LiPA were fully concordant with the nucleotide sequenced.Conclusions Long-term prophylaxis and treatment with GCV in heart and lung transplant recipients may develop HCMV resistant strains. HCMV-LiPA proves to be an alternative method for the resistance genotype analysis of HCMV.
